New polyoxypregnane ester derivatives from Leptadenia hastata.

Six new polyoxypregnane esters [1-6] having sarcostin or deacetylmetaplexigenin as the aglycones, and acetyl, benzoyl, cinnamoyl, and nicotinoyl residues as the ester moieties linked at C-12 and/or at C-20, were isolated from Leptadenia hastata. In addition, three new glycosides [7-9] with D-cymarose and D-oleandrose linked at C-3 were present in the CHCl3 extract. Structures were deduced on the basis of spectral evidence.

Constituents of Werneria ciliolata and their in vitro anti-HIV activity.

The aerial part of Werneria ciliolata afforded a series of new diterpenes: two ent-kaurane derivatives, a norkaurane, an ent-manoyloxide derivative, a dimeric diterpene, as well as a rare diterpene. Their structures were elucidated by spectroscopic methods, including the concerted application of 1D NMR techniques (DEPT and NOEDS) and 2D NMR techniques (1H-1H COSY and HETCOR). In addition, four known kauranes, four coumarins and 6-hydroxytremetone were isolated. All isolated compounds from W. ciliolata and W. dactylophylla were tested in vitro for anti-viral activity against HIV-1, but only 6-hydroxytremetone showed a significant anti-HIV-1 activity.

Traditional phytotherapy in the Peninsula Sorrentina, Campania, southern Italy.

A study on the wild and cultivated medicinal plants used in the Peninsula Sorrentina (southern Italy) is reported; 129 plants distributed among 53 different families beloning to Pteridophytes, Gymnosperms and Angiosperms are used in the treatment of various human diseases. The use of some species is reported for the first time in this paper.

Quinovic acid glycosides from Uncaria guianensis.

From the bark of Uncaria guianensis, two new quinovic acid glycosides, quinovic acid 3 beta-O-beta-D-quinovopyranoside and quinovic acid 3 beta-O-beta-D-fucopyranosyl-(27----1)-beta-D-glucopyranosylester, have been isolated, in addition to known quinovic acid 3 beta-O-[beta-D-glucopyranosyl-(1----3)-beta-D-fucopyranosyl]-(27----1)- beta-D-glucopyranosylester and quinovic acid 3 beta-O-beta-D-fucopyranoside. Their structures were elucidated by spectral and chemical studies.

Antiviral activity of constituents of Tamus communis.

The antiviral activity of the phenanthrene derivatives 1-6, of the spyrostane triglycosides dioscin (7) and gracillin (8), of the furostanol tetraglycosides methylprotodioscin (9), its (25S) epimer methylprotoneodioscin (10), and methylprotogracillin 11, have been tested towards two RNA viruses: vesicular stomatitis virus and human rhinovirus type 1B. All these products were extracted from the rizomes of Tamus communis L; compound 11 was isolated also from Asparagus cochinchinesis, together with pseudoprotodioscin (12), a 20 (22)-unsaturated furostanoside, which was also investigated for antiviral activity. The results were of some interest mainly for the phenanthrene derivatives.

Microphthalmia and chorioretinal lesions in a girl with an Xp22.2-pter deletion and partial 3p trisomy: clinical observations relevant to Aicardi syndrome gene localization.

We present a 4-year-old girl with a maternally derived, unbalanced X;3 translocation resulting in partial Xp monosomy and partial 3p trisomy. She had chorioretinal defects, developmental delay, infantile seizures, and microphthalmia. These findings initially suggested a diagnosis of Aicardi syndrome. However, she had a normal-appearing corpus callosum on CT and magnetic resonance imaging scans of the brain and her retinal findings were not typical for Aicardi syndrome. This represents the 6th reported example of microphthalmia associated with an Xp22 chromosome abnormality. Four of these individuals also had features suggestive of focal dermal hypoplasia (FDH), which was not evident in our patient. The available evidence supports the hypothesis that gene disruption at Xp22 may lead to findings similar to those seen in Aicardi syndrome and FDH, both of which are believed to be X-linked dominant male lethal conditions.

Iridoids and secoiridoids from Guettarda platypoda.

A reinvestigation of the roots of Guettarda platypoda afforded in addition to morronoside and sweroside isolated previously further two iridoids, loganic acid and loganin and a rare secoiridoid, secoxyloganin. The structures were elucidated by NMR techniques. This is the first report of the isolation of such compounds from G.platypoda. Some chemotaxonomic conclusion can be drawn.

De novo 2q+ masquerading as Smith-Lemli-Opitz syndrome.

We report a female infant diagnosed shortly after birth as having Smith-Lemli-Opitz syndrome. Despite previously reported normal G banded karyotypes, a high resolution banded chromosome analysis identified 46,XX,2q+. The importance of attention to established features of clinical syndromes, as well as persistence in investigation when diagnostic uncertainties exist, are discussed.

[Chemical composition and biological properties of Erythraea centaurium Rafn].

The steroidic fraction from the ethereal extract of the aereal parts of Erythraea centaurium Rafn. has showed the presence of the following sterols: beta-sitosterol, stigmasterol, campesterol, brassicasterol and delta 7 stigmastenol, isolated by H.P.L.C. and identified by G.L.C., 1H-N.M.R. and MS. Then the free amino-acids from the aqueous extract of E. centaurium were determined.

Steroidal glycosides of Tamus communis.

From the rhizomes of T. Communis two steroidal glycosides were isolated and identified as dioscin [diosgenin bis-alpha-L-rhamnopyranosyl (1----2 and 1----4) -beta-D-glucopyranoside] and gracillin [diosgenin-alpha-L-rhamnopyranosyl (1----2)-beta-D-glucopyranosyl (1----3)-beta-D-glucopyranoside] by 1H and 13C NMR spectroscopy, MS (FAB) and chemical evidences. It is interesting to note that this is the first report of the occurrence of dioscin and gracillin in the rhizomes of T. communis.

Diosgenin

[Sterols in Stevia rebaudiana Bertoni].

The sterol fraction of Stevia rebaudiana Bertoni contains, essentially, the following sterols: stigmasterol (45,8%), beta-sitosterol (39,4%) and campesterol (13,1%). The individual components were separated, after acetylation, by HPLC with absolute methanol as eluant. The identification of the compounds has been carried out through NMR and MS, while the corresponding percentages have been desumed from the GLC data.

Sterols from Equisetum arvense.

The sterol fraction of Equisetum arvense L. contains, essentially, the following sterols: beta-sitosterol (60.0%), campesterol (32.9%), isofucosterol (5.9%) and cholesterol (trace amounts). The identification of the compounds has been carried out through NMR and MS, while the corresponding percentage have been desumed from the GLC and HPLC data.

Plant metabolites. Structure and in vitro antiviral activity of quinovic acid glycosides from Uncaria tomentosa and Guettarda platypoda.

A reinvestigation of the bark of Uncaria tomentosa afforded, in addition to the major quinovic acid glycosides 1-3, three further glycosides 4-6. The structures were elucidated by spectral and chemical studies. Furthermore, a series of antiviral tests were performed on all these glycosides and on the related glycosides 7-9, previously isolated from Guettarda platypoda.

New polyhydroxylated triterpenes from Uncaria tomentosa.

Three novel polyhydroxylated triterpenes have been isolated from Uncaria tomentosa. Their structures were established as 1, 2, and 3 by detailed spectral studies including 1H-13C correlations via long range couplings using the INAPT pulse sequence, nOeds, and 2D 1H-13C direct chemical shift correlation (HETCOR) nmr techniques.

Plant metabolites. New compounds and anti-inflammatory activity of Uncaria tomentosa.

Bioassay-directed fractionation of the anti-inflammatory extracts of Uncaria tomentosa, using the carrageenan-induced edema in rat paw, has led to the isolation of a new quinovic acid glycoside 7 as one of the active principles. Furthermore, a new triterpene 8 was isolated as its methyl ester. The structures were elucidated by spectral and chemical studies.
