What we remember and what we tell: the effects of culture and self-priming on memory representations and narratives.

Two experiments were conducted to explore culture and self-priming effects on memories of Caucasian and Asian American adults (N=526). In the experimental conditions, either the collective or private self was primed prior to retrieval. Participants then described their earliest childhood memories (Study 1) or recalled a fictional story (Study 2). Systematic cultural differences in memory content were obtained across both memory tasks, independent of priming conditions. Caucasians tended to recall specific, one-moment-in-time events that focused on the individual as the central character. Asians tended to provide memories of general, routine events centering on collective activities and social interactions. Priming effects also emerged: memory content reflected the particular aspect of the self being primed. Findings are discussed in light of the interactive relation between memory representations and memory narratives and the role culture plays in remembering.

Bioinformatics analysis of alternative splicing.

Over the past few years, the analysis of alternative splicing using bioinformatics has emerged as an important new field, and has significantly changed our view of genome function. One exciting front has been the analysis of microarray data to measure alternative splicing genome-wide. Pioneering studies of both human and mouse data have produced algorithms for discerning evidence of alternative splicing and clustering genes and samples by their alternative splicing patterns. Moreover, these data indicate the presence of alternative splice forms in up to 80 per cent of human genes. Comparative genomics studies in both mammals and insects have demonstrated that alternative splicing can in some cases be predicted directly from comparisons of genome sequences, based on heightened sequence conservation and exon length. Such studies have also provided new insights into the connection between alternative splicing and a variety of evolutionary processes such as Alu-based exonisation, exon creation and loss. A number of groups have used a combination of bioinformatics, comparative genomics and experimental validation to identify new motifs for splice regulatory factors, analyse the balance of factors that regulate alternative splicing, and propose a new mechanism for regulation based on the interaction of alternative splicing and nonsense-mediated decay. Bioinformatics studies of the functional impact of alternative splicing have revealed a wide range of regulatory mechanisms, from NAGNAG sites that add a single amino acid; to short peptide segments that can play surprisingly complex roles in switching protein conformation and function (as in the Piccolo C2A domain); to events that entirely remove a specific protein interaction domain or membrane anchoring domain. Common to many bioinformatics studies is a new emphasis on graph representations of alternative splicing structures, which have many advantages for analysis.

Effects of three fluid resuscitation methods on apoptosis of visceral organs in rats with hemorrhagic shock.

OBJECTIVE: To observe the effects of three fluid resuscitation methods on apoptosis of visceral organs in rats with hemorrhagic shock. METHODS: A model of rat with severe hemorrhagic shock and active bleeding was established in 32 SD (Sprague-Dawley) rats. The rats were randomly divided into control group, no fluid resuscitation group (NF group), controlled fluid resuscitation group (NS40 group) and rapid large scale fluid resuscitation group (NS80 group). Each group contained 8 rats. The curative effects were compared. At the same time, the apoptosis in liver, kidney, lung and small intestinal mucosa of survivors after hemorrhage and resuscitation was detected by light microscopy in HE (hematoxylin and eosin) stained tissue sections, flow cytometry and terminal deoxynucleotidyl transferase dUTP nick end labelling (TUNEL). RESULTS: The survival rate of early fluid resuscitation (14/16) was markedly higher than that of NF group (3/8). There was some apoptosis in liver, kidney, lung and small intestinal mucosa of all survivors. Compared with NF and NS40 groups, the apoptosis of liver, kidney and small intestinal mucosa of NS80 group was obviously increased. CONCLUSIONS: Among three fluid resuscitation methods, controlled fluid resuscitation can obviously improve the early survival rate and the apoptosis of liver, kidney and small intestinal mucosa in rats with severe and uncontrolled hemorrhagic shock, and may benefit improvement of prognosis.

Applying informatics in tissue engineering.

OBJECTIVE: To facilitate tissue engineering strategies determination with informatics tools. METHODS: Firstly, tissue engineering experimental data were standardized and integrated into a centralized database; secondly, we used data mining tools (e.g. artificial neural networks and decision trees) to predict the outcomes of tissue engineering strategies; thirdly, a strategy design algorithm was developed, and its efficacy was validated with animal experiments; lastly, we constructed an online database and a decision support system for tissue engineering. RESULTS: The artificial neural networks and the decision trees respectively predicted the outcomes of tissue engineering strategies with the predictive accuracy of 95.14% and 85.26%. Following the strategies generated by computer, we cured 18 of the 20 experimental animals with a significantly lower cost than usual. CONCLUSION: Informatics is beneficial for realizing safe, effective and economical tissue engineering.

[Setup of high voltage direct circuit equivalent circuit model in leakage detection of landfill].

An equivalent circuit model is put forward through analysis of the experiment data under the conditions of applying 400V, 350V and 300V DC supply power to a small-scale simulated landfill. Due to the fact that the existence of the HDPE geomembrane liner endows the model with commutating character, the electrolytic capacity increases as the area of geomembrane liner and the supplying electric voltage increase. The contact electric resistance of the power supply electrode is in direct proportion to earth electric resistance rate, and in relation to the diameter of the electrode and the depth of the earth. Moreover, the contact resistance is mainly determined by the earth electric resistance around the electrode (the influence induced by the earth farther than 10 times electrode radius is less than 10 percent of those involved by all resistance), hence decrease of the earth electric resistance rate around the electrode may result in effective decrease of the contact electric resistance of the electrode.

[Diagnostic value of adenosine (99m)Tc-MIBI myocardial perfusion imaging for detecting coronary artery disease].

OBJECTIVE: The aim of this study was to assess the accuracy of adenosine (99m)Tc-MIBI myocardial perfusion SPECT for the diagnosis of coronary artery disease. METHODS: A total of 89 patients [mean age (57 +/- 9) years,62 men, 27 women] were included in this study. Adenosine was infused intravenously at a rate of 140 microg.kg(-1).min(-1) for 6 minutes. At the end of 3 minute of adenosine infusion, 925MBq of (99m)Tc-MIBI was injected intravenously. SPECT myocardial imaging acquisition was obtained 1 hour after adenosine infusion. Coronary angiography was performed in all patients. RESULTS: Thirty-one patients had significant coronary artery stenosis and 58 had normal coronary angiography. Adenosine myocardial perfusion imaging was abnormal in twenty-two out of the 31 patients with significant coronary artery stenosis (sensitivity, 71%). Fifty-three out of the 58 patients with normal angiography had a normal adenosine myocardial perfusion imaging (specificity, 91%). The positive predictive value and negative predictive value of adenosine (99m)Tc-MIBI myocardial perfusion imaging for detection of coronary artery disease was 81% and 84% respectively. CONCLUSION: Adenosine myocardial perfusion imaging is probably an accurate method for detecting coronary artery disease.

[Characteristics of melting and solidification process of fly ash from refuse incinerator].

This study was conducted to investigate the migration patterns of main compositions of the fly ash from refuse incinerator during melting and solidification process. The experiment was performed in a high temperature melting furnace with temperature controlled. X-ray fluorescence spectroscopy (XRF) and X-ray diffractometer (XRD) were used to analyzed the fly ash treated. The parameters investigated included main compositional contents, phase constituents, alkalinity, vaporation rate, volume reduction rate. The results show: (1) During the melting and solidification process, the contents of CaO, Al2O3, and SiO2 in fly ash increased as the temperature went up, but element Cl and S decreased from initial 20.59%, 10.74% to final 0.15%, 0.22%, respectively. This suggests that higher amount of Cl and S in original fly ash could lead to more vaporation in the form of chloride and calcium salfate in the process, which was further verified by XRD analyzing result. (2) The alkalinity decreased as melting temperature increased, but tended to be stable and maintained around 0.95 after temperature reached flowing temperature. (3) The decomposition and vaporation of the salts in the fly ash mainly occurred in the temperatures between 1150 degrees C and 1260 degrees C, which was approximately 100 degrees C lower than melting temperature.

[Computational study on the electronic absorption transitions for 2,3-diaryl maleic anhydrides].

The electronic absorption transitions for 2, 3-bis (2, 4-dimethylthiophen-3-yl) maleic anhydride were calculated by time-dependent method with 6-31G(d) basis at structures optimized at B3LYP/6-31G(d) level. The S1, S2, and S3 transitions calculated for the open and the closed forms of the compound are 390, 360.5, 339.4; 584.6, 395.8, 370.2 nm, respectively. The results are comparable to the electronic absorption spectrum of 2,3-bis(2,4,5-trimethylthiophen-3-yl) maleic anhydride which bears extra methyl groups that should not have much effects on the spectrum. The oscillator strengths are well correlated to the spectrum peak heights.

[Inhibitory effect of potential metastasis in SGC-7901 cells induced by beta-ionone].

OBJECTIVE: To determine the effect of beta-ionone on the potential metastasis of human gastric adenocarcinoma cell line SGC-7901 and the underlying mechanism. METHODS: Using curve of cellular growth, Zymograms, and RT-PCR assays, we analyzed the growth rate, the activities of two types IV collagenase of Matrix met alloproteinase 9 (MMP-9) and MMP-2 and the expression of nm23-H1 gene, tissue inhibitor of met alloproteinase 1 (TIMP-1) and TIMP-2 in SGC-7901 cells which were treated with progressively increasing concentrations (25, 50, 100 and 200 micromol/L) of beta-ionone for 24 h and 48 h. RESULTS: The growth of SGC-7901 cells was inhibited by beta-ionone. Eight days after treatment with different concentrations of beta-ionone, as mentioned above, the inhibition rates were 25.93%, 28.21%, 74.36% and 90.11%, respectively compared to the negative control. The estimated IC50 value of beta-ionone for SGC-7901 cells was estimated to be 89 micromol/L; beta-ionone did not show any effect on the activities of MMP-9 and MMP-2 in SGC-7901 cells. However, the expression of nm23-H1, TIMP-1 and TIMP-2 mRNA transcripts gradually increased in response to beta-ionone in a dose-dependent manner. CONCLUSION: beta-ionone can inhibit the growth and proliferation of SGC-7901 cells. It may show some effects on the potential metastasis of SGC-7901 cells indicates by its upregulation of nm23-H1, TIMP-1 and TIMP-2 expression. However, beta-ionone may have no effect on the activities of type IV collagenase in SGC-7901 cells. The mechanism by which beta-ionone inhibits the potential metastasis of SGC-7901 cells needs to be studied further.

[Potential antiviral drug Pueraria crude extract and puerarin protect against ethanol-induced cytotoxicity in embryonic mouse hippocampal cultures].

OBJECTIVE: To examine whether Chinese medical herb Pueraria crude extract (CP) and standard of pure puerarin (SP) possess the same neuroprotective effects during concomitant ethanol (EtOH) treatment. METHODS: Hippocampus cultures were prepared from mice at gestational age of 18 day. Cell viability was measured by MTT assay. RT-PCR was employed to determine mRNA expression of superoxide dismutase (SOD). RESULTS: As measured by MTT assay, supplementation with 15 mg/L CP or 10 mg/L SP afforded neuroprotection against all EtOH concentrations (50, 200 and 350 mmol/L, respectively) in embryonic hippocampal culture system. In addition, both 15 mg/L CP and 10 mg/L SP could decrease expression of SOD at mRNA level. CONCLUSION: This study suggests that CP and SP could decrease oxidative stress induced by ethanol treatment by the decreased expression of SOD at mRNA level, and demonstrates antioxidative neuroprotective effect of CP and SP against developmental ethanol exposure in vitro.

[Level of vitamin K-dependent coagulation factors in premature infants and the influence of maternal antenatal administration of vitamin K1 on their activity].

OBJECTIVE: Infants less than 35 weeks of gestational age are susceptible to peri-/intraventricular hemorrhage (PIVH). This may be due in part to low concentrations of vitamin K-dependent coagulation factors. This study was conducted to determine the umbilical cord blood activities of vitamin K-dependent coagulation factors II, VII, IX and X in premature infants to understand whether preterm infants have absence status of these factors the changes of theses factors' activities in premature infants' umbilical blood after vitamin K(1) was given to mothers antenatally and the preventing effectiveness of PIVH after maternal antenatal supplement of vitamin K(1). METHODS: Pregnant women in preterm labor at less than 35 weeks of gestational age were randomly selected to receive antenatal vitamin K(1) intramuscular or intravenous injections 10 mg per day for 2 to 7 days (vitamin K(1) group), or no vitamin K(1) treatment (control group). Dexamethone was antenatally given to both groups of pregnant women routinely. Vitamin K(1) group had 44 infants and the control group had 133 infants. During the same period, thirty full-term neonates' cord blood samples were obtained to determine theses factors to compare with those from the premature infants. The cranial ultrasound was performed by a same physician to understand whether the neonates were complicated with PIVH and its severity. RESULTS: The levels of vitamin K-dependent coagulation factors in umbilical blood in control group were significantly lower than those in full-term infants' cord blood (P < 0.05). However, in vitamin K(1) group, supplement of vitamin K(1) antenatally could significantly increase activities of factors II, VII and X in preterm infants' cord blood (P < 0.05). The total occurrence rates of PIVH in vitamin K(1) group and control group were 31.8% and 52.6%, respectively, (P = 0.017), and the frequency of severe PIVH in vitamin K(1) group and control group was 2.3% and 12.0%, respectively (P = 0.057). CONCLUSION: Preterm infants have absence status of vitamin K-dependent coagulation factors. Administration of vitamin K(1) to pregnant women at less than 35 weeks of gestational age resulted in significantly improved activities of vitamin K-dependent coagulation factors II, VII, and X, and a significantly decreased frequency of PIVH and less severe hemorrhage in preterm infants.

[Speciation analysis of arsenic in traditional Chinese medicine].

Some of traditional Chinese medicine (TCM) contain arsenide, such as realgar. The total amount of arsenic in the TCM exceeds the limits according to related regulations. But the roles of arsenic in TCM or its side-effects depend on its species existed in those therapies, not the total amount of arsenic. Therefore, in recent years, the analysis of arsenic in TCM focuses on the species of arsenic. The present paper summarized some methods and techniques in the speciation analysis of arsenic in TCM, in order that optimal methods can be chosen and the roles of arsenic could be evaluated properly.

[Variation characteristics of leachate quality in a semi-aerobic municipal solid waste landfill].

A large-scale semi-aerobic landfill set (21 m x 3.8 m x 6.0 m) was constructed to study the variation characteristics of pollutants in the landfill leachate. The results showed that after 39 weeks, the concentrations of CODCr, BOD5 and NH3+-N in the leachate declined rapidly, which reached 173, 30 and 15 mg x L(-1) respectively, and the reducing rate of NH3+-N was 99.6%. The pH value was below 7 during the first 2 weeks, but became weak alkaline after three weeks. Based on the experimental data, the attenuation equations of pollutants in the leachate were built.

[Dynamic variation of landfill gas production under different landfill treatment].

Based on the landfill principles, large scale installations of anaerobic and semi-aerobic landfills were established, with the concentrations of CH4, O2 and CO2 detected regularly. The results showed that the average CH4 concentration in semi-aerobic and anaerobic landfill installations was 7%-13% and 35%-50%, respectively, indicating the better effect of semi-aerobic landfill in reducing CH4 production. The CH4 and O2 concentrations in the two landfill installations all presented obvious spatial layer effect, i.e., under-layer > middle layer > upper layer for CH4, while upper layer > middle level > under-layer for O2.

[Histopathologic and ultrastructure changes of corneal dystrophy of bowman layer, type I].

PURPOSE: To explore the histopathologic and ultrastructure changes of corneal dystrophies of bowman layer, type I (CDB I ) in Chinese. METHODS: Cornea buttons were obtained from 4 CDB I patients in 2 pedigrees who underwent lamella or penetrating keratoplasty. Sections with HE and special staining were observed under light microscope. Ultrathin sections were performed and ultrastructure changes were investigated under transmission electron microscope. Two normal cornea specimens were used as control. RESULTS: In the CDB I cornea epithelium, microvillus were decreased or lost. Small clumps of deposits with high electron density were found between basal cells. Some basal cells appeared apoptosis sign. A great quantity of rod-shape deposits were seen in Bowman's layer and the anterior part of stroma. CONCLUSIONS: CDB I patients have characteristic corneal structure changes, and may be one of the reasons of the clinical symptoms.
