Total RNA, extracted from synchronously growing yeast cells by the method of Georgiev-Mantieva, was separated by columns of methylated albumin on Kieselgur (MAK). By comparison with definited RNA's in discelectrophorese and after pulse labelling with [3H]methionine the following fractions are identified: t-RNA, 5.8S RNA, m-RNA, 18S and 25S R-RNA and their precursors 20S, 27S and 35S. High molecular fractions of RNA between 40S and 70S were also found.
N-Methyl-N-beta-chloroethyl-hydrazine-HCl and its benzaldehydhydrazone inhibit growth and multiplication of yeast cells. The DNA-synthesis is partially blocked. After removal of the substances the rate of this synthesis is much more increased than that of RNA and protein. The both substances we tested are more effective than simple alkylhydrazines.
N-methyl-N-beta-chloroethylhydrazine and its benzaldehydhydrazone inhibit the influx of [6-3H]-uracil and L-[U-14C]leucin in yeast cells as well as the incorporation in RNA and protein. Experiments with synchronously and asynchronously growing cells showed the N-methyl-N-beta-chloroethylhydrazine essentially influences the dividing phase while the benzaldehydhydrazone is more effective in the growth phase. The effect of these two substances on respiration, glycolysis and the concentration of fructose-1,6-diphosphate, triose phosphate, and adenosine triphosphate is small.
The poison gyromitrin, found in the edible false morel Gyromitra esculenta Fr. ex Pers., caused in rats an increased diuresis in which urine was produced with a weak alkaline pH, a high excretion of sodium (530%), and potassium (210%). The observed increase lasted for about 12 h and was followed by a rentention with regard to the volume and the Na-excretion for about 72 h. On the basis of [3H] inulin excretion an increased glomerular filtration was determined followed by a decrease 12 h after application of gyromitrin. An increase of creatinine and urea in the serum could not be established during the retention phase. The diuresis as well as the excretion of sodium could be antagonized by injection of an equimolar dose of pyridoxine. The hydrazine derivative N-methyl-N-formylhydrazine (MFH), which is formed rapidly from gyromitrin by hydrolysis, was without any effect on the renal function. In order to explain the effectiveness of the relatively lipophilic gyromitrin, the non-effectiveness of the more hydrophilic hydrazine MFH and the blockade of the gyromitrin effect by pyridoxine, a mechanism involving the central nervous system is discussed.
Isoniazid was tested for genetic activity in the mammalian spot test by treating in utero mouse embryos, derived from the crosses C57 B1/6J X T-stock and NMRI X DBA/2, heterozygous for various loci affecting coat color. Induction of genetic changes is seen by expression of recessive alleles in colored spots within the fur of the adult animal in a non-agouti background. Injecting mothers with isoniazid (100 mg/kg ip), an expression of recessive loci was found in 7.41% of the F1 animals in the cross C57 B1/6J X T-stock, while in the NMRI X DBA/2 cross 2.92% animals with recessive spots were detected. The control frequencies were respectively 0 and 0.78% animals with recessive spots. The positive control substance, ethyl methanesulfonate (100 mg/kg ip), yielded 5.97% and 4.71% animals carrying a recessive spot in the fur, respectively. These studies clearly demonstrate for the first time the induction of genetic damage other than cytological damage in somatic cells of mice by isoniazid.
Provided that the patient selection criteria, surgical procedure, and postoperative care recommended by Moberg are strictly observed, the posterior deltoid-to-triceps transfer is an excellent surgical procedure for improving the level of function of the C5 or C6 quadriplegic. Before surgery, patients should be thoroughly counseled in realistic expectations, the possible functional results, and the long period of time required to attain the goals of surgical treatment.
