Identification of peroxyacetic acid uroporphyrin I in the urine of patients with congenital erythropoietic porphyria by liquid chromatography and mass spectrometry.

A new porphyrin, peroxyacetic acid uroporphyrin I, has been isolated from the urine of patients with congenital erythropoietic porphyria by reversed phase high performance liquid chromatography. The porphyrin was characterized by high resolution mass spectrometry and by typical chemical reactions of a peroxyacid.

Establishment of human myeloma cell line KM-2R and its preliminary application to human-human hybridoma research.

The authors report the establishment of a human myeloma cell line--KM-2R--and its preliminary use in human-human hybridoma research. KM-2R cells are resistant to both 6-TG and ouabain. Their doubling time is about 30 h. They have a modal chromosome number of 81-85. Using ELISA and immunodiffusion techniques, gamma-heavy and kappa-light chains were detected in the concentrated cell culture supernatant. KM-2R cells are sensitive to HAT medium. When fusing with normal human spleen cells, tonsil cells, and B lymphoblasts transformed by EB virus, fusion frequencies of 20-60% resulted.

[Phospholipid components of danggui].

This paper deals with the phospholipid components in Danggui (Angelica sinensis and Levisticum officinale) by using thin-layer chromatographic scanning and the corrective method of absorbance proportional coefficient. Seven known and two unknown phospholipid components were separated and determined. The authors have compared the composition of phospholipids for six Danggui samples.

[Photodynamic therapy of zinc sulfonated phthalocyanine on murine transplanted tumors, its tissue distribution, and damaging effect on DNA of cancer cell].

Zinc sulfonated phthalocyanine (ZnSPc) 10 mg.kg-1 was injected iv into mice bearing S-180 and RA795 lung carcinoma, after 24 h tumor site were irradiated with red light. In mice bearing S-180, tumor regression rate was 31.8-43.5%, tumor growth inhibition rate was 57.4%. The highest concentration was in tumor tissue 24 h after injection of this dye, on d 5 it still retained relatively highest concentration. However, in most other tissues the dye was not detected at this time, disappearance of ZnSPc from plasma was rapid, it showed an open two compartment model, t1/2 alpha 135.8 min, t1/2 beta 70.1 h, Vd 1.92 x 10(-3) L. In blood, most ZnSPc was bound with plasma protein, the peak light absorption showed blue shift. ZnSPc 2.5 micrograms.ml-1 plus light, percent of DNA double strands greatly decreased, this indicated that DNA was one of target sites for ZnSPc photodynamic action.

[Pharmacological variations of Panax ginseng C.A. Meyer during processing].

Based on a comparative study on the documentation concerning the pharmacological actions of the red ginseng and the white ginseng, it has been found out that in promoting blood circulation, anti-thrombosis, fibrin dissolving activity, phagocytosis of reticular endothelium and anti-decrepit function on the red ginseng is stronger than the white one.

Isolation and characterization of beta-hydroxypropionic acid- and hydroxyacetic acid-uroporphyrin I in the urine of a patient with congenital erythropoietic porphyria by high performance liquid chromatography and liquid secondary ion mass spectrometry.

beta-Hydroxypropionic acid- and hydroxyacetic acid-uroporphyrin I have been isolated from the urine of a patient with congenital erythropoietic porphyria by reversed phase high performance liquid chromatography. The compounds were characterized by their chemical behaviour and confirmed by liquid secondary ion mass spectrometry.

Monoclonal anti-human T cell antibody and PAP-s conjugate--preparation and selective cytotoxic properties on leukemic cell.

Pokeweed antiviral protein (PAP-s) was prepared from seeds of Phytolacca americana. Monoclonal antibody against human pan-T lymphocyte Wu71 was linked to PAP-s by a disulfide bond. The results of SDS-PAGE, double immunodiffusion of active monoclonal antibody and PAP-s showed that the conjugate was highly cytotoxic to the human T-leukemic cell line CEM, but not to antigen-negative cell line SP2/O. At a concentration of 10(-9) mol/L, 76.4% of the target cells were killed, as compared with 10.1% at 10(-9) mol/L of free PAP-s. Treatment of the CEM cells with conjugate at 10(-9) mol/L reduced their rate of protein synthesis by 72.4%, as determined with 14C-leucine incorporation. The immunotoxin may be useful for the in-vitro eradication of leukemic cells in autologous bone marrow transplantation to leukemia patients.

[Changes in bile composition in rabbits infected with Clonorchis sinensis].

This article presents the changes in the concentration of bilirubin, cholesterol, phospholipid, bile acid and the activity of bacterial beta-glucuronidase in hepatic bile of rabbits infected with metacercariae of Clonorchis sinensis. We found that the activity of bacterial beta-glucuronidase was significantly higher in the infected group (52.6 +/- 31.6 u/dl) than that in control rabbits (12.5 +/- 20.5 u/dl) (P less than 0.001), and no marked changes in the concentration of bilirubin, cholesterol, phospholipid and bile acid were found. concentration of bilirubin, cholesterol, phospholipid and bile acid were found. By using alcian blue-PAS combined stain method the authors also noticed that the amount of glycoprotein in goblet cells has considerably increased and secreted increasingly to the biliary duct after infection with Clonorchis sinensis. As the increase in bacterial beta-glucuronidase activity and glycoprotein in bile is in favour of the formation of pigment stone, this may explain why clonorchiasis is often complicated with cholelithiasis.

The use of Chinese herb medicine in experimental radiotherapy.

Extracts from a group of destagnative herbs, 764-1, and the effective chemical, 764-3, were tested by an in vitro experiment using a HeLa-S3 cell line. Under aerobic conditions, the shoulder of the cell survival curve diminished or disappeared according to the different doses of 764-1 used, but no change in slope was observed. In nitrogen, when the dose of 764-1 increased to 20 mg/ml (ID20), besides the disappearance of the shoulder, the slope of the curve also showed changes at lower doses (4-8 Gy); up to 10-25 Gy the curve became more flattened. 764-3 showed almost a similar effect by mainly affecting the shoulder of the survival curve. At low drug doses a SER as high as 1.87 might be obtained under hypoxic condition. At the same time 764-1 was used in testing the effect on radiation lung damage. It was found that 764-1 could markedly inhibit the change of alveolar surfactant at 1 and 3 weeks after radiation. A wide field of investigation is thus spread out in front of us on radiosensitization and protection. Further studies on 764-3 are carried out.

Protein extraction with reverse micelles.

Liquid-liquid protein extraction with reverse micelles is a new method for protein separation. We studied the effects of pH, ionic concentration, cation type, and protein molecular mass on performance of an AOT/isooctane reverse micelle system in extraction of lysozyme, trypsin, and pepsin. Extraction was found to be very efficient for low-molecular-mass proteins.

[A research on the constituents of ginger in various preparations].

The constituents extracted by ether from ginger in different preparations were checked up by GC-MS-DS. The results are as follows: 25 constituents were found in fresh ginger; 22 in dry ginger, of which one had not been found in other forms of ginger; 23 in ginger roasted with sand in a pan, of which two had not been found in other forms of ginger; 23 in ginger charcoal, of which three had not been found in other forms of ginger. A comparison has been made of the content of the constituents in the ginger studied, and the result shows that some of the constituents have changed. All of the essential oil constituents in roasted ginger and ginger charcoal were reported for the first time.

Accurate and specific HPLC assay of coproporphyrinogen III oxidase activity in human peripheral leucocytes.

An HPLC method is described for the assay of coproporphyrinogen III oxidase in human leucocytes. The optimal pH for the assay was 6.5-7.0 and the Km for coproporphyrinogen III was 0.12 +/- 0.021 mumol/l. The mean activity in 28 apparently health subjects was 0.249 (2 SD range 0.130-0.368) nmol/h per mg protein. In two patients with hereditary coproporphyria, the activities were 0.029 and 0.078 nmol/h per mg protein.
