Extra microchromosome mosaicism in amniotic cells confirmed in fetal tissues.

Mosaicism for an extra microchromosome was discovered in amniotic cell cultures of a 39-year-old woman. Using G, Q, C bands and silver staining, it was concluded that the extra chromosome was bisatellited. Parents' karyotype was normal. Parents elected for termination of the pregnancy. The presence of the extra microchromosome was confirmed in various tissues of the aborted fetus. The literature on the subject is briefly reviewed.

[CSF cytology using prestained slides (author's transl)].

A new and rapid vital-stain method for the diagnosis of CSF cells demonstrates not only red blood cells but also white blood cell types, predominantly by differentiating nuclear patterns. 20 microliter untreated CSF are put on the prestained slides and a cover slip put over it. Differentiation can be made after 15 minutes. The lower limit is at 3-4 WBC/ml of CSF. At room temperature the preparation keeps for about an hour. The method is reproducible and has the advantage also over other routine methods that the CSF volume is very small and there is neither loss of cells nor cell denaturation.

[Therapeutic experiences using the new podophyllotoxin derivative VP 16-213 in malignant human tumors].

The new semisynthetic epipodophyllotoxin VP 16-213 has been tested for antitumor activity and toxicity in a non-randomized phase II trial. The study included 33 patients with solid tumors and acute leukemias. The drug was given in 5-day courses every 3 weeks, parenterally for the first 2 courses and thereafter orally. Subjectively VP 16-213 was well tolerated in both i.v. and oral administration, but almost universally induced leukothrombopenia and hair loss. The best results were achieved in 12 patients with oat cell carcinoma of the lung (4 good and 4 less good responses). Remissions of lesser quality were also observed in anaplastic lung cancer, ovarian carcinoma and acute myelomonocytic leukemia. VP 16-213 possesses proven, clinically valuable cytostatic activity against various human neoplasms. Complementary studies to establish its optimum dosage and administration, and its place in combination chemotherapy, are in progress.
