Hemodynamic measurements in a sheep model with a hollow fiber artificial kidney containing modified cellulose.

HFAK-RC caused pronounced leukopenia, increase in TXB2 levels in plasma and hemodynamic pressure changes as a reflection of complement activation during EC in sheep. In contrast no increase in TXB2 levels and no changes in hemodynamics are observed with HFAK-MC. The leukopenia and granulocytopenia in the latter is much less pronounced and probably reflects the phenomenon "frustrated phagocytosis".

Arrangement of protofilaments in two forms of tubulin crystal induced by vinblastine.

Tubulin crystals produced by incubation with vinblastine sulphate have been investigated by X-ray diffraction, electron microscopy and density measurement. The results show that crystals produced in vitro from bovine brain tubulin are made of packed helices, each consisting of a single continuous protofilament, whereas crystals produced in vivo in sea urchin eggs are built from pairs of such helices. The symmetry, cell dimensions, percentage protein and composition of the crystals are consistent with either 24 or 30 tubulin monomers per turn of the helix. The sea urchin egg crystals are more highly ordered and thus more suitable for further analysis.

An apparatus for making fine-grained measurements of canine eating behavior.

An apparatus for making fine-grained measurements of eating and drinking in dogs is described. Examples of how this computer controlled measurement system can be used to illustrate eating behavior in one- and two-pan tests are given. Also, very fine-grained measurements are illustrated in illustrating a single meal segment and a single water drinking bout.

The influence of rate of skin indentation on threshold and suprathreshold tactile sensations.

The effects of skin indentation depth and rate on threshold and suprathreshold tactile sensations were investigated. Indentation rates between 0.3 and 10 mm/sec had little effect on the absolute tactile thresholds measured in terms of indentation depth. Slower rates resulted in increased absolute thresholds. Estimates of the growth in intensity of tactile sensations were made as functions of indentation depth and rate. The fastest rate used (10 mm/sec), for a given depth of indentation, produced the most intense sensation; the slowest (0.1 mm/sec), the least intense sensation. The tactile sensation magnitude estimates, with rate as the parameter, could be described by power functions. At the slowest indentation rate the exponent of the function was 1.36. At faster indentation rates (0.4, 1.0, and 10 mm/sec), two functions of markedly different slopes were required to describe the estimates. The exponents of the power functions were between 0.38 and 0.49 for indentation depths up to about 0.9 mm, and between 1.07 and 1.43 for deeper indentation depths.

Factor VIII-related antigen and procoagulant activity in manic-depressive patients.

Factor VIII-related antigen (VIIIR:AG) and procoagulant activity (VIII:C) were measured at weekly intervals in 11 manic-depressive patients during their hospital stay. There were significant correlations between VIII:C (P less than 0.02), VIIIR:AG (P less than 0.05) and the number of hospital admissions over a 10-year period. The patients' VIIIR:AG showed a wider range of results as compared to a control group (P less than 0.01). The age of onset of the disease (but not the age of the patient) was related to VIIIR:AG (P less than 0.02).

Comparative estimation of octopamine in the aortic smooth muscle, plasma and red blood cells of spontaneously hypertensive and Wistar Koyoto control rats.

Octopamine is an alpha agonist, the levels of which vary with age and in pathophysiological conditions. The levels of octopamine were found to be higher in the aortic smooth muscle and red blood corpuscles, but lower in the plasma of spontaneously hypertensive rats (SHR) than in the Wistar Koyoto rats. The importance of these variations are discussed.

Preparation of two-dimensional arrays from purified beef heart cytochrome c oxidase.

A method for preparing two-dimensional crystals from highly purified beef heart cytochrome c oxidase is described. This involves mixing the enzyme with phosphatidylcholine and then extracting excess lipid with deoxycholate. The reconstituted crystals show PI symmetry. Alternating rows of monomers are more closely packed (along the b axis) than in the previously described P121 crystals. However, the monomer structure in projection is the same in the two crystal forms. The P121 crystal form has been reacted with trypsin. This treatment did not alter the crystals but removed most of the impurities present in these cytochrome c oxidase preparation of low purity.

Location of the carboxyl terminus of bacteriorhodopsin in purple membrane.

Purple membrane samples have been prepared by trypsin digestion to have either 10 or 21 residues removed from the carboxyl terminus of the proteins. Electron diffraction of single membranes and x-ray diffraction of unoriented membrane pellets have been carried out on both these specimens and on native purple membranes. the main conclusion from this work is that the carboxyl terminus is almost entirely disordered, being free to take up many positions, and that its removal does not affect the packing in the crystal. The low resolution x-ray diffraction difference map may also suggest the approximate location of the carboxyl terminus.
