Modulation of transcriptional sensitivity of mineralocorticoid and estrogen receptors.

Recent reports describe the ability of factors to modulate the position of the dose-response curve of receptor-agonist complexes, and the amount of partial agonist activity of receptor-antagonist complexes, of androgen, glucocorticoid (GRs), and progesterone receptors (PRs). We now ask whether this modulation extends to the two remaining steroid receptors: mineralocorticoid (MRs) and estrogen receptors (ERs). These studies of MR were facilitated by our discovery that the antiglucocorticoid dexamethasone 21-mesylate (Dex-Mes) is a new antimineralocorticoid with significant amounts of partial agonist activity. Elevated levels of MR, the co-activators TIF2 and SRC-1, and the co-repressor SMRT do modulate the dose-response curve and partial agonist activity of MR complexes. Interestingly, the precise responses are indistinguishable from those seen with GRs in the same cells. Thus, the unequal transactivation of common genes by MRs versus GRs probably cannot be explained by differential responses to changing cellular concentrations of homologous receptor, co-activators, or co-repressors. We also find that the dose-response curve of ER-estradiol complexes is left-shifted to lower steroid concentrations by higher amounts of exogenous ER. Therefore, the modulation of either the dose-response curve of agonists or the partial agonist activity of antisteroid, and in many cases the modulation of both properties, is a common phenomenon for all of the classical steroid receptors.

Genetic and metabolic analysis of folate salvage in the human malaria parasite Plasmodium falciparum.

Antifolate drugs that target the biosynthesis and processing of essential folate cofactors are widely used for treatment of chloroquine-resistant falciparum malaria. Salvage of pre-formed folate can strongly compromise the efficacy of these drugs in vitro and the availability of folate from the human host in natural infections also influences therapeutic outcomes. To investigate how different parasite lines respond to the presence of exogenous folate, we measured the effect of the latter on the susceptibility of parasites to sulfa-drug blockage of folate biosynthesis, utilising the parents and 22 progeny of the HB3-Dd2 genetic cross of Plasmodium falciparum, together with selected unrelated lines. Complete linkage of the folate utilisation phenotype was observed to a DNA sequence of 48.6 kb lying between nucleotide positions 738,489 and 787,058 of chromosome 4 and encompassing the dihydrofolate reductase-thymidylate synthase (dhfr-ts) gene locus. Examination of the putative ORFs on this fragment upstream (3) and downstream (4) of dhfr-ts revealed no plausible candidate genes for folate processing. Similarly, a marked heterogeneity in the 5'-UTR regions of Dd2 and HB3, manifest as a directly repeated 256 bp sequence in the former, also did not correlate with the folate utilisation phenotype nor apparently influence levels of dhfr-ts transcripts or protein products. By contrast, the nature of the coding sequence of the dhfr domain appeared to play a direct role, with the single mutant (S108N) HB3-type utilising folic acid much less efficiently than other allelic variants. We also compared the processing of exogenous folic acid, folinic acid and p-aminobenzoic acid (pABA) in metabolic labelling studies of HB3 and Dd2. These support the view that DHFR is likely to have a low-level folate reductase activity as well as its normal function of reducing dihydrofolate to tetrahydrofolate, and that a significant hurdle in the utilisation of exogenous folic acid is the initial reduction of fully oxidised folic acid to dihydrofolate, an activity that the single mutant enzyme found in HB3 is postulated to perform particularly poorly. This would mirror earlier studies indicating that the DHFR activity of HB3 is also compromised relative to other variants.

Down-regulation and promoter methylation of tissue inhibitor of metalloproteinase 3 in choriocarcinoma.

OBJECTIVE: To assess the differential gene expression in neoplastic and normal trophoblastic cells and evaluate the effect of methylation on tissue inhibitor of metalloproteinase 3 (TIMP3) expression in choriocarcinoma (CCA) cells. METHODS: The Atlas Human Cancer 1.2 Array (Clontech) was used to compare differential gene expression in a trophoblastic cell line (B6) established from first term placenta with two choriocarcinoma cell lines (JAR and JEG-3). The differentially expressed candidate genes in the placental and malignant trophoblastic cells in these cell lines were confirmed by real-time polymerase chain reaction (PCR), Western blotting and immunohistochemistry. Differential expression of a specific gene, TIMP3, was confirmed by immunohistochemistry using clinical specimens of choriocarcinoma and placenta. The involvement of promoter methylation in suppression of TIMP3 expression in choriocarcinoma was examined using methylation-specific PCR (MSP) and demethylation treatment. RESULTS: Differential expression of 23 genes was observed in choriocarcinoma cell lines compared to the placental trophoblastic cells using the cDNA array analysis (Atlas Human Cancer Array, Clontech). Among these differentially expressed genes, down-regulation of TIMP3, PLAB, IGFBP3 and up-regulation of CCNB1 were confirmed by real-time PCR determination. Reduced expression of TIMP3 was further confirmed in clinical samples of choriocarcinoma by immunohistochemical staining. Methylation of TIMP3 promoter was detected in choriocarcinoma cell lines and clinical samples of choriocarcinoma. Treatment with a demethylation drug, 5-aza-2'-deoxycytidine, in choriocarcinoma cell lines restored TIMP3 expression. CONCLUSION: Down-regulation of TIMP3, PLAB, IGFBP3 and up-regulation of CCNB1 were observed in choriocarcinoma cells compared to placental trophoblasts. Down-regulation of TIMP3 expression was confirmed in clinical specimens of choriocarcinoma and may play a role in its pathogenesis. Promoter methylation of the TIMP3 is involved in suppression of TIMP3 expression. Differentiation expression of TIMP3 in choriocarcinoma may have potential application in clinical diagnosis and patient treatment.

Protein thermal aggregation involves distinct regions: sequential events in the heat-induced unfolding and aggregation of hemoglobin.

Protein thermal aggregation plays a crucial role in protein science and engineering. Despite its biological importance, little is known about the mechanism and pathway(s) involved in the formation of aggregates. In this report, the sequential events occurring during thermal unfolding and aggregation process of hemoglobin were studied by two-dimensional infrared correlation spectroscopy. Analysis of the infrared spectra recorded at different temperatures suggested that hemoglobin denatured by a two-stage thermal transition. At the initial structural perturbation stage (30-44 degrees C), the fast red shift of the band from alpha-helix indicated that the native helical structures became more and more solvent-exposed as temperature increased. At the thermal unfolding stage (44-54 degrees C), the unfolding of solvent-exposed helical structures dominated the transition and was supposed to be responsible to the start of aggregation. At the thermal aggregation stage (54-70 degrees C), the transition was dominated by the formation of aggregates and the further unfolding of the buried structures. A close inspection of the sequential events occurring at different stages suggested that protein thermal aggregation involves distinct regions.

The emergence of cultural self-constructs: autobiographical memory and self-description in European American and Chinese children.

This study examined the emergence of cultural self-constructs as reflected in children's remembered and conceptual aspects of the self. European American and Chinese children in preschool through 2nd grade participated (N=180). Children each recounted 4 autobiographical events and described themselves in response to open-ended questions. American children often provided elaborate and detailed memories focusing on their own roles, preferences, and feelings; they also frequently described themselves in terms of personal attributes, abstract dispositions, and inner traits in a positive light. Chinese children provided relatively skeletal accounts of past experiences that centered on social interactions and daily routines, and they often described themselves in terms of social roles, context-specific characteristics, and oven behaviors in a neutral or modest tone. Findings are discussed in light of the self as a constructed meaning system of culture that emerges early in life.

The stories we keep: autobiographical memory in American and Chinese middle-aged adults.

One hundred and eight European American and Chinese adults, aged between 38 and 60, participated in this questionnaire study. They each recalled 20 memories from any period of their lives. Memory content was analyzed as a function of culture (U.S. and China), life period (childhood, youth, early midlife, and peak midlife), and gender (female and male). Across the four life periods, Americans provided more memories of individual experiences and unique, one-time events and focused on their own roles and emotions. In contrast, Chinese were more inclined to recall memories of social and historical events and placed a great emphasis on social interactions and significant others in their memory narratives. Chinese also more frequently drew upon past events to convey moral messages than did Americans. In addition, memory content evidenced age-related increases in both autonomous and social orientations. Findings are discussed in light of the self-definitional and directive functions of Autobiographical memory in the context of culture.

Transfection studies to explore essential folate metabolism and antifolate drug synergy in the human malaria parasite Plasmodium falciparum.

Folate metabolism in Plasmodium falciparum is the target of important antimalarial agents. The biosynthetic pathway converts GTP to polyglutamated derivatives of tetrahydrofolate (THF), essential cofactors for DNA synthesis. Tetrahydrofolate can also be acquired by salvage mechanisms. Using a transfection system adapted to studying this pathway, we investigated modulation of dihydropteroate synthase (DHPS) activity on parasite phenotypes. Dihydropteroate synthase incorporates p-aminobenzoate (pABA) into dihydropteroate, the precursor of dihydrofolate. We were unable to obtain viable parasites where the dhps gene had been truncated. However, parasites where the protein was full-length but mutated at two key residues and having < 10% of normal activity were viable in folate-supplemented medium. Metabolic labelling showed that these parasites could still convert pABA to polyglutamated folates, albeit at a very low level, but they could not survive on pABA supplementation alone. This degree of disablement in DHPS also abolished the synergy of the antifolate combination pyrimethamine/sulfadoxine. These data indicate that DHPS activity above a low but critical level is essential regardless of the availability of salvageable folate and formally prove the role of this enzyme in antifolate drug synergy and folate biosynthesis in vivo. However, we found no evidence of a significant role for DHPS in folate salvage. Moreover, when biosynthesis was compromised by the absence of a fully functional DHPS, the parasite was able to compensate by increasing flux through the salvage pathway.

[Operative treatment of complicated distal femoral fractures].

OBJECTIVE: To explore an effective way for treating severe complicated distal femoral fractures. METHODS: Twenty-six patients with complicated distal femoral fracture who all belonged to 33C3.3 type according to AO/ASIF classification, were treated with a lateral condylar buttress plate or self-designed aliform anatomical plate, and operated on with allogeneic bone grafting. RESULTS: All cases were followed up for an average of 14 months (ranging 5-25 months). Twenty-four wounds were primary healing postoperatively, 2 wounds were infected and healed after dressing change. Twenty-four had bone healing after 4-11 months, 2 needed to operate again because of earlier weight-bearing resulting in fixation failure. According to shelbourne and Brueckmann score, the excellent and good rate was 88.46%. CONCLUSION: The internal fixation for complicated distal femoral fracture by self-designed aliform anatomical plate and lateral condylar buttress plate with a great deal of allograft bone is an effective surgical method. As it has long oval holes and the holes are consecutive, the aliform anatomical plate is more suitable for severe complicated fractures. At the same time, autogenous-ilium transplantation can be substituted by the allograft bone.

METHODS: 72 cases with liver cirrhosis of different stages were classified according to child-pugh's categories A, B, C, in which there were 23 cases in child A, 29 cases in child B, and 20 cases in child C. The plasma levels of uPA, uPAR, PAI-1 and the serum levels of HA, CIV were detected by ELISA. The serum PCIII concentration was determined by radioimmunoassay. RESULTS: With the progression of hepatic fibrosis, the plasma levels of uPA, uPAR and PAI-1 were (1.36+/-0.43) microg/L, (3.03+/-1.48) microg/L and (24.09+/-7.14) microg/L respectively in group A, (1.79+/-0.62) microg/L, (4.80+/-2.22) microg/L and (41.40+/-17.52) microg/L respectively in group B. The highest levels were in child C, whose levels were (1.88+/-0.64) microg/L, (4.82+/-2.02) microg/L and (52.60+/-16.87) microg/L respectively, compared with group A and group B, t value were from 2.81 to 7.38, all of P value were less than 0.01. There was negative correlation between the plasma levels of uPA and the serum PCIII (r=-0.4785, P<0.05) in child A, but, positive correlation between the plasma PAI-1 and the serum HA (r=0.5447, P<0.01) in child C. The value of PAI-1/uPA was significantly decreased in child A, but increased in child B and child C. CONCLUSION: In the late of liver cirrhosis, increased PAI-1 together with uPA, uPAR are associated with overall inhibition of matrix degradation. The plasma levels of uPA and PAI-1 were correlation to the progression of liver cirrhosis.

[Detection of microsatellite instability in primary gastric cancer using fluorescence-labelled primers multiplex PCR].

To assess the role of microsatellite instability(MSI) on chromosome 19 in the carcinogenesis of gastric cancer. 26 microsatellite markers on chromosome 19 were amplified by Fluorescence-Labelled Primers Multiplex PCR in 44 paired gastric cancer tissues for MSI. The amplified fragments ran in PAGE gel by ABI 377 sequencer and the gel images were analyzed by GeneScan and Genotyper. Meanwhile, MSI of four of all microsatellite sites were also analyzed using PCR-SSLP-silver stain method. MSI was observed in 22 of 26(84.62%) microsatellite sites. D19S402 showed the highest frequency(22.73%). The total frequency of MSI was 8.3%. In 44 gastric cancer samples, 34(77.27%) cases showed MSI at one or more than one site. MSI, as a possible mechanism of carcinogenesis, might play an important role in human primary gastric cancer. MSI detections will become an integral part of the routine classification of all gastrointestinal tumors in the very near future.

[The plasma levels of transforming growth factor beta1 and the protein expressions of alpha-SMA, urokinase plasminogen activator and plasminogen activator inhibitor-1 in liver of patients with different grades of hepatic fibrosis].

OBJECTIVE: To measure the plasma levels of transforming growth factor beta1 (TGFbeta1), the protein expression of alpha-SMA in hepatic stellate cells and urokinase plasminogen activator (uPA) and plasminogen activator inhibitor-1 (PAI-1), and study on the relationships between the plasma levels of TGFbeta1, the protein expression and the serum hyaluronic acid (HA) in patients with different grades of hepatic fibrosis. METHODS: Thirty seven cases with hepatic fibrosis of different grades were classified according to HE and VG staining categories from 0 to 4, in which there were 8 cases in grade 1, 9 cases in grade 2, 7 cases in grade 3, 13 cases in grade 4. The plasma levels of TGFbeta1 and the serum levels of HA were detected by ELISA. The protein expressions of a-SMA, uPA and PAI-1 in fibrotic liver tissue were observed by immunohistochemistry. RESULTS: With the progression of hepatic fibrosis, the plasma levels of TGFbeta1 and the protein expression of a-SMA, uPA and PAI-1 in fibrotic liver tissue were increased. In grade 3 and 4, the plasma levels of TGFbeta and the protein expression of a-SMA and PAI-1 in fibrotic liver tissue were significantly increased, but the protein expression of uPA in cirrhosis liver tissue did not increased. CONCLUSION: TGFbeta1, a-SMA, uPA and PAI-1 play an important role in the progression of hepatic fibrosis. Inhibiting the early activation of latent TGFbeta1 or increasing uPA and inhibiting PAI-1 over express may contribute to matrix degradation and retard the progression of hepatic fibrosis.

[Tumor necrosis factor related apoptosis-inducing ligand induces apoptosis and sensitizes resistant hepatic cancer cells to chemotherapy].

OBJECTIVE: To investigate the effective of tumor necrosis factor related apoptosis-inducing ligand (TRAIL) combination with chemotherapeutic agent inducing apoptosis in resistant hepatic cancer cells. METHODS: HepG2 cells resistant to Adriamycin (HepG2/ADR) were induced stepwise. The effects of TRAIL in combination with ADM (0.1 mg/L) on promoting apoptosis in HepG2/ADR were analyzed, the proliferation was observed by MTT assay, the apoptosis of cells was also observed by flow cytometry and TUNEL method. RESULTS: HepG2/ADR was confirmed resisting to ADM. Treated with TRAIL combination with ADM (0.1 mg/L), it showed significant inhibitory effect on the growth of HepG2/ADM, the percentage of apoptosis was increased as comparison with control at 24 h. CONCLUSION: MDR1 might not take part in resistance to TRAIL-induced apoptosis. TRAIL dramatically augmented the sensitivity to chemotherapeutic agents in HepG2/ADR. Combined TRAIL with chemotherapeutic agents treatment could be a novel and attractive strategy to drug-resistant/ TRAIL-resistant tumor cells.

[Functional study on TGF-beta/Smads signaling pathway in human ovarian cancer cells].

Resistance to the growth inhibitory effects of transforming growth factor-beta (TGF-beta) is a characteristic of many transformed cells. The purpose of this study was to determine the response of ovarian cancer cells to TGF-beta1 and to investigate the roles of components of the TGF-beta/Smads signaling pathway in carcinogenesis of ovarian cancer. Three ovarian cancer cell lines, HO-8910, HO-8910PM and SKOV3, were treated with TGF-beta1 and assayed for growth response by MTT assay. Furthermore, expression and subcellular localization of the components of TGF-beta/Smads signaling pathway in these cell lines in the absence or presence of TGF-beta1 were determined by RT-PCR and immunofluorescence analysis. We found that proliferation of SKOV3 cell was not significantly inhibited by TGF-beta1 while it expressed all components of the TGF-beta/Smads signaling pathway. After exposure to TGF-beta1, Smad7 protein in SKOV3 increased transiently and translocated to cytoplasm from nucleus while P-Smad2 translocated into nucleus from cytoplasm. Taken together, the results suggested that the TGF-beta/Smads signaling pathway remained functional in human ovarian cancer cells, HO-8910, HO-8910PM and SKOV3, and the abnormalities of the downstream effectors of Smads proteins might contribute to the resistance of SKOV3 cell to TGF-beta1.

[Predicting human tumor-specific promoter using transcription factor binding sites].

To predict human tumor-specific promoters using computer technology, three data sets of tumor-specific promoter sequences, transcription factor binding site sequences and non-tumor promoter sequences were collected, and the tumor-specific promoter sequences and non-tumor promoter sequences were analyzed for the comparative density of each unique transcription factor binding site. The density of each transcription factor binding site in the corresponding data set was used to derive the density ratio, from which the eigenvector was constructed to identify human tumor-specific promoters. This method proved to be highly accurate in predicting the tumor-specific promoters, with recognition rates of over 90% of the training sequences and over 80% of the testing sequences.

[Clinical analysis of 92 cases of pregnancy with idiopathic thrombocytopenic purpura].

OBJECTIVE: To investigate the diagnosis, prenatal treatment and prognosis of pregnancy with idiopathic thrombocytopenic purpura (ITP). METHODS: The clinical data of 92 cases of pregnancy with ITP in Beijing Obstetrics and Gynecology Hospital during Oct. 1994 to Oct. 2003 were analysed, including the changes of platelet counts, bleeding improvement, and the outcome of mothers and infants after treatment by using glucocorticoids, gamma globulin and transfusion of platelets. RESULTS: The incidence of pregnancy with ITP was 2.4 per thousand. Thirty-four cases (40%) were accompanied with other complications, such as hypertensive disorders complicating pregnancy (7), postpartum hemorrhage (6), premature labor (9), impaired glucose tolerance (19) and fetal macrosomia (10). Among the 92 cases, vaginal delivery and cesarean section were performed in 20 (22%) and 72 (78%) cases, respectively. Twenty-six, 18 and 24 cases were treated by glucocorticoids, glucocorticoids plus gamma globulin, and glucocorticoids plus gamma globulin plus transfusion of platelets, respectively. Significant effects were achieved in 42 cases (62%), general effects in 16 cases (24%), and improvement in 8 cases (12%), respectively. The incidence of postpartum hemorrhage was 8% (6/72 cases). There was no maternal death. The infantile death ratio was 2/94 during perinatal period. The platelet count of peripheral blood in all the newborns was higher than 100 x 10(9)/L. There were no intracranial hemorrhage and other diseases related to platelet decrease. CONCLUSION: Glucocorticoids, gamma globulin and transfusion of platelets are effective methods for the treatment of ITP in mid-late pregnancy. They can increase the platelet counts and decrease the postpartum hemorrhage of the patients, and have no side effects to mother or infant.
