[Efficacy of intra-articular injection of sodium hyaluronate in post-operation treatment of the knee].

OBJECTIVE: To investigate the effects of intra-articular injection of sodium hyaluronate in post-operation treatment of the knee. METHODS: From January 1998 to February 2001, 4 ml of sodium hyaluronate injection was injected into the knee joint of the 134 cases at the end of arthroscope operation, or the 91 cases undergoing open operation of the knee at the time when the drain tube was removed (treatment group). Five days after operation, the hydrarthrosis was removed and 2 ml of sodium hyaluronate was injected into the knee joint. According to the patient's condition, injection of sodium hyaluronate was performed once a week for several weeks. Clinical evaluation was made by evaluating pain visual analog scale (VAS) and painless range of movement (ROM) of the joint at every definite point of time. The 85 patients in control group used nothing at the same time. RESULTS: The VAS score of patients in the treatment group was significant lower than that of the control group. The period to the maximal painless ROM of the joint was 6 days in the treatment group after open operation, while 9 days in the control group. CONCLUSION: Sodium hyaluronate appears effective in relieving post-operation pain of the knee joint.

[The clinical study of adhesion prevention of sodium hyaluronate in flexor tendon surgery].

OBJECTIVE: To evaluate the effects of sodium hyaluronate on adhesion prevention after flexor tendon surgery. METHODS: In 47 cases with the flexor tendon surgery, two kinds of sodium hyaluronate jelly preparations were injected into the tendon sheath before suture. Preparation I (20 mg/2 ml) was for group A (17 patients) and preparation II (20 mg/2 ml) was for group B (16 patients). The control group (group C, 14 patients) were treated in the same way except injection of sodium hyaluronate. The functions of afflicted fingers including flexibility, pain and swelling were measured immediately, at the first, second and the third month after operation. RESULTS: All 47 patients were followed up 1 to 3 months. 64.71% patients in group A and 68.75% in group B showed significant improvement. There were significant difference compared with group C (P < 0.05). There were no significant adverse reactions were observed in all groups. CONCLUSION: Two sodium hyaluronate preparations have effects in adhesion prevention after flexor tendon surgery with safety and expedience.

[The effect of bradykinin B(2) receptor antagonist on cough reactivity in a sensitized guinea pig model].

OBJECTIVE: To investigate the effect of bradykinin B(2) receptor antagonist FR173657 on cough response in guinea pigs sensitized and challenged with ovalbumin. METHODS: 40 normal and 40 sensitized guinea pigs were challenged with the aerosol of ovalbumin. 24 hours later, 10 animals from the normal group and 10 from the sensitized group were intraperitoneally injected either with saline or with 0.03 mg/kg, 0.3 mg/kg and 3 mg/kg of FR173657 respectively, and then cough response to inhaled capsaicin was measured. Specific airway resistance was recorded with a noninvasive technique only in normal, sensitized and FR173657 (0.3 mg/kg)-treated sensitized guinea pigs. RESULTS: FR173657 did not influence cough response and airway resistance in normal guinea pigs. Compared with normal animals, sensitized guinea pigs presented an increased cough frequency and specific airway resistance [(21.7 +/- 3.0) times/3 min vs (8.3 +/- 1.4) times/3 min, (9.4 +/- 0.5) cm H(2)O/s vs (7.9 +/- 0.9) cm H(2)O/s], (P < 0.05) after inhalation of 10(-4) mol/L capsaicin. The cough frequency and specific airway resistance were decreased to [(12.2 +/- 1.3) times/1 min and (7.5 +/- 0.9) cm H(2)O/s] after administration of 0.3 mg/kg of FR173657 (P < 0.05). CONCLUSIONS: Bradykinin B(2) receptor antagonist inhibited increased cough response and airway resistance in guinea pigs sensitized and challenged with ovalbumin. Bradykinin may be an important mediator in cough associated with eosinophilic airway inflammation.

[Effects of microfilaments on the expression of collagenases and tissue inhibitor of metalloproteinases-1 in fibroblasts].

OBJECTIVE: To investigate the roles of microfilaments in the expression of collagenase and TIMP-1 in normal and hypertrophic scar. METHODS: Cell culture and Northern blot hybridization methods were used in this study. RESULTS: After the disruption of microfilaments with cytochalasin B, mRNA contents of collagenase and TIMP-1 both increased significantly in normal and hypertrophic scar fibroblasts. CONCLUSION: The result suggest that the microfilament cytoskeleton may regulate the synthesis and degradation of ECM, which may be a mechanism of gene regulation during wound healing.

Evidence that IL-13R alpha2 chain in human glioma cells is responsible for the antitumor activity mediated by receptor-directed cytotoxin therapy.

The interleukin-13 receptor alpha2 (IL-13R alpha2) chain is a primary IL-13 binding and internalization component of the IL-13R system. Previous studies have shown that human brain tumors, including glioblastoma multiforme (GBM), overexpress IL-13R alpha2 chain, while normal brain cells do not express this protein or express very low levels of it. To target IL-13R on brain tumor cells, the authors have developed an IL-13R-directed cytotoxin termed IL13-PE38QQR to induce specific cancer cell killing. To investigate the role of IL-13R alpha2 chain in GBM, cells were treated with antisense oligonucleotide or siRNA to IL-13R alpha2 chain, and cellular IL-13 binding and sensitivity to IL-13 cytotoxin were assessed. IL-13R alpha2 gene interference in GBM cells showed decreased ligand binding, and consequently IL-13 cytotoxin exhibited less cytotoxicity to these cells. The authors next evaluated the antitumor activity of IL-13 cytotoxin in native IL-13R-expressing tumors and after gene transfer of IL-13R alpha2 by injecting plasmid in U87MG tumors subcutaneously implanted in nude mice. These mice were then treated with IL-13 cytotoxin. Mean tumor size in mice receiving intraperitoneal or intratumoral IL-13 cytotoxin was significantly smaller in control tumors; however, tumor sizes were much smaller in IL-13R alpha2-transfected tumors. Furthermore, convection-enhanced delivery of IL-13R alpha2 cDNA in intracranially established U87MG glioma followed by IL-13 cytotoxin administration by the same route mediated tumor regression and prolonged survival of animals by 164% compared with control. These results indicate that IL-13R alpha2 chain in GBM cells is essential for IL-13 cytotoxin-induced cytotoxicity and that IL-13R alpha2 chain plays a critical biologic role in IL-13 cytotoxin-mediated therapy for GBM.
