Characterization of denatured protein inducers of the heat shock (stress) response in Xenopus laevis oocytes.

In addition to thermal stress, a large variety of physical and chemical treatments are known to induce heat shock gene expression. Denatured protein, thought to result from the stress condition, has been postulated to act as the common signal. Accordingly, of three pairs of native and denatured proteins injected into Xenopus laevis oocytes, only the denatured derivatives induced expression of a reporter gene from a heat shock promoter (Ananthan, J., Goldberg, A. L., and Voellmy, R. (1986) Science 232, 522-525). These observations are extended here. Protein denaturation per se is shown to be insufficient for heat shock induction; although reduced and carboxymethylated bovine serum albumin (rcm-BSA) and alpha-crystallin elicited a stress response, many other denatured proteins had no effect. Methylation of protein lysines, done to prevent ubiquitination, suppressed heat shock induction by rcm-BSA, but enhanced induction by alpha-crystallin. Thus, the potential for a protein to be ubiquitinated is independent of its ability to induce the stress response. Instead, aggregation distinguished the proteins that were effective stress inducers, and the formation of large aggregates correlated with the magnitude of the response. This correlation may derive in part from decreased in vivo degradation rates of the inducer proteins. An apparent requirement for stress response induction that the inducer proteins be injected directly into the oocyte nucleus may relate to this issue of in vivo stability. The dependence of the stress response on the amount of injected protein is non-linear and of a form consistent with the titration of a factor that otherwise suppresses heat shock gene expression.

hsc70 moderates the heat shock (stress) response in Xenopus laevis oocytes and binds to denatured protein inducers.

Injections of hsc70 protein into Xenopus oocytes lowered the stress response to both a thermal shock and to co-injected protein inducers. Binding of hsc70 to native and modified forms of bovine serum albumin (BSA) were tested by two assays. In one, nitrocellulose-bound proteins were incubated with hsc70, cross-linked with glutaraldehyde, and then bound hsc70 was probed with a monoclonal anti-hsc70 antibody. In the second, peroxidase-conjugated hsc70 was employed as a more direct probe for binding to proteins displayed on nitrocellulose membranes. hsc70 binding to the BSA derivatives correlated with their abilities to induce a stress response upon microinjection into oocytes. Reduced and carboxymethylated (rcm) BSA, the most potent stress inducer tested, was bound most tightly by hsc70, whereas hsc70 had moderate affinity for N-methylated rcm-BSA and very little affinity for the native protein. No binding was observed with iodinated BSA. The results suggest a mechanism whereby the cell employs hsc70 or other proteins of the hsp70 family both to trigger the response to an environmental stress and to provide a feedback mechanism to attenuate the response.

Phenotypic characterization of mononuclear cells following anorganic bovine bone implantation in rats.

The purpose of this study was to measure inflammatory changes associated with implantation of anorganic bovine bone and bovine bone/collagen composite grafts, and to compare the response to that obtained following grafting with hydroxyapatite. Anorganic bovine bone, either with or without bovine collagen, as well as granular and block forms of synthetic hydroxyapatite, were implanted subcutaneously in Wistar rats. Saline and turpentine oil were used as controls. Biopsies were obtained after 3 days and at 1, 2, 4, 6, and 8 weeks. A panel of 6 monoclonal antibodies was used to detect monocytes, several distinct macrophage subsets, Ia-antigen expression, and T- and B-lymphocytes. Cells identified by each antibody were counted after immunocytochemical staining, and sera obtained 6 weeks after grafting were used in immunoblotting assays to detect antibodies to bovine serum proteins and collagen. Anorganic bovine bone, bovine bone/collagen, and hydroxyapatite all produced a transient macrophage infiltrate that was maximum 3 days after implantation, but resolved to normal levels within 6 to 8 weeks. Lymphocyte infiltration was not elicited by any bovine graft material, and antibodies to bovine serum proteins or type I collagen were not detected in any of the animals examined. These data indicate that a systemic or local immune response does not develop following implantation with anorganic bovine bone or with anorganic bovine bone/collagen materials. It appears appropriate to explore further the merits of these materials for periodontal regenerative procedures.

Crystal structure of a ubiquitin-dependent degradation substrate: a three-disulfide form of lysozyme.

Covalent attachment of ubiquitin marks substrates for proteolysis, but features that identify ubiquitination targets such as chicken egg white lysozyme are poorly understood. Recognition of lysozyme first requires reduction of Cys-6 Cys-127, one of its four native disulfide bonds, and Cys-6,Cys-127-carboxymethylated (6,127-rcm) lysozyme can mimic this three-disulfide intermediate. The 6,127-rcm form of lysozyme is known to retain a substantially native-like conformation in solution, and we demonstrate that it is this folded structure that is recognized for ubiquitination. Because native lysozyme is not a substrate, differences between the native and three-disulfide structures must include features responsible for selective ubiquitination. The 1.9-A resolution crystal structure of 6,127-rcm-lysozyme, reported here, affords a view of this ubiquitin-dependent degradation substrate. Two conformers of 6,127-rcm-lysozyme were obtained in the crystal. These differ uniquely from crystal forms of native lysozyme by displacement of the C-terminal residues. The structures suggest that localized unfolding at the C terminus of three-disulfide lysozyme allows the complex of E3 alpha (ubiquitin-protein ligase) and E2 (ubiquitin-carrier protein) to bind to a surface that includes Lys-1 and the putative ubiquitination site Lys-13. From this we infer that the N-terminal and internal substrate recognition sites on the E3 alpha.E2 complex are separated by approximately 20 A.

Induction of type 2 cystatin in rat submandibular glands by systemically administered agents.

An inducible type 2 cystatin has earlier been characterized in submandibular glands and kidneys of rats treated with isoproterenol, as well as in kidneys of rats with experimental renal disease. The purpose now was to determine whether giving agents that have systemic toxicity could also be associated with induction of cystatin in rat salivary glands. Female Wistar rats (200-250 g) were given isoproterenol, cyclocytidine, potassium dichromate or turpentine oil. After autopsy, the organs were sectioned, fixed in 10% formalin, and processed routinely. Paraffin sections were processed for both the peroxidase-antiperoxidase and the avidin-biotin-alkaline phosphatase immunocytochemical methods. The submandibular glands of rats given cyclocytidine had generalized, strong staining of acinar cells, as well as occasional weak staining within granular convoluted tubules. Animals given either potassium dichromate or turpentine oil exhibited moderate staining for cystatin in submandibular acini. Rats given isoproterenol as a positive control exhibited strong acinar staining throughout the submandibular gland, while the glands of untreated rats were unreactive. Inducible type 2 cystatin could not be detected in the parotid or sublingual glands, or in trachea, lung, stomach, small intestine, large intestine, spleen, liver and pancreas, after treatment with any of the systemic agents evaluated. The results indicate that elaboration of type 2 cystatin can be induced by a variety of systemically administered agents other than isoproterenol, and suggest that elaboration of type 2 cystatin may represent a more generalized response to tissue injury.

Cellular inflammatory responses to direct restorative composite resins.

As a result of shaping, finishing, or removal of dental composite resin restorations, resin particles may become trapped and embedded in oral tissues. However, tissue reactions induced by entrapped composite resin particles have not been thoroughly examined. To assess the soft tissue inflammatory response to composite restorative materials, three commercial dental composite resin suspensions were implanted subcutaneously in rats. Implantation resulted in a granulomatous inflammatory reaction that persisted 8 weeks after placement. The lesion was characterized by an influx of lymphocytes and the presence of fibroblasts and epithelioid cells. Composite resin particles have the potential to cause persistent inflammation if they are entrapped in oral tissues.

Induction of type 2 salivary cystatin in immunological and chemical kidney injury.

We have previously reported that isoproterenol induces type 2 salivary cystatin in both submandibular glands and kidney tubule cells of rats but not in any other organs examined. In the present study, we investigated whether this salivary protein is induced in other conditions that show kidney tubule injury. Immunocytochemistry, using a monospecific antiserum to this cystatin, revealed specific staining within the proximal tubule epithelium of the cortex as well as in the inner and outer stripe of the medulla of immunologically and chemically injured rats. Cystatin could not be detected in kidneys from healthy rats by means of immunocytochemistry. Weak staining was found in 3/3 kidneys of rats treated with turpentine and in 5/5 animals treated with potassium dichromate. In rats treated with puromycin, cystatin could not be demonstrated in 5/5 animals having proteinuria of less than 100 mg/24 h; however, moderate staining was observed in 4/5 puromycin-treated rats having proteinuria greater than 100 mg/24 h. In Heymann nephritis, cystatin was present in 7/31 kidneys with proteinuria lasting 6 to 15 weeks and in none (0/7) with proteinuria of shorter duration. Strong staining was also observed in 10/10 kidneys from rats with moderate-to-severe chronic serum sickness. This study shows that elaboration of type 2 cystatin in rats is not limited to salivary glands and, with our previous study, suggests that induction of this cysteine inhibitor may represent a local response to generalized tissue injury in both submandibular and renal tissues. These findings further demonstrate that induction of cystatin in salivary glands is not unique to these glands and suggest that induction of this cysteine proteinase inhibitor may represent a local response to tissue injury caused by diverse mechanisms.

1H NMR studies on the catalytic subunit of aspartate transcarbamoylase.

The 1H NMR spectrum of the catalytic subunit of Escherichia coli aspartate transcarbamoylase (EC 2.1.3.2) was simplified by using strains auxotrophic for the aromatic amino acids and a growth medium containing fully deuterated Trp, Phe, and His and partially deuterated Tyr. 1H resonances for Tyr in the catalytic trimer (M(r) = 10(5)) were partially resolved into five peaks at 27 degrees C, which above 50 degrees C were further resolved to give a distinct resonance for each of the eight Tyr residues in the polypeptide chain. Experiments on chemically modified catalytic subunits and on a mutant form in which Tyr-165 was converted to Ser-165 led to the assignment of resonances for Tyr-165, Tyr-240, and Tyr-185. Binding of the substrate, carbamoyl phosphate, caused shifts of two of the unassigned resonances, and the subsequent binding of the aspartate analog succinate perturbed the resonances corresponding to Tyr-165 and Tyr-240. The bisubstrate analog N-(phosphonacetyl)-L-aspartate produced a spectrum differing considerably from that caused by the combination of carbamoyl phosphate and succinate. The NMR spectrum for the Tyr-165-->Ser mutant trimer showed clearly that the single amino acid substitution caused conformational changes affecting the environment of residues remote from the position of the replacement. In contrast, the inactive mutant subunit in which Gly-128 was replaced by Asp exhibited a spectrum virtually identical to that of the wild-type protein. However, addition of the substrate carbamoyl phosphate caused a marked change in the spectrum of the mutant enzyme, whereas that of the wild-type trimer was altered only slightly, showing that the effect of the amino acid substitution was manifested in the NMR spectrum only with the liganded enzyme.

Ubiquitin conjugation to cytochromes c. Structure of the yeast iso-1 conjugate and possible recognition determinants.

Saccharomyces cerevisiae iso-1-cytochrome c was conjugated with ubiquitin (Ub) in vitro in a rabbit reticulocyte extract (Fraction II). By N-terminal protein sequencing, it was found for both the mono- and diubiquitinated products that the major Ub attachment site is on Lys4 (residue 9) of the cytochrome c. Thus, the residue ubiquitinated in iso-1-cytochrome c is identical with that previously determined for the yeast iso-2 form (Sokolik, C. W., and Cohen, R. E. (1991) J. Biol. Chem. 266, 9100-9107). For both cytochromes c, the proportions of diubiquitinated and higher order conjugates are drastically reduced when Ub is replaced with a Lys48----Arg variant, suggesting that the Ub-Ub moieties are linked predominantly through Lys48. Despite close similarities in structure and ubiquitination sites, conjugation to iso-2-cytochrome c is approximately 5-fold faster than for the iso-1 form; vertebrate cytochromes c are even poorer substrates, being ubiquitinated at only approximately 5% of the rate of the iso-2 protein. Comparison of several cytochrome c variants excludes alpha-N-acetylation or the identity of the N-terminal amino acid as the important recognition determinants in these reactions. The results, which include the finding that ferro and ferri-iso-2-cytochromes c are ubiquitinated equally, also are evidence against a simple correlation between ubiquitination efficiency and thermodynamic stability. Rather, the presence of a pair of lysines (Lys4-Lys5) within the relatively unstructured N-terminal extension of the yeast cytochromes c may be responsible for their preferential ubiquitination.

Phenotypic characterization of resident macrophages in submandibular salivary glands of normal and isoproterenol-treated rats.

Macrophages exert a major effect in the stimulation of lymphocytes and the modulation of immunological responses. To determine the presence and phenotypic distribution of the resident cells of the mononuclear phagocyte system in submandibular glands, frozen sections were prepared from five normal rats, and from six rats treated with 20 mg/kg isoproterenol/day for 10 days. A panel of six monoclonal antibodies was used to identify membrane markers associated primarily with circulatory monocytes (ED1), mature tissue macrophages (ED2), lymphoid macrophages (ED3), Ia antigen (OX6), CD5-positive T lymphocytes (OX19) and rat B lymphocytes (OX33). Cells identified by each monoclonal antibody were quantified by averaging the number of positive cells in 10 consecutive random high-power fields. ED2 cells (165 cells/field) were predominant in normal rat submandibular gland, followed by lower numbers of OX6-positive cells (18 cells/field). Cells positive for the remaining markers were also present in smaller amounts. In submandibular glands, treatment of rats with isoproterenol resulted in an increase in ED1-positive cells (from 2 to 39 cells/field), but also in substantial decreases in the number of cells positive for the remaining cell markers. B cells were not detected in any of the submandibular glands examined. These data suggest that isoproterenol induces a mild inflammatory response within rat submandibular glands that is not observed in normal glands. This results in an increase in the relative number of infiltrating monocytes compared to the number of more mature tissue macrophages.

Corrosion potential of steel bird shot in dogs.

Each year many dogs are accidentally or purposely wounded with shotguns. When lead pellets were used exclusively in the past, clinical problems from chronically embedded shot seldom developed except for rare cases of lead toxicosis. However, because expended lead shot ingested unintentionally by waterfowl and other avian species is fatal, the US Fish and Wildlife Service mandated exclusive use of steel shot for waterfowl hunting beginning in 1991. To discover the effects of implanted steel shot in a biological system, in vitro and in vivo studies were performed. Severe surface corrosion was evident when steel shot was placed in physiologic saline solution and sterile canine plasma. Eight laboratory dogs were surgically implanted with sterile steel shot in various superficial locations for intervals of 2 to 26 weeks. Corrosion of implants and tissue inflammation was observed in all biopsy specimens examined. It has been shown that steel shot embedded in tissues will corrode and result in a severe inflammatory response. If the accompanying inflammation is complicated by bacterial contamination, foreign body reactions resulting in infected, draining tracts could develop. Veterinarians and dog owners should be aware that treatment and prognosis for wounds caused by steel shot may differ from those for similar wounds caused by lead shot.
