Isothiazoloquinolones with enhanced antistaphylococcal activities against multidrug-resistant strains: effects of structural modifications at the 6-, 7-, and 8-positions.

We describe the biological evaluation of isothiazoloquinolones (ITQs) having structural modifications at the 6-, 7-, and 8-positions. Addition of a methoxy substituent to C-8 effected an increase in antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA) and a decrease in cytotoxic activity against Hep2 cells. Removal of fluorine from C-6 or replacement of the C-8 carbon with a nitrogen compromised anti-MRSA activity. When the groups attached at C-7 were compared, the anti-MRSA activity decreased in the order 6-isoquinolinyl > 4-pyridinyl > 5-dihydroisoindolyl > 6-tetrahydroisoquinolinyl. The compound with the most desirable in vitro biological profile was 9-cyclopropyl-6-fluoro-8-methoxy-7-(2-methylpyridin-4-yl)-9H-isothiazolo[5,4-b]quinoline-3,4-dione (7g). This ITQ demonstrated (i) strong in vitro anti-MRSA activity (MIC90 = 0.5 microg/mL), (ii) strong inhibitory activities against S. aureus DNA gyrase and topoisomerase IV, with weak activity against human topoisomerase II, (iii) weak cytotoxic activities against three cell lines, and (iv) efficacy in an in vivo murine thigh model of infection employing MRSA.

Generalized groove density equation concerning bending-grating-related imaging properties.

Varied-line-spacing gratings are used in soft-x-ray monochromators and spectrometers because of their ability to correct various aberrations by varying the groove density. A curved grating blank in a Rowland circle mounting could eliminate primary aberrations. Bending a plane constant-groove-density grating into a desirable surface can eliminate higher-order aberrations as a result of the change in surface profile and groove density. This provides a simple means for improving the imaging properties. When used in a monochromator, the grating can be bent to different degrees to minimize aberrations at all wavelengths. General formulas concerning the transformations of groove function, groove density, and groove spacing are presented for the bending grating. Analytic results with a numerical example show the significant properties of the system.

[Study on the myoelectric activity of dilatation muscles of upper airway in patients with obstructive sleep apnea-hypopnea syndrome under asleep condition].

OBJECTIVE: To observe the changes of the electromyography(EMG) of the dilatation muscles of upper airway in patients with obstructive sleep apnea-hypopnea syndrome (OSAHS) under asleep condition and to explore their functions and significance. METHOD: The myoelectronic activity of levator palatini muscle, tensor palatini muscle, genioglossus muscle was measured by ENG in patients with moderate to serious OSAHS before and after induction of sleep and was compared with normal control. RESULT: (1) The myoelectronic activity of dilation muscles of the upper airway in OSAHS patients were higher than that in control under awake condition (P < 0.01). (2) When the patients were asleep, the myoelectronic activity of these muscles dropped significantly (P < 0.01). (3) The decrease of myoelectronic activity of dilatation muscles of upper airway was more dramatic in patients with OSAHS than that in control when turned from awake condition to asleep condition (P < 0.01). CONCLUSION: The compensative raise of myoelectronic activity of dilatation muscles of upper airway under awake condition and the decompensation when asleep in patients with OSAHS were important in the pathogenesis.

[The epidemic survey of children rhinosinal inflammatory diseases and their correlation with asthma in Nanjing].

OBJECTIVE: To realize the epidemic features of rhinosinal inflammatory diseases in children and their correlation with asthma in Nanjing,China. METHOD: A questionnaire survey on children chronic rhinosinusitis (CRS) and airway diseases was conducted among 1087 elementary school children, aged from 9 to 10, of seven schools in Nanjing city by random cluster sampling, from March to September, 2004. The questionnaires include such questions as age, gender, family history, basis diseases, inducements, environment, therapy, correlative lower airway diseases and so on. The survey included screening suspects of CRS and allergic rhinitis (AR) by filling up the questionnaires by their parents and then special examination withtion of asthma of children was obtained by inquiring whether children were diagnosed by physician in last 12 months. The results were dealt with statistics. RESULT: Nine hundred and eighty nine questionnaires were returned. The response rate was 91% and 942 questionnaires were available. The prevalence rate of CRS of children aged from 9 to 10 was 8.8% in Nanjing city. The prevalence rate of AR was 5.1%. The prevalence rate of asthma of children was 5.3%. The incidences of asthma of children with CRS and AR were 19.3% and 39.6% respectively, while that of children without rhinosinal inflammatory diseases was 1. 8%. The results showed that the differences of the incidences of asthma between children with CRS, AR and without rhinosinal inflammatory diseases were statistically significant( P <0.01). CONCLUSION: Chronic rhinosinusitis is a common disease in children. The prevalence rates of CRS and AR in children aged from 9 to 10 in Nanjing city were 8.8% and 5. 1%. The incidence of asthma in children with rhinosinal inflammatory diseases is markedly higher than that in children without these diseases.

Biological evaluation of isothiazoloquinolones containing aromatic heterocycles at the 7-position: In vitro activity of a series of potent antibacterial agents that are effective against methicillin-resistant Staphylococcus aureus.

We synthesized a diverse series of 9H-isothiazolo[5,4-b]quinoline-3,4-diones containing heteroaromatic groups at the 7-position via palladium-catalyzed cross-coupling. Many of these compounds demonstrated potent antistaphylococcal activity (MICs 2 microg/mL) against a multi-drug-resistant strain (ATCC 700699) and low cytotoxic activity (CC(50)>100 microM) against the human cell line Hep2 (laryngeal carcinoma).

Isothiazoloquinolones containing functionalized aromatic hydrocarbons at the 7-position: synthesis and in vitro activity of a series of potent antibacterial agents with diminished cytotoxicity in human cells.

This report describes 9H-isothiazolo[5,4-b]quinoline-3,4-diones (ITQs) containing aromatic groups at the 7-position that were prepared using palladium-catalyzed cross-coupling and tested against a panel of susceptible and resistant bacteria. In general, these compounds were more effective against Gram-positive than Gram-negative organisms. Many of the ITQs were more potent than contemporary quinolones and displayed a particularly strong antistaphylococcal activity against a clinically important, multi-drug-resistant strain. In contrast with ITQs reported previously, several of the analogues described in this Letter demonstrated low cytotoxic activity against a human cell line.

Simplification of vector ray tracing by the groove function.

Tracing rays through arbitrary diffraction gratings (including holographic gratings of the second generation fabricated on a curved substrate) by the vector form is somewhat complicated. Vector ray tracing utilizes the local groove density, the calculation of which highly depends on how the grooves are generated. Characterizing a grating by its groove function, available for almost arbitrary gratings, is much simpler than doing so by its groove density, essentially being a vector. Applying the concept of Riemann geometry, we give an expression of the groove density by the groove function. The groove function description of a grating can thus be incorporated into vector ray tracing, which is beneficial especially at the design stage. A unified explicit grating ray-tracing formalism is given as well.

Low-dosage adrenaline induces transient marked decrease of blood pressure during functional endoscopic sinus surgery.

BACKGROUND: Low-dosage adrenaline contained in local anesthetics is commonly used for hemostasis during functional endoscopic sinus surgery (FESS), but often causes significant hemodynamic side effects that might be neglected. A randomized, controlled, prospective clinical trial was designed to find out these effects after local infiltration of different concentrations and/or different dosages of adrenaline during FESS under general anesthesia. METHODS: One hundred eight adult patients were randomized into one of four groups to receive 2 mL of 2% lidocaine and adrenaline 1:200,000 (group I), 4 mL of 1% lidocaine and adrenaline 1:400,000 (group II), 4 mL of 1% lidocaine and adrenaline 1:200,000 (group III), or 4 mL of 1% lidocaine (group IV) for local infiltration, respectively. Heart rate, systolic blood pressure (BP), diastolic BP, and mean arterial pressure were monitored continuously in radial artery. RESULTS: Significant hemodynamic changes, particularly decrease of BP (p < 0.001) with slightly increased heart rate (p < 0.001) approximately 1.5 minutes after local infiltration, were observed in group I, group II, and group III compared with the baseline, but not in group IV. However, no significant hemodynamic changes were observed between group I, group II, and group III at the same time points (p > 0.05). CONCLUSIONS: Local infiltration of low-dosage adrenaline causes temporary significant hemodynamic changes, particularly marked decrease of BP during FESS, and there were no significant hemodynamic changes between adrenaline 1:200,000 and 1:400,000.
