Integrated assessment of contaminated sediments in the lower Fox River and Green Bay, Wisconsin.

Samples of sediment and biota were collected from sites in the lower Fox River and southern Green Bay to determine existing or potential impacts of sediment-associated contaminants on different ecosystem components of this Great Lakes area of concern. Evaluation of benthos revealed a relatively depauperate community, particularly at the lower Fox River sites. Sediment pore water and bulk sediments from several lower Fox River sites were toxic to a number of test species including Pimephales promelas, Ceriodaphnia dubia, Hexagenia limbata, Selenastrum capricornutum, and Photobacterium phosphorum. An important component of the observed toxicity appeared to be due to ammonia. Evaluation of three bullhead (Ictalurus) species from the lower Fox River revealed an absence of preneoplastic or neoplastic liver lesions, and the Salmonella typhimurium bioassay indicated relatively little mutagenicity in sediment extracts. Apparent adverse reproductive effects were noted in two species of birds nesting along the lower Fox River and on a confined disposal facility for sediments near the mouth of the river, and there were measurable concentrations of potentially toxic 2,3,7,8-substituted polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs), and planar polychlorinated biphenyls (PCBs) both in the birds and in sediments from several of the study sites. Based on toxic equivalency factors and the results of an in vitro bioassay with H4IIE rat hepatoma cells, it appeared that the majority of potential toxicity of the PCB/PCDF/PCDD mixture in biota from the lower Fox River/Green Bay system was due to the planar PCBs. The results of these studies are discussed in terms of an integrated assessment focused on providing data for remedial action planning.

Reduced sensitivity of penile mechanoreceptors in aging rats with sexual dysfunction.

Male sexual activity gradually declines with age in most mammalian species and may be associated with a decrease in vibrotactile sensitivity of the penis. This study was designed to determine if the penile mechanoreceptors in aging rats exhibit reduced sensitivity to vibrotactile stimuli. Electrophysiological and behavioral data was obtained from mature adult (9 months) and old (23 months) rats. Single penile mechanoreceptor afferent fibers from aging animals exhibited a decrease in the ability to transduce high-frequency vibratory stimuli when compared to afferents from mature adult animals. In addition, there was a reduction in single-fiber and sensory nerve conduction velocity. Copulatory performance was reduced in the aging animals as determined by the inability to ejaculate with a receptive female. The results of this study demonstrate that the male rat exhibits an age-related decrease in penile vibrotactile sensitivity mediated by mechanoreceptor afferents which may be partly responsible for the decrease in parameters of sexual activity.

The allometric approach for interspecies scaling of pharmacokinetic parameters.

A long standing problem in pharmacokinetics and toxicology is the extrapolation and correlation between results obtained in different animal species and man. Animal data may be scaled-up to predict PPs in man using the allometric approach. The allometric approach is empirical, but easy, and is based on the fact that the underlying physiological processes such as blood flow, heartbeat duration, breath duration etc. are essentially physical and related to B. This approach is generally applicable to compounds that are essentially renally excreted. For substances that are highly extracted by the liver, Cltot is a function of the LBF among various species. Based on the concept of neoteny, use of brain weight affords a more correct approach to the scaling of Cl(int) of low extraction ratio drugs. By using the invariant pharmacokinetic time, the superficial differences in concentration-time profiles due to chronological time among different species are removed. Finally, as Boxenbaum (1984) has said "parameters to be scaled, independent variables, and the mathematical relationships used in the scaling process are all at the discretion of the investigator. There are no proper or improper approaches; the only limitations are those imposed by the investigator."

Mechanisms of major biliary injury during laparoscopic cholecystectomy.

Laparoscopic cholecystectomy has become the procedure of choice for surgical removal of the gallbladder. The most significant complication of this new technique is injury to the bile duct. Twelve cases of bile duct injury during laparoscopic cholecystectomy were reviewed. Eight injuries were of a classic type: misidentification of the common duct for the cystic duct, resection of part of the common and hepatic ducts, and associated right hepatic arterial injury. Another injury was similar: clip ligation of the distal common duct with proximal ligation and division of the cystic duct, resulting in biliary obstruction and leakage. Three complications arose from excessive use of cautery or laser in the region of the common duct, resulting in biliary strictures. Evaluation of persistent diffuse abdominal pain led to the recognition of ductal injury in most patients. Ultimately, 10 patients required a Roux-en-Y hepaticojejunostomy to provide adequate biliary drainage. One patient had a successful direct common duct repair, and the remaining patient underwent endoscopic dilatation.

Erectile and copulatory dysfunction in chronically diabetic BB/WOR rats.

The pituitary-testicular axis, penile reflexes, and copulatory behavior were studied in male BB diabetic rats from 10 to 40 wk of diabetes. Serum testosterone was diminished from 18 to 28 wk of diabetes, and the responses to human chorionic gonadotropin stimulation were blunted. Serum luteinizing hormone (LH) in diabetic rats did not differ from that of the control rats before or after LH-releasing hormone stimulation. Serum follicle-stimulating hormone and prolactin levels were also similar to controls. After 26 wk of diabetes, androgen-sensitive reproductive accessory organs were significantly reduced in size. This also was true for the androgen-sensitive bulbocavernosus and ischiocavernosus muscles. Penile reflexes in these animals from 20 to 32 wk of diabetes were consistently reduced in number and demonstrated prolonged latency. Copulatory behavior was evaluated in these animals at 25 and 28 wk of diabetes and revealed a reduced number of BB diabetic rats showing normal behavior at 25 wk of diabetes. At 28 wk of diabetes, mount latency, intromission latency, ejaculatory latency, and the postejaculatory interval were all prolonged compared with controls. In addition, the number of diabetic animals showing normal behavior was reduced compared with controls. These studies demonstrate that chronically BB diabetic rats develop diminished testosterone and erectile dysfunction that precedes ejaculatory dysfunction in a similar fashion as impotence in diabetic men. We suggest that further studies in this animal model may be critical to the better understanding and treatment of impotence in diabetic men.

Optimized spectrophotometric determination of aldehyde dehydrogenase activity in erythrocytes.

We describe a reliable and sensitive semiautomated spectrophotometric assay of aldehyde dehydrogenase (ALDH; EC 1.2.1.3) activity in erythrocytes. The hemolysate can be stabilized with sucrose, and the technique involves only microliters of hemolysate on a centrifugal analyzer. The use of microcolumns to remove interfering hemoglobin is avoided, and reproducibility of the assay has been improved by manipulating the inherent lactate dehydrogenase activity of erythrocytes by adding lactate and oxalate to the reaction mixture. These modifications have decreased the analytical imprecision of the assay, allowing a better appraisal of aldehyde dehydrogenase activity in erythrocytes as a biological marker of excess alcohol consumption. Erythrocytic ALDH activity was significantly less in 40 alcoholics than in 145 teetotallers (median activity 128 vs 219 mU/g of hemoglobin, respectively; P = 0.0001), indicating the potential of this assay as a useful marker of excess alcohol consumption.

Anatomic and radiographic study of the lacrimal apparatus of llamas.

The anatomy of the nasolacrimal duct of llamas was examined grossly and radiographically in 2 llamas. Dacryocystorhinography was performed on cadaver heads, using a radiographic aqueous contrast agent. Anatomic casts of the nasolacrimal apparatus were obtained by cannulation of the duct and use of polyurethane cast material. Dacryocystorhinography accurately revealed the nasolacrimal apparatus and compared favorably with gross dissections and polyurethane casts.

Topography and ultrastructure of sensory nerve endings in the glans penis of the rat.

The aim of the present study was to describe the types and location of sensory nerve endings in the glans penis of the rat on the basis of light and electron microscopy. Free nerve endings are abundant throughout the glans and are derived from either thinly myelinated A delta or unmyelinated C fibers. The free nerve endings derived from A delta fibers often extend into the epidermis as far as the granular layer and exhibit buttonlike terminal thickenings. The distal urethra contains many free nerve endings from C fibers. Numerous lamellated corpuscles are present. Each corpuscle, innervated by one or two medium diameter myelinated nerve fibers, consists of 4-10 spiralling and intertwined inner cores enveloped by Schwann cell lamellae. The corpuscles are usually enclosed by a perineural capsule, although some located near the epidermis lack a capsule. Lamellated corpuscles in the superficial dermis are located adjacent to but not directly under a penile spine and change their orientation when the glans is erect. The dermal papillae of the penile spines are devoid of any neural structure. Lamellated corpuscles are also located within the trabeculae of the corpus spongiosum glands and in the lamina propria of the urethra. Occasionally, unmyelinated nerve bundles are seen inside these corpuscles. Ruffini corpuscles are few in number and are located in the deep connective tissue near the os penis. They have an incomplete connective tissue capsule and the nerve terminals are anchored between collagen fiber bundles. No Merkel nerve endings, Meissner, or Vater-Pacini corpuscles were found in any of the sections. We conclude that the glans penis of the rat contains a sensory receptor complement that is generally similar to the human glans penis but is unlike the glabrous and hairy skin.

Cigarette smoking and rate of gastric emptying: effect on alcohol absorption.

OBJECTIVE: To examine the effects of cigarette smoking on alcohol absorption and gastric emptying. DESIGN: Randomised crossover study. SETTING: Research project in departments of medicine and nuclear medicine. SUBJECTS: Eight healthy volunteers aged 19-43 who regularly smoked 20-35 cigarettes a day and drank small amounts of alcohol on social occasions. INTERVENTIONS: Subjects drank 400 ml of a radiolabelled nutrient test meal containing alcohol (0.5 g/kg), then had their rates of gastric emptying measured. Test were carried out (a) with the subjects smoking four cigarettes an hour and (b) with the subjects not smoking, having abstained for seven days or more. The order of the tests was randomised and the tests were conducted two weeks apart. MAIN OUTCOME MEASURES: Peak blood alcohol concentrations, absorption of alcohol at 30 minutes, amount of test meal emptied from the stomach at 30 minutes, and times taken for 50% of the meal to leave the proximal stomach and total stomach. RESULTS: Smoking was associated with reductions in (a) peak blood alcohol concentrations (median values in non-smoking versus smoking periods 13.5 (range 8.7-22.6) mmol/l v 11.1 (4.3-13.5) mmol/l), (b) area under the blood alcohol concentration-time curve at 30 minutes (264 x 10(3) (0-509 x 10(3)) mmol/l/min v 140 x 10(3)) (0-217 x 10(3) mmol/l/min), and (c) amount of test meal emptied from the stomach at 30 minutes (39% (5-86%) v 23% (0-35%)). In addition, smoking slowed both the 50% gastric emptying time (37 (9-83) minutes v 56 (40-280) minutes) and the intragastric distribution of the meal. There was a close correlation between the amount of test meal emptied from the stomach at 30 minutes and the area under the blood alcohol concentration-time curve at 30 minutes (r = 0.91; p less than 0.0001). CONCLUSION: Cigarette smoking slows gastric emptying and as a consequence delays alcohol absorption.

Application of neural computing in pharmaceutical product development.

Neural computing technology is capable of solving problems involving complex pattern recognition. This technology is applied here to pharmaceutical product development. The most commonly used computational algorithm, the delta back-propagation network, was utilized to recognize the complex relationship between the formulation variables and the in vitro drug release parameters for a hydrophilic matrix capsule system. This new computational technique was also compared with the response surface methodology (RSM). Artificial neural network (ANN) analysis was able to predict the response values for a series of validation experiments more precisely than RSM. ANN may offer an alternative to RSM because it allows for the development of a system that can incorporate literature and experimental data to solve common problems in the pharmaceutical industry.

Spinal root origin of the radial nerve and nerves innervating shoulder muscles of the dog.

The ventral spinal root origin of the radial nerve, its muscle branches, and brachial plexus nerves which supply shoulder and thoracic musculature was determined in the dog. Electrophysiological signal averaging techniques measured evoked potential from specific ventral spinal roots to individual muscle nerves. The entire radial nerve received input from the sixth cervical (C6) through the second thoracic (T2) spinal roots. The most significant (p less than .05) input to triceps brachii came from C8 while the deep ramus of the radial nerve received its largest input from C7. The brachiocephalicus, suprascapular, and subscapular nerves all received their most significant (p less than .05) innervation from C6. Approximately 90% of the evoked potential to the axillary nerve originated from C7. The thoracodorsal nerve received most of its innervation from ventral roots C7 and C8. The lateral thoracic nerve which innervates the cutaneous trunci muscle was supplied by ventral roots C8-T2. Examination of innervation patterns suggests that only modest variation of spinal root input to specific nerves occurred between individual dogs.

Rescue of motoneurons from the axotomized state by regeneration into a sensory nerve in cats.

1. We studied the electrical properties of spinal motoneurons, the axons of which had regenerated into a cutaneous nerve. 2. In cats, all or part of the medial gastrocnemius (MG) muscle nerve was cut and directed distally into the caudal cutaneous sural (CCS) nerve, a sensory (primarily cutaneous) nerve. One or 2 yr later, electrical properties [conduction velocity (CV), rheobase (Irh), input resistance (RN), afterhyperpolarization (AHP), and excitatory postsynaptic potentials (EPSPs)] of MG motoneurons that had cross-regenerated into the CCS nerve were determined. These were compared with properties of normal and of axotomized MG motoneurons and with data from previous studies in which MG motoneurons had reinnervated their own or a foreign muscle. 3. Electrical stimulation of the MG-innervated CCS nerve produced no detected mechanical activity, indicating an absence of muscle innervation. Tactile stimulation of skin did not activate these motoneurons; i.e., they did not acquire properties of cutaneous afferents. 4. The CV and Irh of MG motoneurons axotomized 11 mo declined by 48 and 60%, respectively. 5. The CV of MG motoneurons that had regenerated through CCS was only slightly slower than normal, similar to that of MG motoneurons that reinnervated the "slow" muscle soleus (Foehring and Munson 1990). 6. The Irh and RN were also similar to those of MG motoneurons that had regenerated into the soleus muscle. 7. Electrical stimulation of the lateral gastrocnemius-soleus nerve generated EPSPs of normal or almost normal amplitude in MG motoneurons axotomized for 11 mo or cross-regenerated into CCS up to 2 yr.(ABSTRACT TRUNCATED AT 250 WORDS)

Regenerating sprouts of axotomized cat muscle afferents express characteristic firing patterns to mechanical stimulation.

1. In cats, we studied the physiological properties of regenerating sprouts of muscle afferent fibers and compared them with sprouts from cutaneous afferent fibers. 2. Muscle nerves to the triceps surae and cutaneous sural nerves were axotomized in the popliteal fossa, and the proximal ends were inserted into nerve cuffs. Six days later, we recorded action potentials from single Groups I and II muscle and mostly Group II cutaneous afferents driven by mechanostimulation of the cuff. 3. Most muscle afferent sprouts (91%) had a regular slowly adapting discharge in response to sustained mechanical displacement of the cuff, particularly to sustained stretch stimuli, whereas most cutaneous afferents (92%) did not. Muscle afferents were more likely to have a spontaneous discharge and afterdischarge. 4. Group II muscle afferent sprouts had lower stretch thresholds and a higher incidence of spontaneous discharge compared with Group I fiber sprouts, whereas Group I fibers had a higher incidence of high-frequency afterdischarge to mechanical stimuli. 5. We conclude that, 6 days after axotomy, regenerating sprouts of muscle afferents, particularly Group II afferents, have become mechanosensitive in the absence of a receptor target and exhibit physiological properties similar to those found when innervating their native muscle but significantly different from sprouts of cutaneous afferents. Expression of these native muscle afferent firing patterns after the inappropriate reinnervation of hairy skin may be due to inherent properties of the muscle afferent fiber.

Effects of trenbolone acetate on carcass characteristics and serum testosterone and cortisol concentrations in bulls and steers on different management and implant schemes.

The objective of this study was to determine the effects of different implanting schemes on serum testosterone and cortisol concentrations, and carcass traits of bulls and steers implanted with trenbolone acetate (TBA) and zeranol (Z). Twenty Polled Hereford bulls were randomly assigned to one of three treatments after birth. Five calves served as nonimplanted control bulls (NIB). Nine bulls were implanted (IB) with 140 mg of TBA and 36 mg of Z at about 1 mo of age and reimplanted with both compounds 10 wk later. When IB calves were about 21 wk of age, the TBA implant was removed and calves were reimplanted with Z every 10 wk until slaughter. Six calves were castrated at 3 wk of age and implanted (IS) with TBA and Z every 10 wk until slaughter. Blood samples from each animal were obtained at 14-d intervals beginning at 14 wk of age and serum cortisol (C) and testosterone (T) concentrations were determined. The NIB had higher C levels than IB or IS (P less than .05) during the preweaning period. During the finishing period, there were no differences in C concentrations between NIB and IB; however, IS had lower levels (P less than .05) than both bull treatments. Serum T concentrations began to increase about 12 wk later (42 vs 30 wk, respectively) in IB compared with NIB. Testicular size was smaller (P less than .05) in IB than in NIB. No differences (P greater than .05) were observed in carcass characteristics. Taste-panel scores were not different among treatments. In conclusion, implanting schemes using TBA and Z lowered serum levels of C and delayed puberty in bulls; however, they did not alter carcass characteristics or eating quality.

Interspecies scaling of the pharmacokinetic parameters of coumarin among six different mammalian species.

The pharmacokinetic parameters, total body clearance, apparent volume of distribution and the terminal elimination half-life of coumarin were correlated among six mammalian species. It was demonstrated that the total body clearance was highly related to the liver blood flow, liver weight and body weight. The apparent volume of distribution appeared to be related only to body weight. The elimination half-life, which is dependent upon both the apparent volume of distribution and total body clearance, was poorly correlated with all physiologic variables that were evaluated. Hence, it appears that only the primary pharmacokinetic parameters of coumarin display a definite interspecies relationship.

Electrophysiologic studies of the cutaneous innervation of the pelvic limb of male dogs.

The area of skin supplied by the afferent fibers in one cutaneous nerve is called the cutaneous area (CA) for that nerve. The CA of peripheral branches of lumbar and sacral spinal nerves responsive to the stimulation of hair follicle mechanoreceptors were mapped in 27 dogs. The amount of overlap among the CA was similar to that found for other CA of the body. The CA of peripheral branches of the sciatic nerve were restricted to the lateral, cranial, and caudal aspects of the pelvic limb distal to the stifle. The CA of the saphenous nerve was located on the medial side of the limb, except for a small area located on the lateral side of the crus. The distal part of the CA of the saphenous nerve was completely overlapped in the hind paw by branches of the superficial peroneal nerve laterally and the medial plantar branch of the tibial nerve medially. The CA for the deep peroneal nerve was located on the dorsal surface of the webbing between digits 2 and 3 and the adjacent skin of these digits. The CA of the plantar branches of the tibial nerve were small in comparison with the diameter of the nerve, suggesting that these branches contained nerve fibers supplying other, deeper structures in the hindpaw and that damage to these nerves would interfere with cutaneous sensation in only a small region on the plantar surface of the hindpaw. Knowledge of the CA of the various branches of the sciatic nerve allows more accurate localization of injury to the sciatic nerve or its branches by using areas of anesthesia.

Heterogeneity and organization of the ribosomal RNA genes of Cucurbita maxima.

Thirty-six clones were recovered from Cucurbita maxima genomic DNA which had been enriched for rDNA and cleaved at the unique repeat unit Hind III site. Twenty-nine of these, which contain complete rDNA units, were compared to a standard whose intergenic spacer (IGS) nucleotide sequence has been determined. Twenty-one are identical in length and restriction site pattern. Eight which differ from the standard in length do so because of addition or deletion of varying numbers of IGS subrepetitive units of two different classes, with four of the length variants being different in both of these classes. Seven clones were isolated which contain incomplete repeat units, six of which are composites of rDNA and non-rDNA material. They have been cleaved at the unique rDNA Hind III site at one end and at a non-rDNA Hind III site at the other. We consider it most likely that these are derived from the termini of repeat unit tandem arrays, although other explanations are possible. Twelve individual plants of two different cultivars were examined for heterogeneity of IGS length distribution. They all appear to be identical in this regard.
