[Preparation of McAb against lung cancer and its application in tumor radioimmunoimaging].

OBJECTIVE: In order to tracing the lung cancer, A monoclonal antibody was produced. METHOD: The McAb, named SM-1, was specifically reacted with lung cancer confirmed by the immunopathological studies, then the McAb was digested into fragments by ficin and bromelain proteases. RESULTS: Both of the enzymatic methods were very effective and the immuno-reactivity of fragments was retained well. In our localization experiments, the T/NT ratio revealed that radioiodinated McAb and fragments could be specifically uptaked by the tumour tissue and the T/NT ratio was higher in fragments group than intact IgG. After injection of 131I-IgG and fragments, the xenografts of human lung cancer bearing in nude mice were visualized distinctly during 36-48 hrs. Eighteen patients with lung cancer received Tc-99m labeled McAb intravenously, 13 cases showed radioactive accumulation in the foci of tumours. CONCLUSION: These preliminary results suggested that SM-1 and its fragments might be a potential agents for radioimmunoimaging diagnosis of lung cancer.

[Relationship of primary pulmonary carcinoma with P16 protein expression].

This study sought to address the relationship of primary pulmonary carcinoma (PPC) with P16 protein expression. The expression of P16 protein in 65 cases of PPC was examined with immunohistochemical staining. The rate of loss of P16 protein expression in PPC was 36.92% +/- 18.45%. The rate of loss of P16 protein expression in adenocarcinoma (AC) was 28.47 +/- 16.33%, that in squamous cell carcinoma (SCC) was 35.95% +/- 17.36%, and that in adenosquamous cell carcinoma (ASCC) 57.88% +/- 10.18%. There significant differences were in the rate of loss of P16 protein expression between those aged < 60 with SCC and those aged > 60 with SCC, between low differentiated PPC and intermediate differentiated PPC, between AC and ASCC, and between SCC and ASCC (P < 0.05). There were no significant differences in the rate of loss of P16 protein between males and females with PCC, between left and right PPC, and between PPC and metastatic pulmonary carcinoma of lymph node (P > 0.05). These suggest that P16 protein expression may be one of the important standards in estimating the prognosis of patients with PCC.

[Analysis of malate dehydrogenase isozymes in Taihe negro fowl by thin-layer isoelectric focusing polyacrylamide gel electrophoresis].

The isozyme patterns of malate dehydrogenase (MDH) from six tissues (liver, kidney, eye, muscle, brain and heart) of the Taihe negro fowl (Gallus gallus domesticus Brission) were analysed by thin-layer isoelectric focusing polyacrylamide gel electrophoresis. It was found that there were twelve MDH isozyme bands in the tissues of the fowl, and these isozymes displayed a tissue-specific distribution.

[Identification of a pure culture of nodular endophyte from Coriaria nepalensis].

A Frankia-like strain, Cs146, was isolated from nodules of Coriaria nepalensis in southwest China. The isolate is an actinomycete with vesicles and sporangia borne on the hyphae, and shown to have an ability to nodulate the host plant. No aerial hyphae is formed, but the substrate hyphae are usually orange and produce soluble yellow pigment. Cs146 could grow quite slow at 28-30 degrees C on both liquid and solid media. However, the isolate is greatly different from other frankiae in physiology (intermediate physiologocal group between A and B trains), cell wall composition of characteristical amino acids (meso-DAP plus glycine) and whole cell sugar patterns (xylose plus little amount of galactose and arabinose). Cs146 thus appears to be a member of a distinct Frankia lineage.

[Studies on nimodipine sustained-release tablet capable of floating on gastric fluid with prolonged gastric resident time].

Nimodipine was incorporated into poloxamer 188 solid dispersion before formulation, then mixed excipient and solid dispersion were directly compressed into nimodipine floating sustained-release tablet(NM-FSRT). Formulations were optimized using uniform design and variables affecting nimodipine release from matrix were studied. Preliminary in vivo evaluation was carried out in healthy volunteers. Results showed that the optimized formulation could remain floating on gastric fluid for over 10 hours. In vitro release(0.15-6 h) conformed to zero-order kinetics. Hydroxypropylmethylcellulose(HPMC) and polyethylene glycol 6000(PEG 6000) showed the biggest effect on in vitro drug release. Increasing HPMC content and decreasing PEG 6000 content led to decrease of nimodipine release in vitro. NM-FSRT did remain floating on gastric fluid with prolonged gastric resident time(GRT) of 5 hours under fed condition, while GRT was only 3 hours under fasted condition. GRT of nimodipine conventional tablet(NM-CT) under fed and fasted conditions was 3 and 2 hours, respectively. Relative bioavailability of NM-FSRT was 391.46% and MRT was over twice that of NM-CT. NM-FSRT appeared to have prolonged GRT and improved bioavailability.

Effect of 27-hydroxycholesterol on cellular sphingomyelin synthesis and Ca++ content in cultured smooth muscle cells.

The relationships among sphingomyelin synthesis cytosolic free Ca++ level and cytotoxicity in the presence of 27-hydroxycholesterol were investigated by measurement of [methyl-3H] choline incorporation into sphingomyelin, phosphatidylcholine, phosphocholine and cytidine choline 5'-diphosphate, 45Ca++ uptake, cytosolic free Ca++ levels and viable cell numbers. The results indicated that 27-hydroxycholesterol at higher than normal level induced increased [3H] choline incorporation in sphingomyelin accompanied by decreased radioactivity in phosphatidylcholine but did not alter [3H] choline content in phosphocholine and cytidine choline 5'-diphosphate. The increased radioactivity in sphingomyelin by 27-hydroxycholesterol was detected first, followed by detection of the enhanced Ca++ uptake and cytosolic free Ca++. Finally, a decrease in viable cell number occurred. We suggest that the increased transfer from phosphatidylcholine to sphingomyelin by 27-hydroxycholesterol may be related to enhanced cytosolic free Ca++.

Cholesterol metabolism in human umbilical arterial endothelial cells cultured in low magnesium media.

Epidemiological and experimental studies have shown that magnesium is closely related to regulation of lipid metabolism, membrane structure and permeability, ion migration through cellular membranes, endocrine hormone and platelet function. The cause of atherosclerosis induced by magnesium deficiency has been suggested to be due to abnormal lipid metabolism, lipid peroxidation, a decrease of prostacycline produced by endothelial cells, and an increase of platelet aggregation. We found that the plasma from cardiac catheterized patients suffering from chest pains contained higher levels of oxysterols than age and sex matched patients free of chest pain. Studies with cultured arterial cells in media deficient in magnesium or containing oxysterols indicated that both magnesium and oxysterols have an important role in lipid metabolism in patients with coronary heart disease.

Esophageal and gastric cardia epithelial cell proliferation in northern Chinese subjects living in a high-incidence area.

Linxian and the nearby county Huixian, in the Henan province in Northern China, have a very high incidence of esophageal squamous cell carcinoma (SCC). Previous studies from these counties have suggested that increased proliferation of esophageal epithelial cells, morphologically manifested as basal cell hyperplasia (BCH) and dysplasia (DYS), is an early indicator of abnormality in persons predisposed to SCC. A high incidence of gastric cardia adenocarcinoma (AC) was also found in these areas. To determine proliferation patterns of esophageal and gastric cardia epithelia with normal and different severities of precancerous lesions, we measured proliferating cell nuclear antigen (PCNA), Ki-67, and bromodeoxyuridine (BrdU) incorporation and compared the results. Esophageal biopsies (175) and gastric cardia biopsies (41) were collected from symptom-free subjects in Huixian. Of these, 23 esophageal biopsies were incubated with BrdU. The avidin-biotin-peroxidase complex (ABC) method was used to detect PCNA, Ki-67, and BrdU. The number of immunostain-positive cells was counted manually. Intense immunostaining for PCNA, Ki-67, and BrdU was observed in the cell nuclei of tissues with normal and different severities of precancerous lesions. With esophageal biopsies, both PCNA and Ki-67 increased significantly as the epithelia progressed from normal to BCH and to DYS. The number of PCNA- and Ki-67-positive cells was three times higher than that of BrdU incorporation in the same category of BCH. With cardia biopsies, the number of Ki-67 positive cells was lower in normal tissue and increased significantly from chronic superficial gastritis to chronic atrophic gastritis to DYS. Staining patterns for PCNA and Ki-67 were correlated with the histopathology of the esophagus. The correlation was not as clear with gastric cardia. BrdU studies appear to be more complicated. The PCNA and Ki-67 methods may be useful for screening high-risk esophageal and gastric cardia cancer subjects, and for monitoring chemoprevention effects.

[Preparation of leucine-methyl glutamate-glutamic acid copolymers].

The method for preparing leucine-methyl glutamate-glutamic acid copolymer was studied. In the first place benzyl glutamate and methyl glutamate were synthesized respectively. Then N-carboxy anhydrides (NCA) of leucine, benzyl glutamate or methyl glutamate were prepared in a closed container by phosgene-toluene solution method. After copolymerization the copolymers were debenzylated and demethylated by anhydrous hydrogen bromide. The free carboxyl group mole content in side chains of the copolymer was controlled by various standing periods following bubbling HBr. Analysis of infrared spectrogram and ultraviolet asorbance of copolymers indicated that this procedure resulted in the loss of almost all benzyl groups and some methyl groups.

[A study on the relationship between plasma level of ET-1 and cGRP and blood pressure in hemodialysis patients].

Plasma endothelin-1 (ET-1) and calcitonin gene related peptides (cGRP) were studied by immunoradiological method in 60 hemodialysis (HD) patients. The results showed that: (1) Plasma level of ET-1 and cGRP were increased in the HD patients. (2) Plasma level of ET-1 and cGRP varied in different times of HD. (3) A significantly positive relation between plasma ET-1 and blood pressure (BP) existed in the HD patients, while a significantly negative relation between plasma cGRP and BP existed in the HD patients. The results suggest that ET-1 and cGRP are major factors which lead to BP changes in HD patients.

[Clinical valuation color Doppler echocardiography for diagnosis of coronary artery fistula].

The purpose of this study was to investigate the advantages of color Doppler echocardiography in the diagnosis of congenital coronary artery fistula. Twenty-one patients with coronary artery fistula ranging in age from 3 to 28 years underwent color Doppler echocardiographic study 1991.4-1996.8. When a coronary artery fistula was found, the origin, the course and the drainage site were carefully searched for to make a precise diagnosis. Color Doppler echocardiographic diagnosis of all patients were confirmed by cardiac operation on cardioangiography. The results indicate that color Doppler echocardiography can provide a panoramic view of coronary artery and the fistulous vessel with precise definition of the origin, the course of draingae site of the fistula. Therefore, it is the noninvasive method of first choice to diagnose congenital coronary artery fistula.

[Color Doppler echocardiography in the diagnosis for ruptured aneurysm of sinus of Valsalva].

Twenty-five cases with ruptured aneurysm of sinus of Valsava were detected by color Doppler sonography and twenty-four cases undenwent surgical repair. In twenty-three cases, the sonographic findings of the site of the aneurysm and the chambers into which aneurysm ruptured were coincident with the surgical findings. The accuracy was 95.8%. The size of the aneurysm and the diameter of the ruptured site detected by ultrasound were correlated well with surgical findings. The associated lesions obtained by ultrasound were completely identical with surgical findings. Color Doppler flowing imaging has special value in diagnosing the ruptured aneurysm of sinus of Valsalva.

Tat-SF1: cofactor for stimulation of transcriptional elongation by HIV-1 Tat.

Tat may stimulate transcriptional elongation by recruitment of a complex containing Tat-SF1 and a kinase to the human immunodeficiency virus-type 1 (HIV-1) promoter through a Tat-TAR interaction. A complementary DNA for the cellular activity, Tat-SF1, has been isolated. This factor is required for Tat trans-activation and is a substrate of an associated cellular kinase. Cotransfection with the complementary DNA for Tat-SF1 specifically modulates Tat activation. Tat-SF1 contains two RNA recognition motifs and a highly acidic carboxyl-terminal half. It is distantly related to EWS and FUS/TLS, members of a family of putative transcription factors with RNA recognition motifs that are associated with sarcomas.

Elimination of potential sites of glycosylation fails to abrogate complement regulatory function of cell surface CD59.

CD59 is a glycosylphosphatidylinositol-anchored membrane glycoprotein that serves as the principle cellular inhibitor of the C5b-9 membrane attack complex (MAC) of human complement. Approximately 50% of the total apparent mass of CD59 is attributable to glycosylation of a single Asn (Asn18). The deduced amino acid sequences of CD59 homologues identified in Old and New World primates as well as in rat reveal that the motif for N-linked glycosylation at the residue corresponding to Asn18 of human CD59 is invariably conserved, despite considerable sequence divergence elsewhere in the protein. Such conservation suggests that the post-translational modification at Asn18 has importance for either expression or normal function of CD59 at the cell surface. In this study, we specifically examined how deletion or transposition of the site of N-linked glycosylation in the CD59 polypeptide affects its MAC inhibitory function. Our data demonstrate that the inhibitory potency of CD59 is unaffected when glycosylation is transposed from Asn18 to another site in the polypeptide. Furthermore, we show that CD59 retains normal MAC regulatory function when mutated to eliminate all potential sites for N-linked glycosylation. These data suggest that the MAC inhibitory function of CD59 is entirely provided by residues exposed at the surface of the core polypeptide and that this core structure is not influenced by glycosylation at Asn18.

The effect of exercise on ob gene expression.

Expression of the ob gene is subject to nutritional as well as hormonal regulation to control fat storage. In the present study we investigated the effect of acute exercise and long-term exercise training on ob mRNA levels in rat adipose tissue. Northern blot analysis showed that a single bout of exercise significantly decreased ob mRNA levels approximately 30% immediately and 3 hr after exercise. After 4 weeks of exercise training the ob mRNA level was significantly decreased approximately 48% at 2 hr after the last training session compared to the control group. These data demonstrated that exercise has a transient effect on ob gene expression and suggest that regulation of ob gene expression may help control energy balance following exercise.
