BHK monolayer and suspension cell populations maintained in different laboratories were found to vary in their susceptibility to infection with three strains of foot-and-mouth disease virus (FMDV). The susceptibility of parent cell populations was compared with that of individual clones of cells derived from them. The populations tested in this way consisted of a number of cell types, each expressing a different capacity to produce FMDV. The relative numbers of susceptible and insusceptible cells in each population appeared to determine the overall susceptibility of that population. The ability of the FMDV strain Asia 1 Iran 1/73 to multiply was an indicator of the general FMDV susceptibility of a BHK cell population.
The cell and virus productivity of a 3 mm glass sphere based unit process monolayer system was determined at scales of 0.1, 1, 10 and 100 litres medium capacity. Enhanced yields were obtained up to the 10L size when compared with control Roux bottle cultures. Modifications of the operating conditions of the 100L system should bring this into line with the smaller-scale units.
A continuous culture of BHK suspension cells was monitored by a micro-processor. The potential of this system is discussed as well as the results obtained to date on the growth of BHK suspension cells and the susceptibility of those cells to FMDV.

In order to control the four primary variables (cells, serum, medium and physical conditions) in a cell or virus growth system it is important to have a reliable and constant positive control with which to compare any variable of the system components. The use of aliquots of a frozen cell population (stored at -136 degrees C) for this purpose is the subject of this paper. Using such cells, tests have been established for the control of sera, media, cell susceptibility and the quality of serum treated DEAE Sephadex A50 beads.

In view of the advantages which are associated with the use of the BHK monolayer cell for the production of foot-and-mouth disease (FMD) virus, a unit system using glass spheres was developed to grow BHK monolayer cells and to test the susceptibility of such cells to FMD virus. The yield of cells and their susceptibility compares favorably with GREEN BHK monolayer cells which have been grown in Roux bottles.
Methods are described which make possible the production of foot-and-mouth disease (FMD) virus from BHK 21 C13 monolayer cells which have been grown on the surface of serum coated DEAE Sephadex A50 beads. The yield of cells and their susceptibility to infection by FMD virus are equivalent to conventional Roux monolayer systems. The potential for the commercial application of the DEAE Sephadex A50 system is discussed in relation to other unit process monolayer systems and in particular to the system in which cells are cultured in a deep bed of small glass spheres.
Cultures of BHK monolayer and suspension cells obtained from a number of laboratories and a group of cloned sub-lines derived from suspension cells were examined for their susceptibility to three FMD virus strains. It was found that the various cultures were sensitive to the test virus strains to differing degrees. It was shown possible to obtain a clone with high susceptibility to Asia 1 Iran 1/73 virus from a parent culture which had a low susceptibility to that virus. The implication of these findings is discussed.
The production of FMD virus from BHK 21 C13 monolayer and suspension cells was examined under standardized conditions and in different systems. The yields of virus from suspension cells did not significantly exceed the yield from monolayer cells, whereas the monolayer cell was capable of producing virus from some strains of FMD virus which would not grow in suspension cells. When different production systems were examined, the scale of operation did not significantly influence the yield of virus from suspension cells, while the plastic spiral film propagator produced significantly less virus than the other monolayer systems examined. The use of unit monolayer systems on the large scale is discussed.
The effect of changes in the conditions which control the growth of BHK monolayer cells on the surface of DEAE Sephadex A50 beads is examined. For any particular parameter a range of conditions exists which supports the growth of cells to adequate levels.
