Genomic heterogeneity in Chlorobium limicola: chromosomic and plasmidic differences among strains.

Chromosome analysis by pulsed-field gel electrophoresis shows a high level of genetic heterogeneity between the two subspecies of the green sulfur bacteria Chlorobium limicola analysed: C. limicola and C. limicola f. s. thiosulfatophilum. Currently, they are differentiated only by the ability to utilize thiosulfate as photosynthetic electron donor, and by their %G + C content (51% and 58.1%, respectively). However, the capacity to utilize thiosulfate as photosynthetic electron donor does not appear to be a useful criterion to differentiate between some strains of this species, because this ability is encoded by plasmids that are different depending on the thiosulfatophilum strain analysed. In contrast, this study reveals that the comparison of chromosomal restriction patterns is very useful as an additional aid for the differentiation and identification of C. limicola strains.

Induced mutagenesis by bleomycin in the purple sulfur bacterium Thiocapsa roseopersicina.

Cell death and mutagenesis in bleomycin-treated cells of Thiocapsa roseopersicina (a purple sulfur bacterium) was studied by cultivation in a semisolid medium (agar-shake technique). This technique has also proven useful in assessing the frequency of antibiotic mutations by detecting and counting individual colonies of Thiocapsa roseopersicina. The frequencies of spontaneous mutants resistant to ampicillin, rifampicin, cloramphenicol, tetracycline, kanamycin, streptomycin, and neomycin were also studied: they ranged between 2 x 10(-9) and 9 x 10(-8). Bleomycin (4 micrograms/ml) sharply increased the frequency of ampicillin-resistant mutants, from 10(-8) (spontaneous) to 4 x 10(-4) (induced), in 17 h. An inducible, error-prone mechanism of DNA synthesis seems to be responsible for this enhancement of the mutagenic effect. this is the first report on the sensitivity to several antibiotics, and capacity of lethality and mutagenesis by bleomycin has been studied in a purple sulfur bacterium.

Differential characteristics of AIDS patients with a history of imprisonment.

BACKGROUND: AIDS is among the leading causes of death in prisons, but there is little information about AIDS patients with a history of imprisonment. METHODS: AIDS patients diagnosed in Barcelona between 1988 and 1993 were studied. Those with prison histories were compared to those without, with respect to epidemiological variables, including survival analysis. RESULTS: 28.5% of 2336 AIDS patients, 49.4% of intravenous drug users (IVDU) and only 2.6% of homosexuals who were not IVDU had a prison history. Those with prison histories, compared to those without, were younger (median age of 30.6 versus 36.4, P < 0.0001), more often IVDU than homosexuals (87.8% versus 35.8%, OR = 36.9, 95% CI: 22.6-60.8, P < 0.0001), and diagnosed with AIDS because of extrapulmonary tuberculosis (32.0% versus 14.7%, P < 0.001). Among IVDU, those with prison histories were more frequently males (OR: 2.2; 95% CI: 1.6-2.9), lived in the poorest district of Barcelona more frequently than in the richest district (OR: 6.6; 95% CI: 3.4-12.9) and presented with extra-pulmonary tuberculosis more frequently than Pneumocystis carinii pneumonia (OR: 1.7; 95% CI: 1.2-2.4). Longer survival in the prison group did not persist when adjusted for age and AIDS-defining disease. Those with prison histories who presented with AIDS with only extrapulmonary tuberculosis had better probability of survival than those who presented only with P. carinii pneumonia (P < 0.001). CONCLUSIONS: AIDS patients in Barcelona with prison histories tended to be younger, more likely to be IVDU, and to present with extrapulmonary tuberculosis as an AIDS-defining illness than other patient groups. Better survival appears to be related to age and AIDS-defining illnesses in the prison group. The fact that half the IVDU AIDS cases had prison histories has important implications for the care and prevention of HIV, tuberculosis, and drug abuse in comparable prison settings.

Biodegradation of Poly-(beta)-Hydroxyalkanoates in a Lake Sediment Sample Increases Bacterial Sulfate Reduction.

The addition of a poly-(beta)-hydroxyalkanoate (PHA) to incubation bottles containing anaerobic sediment from Lake Ciso (Banyoles, Catalonia, northeast Spain) was correlated with an increase in sulfide production. The concentration of PHA diminished to 51 to 99% of the initial amount. Sodium molybdate (1 mM) decreased the rate of PHA degradation and inhibited sulfide production, thus indicating that PHAs serve as carbon and electron sources for sulfate reduction.

Life as a planetary phenomenon: the colonization of Mars.

Life is a planet-wide phenomenon in which its components incessantly move and interact. Life imperatively recycles its parts at the surface of the Earth in a chemical transformation and physical transport that depends utterly on the energy from a recent star, the Sun. Humanity, entirely dependent on other beings, plays a recent and relatively small part in the great phenomenon of life that transports and transforms the surface of the Earth. Our species accelerates but does not dominate the metabolism of the Earth system. Ironically, during the Apollo days of the sixties, fears were rampant that Martian or other extraterrestrial "germs" might "contaminate" our planet. After Viking, such fears are seen as the manifestation of cultural paranoia. The Viking missions complemented ground-based astronomical observation and yielded definitive evidence for the lack of life on the red planet. The Gaia hypothesis states that the surface temperature, composition of the reactive gases, oxidation state, alkalinity-acidity on today's Earth are kept homeorrhetically at values set by the sum of the activities of the current biota. Life, in other words, not only produces and maintains its immediate environment, but appears on Earth only as a planetary phenomenon. Since the natural tendency of all life is to grow exponentially to fill proximal volume, the question now "can life ecopoietically expand to Mars?" is entirely equivalent to the query of "can Gaia reproduce?"

Analysis of bacterial genomes by pulsed field gel electrophoresis.

The present article reviews the main achievements reached in bacterial genetics through the application of pulsed field gel electrophoresis. In particular, it summarizes the main subjects where the use of this technique has significant relevance for the analysis of bacterial genomes. With this purpose, it sums up some of the most relevant examples which illustrate the applicability of this technique in bacterial taxonomy, in epidemiological work and in the study of bacterial genome organization (including the assessment of the number of genomic elements and the size and topology of all of them). This technique, which makes it possible to make genomic comparisons in an easy, fast and repetitive way, has been demonstrated to be especially useful for those bacteria where the application of classical genetic techniques has not been possible.

Susceptibility of various purple and green sulfur bacteria to different antimicrobial agents.

Several purple and green sulfur bacteria (genera Chromatium, Thiocapsa and Chlorobium) were tested for their sensitivity to different antimicrobial agents by a disc diffusion assay, using thioacetamide as a source of hydrogen sulfide for plate growth. Chlorobium limicola strains were more sensitive to amoxicillin, erythromycin and nalidixic acid, whereas gentamicin and netilmicin were more active against the purple bacteria tested. None of the organisms were sensitive to oxacillin and trimethoprim+sulfamethoxazole. The critical concentrations at the edge of the inhibition zone were also calculated for three organisms and the antimicrobials colistin, mitomycin C, penicillin G, rifampicin, and streptomycin. The results obtained suggest that colistin, mitomycin C, penicillin G would provide selective conditions against the growth of Chlorobium limicola strains, while streptomycin and other aminoglycoside antibiotics would select against purple bacteria.

Transformation of Chlorobium limicola by a plasmid that confers the ability to utilize thiosulfate.

A modified transformation method for Chlorobium limicola confirms the role of an endogenous plasmid in thiosulfate metabolism. The plasmid was present in two forma specialis thiosulfatophilum (Tio+) strains and absent in one non-thiosulfate-utilizing (Tio-) strain. The plasmid (size, 14 kb) was transferred by transformation from Tio- to Tio+. The chromosomal restriction patterns, analyzed by pulsed-field gel electrophoresis, were used to distinguish between the different strains.

Hypersensitivity of Rhodobacter sphaeroides ribosomes to protein synthesis inhibitors: structural and functional implications.

The elongation cycle of protein synthesis systems of purple nonsulfur photosynthetic bacteria Rhodobacter sphaeroides, grown both phototrophically and chemotrophically, was studied using 33 inhibitors with different chemical structures and functional and domain specificities. No functional differences between phototrophic and chemotrophic ribosomal systems were detected. Rhodobacter sphaeroides ribosomes exhibited strong hypersensitivity to nine functional inhibitors when compared with Escherichia coli ribosomes. Most of the R. sphaeroides ribosomal hypersensitivities corresponded to peptidyltransferase inhibitors, implying that this important functional neighborhood must be somehow different in the two organisms.

Six osteocalcin assays compared.

We evaluated comparatively six commercially available osteocalcin kits: ELSA-OST-NAT IRMA (CIS), ELSA-Osteo IRMA (CIS), Osteocalcin IRMA (Nichols), OSTK-PR RIA (CIS), OSCA Test Osteocalcin RIA (Henning), and Osteocalcin RIA (Nichols). All of them presented acceptable values of variance, sensitivity, and serial dilutions. However, only the ELSA-OST-NAT, ELSA-Osteo, and OSCA Test kits had analytical recoveries near 100% for added purified human osteocalcin; the other assays showed important differences in standardization. After treating samples with antibody against amino acids 43-49 of human osteocalcin, only the ELSA-OST-NAT IRMA detected no residual osteocalcin. Stability of serum values at room temperature and at 4 degrees C is good in the ELSA-Osteo and Osteocalcin IRMAs and in the Henning RIA if serum samples are collected in special tubes available with the kit. Differences in stability and after antibody treatment can be due to the degradation of intact osteocalcin molecules in serum and to osteocalcin fragments detected by assays. Results by different assays were well correlated for samples from controls and postmenopausal osteoporotic women but not for patients with impaired renal function or Paget disease of bone, probably because of osteocalcin fragments accumulated in serum of these patients and differences in specificity of the assays.

Composite, large spirochetes from microbial mats: spirochete structure review.

Phenomena previously unknown in free-living spirochetes are reported: large-sized cells with variable diameter (length to 100 microns, width between 0.4 and 3.0 microns), composite structure (smaller spirochetes inside larger ones), and positive phototropic behavior. These bacteria, Spirosymplokos, are compared with all other spirochete genera. The large spirochete, grown in mixed culture, was studied live and by transmission EM. The protoplasmic cylinder was replete with spherical granules 20-32 nm in diameter, and three to six periplasmic 26-nm flagella were inserted subterminally. Comparably granulated and flagellated small spirochetes were located inside the protoplasmic cylinder and in the periplasm of the large ones. When exposed to air, movement became erratic, protoplasmic cylinders retracted to lie folded inside the outer membrane, and refractile membranous structures formed. From one to four structures per still-moving spirochete were seen. Spirosymplokos was enriched from laboratory samples exposed to oxygen-rich and desiccating, but not dry, conditions for at least 4 mo after removal of microbial mat from the field.

Spirosymplokos deltaeiberi nov. gen., nov. sp.: variable-diameter composite spirochete from microbial mats.

Large (up to 100 micrometers long), loosely coiled, free-living spirochetes with variable diameters (from 0.4 to 3 micrometers in the same cell) were seen at least 40 times between August 1990 and January 1993. These spirochetes were observed in mud water and enrichment media from highly specific habitats in intertidal evaporite flats at three disjunct localities, one in Spain and two in Mexico. All three are sites of commercial saltworks. Associated with Microcoleus chthonoplastes the large spirochetes from Spain display phototaxis and a composite organization. Shorter and smaller-diameter spirochetes are seen inside both healthy and spent periplasm of larger ones. Small spirochetes attached to large ones have been observed live. From two to twelve spirochete protoplasmic cylinders were seen inside a single common outer membrane. A distinctive granulated cytoplasm in which the granules are of similar diameter (20-32 nanometers) to that of the flagella (26 nanometers) was present. Granule diameters were measured in thin section and in negatively-stained whole-mount preparations. Based on their ultrastructure, large size, variable diameter, number of flagella (3 to 6), and phototactic behavior these unique spirochetes are formally named Spirosymplokos deltaeiberi. Under anoxic (or low oxygen) conditions they formed blooms in mixed culture in media selective for spirochetes. Cellobiose was the major carbon source in 80% seawater, the antibiotic rifampicin was added, mat from the original field site was present and tubes were incubated in the light at from 18-31 degrees C. Within 1-2 weeks populations of the large spirochete developed at 25 degrees C but they could not be transferred to fresh medium.

Distribution of types of microbial mats at the Ebro Delta, Spain.

The distribution and types of microbial mats of the Ebro Delta (Catalonia, Spain) are described. The studied area is La Banya spit, formed by a narrow sand bar and a peninsula, located south of the main body of the Ebro Delta. Sea water can penetrate into the back shore through channel inlets, cutting the steeper coastal barrier of the open sea side of the spit or through the complex drainage channel system of the low-energy beaches in the inner Alfacs Bay. Sea water can stay in the back shore almost permanently, trapped in isolated depressions and lagoons. The surface distribution of microbial mats in La Banya spit has been studied by means of a detailed interpretation of vertical aerial photographs and field work consisting of mapping and sampling. The observed different ratios of cyanobacteria, as well as the presence and thickness of the layers of anoxygenic sulfur phototrophic bacteria, depend on the moisture content, the system stability, and the age of the microbial mat. Lyngbya, Oscillatoria, and Spirulina are the first cyanobacteria able to colonize the bare sediment. Lyngbya dominates in young microbial mats and in mats exposed to frequent desiccation. Microcoleus is the second most important colonist in the microbial succession. In relation to water, the alternation of emergence and submergence is optimal for the maximal development of Microcoleus-dominated microbial mats. We classify the microbial mats of the Ebro Delta into five main types: (i) Lyngbya-dominated type, in which the anoxygenic phototrophic bacteria are absent and the black layer of sulfate-reducers is very thin; (ii) Spirulina-dominated type; (iii) Oscillatoria-dominated type, which is found only at one site and covers a small area--this type, like the Spirulina-type mat, is not common in the Ebro Delta; (iv) Lyngbya/Microcoleus-transition type, in which Microcoleus and Lyngbya coexist in similar proportions--in the more developed mats of this group a layer of purple bacteria is usually present, and the black layer of sulfate-reducers is usually also well developed; and (v) Microcoleus-dominated type--in La Banya spit, this type is found in localities with relatively stable conditions. These areas are wet during most of the year. After appropriate conditions of diagenesis, the most highly developed microbial mats may be preserved as laminated hard sediments. The field study has been completed with cultivation, isolation, and identification of the main cyanobacterial genera under laboratory conditions.

Endotoxin-induced pulmonary dysfunction is prevented by C1-esterase inhibitor.

In septic shock, hypotension, disseminated intravascular coagulation, and neutrophil activation are related to the activation of the blood coagulation contact system. This study evaluates in dogs the effect of the C1-esterase inhibitor (C1-INH), a main inhibitor of the blood coagulation contact system, on the cardiovascular and respiratory dysfunction associated with endotoxic shock. Two groups were included: controls, which received Escherichia coli endotoxin, and a C1-INH group in which C1-INH was infused before E. coli endotoxin administration. In both groups, endotoxin produced hypodynamic shock; however, the decrease in the systolic index and the ventricular systolic work indexes were greater in controls than the C1-INH group. In controls, the arterial O2 partial pressure decreased by 30% and the alveolo-arterial O2 difference increased by 625%, these parameters remained unchanged in the C1-INH group. Hypoxemia was associated with increased intrapulmonary shunt, decreased blood coagulation contact factors, and decreased C3c. In contrast, C1-INH administration prevented endotoxin-induced hypoxemia, the increase in intrapulmonary shunt, and the decrease in blood coagulation contact factors. This study shows that, in dogs with endotoxic shock, pulmonary dysfunction is associated with an activation of the blood coagulation contact phase system. An inhibition of this system by C1-INH prevented the hypoxemia induced by endotoxic shock.

Diagnosis of brain death: superiority of perfusion studies with 99Tcm-HMPAO over conventional radionuclide cerebral angiography.

The use of technetium-99m hexamethyl-propyleneamine oxime (99Tcm-HMPAO) in the diagnosis of brain death has been evaluated in 41 studies of 37 patients with severe brain injury, who were under the effect of drugs or when other diagnostic methods were equivocal. HMPAO studies were compared with conventional radionuclide angiography performed simultaneously by intravenous administration of HMPAO as a bolus. The ages of patients ranged from 4 months to 75 years. Dynamic flow images and 5-min static uptake images were acquired following bolus injection of 555 Mbq of 99Tcm-HMPAO. All patients showing no brain uptake were confirmed as brain-dead by standard clinical criteria, with no contradictory cases in the static study. In addition, all patients who were not brain-dead showed HMPAO uptake at least in the brainstem. Dynamic flow images were equivocal in five patients, four of whom had no uptake on static images and clinically confirmed brain death. In addition, two other cases showed "mismatched" dynamic and static images: in one case no perfusion was observed on flow images but uptake restricted to the posterior fossa was seen on static images; the other case showed perfusion on the dynamic study and static imaging revealed hemispheric uptake with no posterior fossa uptake. Static perfusion 99Tcm-HMPAO studies offer advantages over conventional brain scintigraphy, better results being due to adequate assessment of posterior fossa activity and avoiding equivocal studies.

Efficiency of human Plasmodium falciparum malaria vaccine candidates in Aotus lemurinus monkeys.

The protective efficacy of several recombinant and a synthetic Plasmodium falciparum protein was assessed in Aotus monkeys. The rp41 aldolase, the 190L fragment of the MSA-1 protein and fusion 190L-CS. T3 protein containing the CS.T3 helper "universal" epitope were emulsified in Freund's adjuvants and injected 3 times in groups of 4-5 monkeys each one. The synthetic polymer Spf (66)30 also emulsified in Freund's adjuvants was injected 6 times. Control groups for both experiments were immunized with saline solution in the same adjuvant following the same schedules. Serology for malaria specific antibodies showed seroconversion in monkeys immunized with the recombinant proteins but not in those immunized with the polymer nor in the controls. Challenge was performed with the 10(5) parasites from the P. falciparum FVO isolate. Neither rp41 nor Spf(66)30 induced protection, whereas 190L induced significant delay of parasitemia. The fusion of the CS.T3 epitope to 190L significantly increased its protective capacity.
