Microfluorometric assessment of sperm maturation in testicular biopsies from men with histologically normal or reduced spermatogenesis.

Spermatogenesis has been studied in testicular biopsies by means of microfluorometric assessment of the DNA-DNP complex using ethidium bromide in 10 men with histologically normal spermatogenesis and in 10 men with histologically reduced spermatogenesis. The expected haploid DNA value was found in the round spermatids, whereas only 70% of the haploid fluorescence value was found in the elongated spermatids and around 60% in the testicular spermatozoa. No difference was found in the mean fluorescence values between the pathological group and the controls. it is suggested that structural changes or an increase in basic nuclear proteins gradually exclude the ethidium bromide from the binding to the DNA molecule and that this phenomenon occurs concomitantly with a decrease in double-stranded RNA. The difference in fluorescence intensity before and after RNAase treatment was regarded as an approximate estimate of the RNA content. There was a significant decrease of these values during spermatogenesis.

Zinc and zinc ligands in human seminal plasma. II. Contribution by ligands of different origin to the zinc binding properties of human seminal plasma.

The zinc binding properties of whole human seminal plasma and various fractions of split ejaculates were analysed with gel chromatographic and dialysis methods. The proportion of dialyzable zinc was strongly correlated to the zinc/fructose ratio in the seminal plasma, i.e. correlated to the relative contributions by the prostate and seminal vesicles, respectively. Zinc originates, however, almost entirely from the prostate gland. The non-diffusible zinc ligands thus seemed to originate from the seminal vesicles. This could also be confirmed in experiments with split ejaculates. The high molecular weight (HMW) zinc ligands from the seminal vesicles have also a higher affinity for zinc than the low molecular weight (LMW) ligands from the prostate. A redistribution of zinc therefore takes place after ejaculation. The HMW zinc ligands from the seminal vesicles were found to have an isoelectric point at 6.1-6.7, to be glucoproteins, and to have an apparent molecular weight of 250,000. The possible physiological implications of a redistribution of zinc after the ejaculation is discussed in view of the fact that zinc is of importance for many functional properties of the human spermatozoa.

Vitamin A in human semen.

Vitamin A concentration has been determined in human semen with and without hydrolysis by fluorometry and high performance liquid chromatography (HPLC). The vitamin A content of human spermatozoa is much lower than that reported for rabbit spermatozoa but approximately equal to that of bull spermatozoa.

Discontinuous synthesis of both strands at the growing fork during polyoma DNA replication in vitro.

In discontinuous polyoma DNA replication, the synthesis of Okazaki fragments is primed by RNA. During viral DNA synthesis in nuclei isolated from infected cells, 40% of the nascent short DNA fragments had the polarity of the leading strand which, in theory, could have been synthesized by a continuous mechanism. To rule out that the leading strand fragments were generated by degradation of nascent DNA, they were further characterized. DNA fragments from a segment of the genome which replication forks pass in only one direction were strand separated. The sizes of the fragments from both strands were similar, suggesting that one strand was not specifically degraded. Most important, however, the majority of the Okazaki fragments of both strands were linked to RNA at their 5' ends. For identification, the RNA was labeled at the 5' ends by [beta-32P]GTP, internally by [3H]CTP, [3H]GTP, and [3H]UTP, or at the 3' ends by 32P transfer from adjacent [32P]dTMP residues. All three kinds of labeling indicated that an equal proportion of DNA fragments from the two strands was linked to RNA primers.

Influence of seminal plasma on the chromatin stability of ejaculated human spermatozoa.

Nuclear chromatin decondensation (NCD) of human ejaculated spermatozoa exposed to sodium dodecyl sulphate (SDS) has been studied. A high proportion of NCD reacting spermatozoa was only found in semen samples with a relatively low activity of some prostatic factor(s) (i.e. zinc/fructose ratio below 0.18) in the seminal plasma. Exposure to SDS for one h was found sufficient to reveal the main proportion of spermatozoa undergoing NCD in such a solution. Addition of seminal plasma with an apparently normal composition to a sperm population with a high NCD reactivity restored the sperm SDS resistance to normal, i.e. blocked the NCD-response. Other results indicated that NCD reactivity was decreased or abolished upon prolonged storage of the spermatozoa in the seminal plasma. The various results indicated that some factor(s) in the seminal plasma can preserve the nuclear chromatin stability of human spermatozoa and that this factor most likely is of prostatic origin.

Secretion of metronidazole into the human semen.

The concentration of metronidazole has been studied in blood and in seminal plasma from apparently healthy volunteers and men with subclinical prostatitis. The volunteers took one dose of 500 mg orally and the patients 250 mg twice daily for two to three days. Therapeutic concentrations of metronidazole were found in blood and semen two to three h after intake. In patients the mean concentration was 8.7 micrograms/ml in blood plasma and 7.0 micrograms/ml in seminal plasma. Split ejaculates were collected in four to six fractions and the concentrations of metronidazole in the different fractions were related to those for zinc and fructose. The results indicated that the drug was excreted from prostate, seminal vesicles, and the ampulla ductus deferens.

Zinc and magnesium in bull and boar spermatozoa.

Mean +/- s.e.m. concentrations (nmol/10(8) cells) of zinc and magnesium in bull spermatozoa were 30.6 +/- 6.6 and 119 +/- 28.8, respectively. Corresponding values for boar spermatozoa were 16.9 +/- 1.98 and 57.1 +/- 4.3. Bull spermatozoa washed twice in a standard buffered salt solution, pH 7.75, lost 72.6% of their zinc and 46.5% of their magnesium. Boar spermatozoa lost 40% of Zn and 18% of Mg, respectively. Addition of albumin (4% final concentration) to the washing solution did not increase the loss of ions from bull spermatozoa but increased the loss of zinc and magnesium from boar spermatozoa to 52% and 41%, respectively.
