Hypocholesterolemic effect of yogurt and milk.

In order to determine the effect of milk products on serum cholesterol, triglycerides, and diet, 54 volunteers were studied for varying periods with dietary supplementation of nonpasteurized yogurt, pasteurized yogurt and 2% butterfat milk. Serum cholesterol was significantly reduced by 5 to 10% after 1 week of supplementation with either nonpasteurized or pasteurized yogurt; 2% butterfat milk reduced serum cholesterol to a smaller and less significant effect. Serum triglycerides were unaffected by the diet and dietary intake studies confirmed that intake of other nutrients remained relatively stable throughout the study. Supplementation of diet with yogurt may have a helpful hypocholesterolemic effect.

Superoxide dismutase activity in nerve cell culture.

Superoxide dismutase (EC 1.15.1.1) activity was assayed in preparations obtained from several clonal lines of nerve cell culture, by enzymatic and nonenzymatic assays. The enzyme was found in dialyzed homogenates of washed cells and in partially purified fractions. The enzyme was also found in cells which had been grown in media containing 5-bromodeoxyuridine, where cell differentiation was observed.

Effect of hyperoxide radicals on bovine-erythrocyte membrane.

1. Bovine erythrocytes exposed to the action of an enzymic source of hyperoxide radicals (hypoxanthine + xanthine oxidase) exhibited hemolysis, which was prevented by the presence of hyperoxide dismutase. 2. Exposing bovine erythrocyte membranes to the source of hyperoxide radicals resulted in a decrease of (Mg2+ + Na+ + K+)ATPase activity which could be partially prevented by addition of hyperoxide dismutase. 3. The damage observed to erythrocyte membranes under the conditions applied is ascribed to toh formed in the Haber and Weiss reaction since a protection by OH scavengers was also observed.

Enzymatic oxidation of diethyldithiocarbamate by xanthine oxidase and its colorimetric assay.

Diethyldithiocarbamate is oxidized in vitro by purified cream xanthine oxidase in the presence of nitro blue tetrazolium and phenazine methosulfate. Disulfiram is formed during the enzymatic or nonenzymatic oxidation. When diethyldithiocarbamate is mixed nonenzymatically with nitro blue tetrazolium, the dye is rapidly reduced, but only in the presence of certain organic solvents. This reaction can be used as a convenient colorimetric assay for diethyldithiocarbamate, which gives a stable color over a wide concentration range.

[A simple photometric test for the routine determination of delta-aminolaevulinic acid in urine for the control of lead exposure].

A simple and accurate method for determining urinary concentration of delta-aminolaevulinic acid (delta-ALA) is described. It is based on an improved application of the principle, first introduced by Tomokuni and Ogata, that a colourless pyrrole derivative is produced by the action of acetic acid ethyl ester on delta-ALA, which develops a violet colour on addition of Ehrlich's reagent. The influence of interfering substances was found to account for 20 plus or minus 3% in all examined urines samples. It is, therefore, recommended that an aqueous delta-ALA standard be prepared which contains only 80% of the concentration used in the calculation. Exact values are obtained in this way and the need for setting up a calibration curve by adding delta-ALA is avoided.

Enzymatic and non-enzymatic assay of superoxide dismutase.

Superoxide dismutase from breef brain and rat liver was assayed in an enzymatic system, using xanthine oxidase, and a non-enzymatic system, based on aerobic reduction of nitro-blue tetrazolium in presence of phenazine methosulphate. The non-enzymatic assay is rapid and simple and permits simulatneous analysis of many samples. Similar results are found by the two methods of assay of superoxide dismutase.
