New cell line from hairy-cell leukemia: confirmation of leukemic cell origin by karyotype and Ig gene analysis.

A hairy-cell leukemia (HCL) line, BNBH-I, was established from the peripheral blood of a 40-year-old male patient with HCL in a relatively stable clinical phase after splenectomy. The cells have since been growing continuously for more than 2 years. Their cell surface immunoglobulin (sIg) was identical with that found on the surface of freshly isolated leukemic cells, consisting of IgG-kappa. The BNBH-I cells were more mature than the original hairy cells in their degree of B-cell differentiation, as reflected by a decrease in sIg expression together with the appearance of some cytoplasmic Ig (cIg)+ cells, loss of EA gamma-rosette formation and reactivity with monoclonal antibody (MAb) FMC7, and an increase in the proportion of MAb PCA-I+ cells. The BNBH-I cells possessed the antigen recognized by Leu-M5, a highly specific MAb for HCL. Epstein-Barr virus nuclear antigen (EBNA) was present. Both the freshly isolated leukemic cells and the cell line had the 14q+ involving q32 chromosomal abnormality, and their Ig gene rearrangements were also identical. Following exposure to 12-O-tetradecanoylphorbol-13-acetate (TPA), both the freshly isolated leukemic cells and the BNBH-I cells adhered to culture dishes and extended long, thin processes, a phenomenon characteristic of HCL. These results indicate that the BNBH-I line was derived from the leukemic hairy cells.

Effects of inositol phosphates on the membrane activity of smooth muscle cells of the rabbit portal vein.

The effects of intracellular perfusion of inositol 1,4,5-trisphosphate (InsP3) or inositol 1,3,4,5-tetrakisphosphate (InsP4) on electrical responses of smooth muscle cell membranes of the rabbit portal vein were studied using the whole cell voltage clamp technique. Depolarisation to 0 mV from a holding potential of -60 mV, evoked inward Ca (Ica), transient outward (ISO), oscillatory outward (IOO) and sustained outward (ISO) currents. Generation of IOO was dependent on the [Ca]o, but it was also generated in 0 mM Ca solution for over 10 min. From amplitude histograms, IOO was divided into two components. Reduction in [Ca]o inhibited the appearance of but not the amplitudes of both IOO components. However, the larger component of IOO was more resistant to a reduction in [Ca]o than the smaller one. InsP3 (10 microM) increased the frequency of both IOO components to a greater extent than their amplitude, but the larger component was more sensitive to InsP3 than the smaller one. The increase in the occurrence of IOO induced by InsP3 did not occur following pretreatment with 3 mM caffeine or 1 nM A23187. In normal PSS, InsP3 was evoked by a depolarising pulse positive to -40 mV, whereas following perfusion with InsP3 (10 microM), IOO was evoked at -60 mV. In normal PSS, intracellular perfusion with 10 microM InsP4 changed neither the frequency nor the amplitude of IOO, and the amplitudes of ICa, ITO and ISO were also unchanged. However, in 10 mM Ca solution, 10 microM InsP4 generated IOO at a membrane potential of -60 mV.(ABSTRACT TRUNCATED AT 250 WORDS)

B cell lymphoma showing clinicopathological features of malignant histiocytosis.

This report describes a case of B cell lymphoma, the clinicopathological features of which are quite similar to those of malignant histiocytosis. The clinical features included fever, anemia, and marked hepatosplenomegaly without lymphadenopathy. Histological findings revealed diffuse and noncohesive proliferation of cytologically atypical cells and benign-appearing histiocytes in the splenic red pulp, where the erythrophagocytosis was frequently found. Immunological studies, however, revealed a B cell nature of proliferating atypical cells. Accordingly, the histology of this patient was interpreted as a neoplastic proliferation of B cells accompanied by marked proliferation and activation of the histiocytes.

A direct radioimmunoassay for serum 11-deoxycortisol.

A simple direct radioimmunoassay for 11-deoxycortisol in serum is described. A new type of bridge heterologous 125I labeled 11-deoxycortisol was prepared and was applied to the direct radioimmunoassay. Low and high levels of 11-deoxycortisol in serum were sensitively determined by the direct radioimmunoassay.

[Distribution of oncotrophoblast antigens defined by monoclonal antibodies].

In order to analyze Oncotrophoblast antigen, I used monoclonal antibodies (MoAbs) TM7-3 and TM3-8, which demonstrated a very strong reaction with choriocarcinoma, particularly cytotrophoblast-like tumor cells, in spite of a very weak reaction in normal chorionic villi. I examined their reaction with other tumors of non-trophoblastic origin. Those examined were 58 malignant cases, 2 low-potential malignant cases and 14 benign cases. Immunofluorescence and immunoperoxidase stainings show that 1) TM7-3 and TM3-8 had similar patterns of distribution. 2) In various normal tissues tested, only a part of the urinary tubules of the kidneys and the duct of the pancreas reacted with these MoAbs. 3) In various neoplastic tissues, ovarian serous cancer (3/6), ovarian clear cell cancer (3/3), ovarian undifferentiated cancer (2/2) and well differentiated uterine corpus cancer (3/3) showed a positive reaction with TM7-3 and TM3-8. On the other hand, no uterine cervical squamous cell cancer (0/20) showed a positive reaction. These results suggest that TM7-3 and TM3-8 may be valuable for research on the relationship between the oncotrophoblast antigen and adenocarcinoma of the ovary.

[Visibility of the inferior horns in computed tomography of normal subjects and epileptics].

In recent years brain CT scan has been so popular that many investigators have been trying to clarify the normal CT images. But little attention has been paid to the inferior horns of lateral ventricles in spite of their importance for judging mesial temporal lobe structures. The present study was designed to elucidate how the inferior horns were visualized on brain CT in normal subjects from childhood to aged group, and to evaluate whether the inferior horns were dilated in the epileptics or not. The subjects of the present study were 502 normal controls (2-79 y, mean 36.1 y) and 163 epileptic patients with normal CT image (4-68 y, mean 26.6 y) including 55 cases of temporal lobe epileptics. CT scans were performed with EMI 1010 scanner, and slices were obtained every 10 mm from the projection of 5-10 degrees angle for orbito-meatal line. Inferior horns were examined at the level of basal cisterns. Because inferior horns were not necessarily visible in our cases, we examined the frequency of clearly visualized inferior horns at each decade, and regarded their frequency as the size of inferior horns at each decade. In normal controls, frequency of visible inferior horns was relatively high in early childhood, and decreased as they grew. In the 3 rd to 4 th decades the visibility frequency became the lowest (30-35%), and then gradually increased as the subjects became older.(ABSTRACT TRUNCATED AT 250 WORDS)

Acetylcholine activates single sodium channels in smooth muscle cells.

Using the whole-cell clamp technique with a patch electrode, single channel activities were recorded in dispersed single smooth muscle cells of the guinea-pig ileum in response to application of greater than 1 microM acetylcholine (ACh) or carbachol (CCh). Under physiological conditions (bath:Krebs, pipette:High K), these channels have a single channel conductance of 20-25 pS at the membrane potentials ranging between -100 and -40 mV and are activated in the membrane potential dependent manner. The amplitude of the channel current showed a strong dependence on the extracellular Na concentration but the reversal potential obtained by the extrapolation of the I-V relationship was not consistent with the equilibrium potential of Na ion. An approximate estimation of permeability ratio based on the independent principle described by Hodgkin et al. gave a value of Na:K = 1.0:0.3-0.4 to this channel. From features of the macroscopic and single channel currents, it is concluded that these muscarinic ACh(CCh)-activated channels mainly pass Na ion and play a major part in the membrane depolarization produced by ACh or CCh in mammalian intestinal smooth muscles.

[Cancer risk by family history--a cohort study in Kanagawa Prefecture].

Cancer family history was studied for 43,117 subjects aged 35-79 at the occasion of stomach cancer mass screening and were followed up for 5 years. 376 cases of cancer were notified out of them by the linkage with records of population based cancer registry covering the prefecture. Significantly elevated risk among those with family history was found only for breast cancer (observed 3, expected 0.82).

The risk of a second primary cancer occurring in five-year survivors of an initial cancer.

Patients with a greater than five-year survival diagnosed with cancer of the stomach, colon, breast and uterus (included in situ) at the Kanagawa Cancer Center from 1963 to 1980 were studied for multiple primary cancers. They were followed up to December 31, 1985, for the occurrence of second primary cancers by means of a record linkage to the population-based cancer registry in Kanagawa Prefecture. The occurrence rate of total second primary cancers was 2.6% (76/2,977). Among the first primary cancer sites, the rate for cancer of the colon was highest: 5.3% (5/94), and the rate for women with cancer of the uterus lowest: 1.4% (16/1,146). The number of observed second primary cancers was compared with the expected number, which was calculated by the person-years method, to obtain an estimate of the risk. The relative risks (RR: observed: expected value) of women with cancer of the breast were significantly high for developing cancer of the colon (RR, 3.7), cancer of the opposite breast (RR, 3.9) and cancer of the ovary (RR, 5.8). Men with cancer of the stomach had a significant risk of developing cancer of the colon (RR, 3.3). It may be considered, at least in part, that some cancer patients, especially those with cancer of the breast, have a high risk of a second primary cancer occurring. These results may provide clues and information to the etiology of predisposition to and prevention of cancer.
