Methylation of elongation factor 1 alpha from the fungus Mucor.
A basic protein from the dimorphic fungus Mucor racemosus, found to be highly methylated, is shown to be protein synthesis elongation factor 1 alpha. This protein is the most abundant protein in hyphal cells but is less abundant in yeast cells. It is post-translationally methylated with the formation of mono-, di-, and trimethyllysine at as many as 16 sites. Nearly 20% of the 44 lysine residues of elongation factor 1 alpha from mycelia are modified while those from sporangiospores are virtually unmethylated.
IgG antibodies: in vitro blocking activity of IgE mediated reactions.
Sera from 177 atopic patients treated by immunotherapy were screened in order to detect IgD antibodies against a purified fraction of Lollium perenne (fractions C). IgD antibodies were found in 23% of the subjects. Eight of these sera were selected and their IgD isolated by means of immunoabsorption. The in vitro biological activity of the IgD antibodies against fraction C was tested by three methods: basophil degranulation, histamine release and RAST inhibition test. In all these assays, the data obtained indicated a possible blocking effect of the IgD antibodies upon the IgE in vitro mediated reactions.
Fractionation and biological characterization of Olea europea pollen extract.
Preliminary fractionation of Olea european pollen extract has been performed. At least 10 antigenic fractions have been found by crossed electrophoresis. After Sephadex gel filtration, two fractions with a molecular weight of 160,000 and 65,000 have been obtained. The fractions were evaluated for allergenic activity by two in vitro techniques: polystyrene tube radioimmunoassay (PTRIA) and basophil degranulation test (BDT), and by skin tests. All tests indicated that the most reactive fractions were those in the 65,000 molecular weight peak. BDT has been shown to be a very reliable method as compared with histamine release and other parameters. Although common antigenic fractions have been found for Lollium perenne and O. europea, no cross-allergenicity has been shown by PTRIA inhibition.

Cytochrome b-245 of neutrophils is also present in human monocytes, macrophages and eosinophils.
A cytochrome b with a midpoint oxidation-reduction potential of -245mV (cytochrome b-245) that is a major component of the microbicidal oxidase system of human neutrophil leucocytes has been identified in human eosinophils, monocytes and macrophages at concentrations similar to that found in human neutrophils. It was absent from a variety of other cells. This cytochrome is present in phagocytic leucocytes and probably plays an important part in the specialized activities of these cells.
The respiratory burst of phagocytic cells is associated with a rise in vacuolar pH.
Neutrophil leukocytes are the body's major defence against bacteria, which they phagocytose and kill. It has been found that phagocytosis and killing are accompanied by a dramatic rise in non-mitochondrial respiration; and that the efficiency of killing is impaired in the absence of oxygen. It is also impaired in neutrophils from patients with chronic granulomatous disease (CGD), where the respiratory burst is absent. This has been difficult to reconcile with their normal content of granule proteins that kill bacteria in vitro. Indeed, CGD cells are essentially normal both morphologically and constitutionally except that they lack a functional very low potential cytochrome b (b-245), which is a component of the oxidase system responsible for the respiratory burst of normal cells. Activation of the oxidase is associated with the generation of various reduced oxygen species which have been widely thought to be responsible for the killing of phagocytosed microorganisms either directly, or by acting as substrates for myeloperoxidase-mediated halogenation. We report here, however, that a major consequence of the defective function of this oxidase in neutrophils and monocytes from CGD patients is an absence of the normal initial rise, and an unusually rapid and extensive fall in pH which is itself associated with the impairment of the killing and digestion of intracellular staphylococci.

Electrocardiogram computer analysis. practical value of the IBM Bonner-2 (V2 MO) program.
One thousand ECGs were selected from those taken from an adult population and analyzed by three cardiologists to assess the performance of the IBM Bonner-2 (V2 MO) program. The sample included 200 ECGs with 248 myocardial infarctions, 200 with conduction abnormalities, 100 with ventricular hypertrophy, 300 normals and 200 with electronic pacemakers. In the MI group, there was a total sensitivity of the program with respect to the readers of 88% (218/248) with 19 (8%) program errors and 90% (248/275) specificity. In 53 ECGs with two MI statements, the sensitivity was 89% (47/53). The sensitivity in conduction abnormalities was 93.4% (183/196) with 98.7% specificity (800/810). The sensitivity in LVH was 90% (74/81) and in RVH 83% (5/6). Among the normals, the specificity was 98.6% (289/293). The sensitivity to PVCs was 84% (56/67), to atrial fibrillation 87% (48/55), to SVCs 58.6% (17/29) and to electronic pacemakers 65% (127/196). Recognizing the limitations of this type of analysis, this study indicates that the V2 MO version compares favorably with the earlier versions of the same program and is a valuable aid in ECG interpretation with rapid acceptance by physicians.
Renin substrate in rat mesenteric artery.
The concentration of renin substrate (RS) was measured in rat mesenteric artery tissue. The concentration of this substrate both in arterial tissue and in plasma was markedly higher in rats 1 day after bilateral nephrectomy than in sham-operated controls, the percentage difference being higher in plasma than in arterial RS. Conversely, the decrease apparently induced 3 days after adrenalectomy (i.e., the difference in RS concentration from sham-operated rats) was greater in arterial tissue than in plasma. This finding may be explained by changes in RS concentrations induced by the sham operation. Sham surgery itself increased plasma RS after 1 day (but not after 3 days) and arterial RS after 3 days (but not after 1 day). There was a positive correlation between arterial and plasma renin substrate concentration for the overall results but not within individual groups. As renin and angiotensin-converting enzyme activity are also present in arterial tissues, all the necessary components for local generation of angiotensin II have now been shown to be present within the wall of resistance vessels.
Effects of tonin on the response to norepinephrine by the aortic strip of the hypertensive rat.
The response to norepinephrine (NE) of arterial smooth muscle from two types of experimental hypertensive rats was investigated. Aortic strips from one-kidney, one-clip hypertensive animals were less responsive to NE than those from their normotensive controls but strips from one-kidney, one-clip hypertensive animals showed no difference from their corresponding controls. The contractility in response to NE was the same in all groups. These results suggest that the mechanisms responsible for lesser reactivity in the one-kidney hypertensive group are not a consequence of elevated blood pressure itself but may be related to changes in the intrinsic sensitivity of aortic smooth muscle. Tonin potentiated the contraction induced by NE in aortic strips from hypertensive and normotensive rats. This effect was more pronounced in the one-kidney, one-clip hypertensive animals, so that although the aortic smooth muscle from these animals is less reactive to NE, the decreased reactivity can be more than compensated by the presence of tonin. The mechanism of potentiation is not yet clear but the fact that Saralasin did not inhibit it suggests that angiotensin II is not generated in situ.
Pressor effect of tonin in anephric animals.
Tonin was injected intravenously to normal rats without effect on blood pressure. Twenty-four hours after bilateral nephrectomy, tonin produced a dose-dependent pressor effect in rats which was abolished by the angiotensin antagonist [Sar1-Ala8]-angiotensin II. Vascular response to angiotensin II was slightly increased after nephrectomy. Plasma angiotensin II increased significantly after injection of tonin and disappeared biexponentially with a half-life of less than 1 min for the fast component and 9 min for the slow component. The change in plasma angiotensin II correlated with the elevation in mean blood pressure. No difference in inhibitory power of plasma on tonin activity could be shown between intact and nephrectomized rats. In vitro, the initial velocity of generation of angiotensin II by tonin acting on plasma increased after addition of semipurified rat renin substrate and was significantly greater in plasma of nephrectomized rats. In nephrectomized rabbits, but not in intact ones, a dose-dependent pressor effect was produced by tonin. These data demonstrate the in vivo production of angiotensin II by tonin in an animal model with elevated substrate levels. Together with the in vitro data, these results suggest a role for substrate concentration in the expression of tonin enzymatic activity in vivo.

Role of T gamma cells in the in vitro IgE response after antigenic stimulation.
Fifteen patients with seasonal allergic pollenosis and five controls were investigated to elucidate the role of the T gamma-cell population in the in vitro IgE response by peripheral blood lymphocytes (PBL). In vitro IgE production by PBL of atopic patients after antigenic stimulation was measured in culture supernatants. The optimal dose for antigenic stimulation was found to be 0.16 micrograms/20 X 10(6) cells of purified antigen. No difference was found when comparing the percentages of T cells (E rosettes) between the two groups: mean per cent for controls was 69.2 +/- 5.76 versus 69.54 +/- 4.42 for the allergic group. With regard to the T gamma-cell population, the values obtained by rosetting with ox erythrocytes sensitized with IgG antibody were 12 +/- 0.71% in normals and 9.8 +/- 1.32% in those with allergic pollenosis. This difference, although significant, may not be enough to explain the different pattern when the in vitro IgE production of both groups investigated was compared. In order to detect the role of T gamma cells in this system, lymphocyte cultures, depleted of T gamma cells, were performed and compared with unfractionated cultures from the same donors. Our results show no differences in the in vitro IgE production when T gamma cells were depleted as compared with the unfractionated cultures.
