[Study of estrogen receptors in hepatocellular carcinoma and its response to anti-estrogen therapy].

OBJECTIVE: To study the usefulness of anti-estrogen therapy for moderately and advanced hepatocellular carcinoma (HCC). METHODS: Estrogen receptors (ER) were examined in 51 cases of HCC by immunohistochemical staining of needle aspiration biopsy HCC specimens. The cases studied were randomly divided into two groups: the control group (N = 25) and the anti-estrogen treated group (N = 26). Patients in the control group were given matelin and immune RNA while those in the anti-estrogen treated group were given Tamoxifen in addition. A 50% reduction of serum AFP level and a 50% decrease in tumor size were considered therapeutically effective. RESULTS: In the control group, the response rate as judged by AFP level (RR-AFP) and by tumor size (RR-TS) in the ER+ patients was 42.9% (3 of 7 cases) and 14.3% (1 of 7 cases), respectively, while the 2 RRs in the ER- patients were lower but the difference was statistically insignificant. In contrast, in the Tamoxifen-treated group, RR-AFP was 83.3% (5 of 6 cases) and RR-TS was 50.0% (3 of 6 cases). The difference in RRs of ER+ patients in the 2 groups was statistically significant. No complete remission was observed. CONCLUSION: Endocrinotherapy may be of value in the treatment of ER+ HCC.

[Long-term results of preoperative chemotherapy for operable breast cancer].

OBJECTIVE: To evaluate the long-term therapeutic results of preoperative chemotherapy for operable breast cancer. METHODS: Patients were divided into preoperative chemotherapy group (group A, 253 cases) and postoperative adjuvant chemotherapy group(group B, 284 cases). The group A patients received preoperative chemotherapy for 4 weeks, followed by radical operation two weeks after chemotherapy. Postoperative adjuvant chemotherapy began within 2 weeks after surgery, with the same chemotherapeutic regimen for 6 or more cycles in both groups. RESULTS: (1) The 5-year overall survival rate (OS) and disease-free survival rate(DFS) were 59% and 54.9%, respectively, for group A in stage III, which were higher than those of group B(28.3% and 20.8%, P < 0.05). (2) For group A in stage II and III, the 8-year OS were 81.4% and 46.9%, and DFS were 76.3% and 40.6%, respectively, which were higher than those of group B(OS: 67.4% and 20.7%, DFS: 62.9% and 13.3%, P < 0.05). (3) The 5-year and 8-year OS were higher for group A than those for group B in patients with T3, T4 or positive nodes > or = 4, (P < 0.05). CONCLUSION: The results show that preoperative chemotherapy can improve the short- and long-term survival of patients with operable, stage III breast cancer and long-term survival of patients with stage II breast cancer.

[Comprehensive treatment of bone metastases of breast cancer: a clinical analysis of 70 cases].

OBJECTIVE: Bone metastasis is common in breast cancer patients and its main symptom is bone pain. A series of methods were tried to relieve bone pain and to improve quality of life. METHODS: 70 cases of bone metastases of breast cancer were divided into 2 groups and treated with either chemotherapy, radiotherapy, endocrine therapy or isotope therapy alone or in combination. RESULTS: Endocrine therapy or isotope therapy given alone was more responsive than chemotherapy or radiotherapy alone. When the treatment modalities were used comprehensively, the response rate was 90% as compared to 56.4% in patients given single modality treatment (P < 0.01). CONCLUSION: Endocrine or isotope therapy given alone is more effective than chemotherapy or radiotherapy alone. Patients positive for estrogen and progesterone receptors, are more suitable for endocrine therapy. Comprehensive treatment is recommended.

[Damage-loss relationships and integrated control of 3rd instar larvae of Actias selene nigpoana Felder in planting area of Cornus officinalis Sieb. et Zucc].

In the present paper, the damage-loss model of Actias selene ningpoana to Cornus officinalis was tested and the results indicated that the yield loss rates obeyed the equation Y = 100 - EXP(4.6042 - 0.0315X). The economic threshold of the 2hd and 3rd instar larvae of A. selene nigpoana was then determined as 22 and 8 insects per tree respecitively. Suggestions for integrated control have been made based on the research of activities of A. selene ningpoana in the forest.

Exon-by-exon screening for RB germline mutations using Heteroduplex-SSCP analysis.

OBJECTIVE: To disclose the nature of RB1 germline mutations in Chinese and to develop a practical and effective way for mutational screening. METHODS: Leukocyte DNA was prepared from 8 Chinese patients with hereditary retinoblastoma. PCR combined with nonisotopic heteroduplex-SSCP analysis was used to screen leukocyte DNA for RB1 germline mutations, exon-by-exon, without the use of restriction endonuclease digestion. The mutations were finally identified by sequencing. In order to testify the effectiveness of this method, the same method was used to detect other 17 samples which have been previously analyzed by other methods. RESULTS: Heterozygous germline mutations were detected in the leukocyte DNA of 6 out of 8 Chinese patients: G del/codon 46, T del/codon 131, CAGAA del/codon 257-258, GCAgta-->GCAgca/donor of exon 16, C-->T/codon 661, and C-->T/codon 787. Heteroduplex-SSCP analysis may detect RB1 germline mutations in 68% (17/25) unselected patients, which is more effective than SSCP(56%) or heteroduplex analysis(64%) alone. The corrected RB1 mutation detection rate may be 80% if the samples were previously analyzed by Southern blotting, which is much higher than those reported on literature. CONCLUSION: Mutations involving a few base pairs in RB1 gene are common in Chinese. Heteroduplex-SSCP analysis is more useful and effective than SSCP or heteroduplex analysis alone for the rapid screening for unknown mutations.

[Cloning, expression and purification of the chaperonin GroESL in Escherichia coli].

The DNA fragment encoding the molecular chaperion GroESL was subcloned into high-expression vector pKC220, and the GroESL were high expressed in the E. coli strain harboring the recombinant plasmid by high temperature induction. The amount of the expressed GroEL and GroES protein were about 40% and 15% of the total cellular proteins, respectively. These two subunits were both purified from E. coli by (NH4)2SO4 salt out DEAE-52 chromography and Sephadex G-50 chromography.

Gravitation model for the magnitude of migration and its application.

"The magnitude of migration is one part of the feasibility study on migration, its result being critical to the yield of positive or negative effect.... The key to the success of migration is that the magnitude of migration must be limited within the capacity of the place of destination with generous room for flexibility. It is therefore necessary to determine the appropriate magnitude by scientific methods." The geographical focus is on China.
