Role of Toll-like receptor 4 in macrophage activation and tolerance during Salmonella enterica serovar Typhimurium infection.

Toll-like receptors (TLRs) play an important role in the innate immune response, particularly in the initial interaction between the infecting microorganism and phagocytic cells, such as macrophages. We investigated the role of TLR4 during infection of primary murine peritoneal macrophages with Salmonella enterica serovar Typhimurium. We found that macrophages from the C3H/HeJ mouse strain, which carries a functionally inactive Tlr4 gene, exhibit marked impairment of tumor necrosis factor alpha (TNF-alpha) secretion in response to S. enterica serovar Typhimurium infection. However, activation of extracellular growth factor-regulated kinase and NF-kappa B signaling pathways was relatively unaffected, as was increased expression of TNF-alpha mRNA. Furthermore, macrophage tolerance, which is associated with increased expression of the NF-kappa B p50 and p52 subunits, was induced by S. enterica serovar Typhimurium even in the absence of functional TLR4. These results indicate that during infection of macrophages by S. enterica serovar Typhimurium, TLR4 signals are required at a posttranscriptional step to maximize secretion of TNF-alpha. Signals delivered by pattern recognition receptors other than TLR4 are sufficient for the increased expression of the TNF-alpha transcript and at least some genes associated with macrophage tolerance.

[Studies on the homing-related adhesion activities of UCB HSPC in short-term ex vivo expansion].

OBJECTIVE: To study the effect of ex vivo expansion on the adhesion activities of umbilical cord blood hematopoietic stem and progenitor cells (HSPC). METHODS: Fresh UCB CD(34)(+) cells were cultured in a serum and stroma-free culture system. At day 7, day 10 and day 14, CD(34)(+) cells were re-selected from the expanded products. The expression of adhesion molecules (CAMs) such as VLA-4, VLA-5, LFA-1, ICAM-1, HCAM, L-selectin and PECAM-1, and the adhesion activity of the expanded CD(34)(+) cells were evaluated and compared with those of precultured fresh CD(34)(+) cells. RESULTS: (1) The CD(34)(+) cells expressing homing-related CAMs were increased (from 15-fold increase for CD(34)(+) CD(54)(+) subset to 72-fold increase for CD(34)(+) CD(49e)(+) subset at day 14). (2) The expressions of CD(49d), CD(44), CD(11a) and CD(49e) on the expanded CD(34)(+) cells were increased or sustained the same levels as those on fresh UCB CD(34)(+) cells, while the expression of CD(62L), CD(54) and CD(31) on expanded CD(34)(+) cells declined with the cultivating. (3) Spontaneous adhesion and SDF-1-induced adhesion tended to be increased in the course of the first 10 day's culture. CONCLUSIONS: The culture system used in this study could substantially support the expansion of HSPCs expressing the above CAMs, and the expanded HSPCs would sustain their intrinsic adhesion potentials.

[Adenovirus-mediated transfer of TIMP-4 gene inhibits neointimal formation after balloon injury].

OBJECTIVE: To investigate the effects of human tissue inhibitor of matelloproteinase-4 (TIMP-4) on vascular smooth muscle cells (VSMCs) migration and the neointimal formation after balloon injury in rats. METHODS: The cultured VSMCs were transfected with an adenoviral vector containing human TIMP-4 gene, AdTIMP-4. Effect of TIMP-4 on VSMC migration was investigated by monolayer cell scrape. AdTIMP-4, control adenoviral vector or PBS was transduced into the rat carotid artery from the adventitial after carotid artery injury. Cell number within the internal elastic lamina 4 days after gene transfer was counted and neointima/media area ratio 28 days after gene transfer was calculated. RESULTS: The migration distance of VSMCs infected with AdTIMP-4 was inhibited markedly. Morphometric analysis demonstrated a statistically significant reduction in the number of cells migrated into neointima compared with controls [(32.5 +/- 4.8) cells per section, (33.8 +/- 7.0) cells per section and (8.2 +/- 2.4) cells per section for uninfected, AdGFP-treated and AdTIMP-4-treated arteries, respectively]. There was a reduction of intima/media ratio of TIMP-4-treated group by 66.5% compared with control groups 28 days after gene transfer (P < 0.01). There was no statistical difference between the control adenoviral vector group and PBS group. CONCLUSION: Adenoviral mediated gene transfer of TIMP-4 inhibits migration of VSMC and significantly reduces neointimal hyperplasia in the rat carotid balloon injury model. The TIMP-4 gene transfer is a potential therapeutic approach to preventing neointimal formation after balloon injury.

[NMR studies on cetirizine hydrochloride].

AIM: To study the NMR phenomena of cetirizine hydrochloride and assign all proton and carbon signals in NMR spectra. METHODS: To record the 1D and 2D NMR spectra of cetirizine hydrochloride while changing the experimental temperature and adding D2O into the solution. RESULTS: More than one NMR signal or broad peak resulting from piperazine and the attached groups with N atom were given in DMSO-d6 solution at room temperature. "Coalescence" or narrowing had occurred for the proton and carbon signals when the experimental temperature was increased or D2O was added into the solution. CONCLUSION: Compared with the NMR "time scale", there are more than one conformation of cetirizine hydrochloride in DMSO-d6 solution at room temperature. The different conformation will be exchanged fast while temperature rise and the stable conformation will be existed while D2O was added into the solution.

[The sinus node itself also plays a role in heart rate slowing down during postnatal development].

The slowing down mechanism of heart rate during growth of the body after birth was studied in isolated rabbit heart and sinus node (SN) preparation with Langendorff perfusion method, conventional microelectrode recording and perforated patch for recording pacemaker current I(f). The radioimmunoassay was also used to measure the concentration of cAMP within SN cells. The results indicate that without the influence of nervous and humoral factors, the spontaneous heart rate would also become slower as the rabbit grew older, which is due to the decrease of spontaneous depolarized rate of phase 4 in SN cells. The negative directed shift of the threshold potential of I(f) and the decrease in cAMP concentration within SN cells may be responsible for the phenomenon. The results obtained suggest that besides the nervous and humoral factors which influence the heart rate, the changes in automaticity of SN cell itself may take part in the slowing down process as the body grows up.

Dissociation energies and charge distribution of the Co-NO bond for nitrosyl-alpha,beta,gamma,delta-tetraphenylporphinatocobalt(II) and nitrosyl-alpha,beta,gamma,delta-tetraphenylporphinatocobalt(III) in benzonitrile solution.

The first two series of Co-NO bond dissociation enthalpies in benzonitrile solution were determined for 12 cobalt(II) nitrosyl porphyrins and for 12 cobalt(III) nitrosyl porphyrins by titration calorimetry with suitable thermodynamic cycles. The results display that the energy scales of the heterolytic Co(III)-NO bond dissociation, the homolytic Co(III)-NO bond dissociation, and the homolytic Co(II)-NO bond dissociation are 14.7-23.2, 15.1-17.5, and 20.8-24.6 kcal/mol in benzonitrile solution, respectively, which not only indicates that the thermodynamic stability of cobalt(II) nitrosyl porphyrins is larger than that of the corresponding cobalt(III) nitrosyl porphyrins for homolysis in benzonitrile solution but also suggests that both cobalt(III) nitrosyl porphyrins and cobalt(II) nitrosyl porphyrins are excellent NO donors, and in addition, cobalt(III) nitrosyl porphyrins are also excellent NO(+) contributors. Hammett-type linear free energy analyses suggest that the nitrosyl group carries negative charges of 0.49 +/- 0.06 and 0.27 +/- 0.04 in T(G)PPCo(II)NO and in T(G)PPCo(III)NO, respectively, which indicates that nitric oxide is an electron-withdrawing group both in T(G)PPCo(II)NO and in T(G)PPCo(III)NO, behaving in a manner similar to Lewis acids rather than to Lewis bases. The energetic and structural information disclosed in the present work is believed to furnish hints to the understanding of cobalt nitrosyl porphyrins' biological functions in vivo.

[The effects of cytokines mediated ex vivo expansion on the cell adhesion molecule expression of cord blood hematopoietic stem and progenitor cells].

OBJECTIVE: To compare the expression of cell adhesion molecules (CAMs) among VLA-4 (CD49 d), VLA-5 (CD49e), LFA-1 (CD11a), L-selectin (CD62L), and PECAM-1 (CD31) which are more related to the homing of hematopoietic stem and progenitor cells (HSPC) on the ex vivo expanded CD34+ subset with that of fresh isolated AC133+ cells. METHODS: AC133+ cells selected from fresh cord blood (CB) samples were cultured in QBSF-60 serum-free media in the presence of Flt-3 ligand + SCF + TPO (FST), with initial addition of IL-3 for up to 2 week. Expansion potential and the expression of above CAMs were evaluated at day 0, day 7, day 10 and day 14. RESULTS: (1) Simultaneously numerical expansion of various HSPC was constantly observed during the culture, and the fold expansion of AC133+ cells and CD34+ cells on day 14 were 33.50 and 64.56 respectively; (2) The number of CD34+ subsets expressing the above adhesions were all increased at different degrees (from 20 fold to 160 fold). (3) The expressions of CD11a, CD49d, and CD49e on ex vivo expanded CD34+ cells were increased as compared to their baseline levels, but the percentage of CD62L+ and CD31+ subpopulations in CD34+ cells were decreased. CONCLUSIONS: Our short-term culture system can not merely support the simultaneous expansion of CB derived AC133+ cells, but the expanded hematopoietic progenitors may well sustained the expression of homing-related adhesion molecules.

Sensitive determination of synephrine by flow-injection chemiluminescence.

It was found that light emission produced by the oxidation of luminol by potassium ferricyanide in basic medium was enhanced by synephrine, an anti-obesity drug. The optimum conditions for this chemiluminescent reaction were studied in detail in a flow injection system and employed in a new, simple and rapid method for the determination of synephrine. A mechanism for this reaction is proposed, based on the chemiluminescence reaction spectra. In the optimum conditions, CL intensity is proportional to concentration of synephrine in the 0.008-1 microg/mL range. The limit of detection is 1.6 ng/mL for synephrine (3sigma), and the relative standard deviation (n = 11) is 2.6% for 0.5 microg/mL synephrine. The method was applied to the determination of synephrine in herbal products, citrus fruit and biological fluids. The recoveries were satisfactory (90-102%). The results given by the proposed method are in good agreement with those given by HPLC-UV.

Chemiluminescence of synephrine based on the cerium(IV)-rhodamine B system.

A new chemiluminescence (CL) method is described for the determination of synephrine. It is based on the reaction between synephrine and Ce(IV) in a nitric acid medium and measurement of the CL intensity produced by rhodamine B used as a luminophore, similar to luminol or lucigenin in basic media, instead of as a sensitizer. In the optimum conditions, the increase of CL intensity was correlated with synephrine concentration over the range 5.0 x 10(-9)-1.0 x 10(-6) g/mL with a detection limit of 1.0 x 10(-9) g/mL. The relative standard deviation (RSD) was 2.9% for 1.0 x 10(-7) g/mL synephrine (n = 11). The method was applied to the determination of a drug in herbal products, citrus fruit and biological samples, with satisfactory results. The results given by the proposed method are in good agreement with those given by HPLC-UV and UV spectrophotometry.

Screening and identification of linear B-cell epitopes and entry-blocking peptide of severe acute respiratory syndrome (SARS)-associated coronavirus using synthetic overlapping peptide library.

A 10-mer overlapping peptide library has been synthesized for screening and identification of linear B-cell epitopes of severe acute respiratory syndrome associated coronavirus (SARS-CoV), which spanned the major structural proteins of SARS-CoV. One hundred and eleven candidate peptides were positive according to the result of PEPscan, which were assembled into 22 longer peptides. Five of these peptides showed high cross-immunoreactivities (approximately 66.7 to 90.5%) to SARS convalescent patients' sera from the severest epidemic regions of the China mainland. Most interestingly, S(471-503), a peptide located at the receptor binding domain (RBD) of SARS-CoV, could specifically block the binding between the RBD and angiotensin-converting enzyme 2, resulting in the inhibition of SARS-CoV entrance into host cells in vitro. The study demonstrated that S(471-503) peptide was a potential immunoantigen for the development of peptide-based vaccine or a candidate for further drug evaluation against the SARS-CoV virus-cell fusion.

Cytotoxic activity of the recombinant anti-mesothelin immunotoxin, SS1(dsFv)PE38, towards tumor cell lines established from ascites of patients with peritoneal mesotheliomas.

BACKGROUND: Mesothelin, a cell surface glycoprotein, is an attractive candidate for targeted therapy given its overexpression, as detected by immunohistochemistry, in mesotheliomas. The goal of this study was to evaluate mesothelin expression in fresh tumor cells obtained from ascites of patients with peritoneal mesothelioma, as well as to determine the sensitivity of these cells to an immunotoxin targeting mesothelin. MATERIALS AND METHODS: Tumor cells were evaluated for mesothelin expression by flow cytometry using the murine anti-mesothelin monoclonal antibody K1. The sensitivity of these tumor cells to SS1(dsFv)PE38, an immunotoxin consisting of the anti-mesothelin Fv linked to a mutated Pseudomonas exotoxin, was evaluated using a cell proliferation assay. RESULTS: Of the 7 tumor cell lines established from ascites of 12 patients with peritoneal mesothelioma, 6 expressed mesothelin while one cell line did not. Cell lines that expressed mesothelin were very sensitive to SS1(dsFv)PE38 with IC50s ranging between 0.08-3.9 ng/ml, while the cell line that was mesothelin-negative was resistant to SS1(dsFv)PE38. CONCLUSION: High expression of mesothelin is seen on tumor cells of patients with peritoneal mesothelioma and correlates with sensitivity to SS1(dsFv)PE38. Clinical studies of SS1(dsFv)PE38 in patients with peritoneal mesotheliomas are ongoing.

Should people be physically active outdoors on smog alert days?

BACKGROUND: Given the importance of physical activity to well-being, there is a need to encourage people to be physically active year-round. At the same time, many people are vulnerable to adverse health effects from air pollution, especially on smog alert days. This study was undertaken to determine when air pollution levels tend to be lowest so that the public can modify strenuous outdoor activity accordingly. METHODS: Existing hourly air pollution data for Toronto were analyzed to determine how pollutant levels varied from hour to hour throughout each 24-hour day, to identify the times when pollution levels are at their lowest on average. RESULTS: Pollutant levels vary throughout the day, with concentrations of some pollutants (such as ozone, particles and sulphur dioxide) being highest during mid-day, and others (such as carbon monoxide and nitrogen dioxide) being highest with morning rush hour. Overall, pollutant concentrations tend to be lowest before seven a.m. and after eight p.m. INTERPRETATION: The public should be encouraged to maintain regular physical activity outdoors while monitoring any air pollution-related symptoms. The intensity of outdoor activity should be reduced, or activities replaced with indoor exercise, at those Air Quality Index (AQI) levels that trigger individual symptoms and when AQI values exceed 50. Where possible, strenuous activity should be taken when and where air pollution levels tend to be lowest, namely early in the morning and in low-traffic areas. More research is required to guide development of health protective advice on exercising when air quality is poor.
