Intrinsic degradation of volatile fatty acids in laboratory-compacted clayey soil.

Volatile fatty acids (VFAs) represent the major organic constituent of landfill leachate and provide the greatest potential for leachate induced organic contamination of groundwater (e.g. as represented by an increase in the concentration of dissolved organic carbon and chemical oxygen demand). Long-term diffusion tests were performed for laboratory-compacted clayey soil plugs exposed to continuous supply of synthetic leachate containing VFAs. Significant microbial activity developed upon exposure of the soil's indigenous microorganisms to these degradable contaminants. The growth of heterotrophic aerobic bacteria (HAB, which include facultative anaerobes), sulfate reducing bacteria (SRB) and methanogenic bacteria carrying out fermentation and mineralization of the VFAs became evident after 30-50 days of testing. The maximum microbial counts of (2-8) x 10(8) and (0.1-1) x 10(8) cfu/g for HAB and SRB were localized in the soil layer at the interface with the source of organic and inorganic nutrients. Regardless of this rapid growth in microbial population, the VFA consumption was small and measurable only after a lag of 140-180 days. It is considered that this lag of otherwise readily degradable organic compounds (such as VFAs) persisted due to a combination of the effects of a high initial concentration of these acids (2.4 g/l as dissolved organic carbon, DOC) applied to carbon starved soil microorganisms and the small pore size of the compacted clay. Once the significant amounts of gas were generated from fermentation, conditions developed for improved mass transport and exchange of the nutrients and bacteria and the outcome of the intrinsic degradation was more apparent. The breakdown of VFAs that followed after the lag was localized near the top of the soil and was characterized by a short half-life of 0.75-5 days for DOC (total VFAs as dissolved organic carbon).

Intromissive stimulation from the male increases extracellular dopamine release from fluoro-gold-identified neurons within the midbrain of female hamsters.

Extracellular concentrations of dopamine in the nucleus accumbens were monitored using microdialysis in ovariectomized female Syrian hamsters hormonally primed with estradiol and progesterone or with a similar regimen of oil injections. Some females in each of these groups had their vaginas occluded with tape, whereas the remaining females' vaginas stayed unoccluded. When exposed to a male, both groups of hormonally primed females showed high levels of lordosis. However, only in the hormone-primed, unoccluded females were there significant elevations of dialysate dopamine during the sexual interactions with the male. There were no significant elevations in dopamine levels in the oil-treated females during interactions with the male. These data suggest that nucleus accumbens dopamine is responsive to stimuli associated with the vaginocervical stimulation received by the female during intromissions by the male. Histological analyses were based on Fluoro-Gold efflux through the probes combined with immunocytochemistry for tyrosine hydroxylase. Probe placements in the rostral accumbens, caudal accumbens, or rostral bed nucleus of the stria terminalis were not distinguishable based on analyses of basal dopamine levels, volume of Fluoro-Gold injection sites, or Fluoro-Gold labeling of midbrain, tyrosine hydroxylase-stained neurons. The number of midbrain neurons containing Fluoro-Gold was positively related to basal dopamine levels, indicating that the amount of dopamine recovered from the nucleus accumbens in microdialysis studies is a function of the number of neurons contributing to the terminal field in the region of the probe.

Effects of serotonin 1A or 1B receptor agonists on social aggression in male and female Syrian hamsters.

Numerous studies have demonstrated that activation of serotonin 5-HT1A or 5-HT1B receptor decreases aggression in male mammals. To determine whether female mammals also show decreased aggression in response to 5-HT1A or 5-HT1B activation, we assessed the effects of the serotonin receptor agonists 8-OH-DPAT (5-HT1A) and CGS-12066A (5-HT1B) on aggression in female Syrian hamsters. Female Syrian hamsters were tested for interfemale aggression 2 days before and 15 min after receiving intracerebroventricular infusions of 8-OH-DPAT (5, 10, 20 microg) or CGS-12066A (5, 10, 20 microg). Neither drug affected aggression as measured by the latency and frequency of attacks or uprights, although the highest dose of 8-OH-DPAT increased general activity. For male hamsters, intraventricular infusions of 10 microg of 8-OH-DPAT essentially eliminated aggression, whereas 5 microg of 8-OH-DPAT or 20 microg of CGS-12066A were without effect. Systemic treatment with 8-OH-DPAT (1 mg/kg body weight) did reduce aggression in females, although there was an attendant increase in symptoms of nonspecific serotonergic activity. There were no behavioral effects of systemic CGS-12066A (4 mg/kg body weight) on female hamsters. These results indicate that there may be sex differences in the neurochemical regulation of aggression and point to a need for more studies directed at this issue.

Dopamine receptor antagonists attenuate conditioned place preference following sexual behavior in female Syrian hamsters.

We assessed the effects of the dopamine D2 receptor antagonists, sulpiride and raclopride, on conditioned place preference produced by sexual behavior in female Syrian hamsters. Female hamsters treated with sulpiride or raclopride showed high levels of sexual behavior (lordosis) that were equivalent to control females receiving vehicle injections. The degree of place preference conditioning for sulpiride-treated females was marginally reduced, whereas females treated with raclopride showed no evidence of conditioning. These results indicate that conditioned place preference is a useful means for probing the appetitive components of female sexual behavior, and that dopamine D2 receptors are involved in this appetitive process.

A stationary hemispherical SPECT imager for three-dimensional brain imaging.

A completely stationary, hemispherical-coded aperture SPECT imaging system was designed to produce three-dimensional images of the brain. The system consisted of a hemispherical multiple-pinhole coded aperture and 20 small (100 x 100 mm crystal area) digital gamma cameras. Reconstructions and measured performance specifications from two laboratory versions of the imager are presented. The reconstructed field of view of these systems was an ellipsoidal region with semi-diameters of 100 x 100 x 50 mm. The reconstructed spatial resolution for a point source in air at the center of this field was found to be 4.8 mm FWHM and the corresponding system sensitivity was 36 cps/microCi. An analysis using an ideal-observer model indicated that the multiplexed projection data suffered a 21% degradation relative to similar, but nonmultiplexed SPECT data. Therefore, by this measure, the effective sensitivity of the brain imager was 79% of the measured value.

A full-field modular gamma camera.

A modular gamma ray camera is described that gives useful image information over its entire crystal face. The lack of dead area on the periphery of the camera is made possible by a unique application of digital electronics and optimal position estimation using maximum likelihood (ML) estimates. The ML estimates are calculated directly from photomultiplier tube responses and stored in a lookup table, so the restriction of calculating the position estimates in separate circuitry is removed. Each module is designed to be optically and electronically independent, so that many modules can be combined in a large system. Results from a prototypical module, which has an active crystal area of 10 cm X 10 cm, are presented.
