Membrane-associated immunoglobulin of rabbit peripheral blood lymphocytes: allelic exclusion at the b locus.

Light chain allotypic determinants were demonstrated on the surface of peripheral blood lymphocytes of rabbits. By means of a combined immunofluorescent and autoradiographic technique, a great majority of the individual lymphocytes of rabbits heteroxygous at the b locus were shown to have detectable quantities of only one allotype. A small proportion of lymphocytes appeared to possess both allelic forms of L chains.

Structural basis of the A14 and A15 allotypic specificities in rabbit immunoglobulin G.

The structural basis for the A14 and A15 allotypic specificities of rabbit immunoglobulin G was investigated with Fe fragments prepared from homozygous rabbits. The presence of threonine in the Cgamma2 region (position 309: Eu numbering) correlates with the A14 allotype; the presence of alanine at this same position correlates with the A15. Now that the sequence of the Cgamma2 region of rabbit IgG has been completely elucidated, a high degree of homology of the constant region domains of rabbit and human gamma chains is apparent (about 60-80%).

Genetic analyses of restriction fragment length polymorphisms using high molecular weight DNA from sperm or lymphocytes embedded in agarose.

A simple and efficient method for determining restriction fragment length polymorphism types on large numbers of individuals using small samples of peripheral blood or sperm cells is described. Whole cells embedded in low gelling/melting temperature agarose were treated with a series of enzyme, detergent, and washing steps to release high molecular weight DNA that was then digested with standard restriction enzymes such as EcoRI and PstI, electrophoresed, blotted, and probed as in normal Southern analyses. The technique should be readily adaptable to any application requiring DNA from small numbers of cells for Southern analyses or pulsed field gel electrophoresis.
