[The role of Hantaan virus serotypes in the etiology of hemorrhagic fever with renal syndrome in the Far East of the USSR].

The employment of two serological tests (indirect fluorescence antibody technique and neutralization test) demonstrated the leading role of Hantaan virus serotype 1 strains isolated from field mice in the pattern of the incidence of hemorrhagic fever with renal syndrome (HFRS). Proofs of the etiological importance of strains of serotypes 3 and 5 occurring in brown rats and Cl. rufocanus were obtained. No data on any association of human HFRS cases with strains of serotype 4 isolated from reed voles could be demonstrated.

[Reservoirs of the causative agent of hemorrhagic fever with renal syndrome in Maritime Territory foci].

Examinations of 6362 small mammals of 18 species revealed the main reservoirs of the causative agent of hemorrhagic fever with the renal syndrome (HFRS) in foci of the Primorsky Krai, and demonstrated the importance of individual species in the epidemiology of the infection. Data on the circulation in the region of antigenically different serotypes of HFRS virus are presented.

[Detection of hemagglutinins in hantavirus serotypes and some of their properties].

Hemagglutinating properties of various serological types of hantavirus isolated in the Primorye area from humans with hemorrhagic fever with the renal syndrome and rodents, natural carriers of the virus, were studied. The strains were found to differ by capacity to accumulate hemagglutinin at multiplication in Vero E-6 cells. A method for elevating hemagglutinin titers by combined treatment of cultural virus-containing fluid with polyethyleneglycol-6000 and Tween-80-ether or acetone-ultrasound is suggested. The hemagglutinating activity of hantavirus strains was found to depend on the method of treatment and pH range.

[Genetic differentiation of hantaviruses using the polymerase chain reaction and sequencing].

Thirty-two hantavirus strains and 8 samples of lung tissue from rodents collected in different regions of Russia have been examined by molecular biological methods. Two methodological approaches have been employed for the study of genetic relationships between the viruses: nested PCR assay and common RT-PCR with subsequent direct sequencing of 200 and 365 base pair of G2 protein encoding regions of M-segment, respectively, and the resultant sequences were compared with those of the prototype hantavirus. The study revealed a mosaic pattern of distribution of different hantavirus genotypes on the territory of Russia.

[The antigenic differentiation of hantaviruses by using monoclonal antibodies].

85 hantavirus (HV) strains from Russia and other counties were examined by indirect fluorescent antibody test using monoclonal antibodies (MAbs) prepared against hantaviruses: Hantaan (Hantaan 76-118, A-9 strains), Seoul (SR-1, R-22 strains) and Puumala (Hallnas 83-223 strain). 68 HV strains were differentiated into five virus type groups: Hantaan, Puumala, Seoul, Prospect Hill and Belgrade-Dobrava. 17 strains were found to be antigenically closely related but distinct from Hantaan type (4 strains), Puumala (6 strains) and Prospect Hill (7 strains). The antigenic characteristics of these 17 strains suggested two supplementary antigenic subgroups of the Hantaan, two of Puumala and two of Prospect Hill.

[Approaches to selecting a strain of hantavirus for developing an inactivated brain vaccine].

Four strains isolated from patients with hemorrhagic fever with the renal syndrome and from field mice, natural hosts of Hantavirus, were examined in order to choose a functionally active strain fit for the development of inactivated Hantavirus brain vaccine. Virulent strain PM-10508/89 isolated from mouse lungs was chosen. The strain was highly virulent for newborn mice and rats: 100% of infected mice aged up to 24 h and 75% of rats developed an acute lethal infection upon challenge. The incubation period of Hantavirus adapted to mouse brain was decreased from 19 to 7 days. The infective titer of the virus in brain suspension was 0.1 to 6.5 lg LD50/ml. The strain was highly immunogenic and possessed a high hemagglutinating activity.

[The ecological aspects of the epidemiology of hemorrhagic fever with renal syndrome in the south of the Far East in Russia].

The data on the relationship between the epizootic process in the population of mice (the source of Hantavirus infection) and the seasonal manifestations of morbidity in hemorrhagic fever with renal syndrome (HFRS) in the southern region of the Russian Far East are presented. The asynchronous course of the epizootic process in the population of rodents causes irregularity in the spread of HFRS in the region and periodicity in the appearance of foci. Seasonal rises in HFRS morbidity coincide with the activity of the epizootic process in the population of mice: spring-summer rises with an increase in the number of infected rodents in the populations of large Japanese field mice and autumn-winter rises in voles.

[Highly purified agar for virological, immunochemical and microbiological research].

A new effective method for the preparation of highly purified agar for microbiological, virological and immunochemical investigations has been obtained and the technology for its production has been developed. The study revealed that the physico-chemical characteristics of newly obtained highly purified agar are much better than those of microbiological agar currently produced in Russia and not inferior to those of the best kinds of agar produced by Difco (USA), Serva and Ferak (Germany). The method of purification is protected by an author's certificate. The production of this agar for medical purposes has been permitted. The manufacturing plant "Medbioagar", created under the auspices of the Research Institute of Epidemiology and Microbiology has now started the serial production of highly purified agar under the name "Primagar". Further increase of the production of the preparation will facilitate its introduction into laboratory practice with the use of modern research methods in the fields of microbiology and virology, molecular biology and genetics, immunology and biochemistry.
