Controlled trial of bromocriptine, quinoestrol, and placebo in suppression of puerperal lactation.

2-bromo-alpha-ergocryptine (bromocriptine) in a dosage of 2-5 mg twice daily caused a rapid fall in plasma prolactin. It was more effective than either a single dose of 4 mg quinoestrol or a placebo in suppressing puerperal lactation, as judged by milk flow and the relief of breast pain and congestion. Patients who received quinoestrol were more comfortable than those who received placebo.

Capillary pH and blood gas determinations in asthmatic children.

Earlobe capillary pH and blood gases in asthmatic children were compared to arterial values. Hyperemia of the earlobe was produced by applying thurfyl nicotinate (Trafuril). Approximately 15 min later capillary blood was obtained simultaneously with a radial arterial sample. Earlobe capillary PO2, PO2, and pH correlated well with arterial values. This technique is simple, safe, and appears to be a satisfactory substitute for arterial blood in normotensive asthmatic children.

Nonimmune rosette formation: a measure of the newborn infant's cellular immune response.

Nonimmune rosette formation, an in vitro measure of cellular immunity, was evaluated in normal newborn infants. Active rosette formation in 14 specimens of cord blood was 18.9 +/- 4.8% compared to 28.1 +/- 5.2% in 15 adult control samples (p less than 0.05). Total rosette formation in 13 cord blood samples was 33.3 +/- 7.6% compared to 55.1 +/- 6.5% in 15 adult control specimens (p less than 0.05). Tritiated thymidine uptake from phytohemagglutinin stimulation was comparable in cord blood and adult control lymphocytes. The importance of these findings is discussed in light of other recent reports suggesting that cord blood thymic derived T lymphocytes may have reduced immune capability when compared to adult lymphocytes.

Some properties of a D-alanine carboxypeptidase in envelope fractions of Neisseria gonorrhoeae.

Envelope preparations of Neisseria gonorrhoeae strain GC1 (a stable, piliated strain of intermediate colony morphology) and type T1 possess a D-alanine carboxypeptidase which releases the terminal alanine residue from the uridine 5'-diphosphate-N-acetyl muramylpentapeptide substrate (isolated from Bacillus cereus T). The D-alanine carboxypeptidase of the GC1 envelopes has a broad pH optimum between pH 8.0 to 10.0. When the molarity of the tris(hydroxymethyl)aminomethane buffer was varied, the activity showed an optimum over the range 0.2 to 0.4 M. Activity was higher (135% of control level) when 20 to 80 mM Mg2+ was present. The Km for the enzyme was 0.25 mM. The D-alanine carboxypeptidase was inhibited by several beta-lactam antibiotics and the 50% inhibitory levels were 10(-8) M penicillin G, 10(-8) M ampicillin, 10(-5) M cloxacillin, and 5 x 10(-7) M methicillin.

Organization and control in the arginine biosynthetic pathway of Neurospora.

Eight enzymes involved in the conversion of acetylglutamate to arginine in Neurospora crassa were studied. The data indicate that of three enzymes early in the sequence, only the first, acetylglutamate kinase, is a nonorganellar enzyme. The next two, N-acetyl-gamma-glutamyl-phosphate reductase and acetylornithine aminotransferase, are in the mitochondrion, which was previously shown to contain the subsequent enzymes: acetylornithine-glutamate acetyltransferase, ornithine carbamyltransferase, and carbamyl-phosphate synthetase A (arginine specific). The last two enzymes of the pathway, argininosuccinate synthetase and argininosuccinate lyase, were previously shown to be cytosolic. All enzymes but one have low amplitudes or repression. Their levels respond little to arginine excess and are about twofold elevated (threefold for ornithine carbamyltransferase) as a result of arginine limitation in the arg-12-8 strain. No restriction of the incorporation of mitochondrial enzymes into mitochondria could be detected when the levels of these enzymes were elevated. Two enzymes, acetylglutamate kinase and carbamyl-phosphate synthetase A, which initiate the synthesis of the ornithine and guanidino moieties of arginine, respectively, show the lowest specific activities in crude extract. These enzymes display special regulatroy features. Acetylglutamate kinase, which has a typically low amplitude of repression, is subject to feedback inhibition. Carbamyl-phosphate synthetase A is wholly insensitive to arginine or citrulline in vitro or in vivo, but displays a very large amplitude of repression (about 60-fold). It is unique in that it can be almost completely repressed by growth of mycelia in excess arginine. These data suggest that mitochondrial localization may be incompatible with a mechanism of feedback inhibition by a cytosolic effector, arginine. Further, they suggest that the high repressibility of carbamyl-phosphate synthetase A compensates for its feedback insensitivity.

Evidence for the existence of endotoxic activity in three strains of Moraxella.

The phenol extraction technique was used to obtain material with endotoxic activityfrom three strains of Moraxella. This material was subjected to partial purification. Biological activity was determined by inoculation of the extracts into the skinof rabbits, and noting the resultant skin reactions. Although containing some cellular contaminants such as nucleic acid from the extraction procedure, the endotin preparations showed biological activity in the microgram range. Killed but unextracted cellsalso elicited skil reactions when injected intradermally into the rabbit. A partial analysis of the biochemical components of the endotoxin revealed that these three strains contain endotoxin whose composition is similar in several respects to that of the endotoxins of the neisseriae and some of the enteric bacteria.

Tissue specificity of nuclear acidic proteins isolated from bovine brain and adrenal medulla.

Nuclear acidic proteins from bovine brain and adrenal medulla demonstrated dissimilar electophoretic and chemical profiles. The amino acid analysis of the acidic nuclear protein fraction isolated from these tissues revealed some variation in the ratio of acidic to basic amino acids. The estimation of free carboxylic acid groups confirmed the more acidic nature of the brain proteins. In comparing the acrylamide gels either visually or optically, several electrophoretically specific bands were apparent. Although the total number of protein bands from each tissue was approximately the same, the adrenal medulla contained a larger proportion of the more positively charged proteins. These observations are interpreted to indicate that the nuclear acidic protein from brain and adrenal medulla may show functional variation.

Radioimmunoassay of serum pancreatic amylase in normal and pancreatectomized pigs (38504).

A double antibody radioimmunoassay has been developed that is capable of assaying and discriminating small amounts of pancreatic amylase even in the presence of amylase isoenzymes. Assays on adult pigs indicate a mean serum concentration of pancreatic amylase of 2.4mug/ml. To date, no other technique can offer such a discrimination of the pancreatic amylase in the circulation. Following pancreatectomy, the amount of radioimmunoassayable amylase in the circulation disappeared within 48 hr. This was reflected by a simultaneous decline in the serum amylase enzyme activity. Within 48 hr after pancreatectomy, the serum amylase enzyme activity had declined 60%. Thus, in pigs, the pancreas contributes significantly to the circulating amylase levels.

The effect of vasectomy and oviduct section on mouse behavior (38554).

The spontaneous movement of vasectomized adult Swiss Webster male mice was 60% less than sham-operated controls 45 days after the operation. A similar trend was recorded for tubectomized frmales. Exploratory behavior as measured by the open field test was not significantly influenced by either surgical proceedure.

Fallopian tube occlusion in rabbits with silicone rubber.

A new system for transcervically blocking the Fallopian tubes with cured-in-place silicone rubber has great potential for fertility control. Good retention, antifertility efficacy, and retrievability were observed in rabbits, and no adverse tissue reactions were seen in oviducts containing silicone elastomer. Continuing studies with rabbits and rhesus monkeys attempt to determine long term efficacy and histologic changes.

Effect of abdominal transplanted mammary tumor on peritoneal fluid cytology in mice.

In this study C3H/HeJ female mice received an intraperitoneal injection of mammary tumor suspended in saline; After 14 days, the presence of mammary neoplasm was detected by a significant change in the cytodifferential counts of aspirated peritoneal fluid. Previous studies suggest that ovarian cancer can be detected in mice and in women by the characteristic changes produced in abdominal cellular fluid distribution.
