Deoxyribonucleic acid polymerase II activity in an Escherichia coli mutator strain.

The polB gene encoding deoxyribonucleic acid (DNA) polymerase II has been located close to a mutator gene, mutT1, in Escherichia coli. We find the DNA polymerase II prepared from mutT1, strains to be normal in reaction requirements, heat stability, and ability to remove mismatched bases at termini. Recombinants formed from a mutant defective in DNA polymerase II (polB100) and mutT1 are deficient in polymerase II and have the same mutator phenotype as mutT1. Our linkage analysis indicates that mutT1 and polB100 are not isoallelic.

Excision of pyrimidine dimers in toluene-treated Escherichia coli.

Toluene-treated cells were used for examining excision of pyrimidine dimers in Escherichia coli strains W3110, DM845 (uvrA-), P3478 (polA-), and KS5064 (polAex1). Excision occurring in toluene-treated cells is rapid, adenosine 5'-triphosphate dependent, and requires the uvrA gene function. In strains lacking either the polymerizing or 5' leads to 3' exonucleolytic activity of deoxyribonucleic acid polymerase I, excision does occur. However, both in vivo and in vitro, the excision in such strains is initially slower than wild type.

DNA repair synthesis dependent on the uvrA,B gene products in toluene-treated cells.

Ultraviolet irradiation of toluene-treated Escherichia coli causes an inhibition of replicative DNA synthesis. This is followed by the appearance of nonconservative DNA repair synthesis which does not require either the polymerase or 5' leads to 3' exonucleolytic activities of DNA polymerase I. The repair synthesis may be catalyzed by DNA polymerase III activity but does not require a functional DNA polymerase II. The ultraviolet-induced synthesis requires ATP and is dependent on a functional uvrA and uvrB gene product. However, other uvr gene products are not required for the synthesis. The recB function is also not required.

Replication and repair of DNA in cells of Escherichia coli treated with toluene.

DNA synthesis has been studied in Escherichia coli cells made permeable to nucleotides by treatment with toluene. Replicative synthesis, as distinguished from repair synthesis, occurs at a rate comparable to that observed in vivo; it is dependent on the presence of all four deoxyribonucleoside triphosphates, but does not require exogenous DNA; and it is stimulated by ATP. Furthermore, replicative synthesis can be abolished at the restrictive temperature in DNA temperature-sensitive mutants. N-ethylmaleimide completely inhibits this type of synthesis, whereas it does not inhibit repair synthesis. Repair synthesis further differs from replicative synthesis in the following points: it does not require ATP; it persists at the restrictive temperature in DNA temperature-sensitive mutants; it can be induced by endogenous or exogenous nuclease activity; and its demonstration requires a Pol(+) strain.
