[Clinical and experimental study on treatment of retention enema for chronic non-specific ulcerative colitis with quick-acting kuijie powder].

OBJECTIVE: To study the clinical effect and mechanism of retention enema with quick-acting Kuijie powder (QAKJP) in treating chronic non-specific ulcerative colitis (CUC). METHODS: A treatment group of 156 patients treated with QAKJP and a control group of 78 patients treated with sulfasalazine orally were established randomly and their scores of main symptoms and signs were measured and compared before and after treatment. Animal experiments were conducted at the same time. RESULTS: The total effective rate in the treatment and the control group was 98.7% and 70.5% respectively, the clinical cure rate was 78.2% and 6.4% and the recurrence rate was 5.3% and 20.0% respectively. The effect of the treatment group was markedly superior to that of the control group, P < 0.01. Experimental study showed QAKJP could rapidly alleviate the congestion and edema of intestinal mucosa, promote the healing of ulcer, inhibit spasm of colon and had significant antidiarrheal action and antagonist effect against allergic mediator histamine. CONCLUSION: Retention enema with QAKJP has good effect on CUC, low recurrence rate and no toxic and side effect.

[A clinical study of mucosa-associated lymphoid tissue lymphoma].

OBJECTIVE: To have a better understanding of the clinical characteristics of mucosa-associated lymphoid tissue (MALT) lymphoma. METHODS: The clinical data of 24 patients with MALT lymphoma were analyzed. RESULTS: The patients were divided into two groups according to the initial location of the lymphoma in the gastrointestinal (GI) tract or not. Eighteen patients (75.0%) had GI involvement and six (25.0%) had involvement of other extranodal sites including salivary gland in three, lung in two and bladder in one. Among the 24 cases, 12 were in stage I, 7 in stage II and 5 in stage III-IV at the time of presentation. Fourteen cases underwent endoscopic biopsy and the pathologic diagnoses were in accordance with those of post-operative specimen in 11 but different in 3. Surgical resection was performed alone in 21 patients, with adjuvant chemotherapy in 7 and with adjuvant radiotherapy in 1. Three patients were treated with chemotherapy only. Results of survival analysis showed that the overall survival rate was 58.8% after 1 year, 35.3% after 3 years, 29.4% after 5 years and 5.9% after 10 years. No significant difference in clinical or biologic characteristics was observed between GI and non-GI patients (P > 0.05). The clinical stage and therapeutic measure had no significant relation with the prognosis. CONCLUSION: MALT lymphoma is an indolent disease that presents in localized extranodal location without any associated adverse prognostic factor in nearly all the patients and that these patients have a high response rate to treatment and a long survival.

[The clinical significance of lung resistance-related protein gene (lrp), multidrug resistance-associated protein gene (mrp) and mdr-1/p170 expression in acute leukemia].

OBJECTIVE: To evaluate the relationship between lrp, mrp, mdr-1/p170 and multidrug resistance in acute leukemia (AL). METHODS: 85 AL patients were divided into three groups: untreated (A), complete remission (B) and relapsed/refractory (C). The expression of lrp, mrp and mdr-1 mRNA was detected with RT-PCR assay and that of p170 measured with immunocytochemistry. RESULTS: The frequency of lrp gene expression in ANLL A, B and C group was 11.11%, 9.09% and 36.36%, in ALL A, B, C group it was 0%, 20.00% and 46.67%; the frequency of mrp gene expression in ANLL A, B, C group was 44.44%, 9.09% and 59.09%, in ALL A, B, C group it was 28.57%, 20.00% and 53. 33%; the frequency of mdr-1 gene expression in ANLL A, B, C group was 0%, 4.54% and 59.09%, in ALL A, B, C group it was 0%, 0% and 33.33%; p170 expression in ANLL C group was (9.45 +/- 14.66) %, it was the highest value in the three groups; statistics showed that there was no correlationship among the expressions of lrp, mrp and mdr-1 gene, P > 0.05; patients with lrp and mdr-1 expression had a lower complete remission (CR) percentage than those who had not in the untreated group; the combination of lrp, mrp and mdr-1 gene detection turned out to be a more sensitive, specific and accurate way in evaluation of multidrug resistance (MDR). CONCLUSION: Overexpression of one or more genes of lrp, mrp and mdr-1/p170 is associated with MDR in AL. Expression of lrp, mrp and mdr-1/p170 is good indicators in clinical MDR. Two or three factors of lrp, mrp and mdr-1 are more valuable in the evaluation of MDR than any one of these three factors. The mechanisms of mrp, lrp and mdr-1 causing MDR are different, it means that the mechanisms of MDR are complicated the combination of lrp, mrp and mdr-1/p170 detection may be the best way to evaluate MDR.

[Studies on the preparation and characters of anti-AFB1 antibody from the yolk of laying hens].

AFB1O-BSA conjugates were injected into four laying hens (A, B, C and D). The yolk anti-AFB1 antibodies A,C and D produced were obviously increased after 90 days of the first injection, and reached the peak after 135 days, and started dropping after 165 days. The indirect ELISA titer of yolk antibodies A,C and D were 1:8000, 1:6000 and 1:6000 respectively. The specificity of the antibody A was good, but a cross-reaction with AFB2, AFG1, AFG2, with an exception of AFM1, was found. The sensitivities of AFB1, AFB2, AFG1 and AFG2 were 6, 25, 125 and 2495 micrograms/L respectively.

[Use of ultrasonography to evaluate carotid atherosclerosis and hemodynamics].

One hundred and one cases of carotid atherosclerosis(CAS) were diagnosed by ultrasonography. The intermediate(IMT) of common carotid endomembrane in CAS group was much thicker than that in the healthy group(P < 0.01). There was no significant difference between the two groups in the peak velocity in systolic stage(MAX), the velocity in diastolic stage(MIN), resistance index(RI) and vessel size. The atheromatous plagues were usually flat plaques, seen at the bifurcation of common carotid artery. They belong to early pathological changes. It seems that the plaques don't affect MAX, MIN and RI of the common carotid artery.

[Study on the relationship between plasma endothelin nitric oxide concentration and renal hypertension and renal function].

OBJECTIVE: To investigate the relationship between plasma endothelin(ET), nitric oxide(NO) levels and, renal hypertension and renal function. METHODS: The plasma concentration of ET-1 was detected by immunofluorescence assay. The plasma concentration of NO was detected by biochemistry assay. RESULTS: 1. In renal disease patients, plasma concentration ET-1 was markedly elevated, and plasma concentration of NO was decreased, compared with the healthy subjects(P < 0.01). 2. Plasma concentration of ET-1 was markedly increased and plasma concentration of NO was decreased in the patients with renal hypertension. 3. Plasma level of ET-1 was higher, and plasma level of NO was lower in the patients with renal function damage than that of those without renal function damage. 4. BP, BUN and Scr were positively correlated with plasma ET-1, but they were negatively correlated with plasma concentration of NO. CONCLUSION: Plasma ET-1 and NO may play an important role in pathogenesis of renal hypertension; the change of their levels may be related to the progress of these renal diseases.

[Clinical effect observation of methods of mandibular fracture treatment].

OBJECTIVE: To observe and search for an adaptative and efficient treatment methods of mandibular fracture. METHODS: 218 cases of mandibular fracture patients were treated with different methods from 1991, then the relationship between methods, and treatment efficiency was appraised objectively. RESULTS: Different ways had different advantages, and the total successful rate was 94.5%. Titanium miniplates was a promising method for mandibular fracture treatment. CONCLUSION: It's crucial to select a right method in the mandibular fracture treatment.

[Studies on microinjection of mouse spermatozoa into rabbit oocytes].

The methods of intracytoplasmic sperm injection (ICSI) and subzonal injection (SUZI) were used to study heterologous fertilization and embryonic development between the mouse and the rabbit. Results were as follows: 1. The mouse sperm nuclei decondensed and formed pronuclei following microinjection into cytoplasm and perivitelline space (PVS) of rabbit oocytes; 2. The hybrid embryos developed to the stage of 8-cell when cultured in vitro; 3. The karyotype analysis showed a normal complement of rabbit oocyte and mouse sperm chromosomes in the 4-cell hybrid embryos; 4. The ultrastructure of 4-cell hybrid embryos was similar to that of normal 4-cell rabbit embryos; 5. The fertilization rate (32.4%) and cleavage rate (22.2%) when 5-10 mouse spermatozoa were injected were higher than those of injection of a single spermatozoon into PVS of the rabbit oocyte, but the difference was not significant (P > 0.05). The fertilization rate (42.3%) and cleavage rate (30.8%) in rabbit oocytes in vitro matured for 11-12 h were higher than those in the oocytes which were in vitro matured for 24-25 h following microinjection of 1-2 mouse spermatozoa into PVS, but the difference was not significant (P > 0.05).

[Study of bacterial adhesion to prosthetic valve materials in vitro].

This article reports a study of the bacterial adhesion to prosthetic valve materials in vitro. The method for assessing the degree of bacterial adhesion to prosthetic valve materials was established primarily. The capacities of staphylococcus aureus(SA), staphylococcus epidermidis(SE), Escherichia Coli(EC) and Pseudomonas aeruginosa(PA) for adhesion to Dacron, Pyrolytic carbon and polytetrofluoroethylene (PTFE) were quantitatively determined by the plate counting and Gamma-ray counting of 125I radiolabeled bacteria in vitro. The results showed that the capacities of four types of bacteria for adhesion to Dacron, Pyrolytic carbon and PTFE coincided with the bacterial-growth curve. The capacities of four bacteria for adhering to Dacron were stronger. The adhesion of SE to Pyrolytic carbon was the strongest. The adhesion of PA Kept up a high level. The capacities of EC and PA for adhering to PTFE were the strongest. The results indicate that the capacities of different types of bacteria for adhesion to the same prosthetic valve material are different, and the capacities of one type of bacteria for adhesion to different materials are also different.

[Separation and analysis of amino acids in secretion of psylla chinesis yang et li].

We used ion-exchange resin column chromatography to separate amino acids in secretion of Psylla Chinese Yang et Li and used capillary gas chromatography to analysis n-butyl-N-trifluoracetyl derivatives of the amino acids. Qualitative analysis was done by comparing with the standard amino acids and quantitative analysis by internal standard method. We determined thirteen amino acids in the secretion.

[Surgical reconstruction of laryngeal and tracheal stenosis].

OBJECTIVE: Reconstruction of laryngotracheal stenosis. METHOD: According to the pathologic condition of the larynx and trachea, various surgical procedures were designed for the treatment of laryngotracheal stenosis. These techniques included endoscopic management, laser therapy and laryngotrachoetomy with autogenous tissue and stent. RESULT: Among 36 patients, 32 were decannulated. The success rate was 88.8%. CONCLUSION: In order to improve the curation effects of laryngjotrachal stenosis, the choice of surgical procedure must be individualized to the pathologic condition of larynx and trachea, as well as the experience of the surgeon. Learning previous successful experience is also very important for a good result.

[Glottal configuration and histological findings in vocal muscles following trauma to cricoarytenoid joint].

OBJECTIVE: The unilateral cricoarytenoid joints of the rabbits were traumatized bluntly to investigate the changes of glottal configuration and histology of the vocal muscles. METHOD: 10 New Zealand white rabbits divided into 4 groups were studied. Under direct laryngoscope, the unilateral cricoarytenoid joint of each rabbit was traumatized bluntly. The glottal configuration and histology of the true cords demonstrated atrophy of the vocal muscles. The remarkable atrophy of the vocal muscles was evaluated after 1, 3, 6 and 9 months. RESULT: In 1 and 3 months after trauma, the glottal configuration of the rabbits showed no difference from that in the period of trauma. The histology of the vocal muscles was approximately normal. The lateral drift and deformity of the fixed vocal cord and wider glottal gap were seen 6 months after trauma. The histological sections of the vocal muscles were noted 9 months after trauma. CONCLUSION: The atrophy of the vocal muscles may occur 6 months after trauma to cricoarytenoid jonint. This pathologic change may cause the lateral displacement and deformity of the vocal cord, which increases the glottal incompetence and affects voice recovery. The laryngeal electromyography may be influenced by the pathologic changes of the vocal muscles.

[Studies on polysaccharides from Holothuria leucospilota].

A powder was obtained from the dried body of Holothuria leucosilota (Brandt). According to IR and UV spectra, paper chromatography and the physical and chemical data, it is showed a sulfated mucopoly-saccharide consisted of galactosamine, glucouronic acid, fucose and sulfate with the molar ratio of 1:0.96:0.78:1.98 respectively. The specific rotation, intrinsic viscoslty and molecular weight of this polysaccharide were all presented.

Alcohol effects on the 5-HT3 ligand-gated ion channel.

1. Alcohols and volatile anesthetics have both potentiating and channel blocking effects on members of the nicotinic ACh-like subfamily of ligand-gated ion channels. 2. The alcohol and anesthetic sensitivity of the 5-HT3 receptor, a member of this receptor subfamily, has been examined extensively by several laboratories. 3. Alcohols and volatile anesthetics potentiate receptor function. 4. This potentiation is characterized by an increase in the apparent potency with which agonists activate the receptor, and an apparent stabilization and favoring of the open channel state. 5. Long-chain alcohols and trichloroethanol also inhibit channel function, perhaps through a channel blocking mechanism similar to that seen at the nACh receptor. 6. Studies of the relationship between receptor structure and alcohol and anesthetic effects are just beginning, but some clues from other members of this subfamily of ligand-gated channels may help to pinpoint areas of the receptor that are important for alcohol effects.

Change in conformation of plasma membrane phospholipid scramblase induced by occupancy of its Ca2+ binding site.

Phospholipid (PL) scramblase is a 35.1 kDa plasma membrane protein that mediates the accelerated transbilayer migration of plasma membrane PL in activated, injured, or apoptotic cells exposed to elevated intracellular Ca2+. We recently identified a conserved segment in the PL scramblase polypeptide (residues Asp273 to Asp284) that is essential for its PL-mobilizing function and was presumed to contain the Ca2+ binding site of the protein (Zhou, Q., Sims, P. J., and Wiedmer, T. (1998) Biochemistry 37, 2356-2360). Whereas the sequence of this peptide segment resembles that of known Ca2+-binding loops within EF-hand containing proteins, it is unusual in being a single such loop in the entire protein and in being closely spaced to the predicted transmembrane helix (Ala291-Gly309). To gain insight into how Ca2+ activates the PL-mobilizing function of PL scramblase, we analyzed conformational changes associated with occupancy of this putative Ca2+ binding site. In addition to activation by Ca2+, the PL-mobilizing function of PL scramblase was found to be activated by other ions, with apparent affinities Tb3+, La3+ >> Ca2+ > Mn2+ > Zn2+ > Sr2+ >> Ba2+, Mg2+. Evidence for coordinate binding of metal ion by the polypeptide was provided by resonance energy transfer from protein Trp to Tb3+, which was competed by excess Ca2+. Metal binding to PL scramblase was accompanied by increased right-angle light scattering and by a prominent change in circular dichroism, suggesting that coordinate binding of the metal ion induces a conformational change that includes self-aggregation of the polypeptide. Consistent with this interpretation, addition of Ca2+ was found to protect PL scramblase from proteolysis by trypsin both in detergent solution as well as in situ, within the erythrocyte membrane. Mutation in the segment Asp273-Asp284 reduced Tb3+ incorporation and attenuated the change in CD spectrum induced by bound metal ligand, confirming that this suspected EF-hand loopike segment of the polypeptide directly contributes to the Ca2+ binding site.
