Acyl-CoA-binding protein/diazepam-binding inhibitor gene and pseudogenes. A typical housekeeping gene family.

Acyl-CoA-binding protein (ACBP) is a 10 kDa protein isolated from bovine liver by virtue of its ability to bind and induce the synthesis of medium-chain acyl-CoA esters. Surprisingly, it turned out to be identical to a protein named diazepam-binding Inhibitor (DBI) claimed to be an endogenous modulator of the GABAA receptor in brain membranes. ACBP/DBI, or proteolytically derived polypeptides of ACBP/DBI, have also been implicated in the control of steroidogenesis in mitochondria and glucose-stimulated insulin secretion. Thus, it appears that ACBP/DBI is a remarkable, versatile protein. Now we have molecularly cloned and characterized the ACBP/DBI gene family in rat. The rat ACBP/DBI gene family comprises one expressed gene and four processed pseudogenes of which one was shown to exist in two allelic forms. The expressed gene is organized into four exons and three introns. There is a remarkable correspondence between the structural modules of ACBP/DBI as determined by 1H nuclear magnetic resonance spectroscopy and the exon-intron architecture of the ACBP/DBI gene. Detailed analyses of transcription of the ACBP/DBI gene in brain and liver were performed to map transcription initiation sites and to examine if transcripts from the ACBP/DBI gene were subject to alternative processing. In both brain and liver, transcription is initiated from two major and multiple minor initiation sites. No evidence for alternative splicing was obtained. The promoter region of the ACBP/DBI gene is located in a CpG island and lacks a canonical TATA box. Thus, the ACDB/DBI gene exhibits all the hallmarks of a typical housekeeping gene.

Characteristics of bovine Staphylococcus aureus with special regard to clumping factor activity.

The occurrence of the following groups of bovine Staphylococcus aureus with characteristic patterns of biochemical reactions and sensitivities to phages was confirmed by the present investigation of 662 isolates from the udders of cows in 53 dairy herds: (1) host-specific variety (v) bovis, (2) not alloted strains with the basic marker, crystal violet type A (group na CV A), (3) group na CV C. The latter was split into three subgroups clearly differing in phage susceptibility patterns: I/119, I without 119 and III, respectively. Lack of clumping factor activity occurred in 323 out of 346 (93%) phage pattern I/119 strains examined while most or all isolates of the other groups of bovine strains proved to be clumping-positive.

A prospective study of thymopentin in severely burned patients.

A randomized prospective double-blind study of thymopentin was performed upon 24 severely burned patients to evaluate its efficacy in correcting postburn immunologic abnormalities and preventing infectious morbidity and mortality. Patients in the treated group received 50 milligrams of thymopentin daily for the first two weeks after injury and three times weekly thereafter until the patient was no longer at risk for having infections develop. The placebo group received saline solution intravenously. The rate of infectious complications was recorded. Immunologic tests used at least weekly were: white blood cell counts, OKT4 to OKT8 ratios, lymphocyte blastogenesis and neutrophil bactericidal index. There were no differences noted in patient mortality, infectious complications or antibiotic use. There were two significant differences noted among the immunologic tests. First, there was a decreased lymphocyte blastogenic response in the treated group at two weeks after injury without a difference in T4 to T8 ratios. Second, there was less leukopenia during the first week after injury in the treated group.

Spread of plasmid-mediated nourseothricin resistance due to antibiotic use in animal husbandry.

After using of the streptothricin antibiotic nourseothricin in animal husbandry for growth promotion, plasmid-borne resistance to streptothricin could be observed in E. coli from nourseothricin fed pigs, from employees in pig farms and from their family members. Moreover, streptothricin resistance plasmids also occurred in E. coli of man without any contact to pig farms (gut flora and even urinary tract infections). However, these individuals live in villages and towns of the territory where nourseothricin was applied to pigs. Similar streptothricin resistance plasmids belonging to different incompatibility groups were found in both E. coli from pigs and E. coli from human beings. As no coselection of resistance to drugs indispensable for therapeutic use in man was observed, the application of nourseothricin in animal husbandry has not clinical implication for human medicine yet. Nevertheless, this problem remains under further investigation.

Identity status and academic achievement in female adolescents.

Theories of Erikson and Marcia were applied to assess the relation between identity status and academic achievement in female adolescents. The differences in identity status between bright high achieving vs. underachieving students were ascertained separately for occupational planning, attitudes toward religion, and peer relations. It was hypothesized that the component of identity most closely linked to high achievement in school is having made commitments to certain goals and values. It was found instead that experiencing and working through crises in beliefs and values are more closely associated with successful academic achievement and formation of identity among adolescent girls than having made specific commitments to career or ideology.

[The system of blood vessels of the small intestine of ruminants (author's transl)].

The vascular system of the small intestine of sheep, goat, and the bovine was investigated by means of corrosion preparations as well as by light and electron microscopic methods. The jejunal villi are shown to contain a single--in the case of broad villi several--centrally placed arterioles. The arterioles show no ramification, they drain into subepithelial capillaries at the apex of the villus. The subepithelial capillaries are not furnished with precapillarious sphincters. In the lower third of the villi several capillaries fuse together, forming a centrally located venule which in turn passes through the base of the villus and--still above the Tunica muscularis mucosae--opens into a transverse running vein. 2 or more of these transverse running veins join together forming a short vascular trunk which opens into collecting veins of the submucosa. Special sphincters of the peripherous blood vessels of the small intestine like arteriovenous marginal loops, arteriovenous anastomoses of the submucosa, vascular sphincters of the muscularis mucosae or veins which are well known in the literature were not found in domestic ruminants.

[Resistance development in human Staphylococcus aureus due to the use of oxytetracycline and tylosin in industrialized swine production units].

Staphylococcus aureus strains from human nasal carriers without vocational contact with pigs could be differentiated from the majority of strains obtained from pathologically affected pigs by determination of host-specific-variety, according to Meyer (1966) and Witte et al. (1977). Less unambiguous findings regarding site variability were recorded from strains in nasal mucosa of pig-handling personnel, clinically intact pigs on farms, and slaughtered pigs. Antibiotics, such as oxytetracycline and macrolides were applied to pigs but failed for all practical purposes to have any bearings on the development of Staphylococcus aureus resistance in human probands with no vocational contact with animals. Personnel of pig units, however, exhibited moderate resistance to oxytetracycline and macrolides of Staphylococcus aureus strains which were typical of man. Macrolide resistance was determined, in that context, by one single-resistance plasmid.

Analytical survey of N-nitroso contaminants in pesticide products.

1. Levels of N-nitroso dialkylamines in dinitroaniline derivatives were high (up to 153 mg/kg) in the past, but have been decreased considerably in current production, due to process changes. 2. Butralin and pendimethalin contain significant amounts of N-nitroso parent compounds. 3. Most dimethylamine salts of phenoxyalkanoic acids presently contain low or non-detectable levels of NDMA. 4. Quaternary ammonium compounds contain NDMA levels up to 16.8 mg/kg. 5. Two morpholine derivatives appear to contain N-nitrosomorpholine. 6. The triethanolamine salt of dinoseb contains over 200 mg/kg N-nitrosodiethanolamine. 7. A large number of amides, carbamates, organophosphates, triazines, urea derivatives and miscellaneous nitrogen-containing pesticides do not contain N-nitrosamines above 1 mg/kg. 8. Most N-nitroso contaminants in pesticides can be avoided by simple process changes and the elimination of nitrite salts in the formulation.

[Resistance against mercury and cadmium by Staphylococcus aureus of diverse ecological origin].

For S. aureus isolates of different ecological origin, MIC-values for HgCl2 and CdSO4 were determined on a synthetical medium. Only strains from outbreaks of suppurative infections in hospitals were resistant to mercury, strains from infections in out-patients, from healthy human nasal carriers, from infections in cattle, sheep, chicken, and pigs were sensitive. Resistance to cadmium occurred in strains from man and from cattle but not in strains from sheep, chicken, and pigs. The majority of the cadmium-resistant strains was also resistant to penicillin, but some strains were only resistant to cadmium and sensitive to antibiotics.

[Staphylococcus aureus infections in domestic fowls. 1. Ecology and epizootiology. (Review)].

The ecological and epizootiological aspects relating to Staphylococcus aureus in domestic fowl are expounded, on the basis of literature, with particular reference being made to five topics: occurrence of these conditionally pathogenic bacteria in domestic fowl, point of attack and routes of infection, conditions for possible occurrence of infections, reciprocal infection between man and animal, as well as aspects of therapy and prophylaxis.
