Structural phase transitions of HfV2 at low temperatures.

We report a high-resolution synchrotron X-ray powder diffraction study on HfV(2), hafnium divanadium, at low temperatures. In this work we show, for the first time, a complete sequence of structural phase transitions of HfV(2) from cubic (Fd3m) to tetragonal (I4(1)/amd) to orthorhombic (Imma) in succession as temperature decreases. Peak splitting and extra diffraction peaks owing to lattice distortion can be clearly distinguished for the low-symmetry phases. The atomic positions and lattice parameters were obtained by Rietveld refinement. The bond lengths and angles of the HfV(2) crystal structure at the low-symmetry phases were correctly determined from the structure refinement. The face-centered cubic (Fd3m) unit cell (Z = 24) transforms to a body-centered tetragonal (I4(1)/amd) phase with a 45 degrees rotation relative to the cubic cell and with a reduced number of atoms (Z = 12) in the unit cell at a temperature of T = 112 K. The orthorhombic phase occurs at T = 102 K and it keeps the body-centered symmetry (Imma) and Z = 12 in the unit cell. The refinement results indicate that there may be a small amount of untransformed cubic phase left over in the lower symmetry phases. The abnormal thermal contraction of both tetragonal phase and orthorhombic phase marks the significance of structural change in HfV(2).

Optical imaging as an adjunct to sonograph in differentiating benign from malignant breast lesions.

The role of near infrared (NIR) diffusive light imaging as an adjunct to ultrasound in differentiating benign from malignant lesions was evaluated in 27 mammography patients with infiltrating ductal carcinomas, apocrine metaplasia, fibroadenomas, radial scar and ductal hyperplasia, cysts, and normal tissues. Conventional ultrasound/mammography images were graded based on BI-RADS assessment categories. The spatial NIR measurements were made at wavelengths of 750 and 830 nm. Functional images, such as relative changes of deoxyhemoglobin (deoxyHb) and total blood concentration, were estimated from the dual wavelength measurements. Maximum relative deoxyHb and blood concentration changes were measured, and spatial correlation of masses in relative deoxyHb and blood concentration images for each breast were calculated. For the five biopsy proven benign lesions, ultrasound/mammography diagnoses were suspicious for malignancy (four cases) and highly suspicious for malignancy (one case). Four lesions showed less than 1.0 V maximum deoxyHb and less than 1.5 V maximum blood concentration levels on average and spatial image correlation showed no correlated masses in both deoxyHb and blood concentration images. For the four biopsy proven malignant lesions, ultrasound/mammography diagnoses were highly suspicious for malignancy. Maximum deoxyHb and blood concentration changes were greater than 2.9 V on average except one lesion which showed smaller deoxyHb signal (maximum 0.85 V) but the deoxyHb mass and blood concentration mass were highly correlated.

The sigma(70) transcription factor TyrR has zinc-stimulated phosphatase activity that is inhibited by ATP and tyrosine.

The TyrR protein of Escherichia coli (513 amino acid residues) is the chief transcriptional regulator of a group of genes that are essential for aromatic amino acid biosynthesis and transport. The TyrR protein can function either as a repressor or as an activator. The central region of the TyrR protein (residues 207 to 425) is similar to corresponding polypeptide segments of the NtrC protein superfamily. Like the NtrC protein, TyrR has intrinsic ATPase activity. Here, we report that TyrR possesses phosphatase activity. This activity is subject to inhibition by L-tyrosine and its analogues and by ATP and ATP analogues. Zinc ion (2 mM) stimulated the phosphatase activity of the TyrR protein by a factor of 57. The phosphatase-active site of TyrR was localized to a 31-kDa domain (residues 191 to 467) of the protein. However, mutational alteration of distant amino acid residues at both the N terminus and the C terminus of TyrR altered the phosphatase activity. Haemophilus influenzae TyrR (318 amino acid residues), a protein with a high degree of sequence similarity to the C terminus of the E. coli TyrR protein, exhibited a phosphatase activity similar to that of E. coli TyrR.

CDP is a repressor of mouse mammary tumor virus expression in the mammary gland.

Mouse mammary tumor virus (MMTV) transcription is highest in the lactating mammary gland but is detectable in a variety of other tissues. Previous results have shown that MMTV expression is suppressed in lymphoid and other tissues through the binding of the homeodomain-containing repressor special AT-rich binding protein 1 to a negative regulatory element (NRE) in the MMTV long terminal repeat (LTR). Another homeoprotein repressor, CCAAT displacement protein (CDP), also binds to the MMTV NRE, but a role for CDP in MMTV transcriptional suppression has not yet been demonstrated. In this paper, we show that the level of CDP decreases during development of the mammary gland and that this decline in CDP level correlates with the known increase in MMTV expression observed during mammary gland differentiation. Moreover, CDP overexpression was able to suppress MMTV LTR-reporter gene activity up to 20-fold in transient-transfection assays of mouse mammary cells. To determine if this effect was due to direct binding of CDP to the promoter-proximal NRE, we performed DNase I protection assays to map two CDP-binding sites from +835 to +845 and +920 to +931 relative to the first base of the LTR. Mutations engineered into each of these sites decreased CDP binding to the proximal NRE, whereas a combination of these mutations further reduced binding. Subsequently, each of these mutations was introduced into the full-length MMTV LTR upstream of the luciferase reporter gene. Analysis of stable transfectants of LTR constructs showed that CDP binding site mutations in the proximal NRE elevated reporter gene expression two- to sixfold compared to wild-type LTR constructs. Thus, MMTV expression increases during mammary gland development, in part due to decreased CDP levels and CDP binding to the LTR. Together, these experiments provide the first evidence that CDP acts as a repressor of MMTV transcription in the mammary gland.

The orphan nuclear receptor Ear-2 is a negative coregulator for thyroid hormone nuclear receptor function.

Thyroid hormone (T3) nuclear receptors (TR) are ligand-dependent transcription factors which regulate growth, differentiation, and development. One emerging hypothesis suggests that TR mediate these diverse effects via a large network of coregulators. Recently, we found that TR-mediated transcriptional responses varied in six cell lines derived from different tissues. We therefore used human TR subtype beta1 (TRbeta1) as bait to search for coregulators in human colon carcinoma RKO cells with a yeast two-hybrid system. RKO cells exhibited T3-dependent and -independent transcriptional activation. One of the three positive clones was identified as Ear-2, which is a distant member of the chick ovalbumin upstream promoter-transcription factors of the orphan nuclear receptor family. The physical interaction between Ear-2 and TRbeta1 was further confirmed by specific binding of Ear-2 to glutathione S-transferase-TRbeta1. In addition, Ear-2 was found to associate with TRbeta1 in cells. As a result of this physical interaction, binding of TRbeta1 to the T3 response elements was inhibited. Using reporter systems, we found that both the basal activation and the T3-dependent activation mediated by TRbeta1 were repressed by Ear-2 in CV1 cells. In RKO cells, however, the T3-independent transcriptional activity was more sensitive to the repression effect of Ear-2 than the T3-dependent transcriptional activity. The repression effect of Ear-2 was reversed by steroid hormone receptor coactivator 1. These results suggest that TR-mediated responses reflect a balance of corepressors and coactivators in cells. These findings further strengthen the hypothesis that the diverse activities of TR are achieved via a large network of coregulators that includes Ear-2.

Modulation of transcriptional activity of p53 by ultraviolet radiation: linkage between p53 pathway and DNA repair through damage recognition.

The increase in the p53 activity in response to DNA damage is thought to be one of the important mechanisms by which p53 contributes to transcriptional activation of p21(wafl), mdm2, and other downstream regulatory genes. To investigate the p53 response to ultraviolet (UV) type of DNA damage, p53 protein level, its transcriptional activity and in vivo ubiquitination were compared in repair-proficient normal human fibroblasts (NHFs) and repair-deficient xeroderma pigmentosum (XP) group A and group C (XP-C) fibroblasts subsequent to irradiation with UV light. Accumulation of p53 protein level was observed with increasing UV doses in all the cell lines; however, discordance between p53 and p21(waf1) and mdm2 levels was observed in NHF and XP-A cells. Induction of p21(waf1) and mdm2 was inhibited by UV irradiation, requiring higher doses in NHF and lower doses in XP-A cells. However, inhibition of p21(waf1) and mdm2 induction was not observed in XP-C cells. Ubiquitin-p53 conjugates could be detected in irradiated or unirradiated NHF and XP-A cells but not in XP-C cells irradiated with 30 and 50 J/m(2) UV light. Using a p53 reporter assay, p53 transcriptional activities were found to be induced by 10 J/m(2) UV exposure and dramatically inhibited with increasing UV doses in NHF cells. Compared with repair-proficient NHF cells, UV inhibition of p53 transcriptional activity was relatively more sensitive in XP-A cells but resistant in XP-C cells. These results indicate that DNA damage by UV, in addition to inducing p53, acts as a trigger for inhibition of p53 transcriptional activity. Overall, recognition of DNA damage links both p53 induction and p53 degradation to DNA repair mechanisms.

Ethanol exposure enhances apoptosis within the testes.

BACKGROUND: Chronic ethanol abuse causes testicular atrophy and male infertility in alcoholic men. It is well known that ethanol exposure disrupts the hypothalamic-pituitary-gonadal axis, adversely affects the secretory function of Sertoli cells, and produces oxidative stress within the testes. It is still not clear what cellular mechanisms are responsible for the morphologic alteration of the testes that results in a reduction of testicular mass as a consequence of ethanol exposure. The hypothesis tested was that ethanol enhances apoptosis of testicular germ cells. METHODS: In the experiments of chronic ethanol exposure, male Sprague Dawley rats (Harlan Sprague Dawley, Inc., Indianapolis, IN) were fed Liber-Decarlie liquid diet for 9 weeks. In the experiments of acute ethanol exposure, a small volume of 20% ethanol solution was administered by intratesticular injection. Both 3'-end labeling of isolated testicular deoxyribonucleic acid (DNA) and labeling of apoptotic cells in situ by the terminal deoxynucleotidyl transferase-mediated deoxyuridine 5'-triphosphate nick end-labeling method were used to determine apoptosis rates within the testes. The expression of proteins involved in apoptosis was assessed by reverse transcription-polymerase chain reaction and by Western blotting. RESULTS: The testes of rats that were fed an ethanol-containing liquid diet had more testicular DNA fragmentation than did those of animals that were fed an isocaloric control diet. Ethanol increased the number of apoptotic spermatogonia as well as spermatocytes. Direct intratesticular injections of ethanol solution enhanced testicular DNA fragmentation, suggesting an increase in apoptosis. Moreover, Fas ligand levels were increased within the testes of rats that were chronically fed ethanol. In vitro, ethanol treatment of cultured Sertoli cells enhanced the production of Fas ligand. In addition, testicular levels of p53 messenger ribonucleic acid were increased in rats that were chronically fed ethanol. CONCLUSIONS: All of these observations suggest that ethanol enhances testicular germ cell apoptosis.

In situ hybridization analysis of transforming growth factor-beta 1 RNA expression during mouse tooth development.

OBJECTIVE: The aim of this study is to investigate the temporal and spatial expression and localization of transforming growth factor-beta 1 mRNA during mouse tooth development. METHODS: The distribution pattern of TGF-beta 1 mRNA during tooth development was analyzed on 5 microns serial sections of paraformaldehyde-fixed and paraffin-embedded embryonic mouse heads or mandibles. RESULTS: The results show that TGF-beta 1 mRNA is expressed during mouse tooth development in a temporally and spatially regulated fashion. Local expression of TGF-beta 1 mRNA in the dental epithelium at budstaged (E13) and capstaged (E15) teeth were observed. During bell stage (E16-18), TGF beta 1 mRNA was very abundant in the ameloblast layer and dental papilla cells. The expression of TGF-beta 1 increased in the layer of odontoblast and ameloblast with the differentiation of these cells. CONCLUSION: These results suggest that TGF-beta 1 may have an important role in odontogenesis and that it acts as a paracrine and autoinducing factor.

Immunohistochemistry of bone sialoprotein and osteopontin during reparative dentinogenesis in vivo.

OBJECTIVE: To investigate the stage-specific and tissue expression of bone sialoprotein (BSP) and osteopontin (OPN) during reparative dentinogenesis in vivo. METHODS: Direct pulp-capping with Ca(OH)2 developed a model used for the investigation of reparative dentinogenesis in the exposed dental pulp. In this model, standardized class V cavities were prepared close to the gingival margin on the buccal surface of each tooth. Animals were sacrificed 3, 7, and 10 days post-operation. Immunohistochemical staining determined the tissue-specific expression of BSP and OPN during the process. RESULTS: Odontoblast-like cells and reparative dentin reacted positively with BSP and OPN. The expression of BSP reached a peak at day 7, then gradually decreased, while the expression of OPN showed no significant difference between day 7 and day 10. CONCLUSION: BSP and OPN may play different roles in reparative dentinogenesis. BSP may serve as a nucleator of hydroxyapatite crystal formation.

Immunolocalization of heat shock protein 70 during reparative dentinogenesis.

OBJECTIVE: To investigate the immunolocalization of heat shock protein 70 (hsp 70) during reparative formation and to discuss the role of heat shock response in dental pulp injury and repair. METHODS: A single cavity was prepared in the mesial surface of the first molars of both maxilla and mandible in Wistar rat. The animals were sacrificed at 3, 15, and 30 days post-operation. After the histological process, the paraffin sections were reacted with monoclonal antibodies against rat hsp 70 using the strept-avidin-biotin-peroxidase complex method. RESULTS: Immunolocalization demonstrated heavy staining for hsp 70 in normal pulp and at different stages of dental pulp repair. In normal pulp, immunoreactivity was visualized in the odontoblasts and the pulp fibroblast. In the group sacrificed at 3 days, heavy staining was located in the odontoblast process and cytoplasm. After 15 days, the newly formed odontoblast-like cells were strongly stained. At 30 days, the same staining intensity was observed in odontoblast-like cells and in pulp cells. No staining was seen in reparative dentin. CONCLUSION: These results demonstrated that heat shock protein 70 might play an important role as a molecular chaperone during reparative dentin formation.

[Recombinant human IGF-1 prevents type 1 diabetes in female non-obese diabetic mice].

OBJECTIVES: To study whether recombinant human (rhIGF-1) can prevent the onset of type 1 diabetes in female non-obese diabetic (NOD) mice, and its influence on their immune system. METHODS: Ten mice were injected intraperitonally (i.p.) rhIGF-1 37.5 micrograms.kg-1.d-1 at 4 weeks old for 2 weeks at treated group; and the same volume of normal solution (NS) was used in 10 mice as control group. All mice were killed at 40 weeks old or 2 weeks after the diagnosis of diabetes. Blood glucose of tail vein blood was measured weekly using a glucometer. At the end of the experiment, serum C-peptide levels and antibodies to glutamic acid decarboxylase (GAD) were detected with RIA and ELISA methods, respectively. T cell subsets of spleen were undertaken two-color FACS analysis using Cy-chrome anti-CD4 and FITC-anti-CD8 monoclonal antibodies; pancreatic histopathology and immunohistochemistry of T cell were conducted. RESULTS: Treatment with rhIGF-1 at 4 weeks old reduced the total incidence of diabetes (40% vs 90%, P < 0.05) and delay its onset [(26.25 +/- 3.68) weeks vs (21.6 +/- 6.7) weeks] compared with control group (P < 0.05). RhIGF-1 showed the growth effect on NOD mice, because the wet weight of spleen and pancreas of groups treated with rhIGF-1 were higher than that of control mice (P < 0.01, respectively). Only 3.24 (0.92 islets/mouse) was observed in the control group at death, but they increased to 13.2 (3.14 islets/mouse) in the rhIGF-1 group. The insulitis score decreased from 0.738 +/- 0.194 to 0.098 +/- 0.035 in the later group. A marked reduction of insulitis severity of pancreatic glands was also observed. A strong proliferative response of CD8+ T subsets in splenocytes occurred in rhIGF-1 treated mice, and the imbalance of CD4+/CD8+ subgroup in control mice (2.03 +/- 0.72) was corrected in the later group (0.53 +/- 0.22, respectively, P < 0.01). The lymphocytes infiltrating islets in the rhIGF-1 group showed a reduction of CD4+ cells, and they existed only outside of islets. CONCLUSIONS: These data demonstrated the preventive effects of rhtIGF-1 on the onset of type 1 diabetes and the severity of insulitis in female NOD mice. The mechanisms of these effects may be related to the induction of a CD8+ Ts cell response that inhibits the spontaneous development of autoreactive CD4+ T subsets response, or inhibiting the homing of CD4+ T cells to islets.

[A primary study on chemical bound forms of copper and zinc in wheat and rape].

Sequential extraction method was used to analyze and distinguish various chemical bound forms of copper and zinc in rape and wheat. The results show that in these two crops, copper was mainly in the form of wate soluble and ethanol soluble, which can be easily transferred in crops. The total content of various chemical bound forms of copper was higher in aboveground part than in underground part, and their content was decreased in the order of water soluble form (W.S. form) > residual form (Re. form) > ethanol soluble form(Eth. S. form) > acid soluble form (A.S. form). Zinc was mainly in the form of acid soluble, which is hard to be transferred in crops. The total content of various chemical bound forms of zinc was lower in aboveground part than in roots, and their content was decreased in the order of A.S. form > Re. form > W.S. form > Eth. S. form. In comparing with copper, a large amount of zinc was enriched in seeds and pods of rape.

[Chemotherapy of non-small-cell lung cancer (NSCLC) and changes in serum sAPO-1/Fas and nitric oxide (NO) levels].

OBJECTIVE: To investigate the therapeutic efficacy, toxic side effects, and changes in serum level of sAPO-1/Fas and nitric oxide(NO) in NSCLC patients treated with VM-26, epirubicin (EPI) and cisplatin (VED regimen). METHODS: Forty-two cases of advanced NSCLC patients (squamous-cell carcinoma, 28; adenocarcinoma, 12; undifferentiated cancer, 2) were treated with 2 cycles of VED. Serum sAPO-1/Fas level was determined by ELISA method, and NO level by enzymatic method before and after treatment. RESULTS: The overall response rate was 47.6% (20/42). Serum sAPO-1/Fas level in patients with squamous-cell carcinoma who responded to treatment was significantly lower than that before treatment. The post-chemotherapy serum NO level was significantly higher than that before chemotherapy in responsive patients with squamous-cell cancer as well as in adenocarcinoma. CONCLUSION: Combination chemotherapy with VM-26, EPI and DDP can be useful in the treatment of patients with advanced NSCLC. Serum sAPO-1/Fas and NO levels could be used to monitor treatment effect.

[The relationship between sensitivity to arsenic trioxide and antioxidative capacity of malignant hematopoietic cells].

OBJECTIVE: To detect the relationship between sensitivity of malignant hematopoietic cells to arsenic trioxide (As2O3) and cellular capacity against oxidation. METHODS: Nine cell lines derived from hematopoietic malignancies were treated with As2O3 in vitro. Apoptosis was assessed by cellular viability, cytomorphology and flow cytometry. The As2O3-treated cells were examined for glutathione (GSH) level and activity of catalase (CAT), superoxide dismutase (SOD), glutathione S-transferase (GST) and glutathione peroxidase (GPX). RESULTS: Of the 9 cell lines examined, the acute promyelocytic leukemia cell line NB4 and its retinoic acid-resistant subclones R2 and MR2, the Burkitt lymphoma cell line Namalwa and multiple myeloma cell line RPMI 8226 were sensitive to As2O3-induced apoptosis while the acute T cell leukemia cell line (Jurkat) and 3 cell lines (HL-60, U937, K562) of myelogenous leukemia origin were not. In comparison with the sensitive cell lines, higher activity of CAT was found in HL-60 and U937, higher level of GSH in Jurkat, and both in K562. However, activity of GPX, GST and SOD did not significantly correlate with their resistance to As2O3-induced apoptosis. CONCLUSION: Intracellular GSH level and/or catalase activity are important factors to determine sensitivity of malignant hematopoietic cells to As2O3-induced apoptosis.

[Clinical diagnosis of apical hypertrophic cardiomyopathy].

OBJECTIVE: To evaluate the clinical features of apical hypertrophic cardiomyopathy (AHCM) and the value of ECG for a reliable clinical diagnosis as compared with two-dimensional echocardiography. METHODS: 29 cases with AHCM were studied by with clinical features, ECG, echocardiography, myocardial scanning, coronary angiography and left ventriculography (LVG). RESULTS: The major clinical feature of AHCM was myocardial ischemia; ECG showed increased R amplitude and inverted T in chest leads. The increase of R amplitude was in such a pattern that RV(4)>or=RV(5) > RV(3). Thickening of left ventricular apical wall was found with echocardiography and radionuclide myocardial scanning. Exercise electrocardiogram showed myocardial ischemia in 20 cases. LVG and coronary angiography demonstrated apical hypertrophic cardiomyopathy with normal coronary arteries in 16 cases. CONCLUSION: The characteristic inverted T waves and increased R wave amplitude with RV(4)>or=RV(5 > RV3) in the ECG are the important signs for clinical diagnosis of AHCM.

[Normal anatomy and related pathological changes of shoulder on MRI].

OBJECTIVE: To describe the normal anatomy and common abnormal changes of rotator cuff impingement and tears and recurrent anterior instability of shoulder joint in MRI pictures. METHODS: MRI was compared in 285 patients with shoulder diseases and 20 patients with symptomatic shoulder diseases. RESULTS: On oblique coronal image, the supraspinatus presented moderate signal intensity and low signal intensity in its tendon-muscle conjunction ranging from the humeral head to the greater tuberosity. The MRI manifestations of impingement lesion of the rotator cuff were as follows: high signal intensity of tendons, changes of their shapes, retraction of tendon-muscle conjunction, and muscle atrophy with high signal intensity. On T1-weighted axial image, the anterior and posterior glenohumeral labrum, the long head biceps tendon were displayed in low signal intensity. The anterior labrum manifested a sharp triangle contour and the posterior labrum a round one. The whole four muscles of the rotator cuff manifested on oblique sagital image. However, it was of less value in detecting the abnormalities of the rotator cuff and the glenohumeral labrum on sagittal imaging. The sensitivity in demonstrating rotator cuff complete tear was 95% for MRI and 91% for arthrography; the specificity was 88% for MRI and 100% for arthrography. The sensitivity and specificity of MRI were 96% and 75% for detecting glenoid labrum abnormalities, and 78% and 88% for detecting labrum tear in anterior recurrent dislocation of the shoulder. CONCLUSIONS: Magnetic resonance imaging with its excellent contrast resolution in multiple anatomic planes allows noninvasive visualization of bone and soft tissues in the rotator cuff and labrum.

[Molecular cytogenetics of bone tumors].

OBJECTIVE: To study alterations of molecular cytogenetics of bone tumor serially. METHODS: Chromosomal analysis, immunohistochemistry, molecular hybridization, nested-PCR/SSCP, DNA sequence analysis, and gene transfection techniques were used to investigate the common bone tumors. RESULTS: Chromosomal aberrations were found in 50 cases of aggressive bone tumors, with a chromosome number aberration rate of 86.96% and a structure aberration rate of 93.94%. However, there were differences among different kinds of bone tumors. The incidence of chromosomal instability in 53 patients with bone tumors was higher than that in normal controls. Meanwhile, the fragile sites of chromosomes of the patients were closely correlated with the high nonrandom chromosome breakpoints of tumor cells and related oncogene loci. The data on p53 and Rb from 305 cases and 89 cases, respectively, indicated that abnormalities of p53 and Rb were common events in bone tumors. The accumulation of p53 protein and loss of Rb protein were more common in aggressive bone tumors. Structures and their products of related proto-oncogenes (ras, myc, fos, bcl-2, met) were studied in 124 cases. Bone tumors presented multi-oncogenes abnormalities with differences in different pathological kinds. Exogenous wild-type p53 gene transfected into osteosarcoma cell line induced growth inhibition and apoptosis of tumor cells. CONCLUSIONS: Abnormalities of chromosomes, proto-oncogenes, and tumor suppressor genes frequently exist in bone tumor cells and are correlated with oncogenesis.
