Nitric oxide attenuates endothelin-1-induced vasoconstriction in canine stomach.

We previously observed that endothelin-1 (ET-1)-induced gastric vasoconstriction is enhanced after ischemia-reperfusion. The purpose of our present study was to examine the role of nitric oxide in regulating ET-1-induced vasoconstriction under normal conditions and after ischemia-reperfusion. Using a mechanically perfused stomach segment from chloralose-anesthetized dogs, we examined 1) responses to NG-nitro-L-arginine methyl ester (L-NAME) alone and in combination with L-arginine, 2) whether L-NAME affects ET-1-induced vasoconstriction under normal conditions and after ischemia-reperfusion, and 3) if spermine NONOate {1,3-propanediamine-N-[4-1-(3-aminopropyl)-2-hydroxy-2-nitrosohydrazi no] butyl; a nitric oxide donor} attenuates the augmented response to ET-1 after ischemia-reperfusion. Our results show that 1) L-NAME significantly increased baseline vascular resistance and this response was reduced by L-arginine, 2) ET-1-induced vasoconstriction was enhanced by L-NAME, and 3) administration of spermine NONOate during reperfusion largely attenuated the vasoconstrictor response to ET-1 after ischemia-reperfusion. Our findings are consistent with the hypothesis that nitric oxide modulates responses to ET-1 under normal conditions, and loss of this vasodilator after ischemia-reperfusion results in an augmented response to ET-1.

Changes in heat shock protein-90 and -70 messenger ribonucleic acid in uterine tissues of the ewe in relation to parturition and regulation by estradiol and progesterone.

Steroid receptors, including estrogen receptor (ER) and progesterone receptors (PR), form associations with heat shock proteins (Hsps). Dissociation of Hsps activates PR, whereas retention of Hsp90 in vitro stimulates ER. Progesterone and estrogen, interacting with their receptors, regulate myometrial contractility throughout pregnancy and during parturition. We hypothesize that uterine ER and PR changes concurrent with changes in Hsp90 and -70 abundance could alter uterine function. We quantified changes in Hsp90 and -70 messenger RNA (mRNA) abundance in pregnant sheep myometrium, endometrium, and fetal placenta during glucocorticoid-induced preterm and spontaneous labor. The effects pf estradiol and progesterone on Hsp90 and -70 mRNA in myometrium and endometrium were examined in ovariectomized nonpregnant ewes. Hsp90 and -70 mRNA distribution was evaluated by in situ hybridization in myometrium and endometrium. Dramatic tissue-specific increases in Hsp90 and -70 mRNA were observed in myometrium and endometrium (P < 0.05) during spontaneous and glucocorticoid-induced labor. Hsp90 and -70 mRNA localized in myometrial, arterial smooth muscle, and endometrial gland epithelial cells. Estradiol increased Hsp90 and -70 mRNA in myometrium and endometrium of nonpregnant ewes. Progesterone did not affect Hsp90 and -70 mRNA abundance, but inhibited the estradiol-stimulated increase. These data support our hypothesis that at term, increased abundance of Hsp90 and -70 may inhibit uterine PR and stimulate ER function in uterine tissues. Similar changes, if present, would be of importance in species showing no progesterone withdrawal before labor, such as primates, including pregnant women.

The expression of cytosolic phospholipase A2 and prostaglandin endoperoxide synthase in ovine maternal uterine and fetal tissues during late gestation and labor.

Gene expression of cytosolic phospholipase A2 (cPLA2) and prostaglandin endoperoxide synthase (PGHS) 1 and 2 was studied in ovine maternal uterine and fetal tissues during glucocorticoid induced premature labor and spontaneous labor and compared with gestational age matched controls not in labor. Northern blot analysis demonstrated that the messenger RNA (mRNA) abundance of cPLA2 and PGHS-2, but not PGHS-1, increased significantly in the endometrium during both glucocorticoid induced premature labor and spontaneous labor. Protein levels of the two enzymes measured by Western blot analyses were also elevated in the endometrium during glucocorticoid induced premature labor. cPLA2 mRNA was detected in the myometrium, but no difference was observed between ewes in labor and their gestational age matched controls not in labor. While the level of PGHS-2 mRNA was below detection in the myometrium with the methods used, the enzyme protein for PGHS-2 in the myometrium was significantly increased in both glucocorticoid induced premature labor and spontaneous labor. Using immunocytochemical methods, PGHS-2 enzyme protein was identified in the endometrial gland epithelial cells and myometrial cells. PGHS-2 mRNA was also detected in the ovine fetal cotyledons in which no differences for PGHS-2 mRNA existed between tissues collected from ewes in labor and ewes not in labor. In contrast, there were no detectable signals for cPLA2 or PGHS-2 mRNA in either amnion or chorion. We conclude that the ovine endometrium and myometrium, but not fetal membranes, are important tissue sites of prostaglandin biosynthesis involved in the process of ovine parturition.

Regulation of rat liver glucose-6-phosphatase gene expression in different nutritional and hormonal states: gene structure and 5'-flanking sequence.

The mRNA level of the catalytic subunit of rat liver glucose-6-phosphatase (Glu-6-Pase) was regulated by hormones commensurate with activity changes in vivo. Insulin exerts a dominant negative effect on the mRNA levels of Glu-6-Pase. Both mRNA levels and activities of the enzyme are low in the fed and refed state where insulin levels are elevated. Insulin administration to diabetic rats also decreases levels of mRNA and Glu-6-Pase activity. Insulin at a concentration of 1 nmol/l completely overcomes the stimulatory effect of glucocorticoids on Glu-6-Pase message levels in FAO hepatoma cells. The stimulatory response to glucocorticoid in FAO cells is biphasic, with maxima seen at 3 and 18 h after hormone addition (respectively 1.6- and 3.3-fold). 8-(4-chlorophenylthio)-cAMP (CPT-cAMP) causes a fourfold increase in Glu-6-Pase mRNA at 3 h in FAO cells. The gene of rat liver Glu-6-Pase is 13 kilobases in length and comprised of 5 exons. The exon-intron structure is completely conserved when compared with the mouse and human genes. A 0.5-kb 3'-untranslated region, which is present in rat and mouse liver Glu-6-Pase cDNA, is absent in the Glu-6-Pase gene reported here, indicating the possible duplication of either the terminal fifth exon or the entire gene. The promoter region contains a consensus core CCAAT element at position -207 and a TATAAA at position -31. Several possible response elements have been identified in the 5'-flanking region (from a HindIII site at position -1641). A consensus glucocorticoid response element is located at base pair -1552, a 9/10 match of the insulin response sequence is located at position -1449, and a 7/8 match of the cAMP response element is located at position -164.

Dynamic changes of constituents in bronchoalveolar lavage fluid in experimental silicotic rats.

Bronchoalveolar lavage (BAL) is a useful and safe method for sampling cellular and biochemical components from the lung. Analysis of bronchoaveolar lavage fluid (BALF) constituents is useful for defining the stage of disease, and for assessing disease progression and the response to therapy in lung disorders. We studied the dynamic changes in various indices for BALF and the accompanying silicotic changes in the lungs of rats at different times after quartz instillation. Total cell counts, LDH activity, protein concentration, and lipoperoxide (LPO) in the BALF of experimental silicotic rats were significantly higher than those of control rats (P < 0.05 or 0.01). After instillation, quartz content, total cell counts, LDH activity and protein concentration in BALF tended to decrease over time. These findings suggested that in acute silicosis, quartz can induce serious inflammation and damage the lung, with acute lung proteinosis seen as the main change in this stage.

Non-invasive assessment of arterial disease: a comparison of Quickscan with intra-arterial digital subtraction angiography.

The ability of Quickscan to identify haemodynamically significant lesions in the arteries of the lower limb was studied. Quickscan was prospectively compared with intra-arterial digital subtraction angiography (IADSA) of the iliac, femoral and popliteal arteries. In 155 patients, 631 arterial segments were evaluated. A Quickscan frequency ratio of 1:3 or greater had a sensitivity of 93 per cent and a specificity of 85 per cent in detecting a stenosis of more than 50 per cent diameter reduction (67 per cent area reduction) in the iliac artery, as shown on IADSA. The sensitivity and specificity in the femoral artery were 85 and 96 per cent respectively, and in the popliteal artery 82 and 98 per cent respectively. In the detection of occlusion, Quickscan had a sensitivity and specificity of 84 and 98 per cent respectively in the iliac artery, 94 and 97 per cent respectively in the femoral artery, and 94 and 85 per cent respectively in the popliteal artery. Quickscan is an inexpensive, quick and non-invasive method of screening for peripheral vascular disease in the lower limb.

[A PCR marker-based selection for resistance to bacterial blight in rice].

Molecular marker-based selection in plant breeding requires not only suitable molecular markers closely linked to the known genes, but a simple, economic and reliable analyzing technique. We report here a useful PCR marker for genetic diagnostics in breeding for resistance to rice bacterial blight. Xa21 is a newly found gene of rice which confers resistance to bacterial blight caused by Xanthomonas oryzae pv. oryzae. We produced two F2 populations between one resistant line IRBB21 containing Xa21 and two susceptible varieties, respectively. One PCR marker, PB78, detected polymorphism between the susceptible varieties and the resistant line. Cosegregation between Xa21 and PB78 was studied in the two F2 populations. The results showed that Xa21 was closely linked to the molecular marker, the crossing over value was 2.48%. Marker-based selection revealed that 100% of the plants with homozygous resistant genotype of PB78 showed resistance to bacterial blight. The available approaches detecting molecular markers in plant breeding are also discussed.

Detection of congenital color vision defects using heteroduplex-SSCP analysis.

Gene deletion and hybrid gene formation result in congenital red and green color vision defects. The innumerable variations in these defects require the development of appropriate methods for detailed investigation. We used heteroduplex-single-strand conformation polymorphism (SSCP) analysis to detect the gene variations in subjects with congenital red and green color vision defects. For comparison, we also used polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) to detect these defects. In all 13 protan and 9 of the 15 deutan subjects, defects of the red or green pigment gene could be identified by either heteroduplex-SSCP or PCR-RFLP analysis. Through heteroduplex-SSCP analysis, sequence polymorphism was detected in exon 5 of the green pigment gene. One polymorphic sequence was present in 8 of 21 trichromats but in only one of 17 dichromats, showing a biased distribution. A specific variation was found in one protan by SSCP analysis. Heteroduplex-SSCP procedures are useful for objective clinical diagnostic testing and for further study of color vision variations because of their simplicity and reliability and because they provide more information.

[Prevention and treatment effect of niacin on FES: experimental study].

OBJECTIVE: On the basis of fat embolism syndrome animal model injected with homologous dog bone marrow fat, we probed into the prevention and treatment effect of niacin on FES. METHODS: Fourteen dogs were divided into experimental group and control group. In the experimental group, the dogs were injected with homologous dog bone marrow fat accompanied by niacin injection 4 mg/kg. 8 hr intravenously for 48 hours, while in the control group, only bone marrow fat was injected. PaO2, FFA, triglyceride, platelet counts, fat drops in the blood clots of vein were examined at different times. Finally, morphology and pathology of the lungs were observed. RESULTS: There were a slower decrease in PaO2 and platelet counts and no obvious increase in triglycerides and FFA in the experimental group. The amount and diameter of fat drops in the blood clots of vein and in the lungs were less in the experimental group than in the control. Pathological examinations of the lungs demonstrated much less injuries in the experimental group. CONCLUSION: Good prevention and treatment effect of niacin on fat embolism syndrome is suggested.

Linoleate, alpha-linolenate, and docosahexaenoate recycling into saturated and monounsaturated fatty acids is a major pathway in pregnant or lactating adults and fetal or infant rhesus monkeys.

Carbon recycling and desaturation and elongation of linoleate, alpha-linolenate and docosahexaenoate in ten fetuses and two nursing infants of chow-fed rhesus monkey mothers were studied in vivo using uniformly labeled tracer molecules and high precision mass spectrometry. Doses of [U-13C]-18:2n-6, [U-13C]-18:3n-3 or [U-13C]-22:6n-3 free fatty acids were infused intravenously to the adults, and milk, maternal plasma, fetal plasma and tissues, and infant plasma were analyzed for enrichment in fatty acids of length C14 to C22. Conversion of tracer fatty acids to palmitic, stearic, oleic, and long chain polyunsaturated fatty acids was observed in fetal liver, brain, and retina ca. 5 days after dosing, and in milk and infant plasma 1 and 7 days after dosing. Animals dosed with [U-13C]-22:6n-3 accumulated more label in the fetal organs compared to the animals dosed with [U-13C]-18:3n-3 or [U-13C]-18:2n-6. The greatest fractions of doses were found in the fetal brains at levels of 0.21%, 0.24% and 1.7% for the [U-13C]-18:2n-6, [U-13C]-18:3n-3, and [U-13C]-22:6n-3, dosed mothers, respectively. Label was found in saturated and monounsaturated fatty acids in liver, brain and retina (0.05-1.5 ppm dose/mg lipid) for all doses. These results demonstrate that 1) recycling of carbon from 18:2n-6, 18:3n-3, and 22:6n-3 into saturates and monounsaturates is a major metabolic pathway in chow-fed primates in the perinatal period; 2) less than 2% of the n-3 doses are found in brain fatty acids of developing fetuses from chow-fed mothers; and 3) [13C]-22:6n-3 accumulates in retina and brain at an order of magnitude higher level when provided as preformed [13C]-22:6 n-3 compared to [13C]-18:3n-3.

PCR analysis of half-seeds of cereal crops and its application in marker-assisted selection and breeding.

A simple and rapid PCR method was adapted to DNA analysis of cereal seeds. Half-seeds of rice, wheat, and maize were treated with an extraction buffer and the resulting supernatants were used in PCR and RAPD reactions. PCR products amplified from the half-seed DNA extracts were identical with that from leaf tissue DNA extracts when the same specific primers were used in PCR reactions. The remaining half-seeds with embryos could still germinate normally. The half-seed PCR analysis was applied to the identification of resistant genotypes of rice bacterial blight, and it was proved effective in plant breeding and genetic studies.

Endoscopic ultrasonography assessment in preoperative staging for carcinoma of ampulla of Vater and extrahepatic bile duct.

OBJECTIVE: To evaluate preoperatively the extent of primary tumor, involvement of regional lymph node, and distant metastasis of ampullary carcinoma and extra hepatic bile duct carcinoma. METHODS: 28 patients with ampullary carcinoma and the 18 patients with extrahepatic bile duct carcinoma were subjected to endoscopic ultrasonography (EUS). The results were compared with those of surgical explorations and pathological findings of the resected specimens for evaluating the accuracy of preoperative staging of EUS. 46 patients underwent surgical explorations. Radical resection with detailed pathological study was done for 22 patients with resectable ampullary carcinoma and 18 patients with extrahepatic bile duct carcinoma. Carcinomas of ampulla of Vater and extrahepatic bile duct were staged according to the tumor, nodes, metastasis (TNM) classification. RESULTS: The accurate rate of EUS in assessing the extent of cancer invasion was 81.8% for ampullary carcinoma, and 72.2% for extrahepatic bile duct carcinoma. The accuracy of EUS in predicting regional lymph node metastasis was 59% for ampullary carcinoma, and 61.1% for extrahepatic bile duct carcinoma. Invasion of portal vein was correctly predicted by EUS in 2 of 3 patients, but the 3 cases of liver metastasis were not detected by EUS. CONCLUSIONS: EUS is an effective method for the evaluation of the extent of invasion of ampullary carcinoma and extrahepatic bile duct carcinoma as well as the involvement of regional lymph node before operation. Because of its limited penetration depth, however, EUS is inadequate in the assessment of liver metastasis.

Cataract free zone and primary health care approach to prevention of blindness in Shunyi county of Beijing.

OBJECTIVE: To establish a three-level eye care network, together with a referral and monitor system for the prevention of blindness, with the aim of creating a cataract free zone. SUBJECTS AND METHODS: Shunyi County with a population of 548364 was chosen as the target site. Our activities included (1) establishment of a County Guiding Committee for the prevention of blindness: (2) training local ophthalmologists and primary eye care personnel: (3) extensive publicity of knowledge for the prevention of blindness: (4) screening for blindness and low vision, (5) cataract surgery as the chief measure for creating a cataract free zone. RESULTS: A referral and monitor system for cataract blindness was established in 1987. Seven local ophthalmologists, 449 primary eye care health workers and six optometrists were trained. 815 cataract-blind patients were identified. Up to December 1991, 667 patients (737 eyes), accounting for 81.84% of the total, were treated with cataract surgery, 90.64% of these eyes had their sight restored, and 59.66% patients resumed work. A cataract free zone has been created in Shunyi County. CONCLUSIONS: The prevention of blindness with cataract surgery as the chief measure can dramatically decrease the prevalence of blindness in a relatively short period of time.

[Effect of dihydroetorphini hydrochloride on mice parturition].

OBJECTIVE: To determine the effect of dihydroetorphini hydrochloride (DHE) on parturition of mice. METHODS: 180 pregnant mice were divided equally into 6 groups. DHE was administered intramuscularly (i.m.) to mice group A, B and C at the dose levels of 1, 2 and 4 micrograms/kg respectively. Morphine hydrochloride 200 micrograms/kg (hypodermic) was given to group D and procaine hydrochloride 1000 micrograms/kg i.m. for group E, while for group F as control distilled water was given. Each preparation was given to the pregnant mice 3 times, i.e. 18 hours before the date of confinement, during labor and 2 hours postpartum. RESULTS: No effects on mice parturition were found in group A, B, E, and F. The percentage of cyanosis in newborn mice of group C and D was 13.8% and 22.8% respectively, which was significantly higher than that (7.4%) in group F. CONCLUSION: DHE is safer than morphine and procaine for analgesia during labor, and no adverse effects of DHE at dose levels of 1 microgram/kg and 2 micrograms/kg were observed on mice parturition.

[Urine level of trans, trans-muconic acid used as an index of internal dose of exposure to benzene].

Urine trans, trans-muconic acid (TTMA) in population exposed to benzene was determined with a high performance liquid chromatography-ultra violet detection system. Result showed urine TTMA increased significantly with increasing of air benzene levels, showing a good correlationship between them (r = 0.940, P < 0.05). Even in relatively low air benzene levels (with an average of 2.46 mg/m3), urine TTMA in exposed group was higher, as compared with that in controls, and their difference reached statistically significant level. It suggests that urine TTMA level can be used as a sensitive and specific indicator for biological monitoring, especially for low exposure to benzene. There was no significant difference in urine TTMA between males and females exposed. Urine TTMA formation can be inhibited by toluene, and confounding of toluene can be roughly adjusted by analysis of covariance.

Colchicine reduces hepatic fibrosis in mice infected with Schistosoma japonicum.

OBJECTIVE: Colchicine has been used clinically to treat hepatic cirrhosis caused by multiple etiologies. MATERIALS AND METHODS: The effect of colchicine on the liver fibrosis of mice infected with Schistosoma japonicum were studied. The morphological observation and morphometric analysis of the infected and untreated, and infected and treated groups were proceeded under the light microscopy, and morphological observation was made under electron microscopy. RESULTS: The results of morphometriic analysis in histological sections showed that the collagen fibers area per schistosome's egg was decreased in the infected and treated groups 10 to 13 weeks after infection. The decrease in the group of 13 weeks was significant statistically. The egg numerical density on area increased continuously from 10 to 13 weeks in both untreated and treated groups in similar degree. The ultrastructural study showed the accumulation of collagen fibrils around the hepatocytes and in the space of Disse decrease and ultrastructural lesion of hepatocytes get recovery in the treated group. CONCLUSIONS: These findings suggest that colchicine has therapeutic effect on schistosomal liver fibrosis, through not only killing off the schistosome but interfering the metabolism of collagen.

[Increase of gentamicin uptake in cultured mouse peritoneal macrophage and rat hepatocytes when used in the form of nanoparticles].

The polybutylcyanoacrylate nanoparticles of 3H-labeled gentamicin were prepared in order to investigate the possibility of gentamicin nanoparticles as an intracellular drug delivery system for intracellular chemotherapy. The 3H-labeled gentamicin nanoparticles were incubated with mouse peritoneal macrophage (MPM) or rat hepatocytes (RH) for some period, then the cells were separated from the nanoparticles, and finally the radioactivity (cpm) of 3H in the cells were measured by a liquid scintillation counter. By comparison with the solution of 3H-labeled gentamicin, a 6.34 times increase of cpm value in MPM after 30 min incubation, and 27.74, 9.03 and 8.36 times increase of MPM values in RH after 1, 12, and 24 h incubation respectively, were observed by binding gentamicin with polybutylcyanoacrylate nanoparticles. The particle size, surfactant coating, stabilizer and the gentamicin concentration were found to have some effect on the uptake of nanoparticles by two kinds of cells. This study provided a basis for the screening of intracellular drug delivery system.
