In vitro selection of antisense oligonucleotides targeted to a hairpin structure.

Antisense oligonucleotides are widely used to selectively prevent pre-RNA splicing, mRNA translation or cDNA synthesis from a retroviral RNA template. However, intramolecular folding of the RNA chain can sequester the target sequence into a stable structure. Consequently, the antisense effect can be greatly reduced or even abolished. Hydrogen donor and acceptor sites are still available on nucleic acid bases involved in secondary structures. However, the rational design of antisense sequences able to recognize the three dimensional array of these sites is not available. We used an in vitro selection procedure to fish out aptastrucs, i.e., oligomers able ("apte") to bind to a structure. A population of randomly synthesized oligonucleotides was mixed with the structure of interest and oligodeoxynucleotide sequences bound to the target were selected and amplified. The selection involves the destruction of the unbound candidates by a restriction enzyme. This procedure can be used both for RNA and DNA target structures and does not require the purification of the bound oligonucleotides at each cycle of selection. Several cycles of selection-amplification, followed by cloning and sequencing, allowed us to identify three oligonucleotides able to form a complex with a DNA hairpin. Due to the sequence of the selected candidates, these aptastruc-hairpin complexes involve very likely non-canonical interactions between the two partners.

Dopamine receptor modulation by a highly rigid spiro bicyclic peptidomimetic of Pro-Leu-Gly-NH2.

A peptidomimetic analogue of Pro-Leu-Gly-NH2 (PLG), compound 3, has been synthesized that contains a highly constrained spiro bicyclic type-II beta-turn mimic. Peptidomimetic 3 enhanced the binding of the dopamine receptor agonist ADTN to dopamine receptors by 40% at 10(-6) M. At this same concentration PLG enhanced the binding of ADTN by 26%. Like PLG, 3 exhibited a bell-shaped dose-response curve with the maximum effect occurring at a concentration of 10(-6) M. Because of the highly rigid nature of the spiro bicyclic type-II beta-turn constraint found in 3, these results lend strong support for the hypothesis that the biologically active conformation of PLG is a type-II beta-turn.

Bicyclic thiazolidine lactam peptidomimetics of the dopamine receptor modulating peptide Pro-Leu-Gly-NH2.

Bicyclic thiazolidine lactam peptidomimetics 3-5 have been synthesized as potential analogues of the dopamine receptor modulating peptide Pro-Leu-Gly-NH2 (PLG). Peptidomimetics 3 and 4 were designed to constrain two, psi 2 and phi 3, of the four torsion angles that define a beta-turn to values approximating those found for a type-II beta-turn, while 5 was designed as a compound that could not achieve a beta-turn conformation. Peptidomimetics 3 and 4 were found to enhance the binding of the dopamine receptor agonist ADTN to the dopamine receptor, while 5 was found to be inactive. Like PLG the dose-response curves for 3 and 4 were bell-shaped in nature with the maximum effect occurring at a concentration of 1 microM. Both 3 and 4 were more effective than PLG in enhancing the binding of ADTN to dopamine receptors. The 5,5-bicyclic thiazolidine lactam peptidomimetic 3 enhanced the binding of ADTN by almost 200%, while the 6,5-bicyclic thiazolidine lactam peptidomimetic 4 enhanced the binding of ADTN by about 75%. These results provide further evidence in support of the hypothesis that the bioactive conformation of PLG is a type-II beta-turn.

High- and low-affinity states of dopamine D1 receptors in schizophrenia.

Using SKF-38393/[3H]SCH-23390 dopamine D1 receptor agonist/antagonist competition binding technique, we showed that schizophrenics distinguished from the neuro-psychiatric and normal controls by exhibiting a significant increase in the density of the low-affinity state, and a concomitant enhancement in the binding affinity of the high-affinity state of D1 dopamine receptors in caudate nucleus. The results suggest that perturbation in dynamic equilibrium of high- and low-affinity states of D1 receptors underlies the pathogenesis of schizophrenia.

Synthesis and dopamine receptor modulating activity of lactam conformationally constrained analogues of Pro-Leu-Gly-NH2.

A series of analogues of the potent analogue of Pro-Leu-Gly-NH2 (PLG), 2-oxo-3(R)-[(2(S)-pyrrolidinylcarbonyl)amino]-1-pyrrolidineacet amide (2) were synthesized in which the (R)-gamma-lactam residue of 2 was replaced with a (R)-beta-lactam, (R)-aminosuccinimide, (R)-cycloseryl, (R)-delta-lactam, (R)-epsilon-lactam, or (S)-epsilon-lactam residue to give analogues 3-8, respectively. These substitutions were made so as to vary the psi 2 torsion angle. The analogues were tested for their ability to enhance the binding of the dopamine receptor agonist ADTN to the dopamine receptor. Analogues 3-6 and 8 exhibited dose-response curves that were bell-shaped in nature with the maximum effect occurring at a concentration of 1 microM. Analogue 7 was inactive. Analogues 3 and 4 were found to be as effective as PLG, while analogues 5, 6, and 8 appeared to be more effective than PLG in terms of enhancing the binding of ADTN to dopamine receptors. The activity of analogues 3-6 and 8 with their psi 2 angles in the vicinity of that observed in a type II beta-turn is consistent with the hypothesis that this type of turn is the bioactive conformation of PLG.

Desensitization of D1 dopamine receptors down-regulates the Gs alpha subunit of G protein in SK-N-MC neuroblastoma cells.

The effect of long-term (72 h) treatment with dopamine D1 receptor agonists, SKF 38393 and dopamine on D1 dopamine receptor and G-protein (Gs alpha) was investigated in SK-N-MC neuroblastoma cells. The prolonged treatment of cells with 10 microM SKF 38393 or 10 microM dopamine resulted in a decrease in dopamine D1 receptor by 41 and 81%, respectively, as measured by specific antagonist [3H]SCH 23390 binding. Similarly, the prolonged treatment of SK-N-MC cells with 10 microM SKF 38393 or 10 microM dopamine resulted in a reduction of the level of Gs alpha subunit of G-protein. The results indicate that agonist-induced down-regulation of D1 dopamine receptor may also modulate G-proteins.

Identification of novel catecholamine absorbing proteins in the central nervous system.

Several pharmacologically active catecholamines have been shown to react covalently with CNS proteins, namely species of 47, 40, 22, and 20 kDa. Of these, the 47-kDa protein showed the greatest incorporation of tritium following treatment with [3H]dopamine, [3H]ADTN, or [3H]N-propyl-norapomorphine. Labeling was accomplished by incubating the tritiated ligands with crude membrane preparations in the absence of reducing agents. These proteins displayed several unique characteristics: 1. The proteins are distributed throughout the CNS, but no evidence was found for their presence in other tissues; 2. The proteins have a unique pharmacological profile, interacting with dopamine, ADTN, N-propyl-norapomorphine, and apomorphine, but not with ligands specific for other proteins known to interact with these compounds; 3. The labeling of these proteins is not inhibited by several similar catecholamines and other catechols, suggesting specific structural requirements; and 4. These proteins exhibited stereoselectivity with respect to this labeling. These results demonstrate the existence of novel CNS proteins capable of covalently absorbing several physiologically important catecholamines in vitro.

Mechanism of initiation of transcription by Escherichia coli RNA polymerase on supercoiled template.

DNA supercoiling is known to influence the pattern of gene expression in prokaryotes. Thus the mechanism of transcription initiation and the topological state of the template are intimately related. Using in vitro reconstituted transcription assays, composed of purified RNA polymerase and promoters in their natural topological state, we have conducted a detailed study of transcription initiation from T7 early promoters including the following steps: the formation of ternary complexes, acquisition of rifampicin resistance, release of sigma factor and the capacity for RNA chain elongation in complexes. We determined the order of these events and the length of the transcripts when each step occurred during initiation of transcription on supercoiled templates. The length of the transcripts varied in a promoter-specific manner. Analysis of abortive products formed during the initiation showed that stronger promoters go to the elongation mode at transcript lengths shorter than that required for weaker promoters.

Ontogeny of dopamine D2 receptor mRNA in rat brain.

The postnatal development of rat brain dopamine D2 receptor gene expression was investigated in animals 1 day to 1 year old. The level of expression of the striatal D2 mRNA was appreciable at birth (day 1), steadily increased to a maximum at day 28, and showed declines at ages 6 months and one year. The mRNA development profile was similar to that of [3H]spiroperidol binding in striatal membranes except that there was a lack of correlation between mRNA levels and [3H]spiroperidol binding during the early developmental periods. For example, although the mRNA expression at day 1 is about 75% of the 28-day value, the corresponding level of [3H]spiroperidol binding is only 15% of the value observed at day 28. Polymerase chain reaction (PCR) analysis of alternatively spliced forms of D2 receptor mRNA showed that the developmental expression of the two isoforms proceeded in parallel as the ratio of D2L and D2S mRNAs remained more or less constant in different age group of rats. In situ hybridization revealed a differential developmental profile of D2 mRNA for major dopaminergic regions of rat brain such as caudate putamen, nucleus accumbens, olfactory tubercle and substantia nigra.

Effects of chronic treatment of haloperidol and clozapine on levels of G-protein subunits in rat striatum.

Chronic administration of typical neuroleptic drugs, such as haloperidol, causes the supersensitivity of brain dopamine D2 receptor in striatum and limbic regions, while the atypical neuroleptic clozapine does not. In order to understand the mechanism of their action at a molecular level, studies were carried out to assess the effects of chronic treatment of these drugs on the levels of G-proteins in the rat striatum using the Western blot method. Results of the present study demonstrate that the treatment with haloperidol or clozapine, respectively, down-regulate or up-regulate the levels of G proteins. Quantitative immunoblotting, using site-directed specific antisera, demonstrated that chronic treatment with haloperidol down-regulates Gi alpha, Gs alpha, and beta subunits while chronic treatment with clozapine upregulates Gi alpha, Gs alpha, and beta subunits. Neither of these drugs has any effect on the levels of Go alpha.

Function of Photosynthetic Apparatus of Intact Wheat Leaves under High Light and Heat Stress and Its Relationship with Peroxidation of Thylakoid Lipids.

Effects of high light and temperature stress on the structure and function of the photosynthetic apparatus of wheat (Triticum aestivum) were studied. There was a decrease in the electron transport activity of chloroplasts isolated from photoinhibited and heat-stressed leaves. Chlorophyll fluorescence was measured in photoinhibited and heat-stressed leaves and the decrease in variable fluorescence and variable to maximum fluorescence ratio of the stressed leaves indicated a loss in the quantum yield of photosynthesis. The decrease in electron transport activity was accompanied by an increase in peroxidation of thylakoid lipids. Lipid peroxidation indicated the oxidative degradation of polyunsaturated fatty acyl residues of the thylakoid lipids. A negative correlation was observed between electron transport activity and lipid peroxidation. The electron transport activity was completely lost as the peroxidation level reached a threshold equivalent to 0.6 micromoles malondialdehyde. The threshold of lipid peroxidation for complete loss of activity was the same for both photoinhibition and heat treatment, suggesting that the nature of the environmental stress may be less important with respect to the relationship between electron transport and lipid peroxidation. Thus, it seems likely that lipids are required for sustaining the photosynthetic activity under environmental stress, and a loss in activity is observed as the lipids are degraded either by high light or high temperature stress.

Quantitative immunological status of males of unexplained infertility in eastern U.P.

126 males of unexplained infertility and 75 fertile males were studied during a period of more than 2 years. Statistically significant increase in the levels of serum IgM was observed in cases of males of unexplained infertility. Level and percentage of cases showing semen IgM was more in study group (134.61 +/- 25.5 mg %, 41.26%) than control (24%, 99.3 +/- 8.3%). Semen IgA was found only in study group in 30.15% cases. Though definite correlation between serum and semen immunoglobulin was not observed, semen immunoglobulin was found to be 1-2 fold lower than serum. Results of the present study indicate towards a possible immunological role in cases of unexplained infertility in males.

Localization of cholecystokinin binding sites in canine brain using quantitative autoradiography.

1. CCK receptors have been characterized and localized in various mammalian species and significant species-specific differences in their distribution have been identified. In the present study, we report the first autoradiographic localization of CCK binding sites in the canine brain. 2. High densities of [125I]BH-CCK-8 binding sites were found in the cortex, cerebellum, hippocampus, caudate nucleus, olfactory bulb and nucleus accumbens. Moderate densities were present in the putamen, amygdala, and substantia gelatinosa. Low binding densities were observed in the globus pallidus, inferior colliculus, hypothalamus and thalamus. 3. Although the distribution profile of CCK binding sites in canine brain is similar to those previously reported in the rodent, primate and human brain, notable differences were observed in the hippocampus, cortex and cerebellum.

Mortality events amongst non insulin dependent diabetes mellitus patients in Orissa.

In a study over one year, it was observed that mortality amongst hospitalised patients with non insulin dependent diabetes mellitus (NIBDM) was nearly 20%. Those dying within 24 hr were classified as group A, between one day and one week as B, between one week and one month as C, and those after one month as D. There were 31 patients each in groups A and B, 14 in C, and 4 in D. The mean age at death was 61 years in the first three groups. The prevalence of cerebro-vascular accident as a terminal event was similar i.e. 32.2, 35.5 and 35.7 per cent in groups A, B and C respectively; 48% of patients in group A suffered from ischaemic heart disease. Diabetic ketoacidosis was equally prevalent amongst groups A, B and C. Infection was significantly more common in group B (45.2%) than A (P less than 0.05). Nephropathy was observed in 57% of patients in group C as compared to 22.5% in A (P less than 0.02). Cerebrovascular accident and infection were the major causes of mortality in groups B and C (80.7% and 71.4%), whereas ischaemic heart disease and cerebrovascular accident accounted for 80% of deaths in group A.

Correlation between the DNA supercoiling and the initiation of transcription by Escherichia coli RNA polymerase in vitro: role of the sequences upstream of the promoter region.

Binding of Escherichia coli RNA polymerase and the abortive initiation of transcription at the A2 promoter of bacteriophage T7, separately cloned in pBR322, was found to be strongly dependent on the degree of supercoiling of the plasmid. Supercoiling does not seem to play any role in the initiation of transcription at the T7A1 promoter under identical conditions. Plasmid containing T7A2 promoter was found to be less amenable to S1 nuclease in comparison to that having T7A1. Sequence comparison reveals a high G/C content upstream to the -35 region of T7A2 which by extra duplex stability probably renders the initiation of transcription more dependent on the state of supercoiling of the template.

Solubilization and reconstitution of dopamine D1 receptor from bovine striatal membranes: effects of agonist and antagonist pretreatment.

The bovine striatal dopamine D1 receptor was solubilized with a combination of sodium cholate and NaCl in the presence of phospholipids, following treatment of membranes with a dopaminergic agonist (SKF-82526-J) or antagonist (SCH-23390). The solubilized receptors were subsequently reconstituted into lipid vesicles by gel-filtration. A comparison of ligand-binding properties shows that the solubilized and reconstituted receptors bound [3H]SCH-23390 to a homogeneous site in a saturable, stereospecific and reversible manner with a Kd of 0.95 and 1.1 nM and a Bmax of 918 and 885 fmol/mg protein respectively for agonist- and antagonist-pretreated preparations. These values are very similar to those obtained for membrane-bound receptors. The competition of antagonists for [3H]SCH-23390 binding exhibited a clear D1 dopaminergic order in the reconstituted preparation obtained from either agonist or antagonist-pretreated membranes, except that (+)butaclamol was about four-fold more potent than cis-flupentixol in displacing [3H]SCH-23390 binding in preparation obtained from agonist-pretreated membranes compared to antagonist-pretreated membranes. The agonist/[3H]SCH-23390 competition studies revealed the presence of a high-affinity component of agonist binding in both the reconstituted receptor preparations. The number of high-affinity agonist binding sites, however, is 40-80% higher in reconstituted preparation obtained from antagonist-treated membrane compared to that obtained from the agonist-treated membrane. In both the preparations, 100 microM guanylylimidodiphosphate (Gpp(NH)p) completely abolished the high-affinity component of agonist binding compared to partial abolition in the native membranes, indicating a close association of a G-protein with the solubilized receptors. Whether the receptor was solubilized following agonist or antagonist preincubation of the membranes, the receptor-detergent complex eluted from a steric-exclusion HPLC column with an apparent molecular size of 360,000. Preincubation of the solubilized preparations with Gpp(NH)p had virtually no effect on the elution profile suggesting a lack of guanine nucleotide-dependent dissociation of G-protein receptor complex.
