The immune status of various leprosy patients was evaluated by using a micromethod to evaluate lymphocyte responses to phytohemagglutinin (PHA). In our study, whole blood was used and the degree of response to PHA stimulation was expressed in terms of unit volume of blood. A markedly decreased response to PHA stimulation was noted in patients with active lepromatous leprosy. Patients with active lepromatous leprosy who have been proved drug (DDS) resistant showed less response than did those of drug sensitive patients with active lepromatous disease, while the patients with active lepromatous leprosy complicated by erythema nodosum leprosum (ENL) showed higher response than did those of patients with no complicated ENL. Comparing the results obtained to those obtained using other methods for T cell analysis indicates that these results reflect the number of T lymphocytes in the leprosy patient. Thus, this simple method is of value in assaying the presence and responses of T lymphocytes in the leprosy patient.
We studied the expression of gamma delta T cell receptors (TCR) on T-cell lines and clones derived from peripheral blood lymphocytes (PBL) from certain patients with primary immunodeficiency disorders and normal donors. Immunoprecipitation with the anti-Leu 4, anti-gamma-chain and/or anti-delta-chain monoclonal antibodies followed by SDS-PAGE analysis revealed that 7 of 13 (54%) T-cell lines and clones developed from PBL of patients with primary immunodeficiency disorders expressed non-disulfide-linked gamma delta TCR, utilizing either the C gamma 2abc or the C gamma 2bc gamma-chain constant region gene segment. 5 of 13 (38%) T-cell lines/clones expressed disulfide-linked gamma delta TCR, whereas an additional T-cell line was comprised of T cells expressing either disulfide-linked (C gamma 1) or non-disulfide-linked (C gamma 2bc) gamma delta TCR. T-cell lines and clones developed from four of light patients with primary immunodeficiency disorders exhibited exclusively non-disulfide-linked gamma delta TCR utilizing either the C gamma 2abc or the C gamma 2bc gamma-chain segment. T-cell lines derived from a fifth patient exhibited primarily non-disulfide-linked gamma delta TCR, bringing to five of eight the numbers of patients that expressed exclusively or primarily non-disulfide-linked gamma delta TCR. T-cell lines/clones derived from the remaining three patients exhibited exclusively disulfide-linked gamma delta TCR. The age of these patients varied over a wide range and there was not an association between their age and the type of gamma delta TCR expressed on T-cell lines derived from their PBL. In contrast, to these findings 14 of 16 (87.5%) T-cell clones derived from PBL of normal donors expressed disulfide-linked gamma delta TCR, whereas only 2 of 16 (12.5% expressed non-disulfide linked gamma delta TCR. Among the T-cell clones from normal donors which express disulfide-linked gamma delta TCR two different types were identified. Those exhibiting under reducing conditions on SDS-PAGE two completely resolved polypeptide chains in the range of 37 kD to 44 kD, and those exhibiting under the same conditions indistinguishable overlapping gamma- and delta- chains in the range of 40-42 kD. Several T-cell lines and clones from normal donors or patients with primary immunodeficiency that expressed either disulfide- or non-disulfide-linked gamma delta TCR were delta TCS1+, demonstrating that the delta TCS1 determinant is expressed on both types of gamma delta TCR.(ABSTRACT TRUNCATED AT 400 WORDS)

In cancer and other therapeutic research, an interpretation of median survival times can and should take cure into account. With this qualification, an analysis of recently published data provides further statistically significant evidence in favor of cancer chronotherapy as compared to homeostatic therapy.

Large-amplitude circadian rhythms were observed in the urinary excretion of polyamines by rats bearing an immunocytoma. Control animals excreted polyamines at a lower rate but also with marked circadian variation. In confirmation of earlier observations, light-chain excretion by the tumor-bearing rats also exhibited a circadian rhythm, superimposed on an increasing trend. The potential of these rhythms as markers for the chronotherapy of cancer is noted.

Rapid and reproducible detection of RNA in cells and tissue sections is routinely accomplished using in-situ hybridization technique provided that the target number of mRNA copies is above a minimum number. Detection of low copy transcripts is problematic when threshold detection occurs below clear signal resolution or alternatively, when technical problems result in background noise which occludes clear signal. RT in-situ PCR methodology utilizes both the power and specificity of PCR to amplify target whose localization is subsequently detected at the cellular level. RT in-situ PCR methods routinely involve a two-step methodology. mRNA copies are initially transcribed into cDNA. This step is followed by a separate PCR step wherein amplification of the newly synthesized cDNA takes place. A simplified one-step procedure biochemically compartmentalizes these sequential steps within a single applications methodology using the enzyme rTth. This method was successfully applied to detect and localize mRNA transcripts for Fas ligand within the immune privileged placental environment and to provide verification of immunohistochemical localization of gene product.
