Transgene correction maintains normal cochlear structure and function in 6-month-old Myo15a mutant mice.

The shaker2 (sh2) mouse is a murine model for human non-syndromic deafness DFNB3. The mice have abnormal circling behavior suggesting a balanced disorder, and profound deafness. The insertion of a bacterial artificial chromosome (BAC) transgene containing the Myo15a gene into sh2/sh2 zygotes confers hearing capability and abolishes the circling behavior in 1-month-old transgenic animals. In this study, we investigated both the hearing and the morphology of the cochlea in Myo15a mutants carrying this BAC transgene at two, four, or six months of age. The hearing threshold of these mice is normal, with no physiologically significant differences compared to age-matched heterozygous sh2J mice (with or without the BAC transgene). In six-month-old transgenic mice with the BAC, the morphology of hair cells in the apical and upper basal turns of the cochlea is normal. Hair cells of lower basal turn, however, were missing in some mutant animals. This study demonstrates that BAC transgene correction cannot only maintain normal morphology but also confer stable hearing function in Myo15a mutant mice for as long as 6 months. In addition, excess Myo15a expression has no physiologically significant protective or deleterious effects on hearing of normal mice, suggesting that the dosage of Myo15a may not be problematic for gene therapy.

Host range, growth property, and virulence of the smallpox vaccine: vaccinia virus Tian Tan strain.

Vaccinia Tian Tan (VTT) was used as a vaccine against smallpox in China for millions of people before 1980, yet the biological characteristics of the virus remain unclear. We have characterized VTT with respect to its host cell range, growth properties in vitro, and virulence in vivo. We found that 11 of the 12 mammalian cell lines studied are permissive to VTT infection whereas one, CHO-K1, is non-permissive. Using electron microscopy and sequence analysis, we found that the restriction of VTT replication in CHO-K1 is at a step before viral maturation probably due to the loss of the V025 gene. Moreover, VTT is significantly less virulent than vaccinia WR but remains neurovirulent in mice and causes significant body weight loss after intranasal inoculation. Our data demonstrate the need for further attenuation of VTT to serve either as a safer smallpox vaccine or as a live vaccine vector for other pathogens.

Recognition of gene acceptor site based on multi-objective optimization.

A new method for predicting the gene acceptor site based on multi-objective optimization is introduced in this paper. The models for the acceptor, branch and distance between acceptor site and branch site were constructed according to the characteristics of the sequences from the exon-intron database and using common biological knowledge. The acceptor function, branch function and distance function were defined respectively, and the multi-objective optimization model was constructed to recognize the splice site. The test results show that the algorithm used in this study performs better than the SplicePredictor, which is one of the leading acceptor site detectors.

Protein secondary structure prediction using dynamic programming.

In the present paper, we describe how a directed graph was constructed and then searched for the optimum path using a dynamic programming approach, based on the secondary structure propensity of the protein short sequence derived from a training data set. The protein secondary structure was thus predicted in this way. The average three-state accuracy of the algorithm used was 76.70%.

[Effects of prenatal methylmercury exposure on learning and memory ability of mice and ultrastructure of hippocampus neurons in mice].

OBJECTIVE: To study the effects of prenatal methylmercury exposure on learning and memory ability of mice and ultrastructure of hippocampus neurons in mice. METHODS: Methylmercury in doses of 0 and 4mg/(kg x d) were fed to pregnant mice at 7th-9th day after conception. Learning and memory ability of the mice offspring was detected. The ultrastructure of hippocampus neurons was observed by electron microscope after the completion of the experiment. RESULTS: The learning and memory ability of the mice offspring of 4 mg/(kg x d) dose group was lower than those of 0 mg/(kg x d) dose group (P < 0.05). Changes in the ultrastructure of hippocampus neurons were obvious in 4 mg/(kg x d) dose group as compared with the 0 mg/(kg x d) dose group. CONCLUSION: Methylmercury could affect the growth and development of central nerve system. The learning and memory ability of mice offspring was greatly damaged and the ultrastructure of hippocampus neurons was changed by methylmercury.

Early diagnosis of bacterial and fungal infection in chronic cholestatic hepatitis B.

AIM: To investigate the early diagnostic methods of bacterial and fungal infection in patients with chronic cholestatic hepatitis B. METHODS: One hundred and one adult in-patients with chronic hepatitis B were studied and divided into 3 groups: direct bilirubin (DBil)/total bilirubin (TBil) > or = 0.5, without bacterial and fungal infection (group A, n=38); DBil/TBil <0.5, without bacterial and fungal infection (group B, n=23); DBil/TBil> or = 0.5, with bacterial or fungal infection (group C, n=40). The serum biochemical index and pulse rate were analyzed. RESULTS: Level of TBil, DBil, alkaline phosphatase (ALP) and DBil/ALP in group A increased compared with that in group B. The level of ALP in group C decreased compared with that in group A, whereas the level of TBil, DBil and DBil/ALP increased (ALP: 156+/-43, 199+/-68, respectively, P<0.05; TBil: 370+/-227, 220+/-206, respectively, P<0.01; DBil: 214+/-143, 146+/-136, respectively, P<0.01; DBil/ALP: 1.65+/-1.05, 0.78+/-0.70, respectively, P<0.001). The level of DBil and infection affected DBil/ALP. Independent of the effect of DBil, infection caused DBil/ALP to rise (P<0.05). The pulse rate in group A decreased compared with that in group B (63.7+/-6.4, 77.7+/-11.4, respectively, P<0.001), and the pulse rate in group C increased compared with that in group A (81.2+/-12.2, 63.7+/-6.4, respectively, P<0.001). The equation (infection=0.218 pusle rate +1.064 DBil/ALP -16.361), with total accuracy of 85.5%, was obtained from stepwise logistic regression. Pulse rate (> or =80/min) and DBil/ALP (> or =1.0) were used to screen infection. The sensitivity was 62.5% and 64.7% respectively, and the specificity was 100% and 82.8% respectively. CONCLUSION: Bacterial and fungal infection deteriorate jaundice and increase pulse rate, decrease serum ALP and increase DBil/ALP. Pulse rate, DBil/ALP and the equation (infection=0.218 pusle rate+1.064 DBil/ALP-16.361) are helpful to early diagnosis of bacterial and fungal infection in patients with chronic cholestatic hepatitis B.

[Preparation and characterization of rabbit antibody against human selenoprotein P].

AIM: To prepare rabbit antibody against human selenoprotein P (HSelP) by using HSelP expressed in the prokaryotic expression system and identify its specificity. METHODS: HSelP gene fragment was expressed in E.Coli via IPTG induction and purified through DEAE and Ni-NTA columns sequentially. The purified HSelP as immunogen was used to immunize rabbit. The specificity of rabbit anti-HSelP antibody was analyzed by Western blot. RESULTS: HSelP protein had been expressed and purified successfully. The polyclonal anti-HSelP antibody could specifically recognize natural HSelP from human serum. CONCLUSION: Polyclonal anti-HSelP antibody with high specificity and purity has been prepared by using purified HSelP as immunogen, which lays the foundation for further research on the function and distribution of HSelP, as well as developing a simple method for detecting HSelP.

[Study rat nephrotoxicity of Longdan Xiegan Tang].

OBJECTIVE: To investigate the nephrotoxocity of Longdan Xiegan Tang in rats, and to test its safety. METHOD: Sprague-Dauley rats were given Longdan Xiegan Tang 4.5 mL x (100 mg)(-1) Bid for thirty days, and the control group was given NS. MTP, BUN, Cr, MDA, MTP/Ucr and SOD were measured on 1st, 2nd, 3rd, 4th week. The kidney tissues were viewed with light microscopy and electron microscope. RESULT: MTP and MTP/Ucr were obviously higher than controls ( P < 0.01), and the other index had no difference (P > 0.05). No remarkable structural change could been seen with light microscopy, but with electron microscope we could find that the basal membranes were thickened and some of foot process were infused. CONCLUSION: Longdan Xiegan Tang will result in injury of kidney function.

[The effect of huanglian jiedu tang on Alzheimer's disease and its influence on cytokines].

OBJECTIVE: To investigate the effect of huanglian jiedu tang on Alzheimer' s disease, and its influence on cytokines. METHOD: AD rats models were made by A beta injection, and then water-maze tests were made to investigate the study and memory ability. TNF-alpha, INF-gamma and IL-2 were examined by ELISA. After treatment with huanglian jiedu tang, the rats were evaluated again. RESULT: After A beta injection, the study and memory ability of the rats was decreased. By the treatment with huanglian jiedu tang, it was improved (P < 0.01); TNF-alpha, INF-gamma and IL-2 levels of AD rats were higher than those in control group (P < 0.01); after treatment the levels came down (P < 0.01). CONCLUSION: Huanglian jiedu tang can improve the study and memory ability of AD rats by changing their immune state.

[The apoptosis-inducing activity of human selenoprotein P shorter isoform].

OBJECTIVE: Human selenoprotein P (HSelP) is unique protein that contains 10 selenocysteines encoded by 10 inframe UGA, which typically function as stop codon. The function of HSelP remains unclear, in part due to the inability to express it by gene recombinant technique. This study is to investigate expression and purification of recombinant HSelP in prokaryotic expression system, and its activity to induce apoptosis in vitro. METHODS: The shorter HSelP isoform was cloned. After the selenocysteine (SeCys) at 40th position from N terminus of the HSelP shorter isoform was mutated into cysteine by PCR, it was expressed in E. coli. The expressed product was purified with DEAE column and identified by Western blot. Subsequently, its function on induction of mitochondrial apoptotic activity was studied. RESULTS: The mutant HSelP shorter isoform expressed in prokaryotic system was purified by DEAE column to 90% homogeneity. The purified product, HSelP280m, induced the opening of mitochondrial permeability transition pore (PTP) and decreased the transmembrane potential in a dose-dependent manner. These events could be abolished by PTP specific inhibitors. CONCLUSION: HSelP280m can induce the opening of mitochondrial PTP, which provides a basis for investigating the structure and function of recombinant HSelP.

[The study of yi-shen-huo-xue fang's effects on expression of GMP-140 and cleaning out the oxygenic free radicle on rabbits blood stasis model].

OBJECTIVE: To evaluate the effects of Yi-Shen-Huo-Xue Fang on expression of GMP-140 and cleaning out the oxygenic free radicle on rabbits blood stasis model. METHOD: Thirty rabbits were divided randomly into five groups as the normal group, model group, large dose of "Yi-Shen-Huo-Xue Fang" group, small dose of "Yi-Shen-Huo-Xue Fang" group and "Xue-Shuan-Xin-Mai-Ning" group. After being treated respectively, granule membrane protein 140(GMP-140), erythrocyte sueroxide dismutase (E-SOD), erythrocyte lipid peroxide(E-LPO), plasma lipid peroxide(P-LPO) were checked up. RESULT: The GMP-140, E-SOD, E-LPO, P-LPO in normal control were compared with those in model groups, With the difference(P < 0.01), model control group was compared with large dose group and small dose group (P < 0.01), with "Xue-Shuan-Xin-Mai-Ning" group(P < 0.05), large dose group was compared with "Xue-Shuan-Xin-Mai-Ning" group(P < 0.05), and large dose group were compared with small dose group (P > 0.05). CONCLUSION: The model was made successfully. Large dose group, small dose group and "xue-shuan-xin-mai-ning" group can inhibit expression of GMP-140, enhence SOD activity and decrease LPO content on blood stasis rabbit model. Large dose group and small dose group have stronger effect than "xue-shuan-xin-mai-ning" group.

Mice deficient in fractalkine are less susceptible to cerebral ischemia-reperfusion injury.

Fractalkine (FKN), also known as neurotactin, is a CX(3)C chemokine that exists in both secreted and neuronal membrane-bound forms and is upregulated during brain inflammation. There is accumulating evidence that FKN induces chemotaxis by binding to its receptor CX(3)CR1 on leukocytes and microglia. We generated FKN-deficient mice to study the role of FKN in postischemic brain injury. After transient focal cerebral ischemia, FKN-deficient mice had a 28% reduction in infarction size and lower mortality rate, when compared to wild-type littermates. The findings of this study indicate a possible role for FKN in augmenting postischemic injury and mortality after transient focal cerebral ischemia.

Fluorescence in situ hybridization analysis of alien genes in Agrobacterium-mediated Cry1A(b)-transformed rice.

The transgene in Agrobacterium-mediated Cry1A(b)-transgenic rice plants has been detected and its chromosomal location determined by fluorescence in situ hybridization (FISH). Eight of the nine transgenic lines tested showed hybridization signals. Signals were located on regions of the chromosome in which fraction length (FL) values varied from 26.2 (near the centromere) to 95.2 (distal regions). No signal was found on regions where the fraction length was less than 26.2, while six of the nine signals detected were located on regions with FL values of 75.3 or over. This demonstrates that Agrobacterium-mediated genes can integrate into multiple sites distributed in different parts of the chromosome, but that distal regions are the preferred sites and regions near the centromeres are colder for T-DNA integration. The donor DNA of the transformation was divided into two parts, labelled separately as probes for two-colour FISH. Results show that the transformed DNA sequences remained linked in the recipient genome. The relationship between integration position and transgene silencing, known as the 'position effect', is discussed.

[Effect of alpha-difluoromethylornithine on growth characteristics and expression of ALT-04ag gene of human lung carcinoma cells].

OBJECTIVE: To study the effect of polyamine biosynthesis inhibition on growth characteristics of human lung carcinoma cells and its correlation with the expression of human lung carcinoma associated antigen ALT-04ag gene. METHODS: The gene expression was detected by RT-PCR and immunocytochemical tests. The cell growth characteristics were studied by cell growth curves, morphological observation, FCM analysis and DNA electrophoresis. RESULTS: Human lung squamous carcinoma cells L78 treated with 5 mmol/L alpha-difluoromethylornithine (DFMO) for 5 days showed significant growth inhibition and apoptosis induction. The mRNA and protein expressions of ALT-04ag gene in the cells were downregulated, while these changes resulted from DFMO treatment were prevented by provision of DFMO along with exogenous putrescine. CONCLUSION: The effect of polyamine biosynthesis inhibition induced by DFMO restrains the growth characteristics and promotes apoptosis of human lung carcinoma L78 cells, which is associated with down regulation of ALT-04ag gene expression.
