Molecular characterization of angiotensin II type II receptors in rat pheochromocytoma cells.

The binding sites and biochemical effects of angiotensin (A) II were investigated in rat pheochromocytoma (PC12W) cells. Sarcosine1, [125I]-tyrosine4, isoleucine8-AII ([125I]-SI-AII) bound to a saturable population of sites on membranes with an equilibrium dissociation constant (Kd) of 0.4 nM and a binding site maximum of 254 fmol/mg protein. Competitive displacement of [125I]-SI-AII by agonists and antagonists elucidated a rank order of potency of AIII greater than or equal to AII greater than PD 123177 greater than AI greater than [des-Phe]AII [AII(1-7)] much greater than DuP 753. The stable guanine nucleotide analog 5'-guanylyl imidodiphosphate did not alter the binding affinity or slope of the inhibition curves for AI, AII, AIII, or AII(1-7). Treatment of PC12W cells with AII or AIII did not affect the free intracellular calcium concentration, phosphoinositide metabolism, arachidonate release, cyclic GMP, or cyclic AMP concentrations. [125I]-AII binding sites remained on the cell surface and were not internalized after 2 h at 37 degrees C. Angiotensin II did not stimulate tyrosine, serine, or threonine phosphorylation. Northern analysis of PC12W mRNA with an AT1 receptor gene probe failed to produce an RNA:DNA hybrid at low stringency. These data indicate that PC12W cells express a homogeneous population of AT2 binding sites which differ significantly from AT1 receptors in signal transduction and molecular structure. AT2 sites may act via potentially novel, biochemical pathways or, alternatively, be vestigial receptors.

Peptidases and smooth muscle cell angiotensin II receptor pharmacology.

Angiotensin (A) II receptors on rat aortic smooth muscle (RASM) cell membranes were characterized using the radioligand [125I][Sar1Ile8]AII ([125I]SI-AII). Angiotensin I, AII, and AIII inhibited specific [125I]SI-AII binding, and their rank order of potencies, and Ki values (nM) were: AII (3.7) > AI (32.5) > or = AIII (54.0), which differed from that observed for rat adrenal cortex: AII (0.85) > AIII (3.3) >> AI (100). Similar results were observed for RASM membranes in the presence of guanine nucleotides, and for intact cells in the absence or presence of an internalization inhibitor. Lowering the incubation temperature from 37 degrees C to 4 degrees C, or inclusion of PMSF (1 mM), and preparing membranes in the presence of EGTA (1 mM) altered the rank order of potencies and Ki values (nM) of the angiotensin peptides to: AII (1.1) > AIII (7.0) >> AI (144). [125I]Angiotensin I was metabolized completely over the course of 90 min to small (<tetrapeptide) fragments as measured by HPLC. There was no evidence for formation of AII or AIII from AI, which would have explained the unusually high potency of AI. [125I]Angiotensin I metabolism could be attenuated by inhibitors of serine proteases PMSF, aprotinin, and chymostatin. The beneficial effects of PMSF and EGTA suggested that serine protease(s) and metalloproteases contribute to the observed anomalous pharmacological characteristics of AI and AIII, respectively. The RASM cell membranes contained a homogeneous population of binding sites for losartan, and its Ki value differed in the absence (50 nM) or presence (16 nM) of protease inhibitors, which suggests that the receptor may also be a target for these peptidases.(ABSTRACT TRUNCATED AT 250 WORDS)

Fracture strength of Class 2 amalgams with various cavity-lining materials.

UNLABELLED: This in vitro study compared the fracture resistance of class 2 amalgam restorations placed over seven materials: three resin-modified glass-ionomer cements (Fuji II LC, Vitrebond, and Vitremer), one polyacid-modified composite resin (VariGlass VLC), two conventional glass-ionomer cements (Ketac-Bond and GlasIonomer Cement), and one calcium-hydroxide material (Dycal). Eighty maxillary molars with flattened occlusal surfaces were divided into 14 experimental groups and two control (no liner) groups. One standardized class 2 amalgam cavity preparation was completed per tooth. Lining materials standardized at a thickness of 0.5 mm were placed in the approximal box portion of 10 test specimens per experimental group. Spherical amalgam was hand condensed into each cavity preparation. At 1 hour and again at 7 days, five samples from each group were fractured in compression using an Instron Universal Testing Machine. The force was directed at 10 degrees to the long axis of the tooth, 2.0 mm inside the approximal portion of the restoration. Results were analyzed using a two-way ANOVA for time and material. No statistically significant differences were found among the materials and controls at either time interval tested (P > 0.05). A statistically significant difference was found (P < 0.05) when comparing 1-hour and 7-day strengths. The 7-day specimens were more resistant to fracture than the 1-hour specimens. CONCLUSION: The fracture resistance of amalgam restorations was not affected by the presence of a material 0.5 mm thick placed in the approximal box when 3 mm of bulk of amalgam remained over it.
