[Studies on cellular protein synthesis in synovial fluid following intraarticular gold administration (author's transl)].

Na-aurothiomalate was applied intraarticularly in the conentration range between 2 and 40 mg to joints of patients with rheumatoid arthritis. Between 2 and 20 mg a constant inhibition of 3H-aminoacids incorporation in proteins was observed. At a dosage of 40 mg of this gold salt a strong inhibition could be detected. In the low concentration range an additional degradation of structurally changed proteins could be the cause of the lower incorporation rate of 3H-aminoacids. From 40 mg on a real inhibition of protein synthesis seems to occur.

[Plasma amino acids in ankylosing spondylitis (author's transl)].

The plasma amino acids of male patients with ankylosing spondylitis (ASp) and male controls were analysed and the concentrations statistically evaluated. The total concentration of all 28 amino acids in ASp-patients did not differ from the controls, but there were some distinct differences in the levels of individual amino acids, e. g. arginine and isoleucine which showed raised concentration (29% and 27%). A higher concentration was also detectable with rare amino acids, e. g. alpha-amino-adipinic acid (23%) and 1-methyl-histidine (32%). Correlation between the amino acid concentration and age was detected only in the case of citrulline. Some amino acids showed a significant correlation to one another which was sometimes evident in both groups and other times was noted in the control- or ASp-group only. If such correlations were found to be disease-dependent this finding could be helpful in the diagnosis of certain diseases.

[Investigations on the plasm amino acids of old patients with rheumatoid arthritis (author's transl)].

The amino acids of blood plasma of 43 patients with rheumatoid arthritis (RA) and 19 controls were investigated by ion exchange chromatography. It was proofed simultaneously whether human more than 60 years old exhibit significantly different amino acid concentrations from younger individuals. It was found that RA patients more than 60 years differ significantly in 23 out of 30 amino acids. Only Asparagine was decreased, all other amino acids were increased. The controls did not show the same uniform direction of results. Older individuals exhibited differences in only 14 amino acids. Phosphoserine, Cystathionine and Arginine were increased to a great extent while Threonine, 1-Methylhistidine and Tryptophane were apparently diminished in this group of age.

Double blind trial involving: Teorema, a new antirheumatic, vs. indometacin in rheumatoid diseases.

Teorema and indometacin were administered to patients with degenerative and inflammatory joint diseases and degenerative vertebral column diseases for a period of 21 days. 24 patients were included in the 3 groups distinguished by diagnostic criteria. The daily Teorema dose was 420 mg in 21 cases and 210 mg in 3 cases; that of indometacin was 150 mg in 22 cases and 75 mg in 2 cases. Both drugs were given orally as capsules (25 mg indometacin/capsule and 70 mg Teorema/capsule). Both compounds produced a highly significant improvement of symptoms. Laboratory tests at regular intervals failed to reveal any substantial deviations from normal in both treatment groups. In conclusion, the results of the double blind trial suggest that Teorema, a new anti-inflammatory substance, when administered at a dose of approximately 420 mg/day, produces a beneficial effect in many rheumatoid conditions, while causing remarkably few side effects.

[The influence of drugs on the purine salvage metabolism in human blood cells (author's transl)].

The influence of 1-phenyl-2-(p-hydroxyphenyl)-3,5-dioxo-4n-butylpyrazolidine-monohydrate (oxyphenbutazone) and N-(alpha,alpha,alpha-trifluoro-m-tolyl)-anthranilic acid (flufenamic acid) on the purine salvage pathway of erythrocytes and lymphocytes following incubation of heparinized whole blood with the drugs was investigated. As described in earlier experiments in vitro with enzymes derived from cell homogenates, the enzyme activities of whole cells were also decreased. Lymphocytes exhibited a reduction of purine-phosphoribosyltransferase (purine-PRT) activities (adenine-, guanine- and hypoxanthine-PRT) of about 20%. The effect on the enzyme activities in red blood cells was different. The A-PRT-activity was decreased to about 33%, the GH-PRT on the other hand, to about 10%. The possible significance of these results in vivo is discussed.

[Treatment of interstitial pulmonary fibrosis with D-penicillamine in progressive scleroderma: a long-term study (author's transl)].

5 patients suffering from progressive scleroderma with interstitial pulmonary fibrosis have been treated since 1971 with D-penicillamine (Artamin) as basic therapeutic agent. All patients showed a significant tendency towards normalization of the pathological findings in regard to functional capacity of the lungs and scintigraphic and radiological assessment. These results, in our opinion, justify the recommendation of a new indication for D-penicillamine therapy, namely in cases of progressive scleroderma with interstitial pulmonary fibrosis.
