Autophosphorylation in vitro of recombinant 42-kilodalton mitogen-activated protein kinase on tyrosine.

Mitogen-activated protein kinase (MAP kinase) is a serine/threonine protein kinase that becomes enzymatically activated and phosphorylated on tyrosine and threonine following treatment of quiescent cells with a variety of stimulatory agonists. Phosphorylation on both tyrosine and threonine is necessary to maintain full activity, and these two regulatory phosphorylations occur close to each other, separated by a single glutamate. To study the mechanisms by which MAP kinase becomes phosphorylated and activated, we have cloned a full-length cDNA encoding MAP kinase and have expressed the enzyme in Escherichia coli as a soluble nonfusion protein. We find that the enzyme displays a basal, intramolecular autophosphorylation on tyrosine-185 that is accompanied by activation of the enzyme's kinase activity towards an exogenous substrate. The tyrosine-phosphorylated protein displays a small fraction of the activity seen with the fully activated, doubly phosphorylated enzyme isolated from mammalian cells but is activated 10- to 20-fold relative to the unphosphorylated enzyme. These findings raise the possibility that regulation of MAP kinase activity in response to agonist stimulation could occur in part through the enhancement of autophosphorylation on tyrosine.

[Adenocarcinoma of the duodenum associated with skin neurofibromatosis and Caroli's disease].

A case of primary adenocarcinoma of the duodenum associated with Recklinghausen's disease and Caroli's disease provides the opportunity to review the literature. Malignant tumours of the duodenum are extremely rare and even rarer is their association with R's and Caroli's diseases. It should not be forgotten that in Recklinghausen's disease gastro-intestinal involvement is possible. Caroli's disease is diagnosed anatomopathologically. Surgical resection remains the sole satisfactory treatment when the tumour has not spread locally or at a distance. If it has, treatment must be palliative.

[Corrado Tommasi-Crudeli, hygienist].

To celebrate the centennary of the death of Prof. Corrado Tommasi-Crudeli, the founder and first director of the "Istituto di Igiene Sperimentale" in Rome (1885), the authors remember this eminent scientist who spent his life not only working in scientific experimental research but also was a member of the Parliament and promotor of important laws regarding social and health problems. It should be remembered that, prior to 1870, Hygiene was not a discipline in itself, but was part of Medicine and taught in theoretical courses such as Public Health Inspection and Legal Medicine. On the basis of the discoveries by Pettenkofer in Munich (1883) and Koch in Berlin (1885), the Prof. Tommasi-Crudeli created the foundations of Hygiene in Rome as an independent subject, strictly linked to experiments thus realizing a theoretic/practical discipline. From that time on words, Hygiene has evolved and grown, to become a specialised subject whose objectives are epidemiological studies, prevention of diseases and promotion of health, and whose important contributions to other branches such as Microbiology, Parasitology and Medical Statistics have helped them to become independent disciplines.

[Menetrier disease. Clinico-pathologic study of a case].

A classic case of Ménétrier's disease in an adult male, diagnosed by histological examination of operative biopsy is reported. Stress is laid on the clinical aspects of the disease and on the histological aspects of that particular type of hyperplastic gastropathy that underlies it. Brief mention is also made of the states of pseudohyperplastic gastropathy that may represent a source of diagnostic confusion, particularly from the radiological viewpoint.
