Physiologic inactivation of fluid phase C3b: isolation and structural analysis of C3c, C3d,g (alpha 2D), and C3g.

The fragments that result from the inactivation of C3b have not been completely characterized. Initial inactivation is catalyzed by the protease factor I, which, in the presence of its cofactor (factor H), cleaves two peptide bonds in the alpha'-chain of C3b. This results in the release of a small peptide (C3f, Mr 3000) from iC3b, which consists of the C3 beta chain covalently bonded to two alpha'-chain-derived peptides (Mr 68,000 and Mr 43,000). Surface-bound iC3b is cleaved at a third site by factor I to produce C3c and C3d,g (or alpha 2D). The factor I cofactor for this cleavage is the C3b receptor that is present on erythrocyte and leukocyte membranes. This report describes the isolation and initial structural characterization of C3c and C3d,g generated in whole blood after complement activation with cobra venom factor. These fragments were compared with the C3 fragments isolated from the serum and plasma of a patient with complement activation in vivo. The fragments were isolated with two solid phase monoclonal antibodies, one of which recognizes a determinant on C3g (clone 9) and one of which recognizes a determinant on C3c (clone 4). C3c isolated from normal blood showed three polypeptides that had apparent m.w. of 75,000, 43,000, and 27,000. The C3d,g consisted of a single polypeptide chain with a m.w. of 40,000. Amino terminal sequence analysis showed that the Mr 27,000 peptide from C3c is derived from the amino terminal portion of the alpha'-chain of C3b, whereas the Mr 43,000 peptide is derived from the carboxy terminus of the same chain. Amino terminal sequence analysis showed also that C3g is derived from the amino terminus of C3d,g. The C3 fragments isolated from a patient with partial lipodystrophy, nephritic factor activity, low serum C3 levels, and circulating C3 cleavage products showed a more complicated pattern on SDS-PAGE. The fragment isolated with clone 9 had an apparent m.w. of 40,000, identical to C3d,g generated in vitro, and it had the same amino terminal sequence as C3d,g generated in vitro. The eluate from insolubilized clone 4, however, showed prominent bands with Mr of 75,000, 56,000, 43,000, and 27,000, together with a triple-banded pattern at 68,000 and a minor band at 80,000. This eluate thus appears to contain C3c, and iC3b or an iC3b-like product. The origin of the Mr 56,000 and Mr 80,000 peptides have not yet been determined. These studies, with previous data, definitively order the C3c and C3d,g peptides in the alpha-chain of C3.(ABSTRACT TRUNCATED AT 400 WORDS)

Human C1 inhibitor: improved isolation and preliminary structural characterization.

An improved procedure for the isolation of the C1 inhibitor (C1-INH) component of human complement is reported. Following preliminary steps to remove plasminogen, fibrinogen, and aggregated material, three conventional chromatographic steps are used to isolate C1-INH in high (70%) overall yield. An extinction coefficient (E 1%, 1 cm 280nm) of 3.60 has been determined. The isolated protein exhibits a single band on sodium dodecyl sulfate-polyacrylamide gel electrophoresis, with a mobility corresponding to an apparent molecular weight (Mr) of 105 000. After removal of carbohydrate, the protein shows an increased mobility, corresponding to an apparent Mr of 78 000. A total carbohydrate content of 33% has been calculated, and from this and the size of the deglycosylated polypeptide, a true molecular weight of 116 000 was estimated. Further analysis of the carbohydrate has indicated a galactose:mannose ratio of 2:1 and approximately equimolar amounts of N-acetylglucosamine and N-acetylgalactosamine. This composition is unusual for a plasma protein and suggests that much of the carbohydrate is contained in linkages other than the typical N-glycosidic structures. Values found for the amino acid composition are compared with those reported previously. The amino-terminal sequence (40 residues) of C1-INH is also reported. Asparagine lies at the amino terminus. Neither high-performance liquid chromatography of the released phenylthiohydantoin derivative nor back-hydrolysis of the thiazolinone permitted identification of the residue contained at position 3. The sequence around this position is compatible, however, with an N-glycosidic linkage to residue 3.(ABSTRACT TRUNCATED AT 250 WORDS)

Long-term infection of Schistosoma mansoni in a vervet monkey (Cercopithecus aethiops).

Schistosoma mansoni infection in Cercopithecus species monkeys has been previously monitored for up to five years. This is an account of observations made during an autopsy on a male vervet monkey which had harboured the parasite for 15 years. Viable eggs had been recovered from the faeces throughout the infection, being hatched to observe the emergence of miracidia. Apart from a nodular appearance of the liver capsule, which was noted before the perfusion, and localized haemorrhaging in the large bowel there was comparatively little pathology attributable to the parasite. 86 worms were recovered from the mesenteries by perfusion. The distribution of worm eggs in the tissues was mainly in these two organs, although with the remarkably low incidence of granuloma it is likely that the schistosome infection would have been overlooked in a routine autopsy. It is stressed that these observations have been made on an individual animal, but it is felt that there is real significance with respect to the aetiology of natural transmission.

Retarded development of Schistosoma mansoni in immunosuppressed mice.

Previous observations, which showed that treatment of mice with hydrocortisone acetate around the time of their infection with Schistosoma mansoni reduced the mature worm burden, have been confirmed. The number of eggs produced by the surviving worms during early patency was also significantly reduced. Cyclophosphamide and another steroid immunosuppressant, betamethasone, also caused a reduction in fecundity of S. mansoni when given at the time of infection, as did T-cell deprivation of the mice by adult thymectomy and injection of anti-thymocyte serum 1 month before infection. There was no effect of these three treatments on worm numbers. In contrast, injection of mice with anti-thymocyte serum at the time of infection marginally increased the size of the mature worm burden. The deleterious effects of hydrocortisone acetate on S. mansoni worm numbers and fecundity were only apparent if the steroid was given within 1 week of infection. Indomethacin, a compound which inhibits tissue inflammatory reactions by inhibiting prostaglandin synthesis, and which therefore mimics one of the actions of corticosteroids, had no effect on S. mansoni worm maturation and fecundity.

The effect of enteral nutritional support on skeletal muscle protein synthesis and whole-body protein turnover in fasted surgical patients.

Whole-body and muscle protein turnover have been studied in surgical patients, before abdominal surgery after an 18-h fast, using infusion of 1-[13C]leucine. Four patients received an unmodified hospital diet and four received extra nutritional support (1800 kcal, 70 g protein) by naso-jejunal tube for 7-10 d previously. In patients who received nutritional support there were significantly greater values of whole-body protein synthesis (+42 per cent, P less than 0.05) and breakdown (+45 per cent, P less than 0.01) and muscle protein synthesis (+36 per cent, P less than 0.05) than in those who received the unmodified hospital diet.

Bovine serum albumin, sperm motility, and the "dilution effect'.

Epididymal spermatozoa from rabbit and ram were washed either once or twice using an efficient washing procedure and were then diluted in various media to a final concentration of approximately 1.4 x 10(7) cells/ml and incubated at 30 degrees C for up to 12 hours. Bovine serum albumin (BSA), either untreated or defatted, was found to be better than polyvinylpyrrolidone, ovalbumin, or alpha-lactalbumin, both at stimulating and maintaining motility levels and at reducing the tendency of the washed spermatozoa to stick to glass. BSA was effective in all media tested, being independent of Ca2+, PO4(3-), HCO3-, and ionic strength). BSA has a reversible stimulatory effect on motility. If BSA was added to sperm suspensions 3 1/2 hours after they had been washed and diluted in protein-free medium, motility was stimulated to levels not significantly lower than those observed in samples that had been washed and diluted in the presence of BSA. However, samples washed into BSA and then washed free of it behaved essentially as though they had never been in contact with protein. The motility, survival, and response to BSA of twice-washed spermatozoa were the same as those of once-washed spermatozoa, showing that epididymal plasma factors are not required for survival in vitro. It was concluded that dilution is not essentially detrimental to rabbit and ram spermatozoa. However, severe dilution of semen may result in levels of male reproductive tract fluids insufficient either to stimulate motility or to prevent sticking of motile cells to container surfaces. Few motile spermatozoa are recovered from samples of such diluted semen.

The interaction of ram proacrosin and acrosin forms with antiserum raised against ram m beta-acrosin.

An antiserum was raised in rabbits to pure m beta-acrosin, the stable form of active acrosin from ram spermatozoa. By use of immunoprecipitation with Protein A-bearing Staphylococcus aureus cells, so as to react antigen with antibody and isolate the complex rapidly, it could be demonstrated that the anti-m beta-acrosin cross-reacted strongly with native proacrosin, with a degenerate proacrosin form, and with at least one form of active acrosin other than m beta-acrosin. Sperm proteins unrelated to acrosin were not recognized. It is concluded that rabbit anti-m beta-acrosin will react specifically with all the major ram acrosin forms; the antiserum can therefore be used with confidence in immunocytochemical studies to locate both proacrosin and acrosin in ram spermatozoa.
