Alternative moving window factor analysis for comparison analysis between complex chromatographic data.

In this investigation, a novel chemometric method is developed for the analysis of five possible relationships of components or spectral features between two correlative but different hyphenated chromatographic systems. It is very helpful for comparison study of components present in different complex systems in both chemistry and systems biology. The proposed method, named alternative moving window factor analysis (AMWFA), could be utilized to determine the number of common components between different samples and then to identify their corresponding spectra half-automatically. AMWFA can alternatively be employed to mind for the selective information hiding in anyone of the two compared data X and Y, and to self-verify the resolution results by changing the extracted target matrices in analysis. From the results of comparison of simulated hyphenated chromatographic data, volatile chemical components in drug pair rhizoma ligustici chuanxiong-radix paeoniae rubra (RLC-RPR) and its single herbal medicines, and analysis of Angelica oral solution and its plasma sample after oral intake to rabbit, powerful ability of the proposed method is shown.

Development, optimization and validation of a fingerprint of Ginkgo biloba extracts by high-performance liquid chromatography.

A four-step development, optimization and validation strategy for high-performance liquid chromatography (HPLC) fingerprints of Ginkgo biloba extract is described. A suitable chromatographic system was selected first. The following step was performing a screening design to select important parameters. After selecting some controllable parameters and their range to further optimize, gradient optimization with uniform design was done. At last, method validation including determination of injection precision, repeatability, and a sample stability test, was performed. Through this effective and integrated four-step method, a feasible and reliable HPLC fingerprint to identify and assess the Ginkgo biloba quality can easily be established using a linear gradient elution with acetonitrile/0.1% phosphoric acid (from 14/86 to 30/70, v/v, in 40 min) as mobile phase, a column temperature of 30 degrees C and a detection wavelength of 350 nm. The strategy can also be applied for the development of fingerprints in the quality control of other herbal medicines.

Two-step multivariate adaptive regression splines for modeling a quantitative relationship between gas chromatography retention indices and molecular descriptors.

The relationship between retention indices and molecular descriptors of alkanes is established by two-step multivariate adaptive regression splines (TMARS). TMARS combines linear regression with multivariate adaptive regression splines (MARS). It is demonstrated for the present data set that using linear regression or MARS modeling alone causes lack of fit. TMARS avoids lack of fit and appreciably improves the prediction ability for the model. The use of this combined approach permits the development of additional understanding of the adaptive nature in MARS modeling.
