[Effect of superoxide dismutase on an arthritis model in the rabbit. A scanning electron microscopy study].

The effect of superoxide dismutase (SOD) was studied in an experimental arthrosis model (fixation arthrosis in rabbits). Out of a total of 40 animals, 20 of which were to be treated with SOD and 20 with placebos, 5 animals each were given padded stiffened bandages to immobilize their right knee joints for 1 week, 2 weeks, 3 weeks and 4 weeks respectively. After removing the bandage and a two-week recovery period the SOD or placebo treatment respectively was begun. During this period the animals were allowed to move freely. In 20 animals 0.5 mg SOD in 0.2 ml of a 0.5% xylocaine solution were applied intraarticularly in both popliteal spaces once a week for a total of 4 weeks. 20 control animals likewise received 0.2 ml of a 0.5% xylocaine solution (placebo) in both popliteal spaces in the same time intervals. After treatment the animals were killed and examined. A further 5 animals were not given a stiffened bandage, but for 4 weeks they received intraarticular injections once a week of 0.2 ml of a 0.9% saline to act as controls. The evaluation of the changes in the joints was by histological examination and examination by scanning electron microscope. In comparison to the placebo treatment the SOD treatment in this animal experiment did not lead to a significant change in the arthrotic process.

Plasma amino acid level in rheumatoid arthritis and ankylosing spondylitis and its variation during age.

28 plasma amino acids of 40 female patients with rheumatoid arthritis (RA), 24 male patients with ankylosing spondylitis (ASp) and 19 controls (14 females and 5 males) were investigated. In RA-patients 19 amino acids showed statistically significant differences to healthy people of which 18 were decreased. In ASp-patients 14 amino acid concentrations were statistically altered whereby 10 showed enhanced values. In female RA-patients and controls a linear dependency between distinct amino acids (threonine, glutamic acid, proline, alanine, citrulline, tyrosine, phenylalanine, ornithine, lysine and 3-methylhistidine) - and advanced age could be demonstrated.

Leucocyte migration inhibition with collagen type I and collagen type III in rheumatoid arthritis and degenerative joint diseases.

The leucocytes of 22 patients with classic rheumatoid arthritis were investigated with the direct leucocyte migration inhibition technique in agarose as described by Clausen. The leucocytes of 9 patients with degenerative joint diseases and of 9 healthy persons served as controls. Collagen type I and collagen type III were used as antigens. The leucocytes of 22 patients with rheumatoid arthritis showed in 15 cases a migration inhibition with collagen type III and in 10 cases with collagen type I. The inhibition with collagen type III was stronger and more frequent than with collagen type I. The frequent leucocyte migration inhibition with collagen type III in classic rheumatoid arthritis seems to be an expression of possible cell mediated reactivity against that type of collagen which appears especially in rheumatoid synovial tissue.

[Determination of the phagocytic ability of leukocytes in patients with chronic polyarthritis].

The phagocytic function of leukocytes from peripheral blood of patients with classical rheumatoid arthritis was investigated using nitroblue-tetrazolium reduction of formazan. One control group (A) and two groups of RA-patients were compared. Group B was treated with nonsteroidal antirheumatics, the third group C additionally with low dose steroids (5,0-7,5 mg/d). Reduction to formazan was increased in group B in relation to the control, whereas treatment with cortico-steroids produced decreased phagocytic function of peripheral leukocytes.

[Effect of D-penicillamine on zinc levels in blood and urine of patients with chronic polyarthritis].

In 6 female patients with classical rheumatoid arthritis D-penicillamine was administered in a daily dose of 250--500 mg for 3 weeks. The only additional antiphlogistic treatment was Indomethacin (100--150 mg daily). At the beginning and during the trial blood levels and the urinary excretion of zinc were determined using both neutron-activation analysis and atomabsorption. The results showed an increase of the urinary zinc excretion during the whole trial, a decrease of blood concentration of zinc until the 2nd week of treatment followed by an increase during the 3rd week above the level of the controls obtained before D-penicillamine therapy was started.

[Anemia in chronic polyarthritis].

In 216 patients, the correlation of anemia and activity of rheumatoid arthritis was studied. In 27% a concomitant anemia was found. These findings are congruent with those of other authors. There is a significant correlation between activity of rheumatoid disease and evolution of anemia as well as changes of serum iron level. With reference to the literature possible pathogenetic mechanisms of this type of anemia are reviewed. Therapeutic possibilities are briefly discussed.

[Histamine in rheumatic diseases].

Histamine, the decarboxylation product of histidine, was determined in serum, leucocytes, synovial fluid and synovial tissue rheumatic diseases. Histamine level was variable in most of the biological samples. In serum of patients with rheumatoid arthritis histamine was more frequent (50%) than in controls (10%). Leucocyte preparations containing basophile cells did not differ in their histamine concentration. Synovial fluid of rheumatoid arthritis patients showed histamine levels in the range of 1,0 to 23,4 ng/ml. The results were discussed by means of the pertinent literature.
