Isolation of a transcriptionally active chromosome from chloroplasts of Euglena gracilis.

A transcriptionally active chromosome has been isolated in highly purified form from choroplasts of Euglena gracilis, It contains chloroplast DNA, DNA-dependent RNA polymerase, and other proteins. Transcription occurs at low levels of endogenous DNA, and is indifferent to high levels of exogenous DNA. RNA chain elongation continues for several hours in vitro, and RNA chain initiation, determined by [gamma-32P]ATP incorporation, is continuous for at least 1 h in vitro. Maximal rates for RNA synthesis require only a divalent cation and the four ribonucleoside triphosphates. Apparent Km values for adenosine triphosphate, cytidine triphosphate, guanosine triphosphate, and uridine triphosphate are 4.0, 0.6, 2.5, and 2.3 muM, respectively. As would be expected for a DNA-dependent RNA polymerase, RNA synthesis is inhibited by actinomycin D. However, rifampicin and streptolydigin, inhibitors of procaryotic RNA synthesis, and alpha-amanitin, an inhibitor of eucaryotic nuclear RNA polymerases II and III, do not inhibt the RNA synthesis reaction. Heparin, which is a potent inhibitor of the initiation of RNA synthesis by a nontemplate bound RNA polymerase, also does not inhibit RNA synthesis. Isolation of transcriptionally active chromosomes should prove to be a useful method to study the mechanism of selective RNA transcription of eucaryotic chromosomes.

Changes in the expression of the chloroplast genome of Euglena gracilis during chloroplast development.

The transcription program from the chloroplast genome of Euglena gracilis Z during light-induced chloroplast development has been characterized by hybridization of total cell RNA to 3H-labeled chloroplast DNA. Pancreatic DNase activated, purified Euglena chloroplast DNA was enzymatically labeled by Escherichia coli DNA polymerase I with [3H]TTP as a substrate. The [3H]DNA 'hybridization probe" was characterized by the kinetics of its renaturation with purified chloroplast DNA, and the thermal stability of [3H]DNA-DNA, and [3H]DNA-RNA hybrids. The [3H]DNA was hybridized in trace amounts to total cellular RNA extracted from Euglena cells 0, 4, 8, 12, 24, 48, and 72 h after the onset of chloroplast development. A large percentage (17%) of the chloroplast genome was found to be transcribed in dark adapted cells. Development is marked by an initial decrease in the fraction of the genome transcribed followed by an increase to 23% transcribed at the end of 72 h of light growth. Chloroplast RNA transcripts were also characterized by the kinetics of their hybridization to chloroplast DNA. The chloroplast specific RNA population is composed of three abundance classes, and the R0t1/2 for each class varies during the early stages of chloroplast development.

Vitamin D3-stimulated template activity of chromatin from rat intestine.

Administration of vitamin D to rats deficient in this vitamin markedly increases the template activity for DNA-dependent RNA synthesis by rat intestinal mucosal chromatin. The maximum stimulation of template activity occurs three hours after a dose of 2000 international units of vitamin D(3) is given. These results support the concept that vitamin D functions by initiating the transcription of DNA into mRNA, which codes for functional protein(s) involved in calcium transport in the small intestine.
