[Purification and characterization of cyclomaltodextrin glucanotransferase from alkaliphilic bacteria].

A cyclomaltodextrin glucanotransferase from alkaliphilic bacteria was purified to PAGE homogenous by ammonium sulfate precipitation and DEAE-cellulose DE-52 chromatography with 11.5 fold purification and 5.7% recovery. Mr estimated with concentration gradient PAGE was 151,000. The optimum conditions for activity were pH 7.0, temperature 60 degrees C, stable in the range 6.0-9.0 and below 50 degrees C. The enzyme activity was activated by salicin and maltitol, slightly inhibited by Sn2+, Mn2+, inositol and pullulan, strongly inhibited by Ag+, Cu2+, Al3+, Fe2+, Pb2+, Hg2+ and Zn2+. The maximum absorption was at 270 nm in ultraviolet spectrum, the maximum emission wavelenghth in the excitation spectrum was 283 nm, the maximum emission spectrum determined at 283 nm was 335 nm. The effects of some protein modification reagents on the CGTase activity have been studied. Histidine and tryptophan residues may be essential for activity, carboxyl groups might have some effects on the activity.

[Study on the physiological activities of the metabolites from Paecilomyces arovirens].

The metabolites of Paecilomyces arovirens exhibited insecticidal activities to many inscct pcsts, including cotton aphid, Aphis gossypii, hawthorn Ispidcr mitc, Tetranychus viennensis and larvae of imported cabbage worm, Pietis rapae. A comparison was made between the metabolites of P. arovirens and naphthyl acetic acid (NAA) in wheat (Triticum aestivum) and water melon (Cucumis sativus) bioassays on their physiological effects. Results demonstrated that some of the metabolites physiological activities of P. arovirens are same to that of NAA, some are different between them. It indicate that there is a new kind or many kinds of plant growth regulator in the metabolites and need further analysized. The results can offer reference for exploiting a new insecticid which not only can control insect pests, but also increase plant growth.

[Analysis of rhodopsin and peripherin/RDS genes in Chinese patients with retinitis pigmentosa].

PURPOSE: To disclose the mutation of rhodopsin and peripherin/RDS genes among Chinese patients with retinitis pigmentosa as there was no identified mutation through sequencing reported in Chinese. METHODS: Genomic DNA was prepared from the peripheral lymphocytes. Gene fragments of the rhodopsin and peripherin/RDS genes were amplified by the polymerase chain reaction. The PCR products were analyzed by Heteroduplex-SSCP technique. PCR samples with aberrant migrational bands were identified through direct sequencing or cloning sequencing. RESULTS: Three different mutations in the rhodopsin gene were found in 3 of the 83 patients with retinitis pigmentosa(Va1104Phe, Lys311Glu, Pro347Leu). Two of the three mutations have not been reported before. One of the two (heterozygous, Va1104Phe) was found in an isolated patient and the other (homozygous, Lys311Glu) in a family with autosomal recessive retinitis pigmentosa. Mutation of peripherin/RDS gene was not found in the 83 patients. CONCLUSION: Mutation in the rhodopsin gene is the common cause in Chinese patients with retinitis pigmentosa, either autosomal dominant, recessive or sporadic.

Color vision defects with variation in the exon 5 of red and green pigment genes.

PURPOSE: To investigate correlation of variation in the exon 5 of red and green pigment genes with color vision defects. METHODS: Exon 5 of the red and green pigment genes in 11 protans, 19 deutans and 38 normal controls were analyzed by heteroduplux-SSCP analysis. RESULTS: In all 11 protans and 8 of the 19 deutans, defects of the red or green pigment gene could be identified. The C polymorphism (A/C at codon 283) in green pigment gene was present in 8 of 44 trichromats and 5 of 24 dichromats. Specific electrophoretic bands were found in 2 normal controls and a deutan. CONCLUSIONS: Variation in the exon 5 of the red and green pigment genes is the most common cause for color vision defects. Heteroduplex-SSCP analysis is a suitable way in screening specific variation in visual pigment genes.

Dacryology in China.

PURPOSE: International Society of Dacryology held the IV International Congress in Stockholm on June 1996. I was warmly invited as a guest of honor beforehand to deliver a lecture on a special topic of "Dacryology in China" there, so that the world might understand the dacryology in China, and thus the international growth of dacryology as a discipline would be promoted. METHODS: In China tear lab research and clinical research of LG are nearly empty except a few papers published. We had studied 3 items on LS for 28 years before 1979, and put our attention mainly on dacryology since 1979 by 3 stages: (1) collecting literatures (1979-81); (2) lab research (1981-84) and (3) clinical research, put DE as the key point by 4 steps: DE diagnosis (1984-86); DE treatment (1987-91); DE Condition Judgement (1989-92) & DE cause & mechanism (1991-95). RESULTS: In the past 44 years, esp. from 1979 on, in a planned way, we have systematically researched the dacryology with main results below: created new word "Leiyexue" in Chinese & its English words, dacryology & lacrimology by adding-ology to dacrya & lacrima; made LS consisted of reflex LG & LP, and basic LG besides; found out the normal & abnormal data of tear and serum LF & its iron bindingcapacity; MDH, LDH & isoenzymes; beta 2 m; MDA & antioxidants; trace elements, prostaglandins, intramuscular gentamicin passing the blood-tear barrier; bacteria, fungi, anaerobe, virus & parasite; Ig, T cell & subsets, Langerhan cell, HLA antigen; in cidence (KCS, 0.3-3%, trancomatous DE 0.003%); the parallel correlations between DE graded by rb, BUT & Slt; and in SS, lymphocytes infiltration in LG; Lab G & conjunctiva; LF (< 100 mg%) & beta 2 m (> 1.7 mg%); and testosterone receptors in LG; negated presence of tear Beta-lysim; set up a series of models for researching & a complete set of measures to diagnose, grade, judge and treat DE, deviced new operations. AT & prescription for DE; proved SS being autoimmune & EB virus, its one cause. CONCLUSIONS: This relatively, overall & systematical research of dacryology already have been widely applied to practice with over 100 papers published. It indicates that LS & its disease as a scientific discipline under the term of dacryology to grow up is entirely possible and extremely needful.

[Studies on the oxidation reaction of octanol-2 with nitric acid by infrared spectroscopy].

In this paper, the reaction process of oxidation of octanol-2 with nitric acid has been studied by IR spectroscopy. It is found that the main components of non-sapoifiable matter are different in different oxidation degrees. The relation between oxidation products and the amount of nitric acid are investigated, the reaction mechanism has also been studied. Experimental results show that the oxidation process of octanol-2 is as follows: first, octanol-2 is oxidated to octanone-2, or to nitrate, nitrite and nitrile copmpounds, then these compounds are reoxidated to caproic acid in the meantime some by-products, such as valeric, enanthic acids are also found in oxidated products.

[Spectral study of the conformational change of yeast alcohol dehydrogenase induced by alcohol].

The conformational change of Yeast Alcohol Dehydrogenase (YADH) at the different concentration of ethanol, n-propanol and ethylene glycol was studied by means of ultraviolet spectrum, fluorescence spectrum and circular dichroism spectrum. The results showed that the ultraviolet absorbance at 220nm and 280nm as well as the relative fluorescence intensity at 336nm of YADH increased with increasing alcohol concentration. The negative peakes at 208nm and 220nm of YADH in circular dichroism spectrum with the solvent of ethanol, ethylene glycol were obviously intensified, but the 220nm peak of YADH was increased in the presence of n-propanol while the 208nm peak was decreased and red-shifted in position as to completely lost. According to the data above, it indicates that the conformation of YADH was changed with losing activity at the various concentration of alcohol.

[Spectroscopic investigations on sol-gel transition of alginate solutions by addition of cupric ion].

Viscosity measurements and 13C-NMR, UV/VIS, and Raman spectroscopic studies have been carried out of various concentration of alginate solutions undergoing sol-gel transition induced by addition of cupric cation. From analyses of the spectra, a qualitative explaination was suggested of the interaction mode of the coordination between functional groups and cupric ions, the conformational change of polychains and the behaviour of solvent molecules druing the process of sol-gel transition. The mechanisms of phase transition of our system induced by the heavy metal ion have been discussed.

Expression and distribution of adhesion molecule CD44 in healing corneal epithelia.

PURPOSE: To study isoform expression and cellular distribution of CD44, a cell surface glycoprotein thought to be an adhesion molecule in cell-cell and cell-substratum interactions, during corneal epithelial wound healing. METHODS: Reverse transcription-polymerase chain reaction was performed to determine alternatively spliced rat CD44 isoforms. In situ hybridization was carried out on frozen sections of the rat corneas obtained at different time points after epithelial debridement. 35S-Labeled sense and antisense cRNA that recognizes rat CD44 standard form was used as a probe. Immunofluorescence was used to assess expression and localization of CD44 in the rat corneas during reepithelialization. RESULTS: Corneal epithelia contained several alternatively spliced CD44 variants. Four large CD44 variants with inserts V1 through V10, V2 through V10, V3 through V10, and V4 through V10 were differentially expressed in migratory epithelia. The silver grains, indicating CD44 transcripts, started to increase in the epithelial cells surrounding the wound margin 3 hours after wounding and peaked at 18 hours in the basal epithelial cell layers, at which time the epithelia were actively migrating. As the cells began proliferation after wounding, the density of CD44 mRNA label declined but was still significantly higher than that in control specimens. The label returned to basal level as epithelial cells reverted to their normal phenotype. The location of CD44 on cell surfaces during corneal reepithelialization was consistent with the pattern of mRNA production. In the corneas at 18 hours after wounding, CD44 immunoreactivity was elevated in the entire epithelium, from the leading edge to the limbal-corneal border. As happened for the mRNA, the cell surface CD44 declined as cells differentiated to reestablish the multilayered epithelium. CONCLUSIONS: The expression of CD44 correlates with corneal reepithelialization, suggesting that CD44 may be involved in cell-cell interactions that provide adhesive strength for the much-stressed epithelial sheet and in the cell-substratum interactions that mediate cell migration during reepithelialization.

Single and multiple dose pharmacokinetic studies of oral sustained release and non-sustained release formulations of isosorbide-5-mononitrate in healthy volunteers.

The pharmacokinetics of a new sustained release tablet (40 mg, "test") of isosorbide-5-mononitrate (CAS 16051-77-7, IS-5-MN) was investigated together with a reference preparation (20 mg, "reference") after single and multiple oral administration in ten healthy human subjects using an open, randomised two-way crossover experimental design. Based on the statistical evaluation of the area under the plasma concentration-time curve (AUC), the two tablet formulations are judged to be the same with regard to the amount absorbed. Pharmacokinetic data showed that with the test tablet significantly lower and delayed mean peak plasma levels (Cmax) were reached compared with the reference preparation in both single and multiple dose studies. The test formulation also produced lower minimum plasma concentration (Cmin). However, there was no statistically significant difference for other pharmacokinetic parameters, including the elimination rate constant (Kel), the elimination half-life (t1/2) and the peak-trough fluctuation constant (PTF) between the two treatments. It was demonstrated that the new sustained release formulation of isosorbide-5-mononitrate could be useful in clinical practice for the treatment of angina pectoris and congestive heart failure.

Contribution of opioid receptors on primary afferent versus sympathetic neurons to peripheral opioid analgesia.

Opioid receptors are synthesized in dorsal root ganglia and transported into peripheral terminals of primary afferent neurons. Activation of such receptors results in antinociceptive effects that are most prominent in inflammation. In addition, opioid receptors located on sympathetic postganglionic neuron terminals may be involved in these effects. This study investigates the peripheral analgesic efficacy of the mu, delta and kappa receptor agonists [D-Ala2,N-Me-Phe4,Gly-ol5]-enkephalin, [D-Pen2,5]-enkephalin and trans-(+/-)3, 4-Dichloro-N-methyl-N-[2-(1-pyrrolidinyl)-cyclohexyl]-benzeneacetamid e, the effective number of peripheral mu, delta and kappa receptors in relation to the development of inflammation and the contribution of sympathetic vs. sensory neurons by use of capsaicin and 6-hydroxydopamine, respectively. In Wistar rats with Freund's adjuvant-induced hindpaw inflammation, antinociceptive effects of intraplantar [D-Ala2,N-Me-Phe4,Gly-ol5]-enkephalin (1.0-32 microg), [D-Pen2,5]-enkephalin (10-100 microg) and trans-(+/-)3, 4-Dichloro-N-methyl-N-[2-(l-pyrrolidiny)-cyclohexyl]-benzeneace tam ide (10-100 microg) were evaluated by paw pressure test. These effects increased linearly between 6 and 24 hr, but did not change between 24 and 96 hr of inflammation, whereas the doses of the irreversible antagonists beta-funaltrexamine, [D-Ala2,Leu5,Cys6]enkephalin or (+/-)-(5beta,7a,8beta)-3, 4-dichloro-N-[3-methylene-2-oxo-8-(1-pyrrolidinyl)-1-oxaspir[4, 5]dec-7-yl]benzeneacetamide required to abolish the respective agonist effects increased between 12 and 96 hr. Pretreatment with capsaicin (30, 50, 70 mg/kg s.c. over 3 days) but not with 6-hydroxydopamine (75 mg/kg i.p. over 3 days) reversed the hyperalgesia in inflamed paws and almost abolished antinociceptive effects of all three agonists. These results suggest that the increased opioid agonist efficacy is due to an increased number of peripheral opioid receptors at later stages of inflammation and that peripheral opioid antinociceptive effects are primarily mediated by mu, delta and kappa opioid receptors on primary afferent neurons.

Modified pentamer formation assay for measurement of tacrolimus and its active metabolites: comparison with liquid chromatography-tandem mass spectrometry and microparticle enzyme-linked immunoassay (MEIA-II).

A modified pentamer formation assay (PFA) for quantification of tacrolimus and active metabolites after extraction from whole blood is described. The lower limit of detection was 2 microg/L. Intraassay precision (CV) was 5.7-13.7%, and the interassay CV was 6. 1-14.9%. Tacrolimus trough concentrations in 104 whole blood specimens from liver and kidney transplant recipients were compared with results from HPLC-tandem mass spectrometry (LC/MS/MS) and microparticle enzyme immunoassay (MEIA-II). Data were analyzed by difference plots and are presented as median (95% confidence intervals) of the method differences. MEIA-II results were on average 2.00 microg/L (range, -0.08 to 5.17 microg/L) higher than LC/MS/MS, whereas PFA results were only 1.07 microg/L (range, -2.62 to 5.33 microg/L) higher. Of 104 specimens tested, 25 displayed differences >/=3 microg/L between MEIA-II and PFA: median difference, 4.65 microg/L (range, 3.01-8.79 microg/L). The corresponding median difference between PFA and LC/MS/MS was -0.91 microg/L (range, -4.11 to 0.85 microg/L), and the difference between MEIA-II and LC/MS/MS was 3.67 microg/L (range, 1.88-6.34 microg/L), suggesting the presence of inactive metabolites that caused a positive bias in the immunoassay. In contrast, similar median differences were observed for the remaining 79 specimens: MEIA-II minus LC/MS/MS, 1.78 microg/L (range, -0.45 to 4.11 microg/L); PFA minus LC/MS/MS, 1.90 microg/L (range, -1.70 to 5.50 microg/L). Active tacrolimus metabolites may have contributed to the higher apparent tacrolimus concentrations in these specimens.

Molecular cloning and characterization of P113, a mouse SNF2/SWI2-related transcription factor.

A full-length cDNA encoding a 113-kDa transcription factor, named P113, was cloned from mouse preadipocyte line 30A5. P113 binds to a 7-bp consensus TNF-response element and a 30-bp fragment from mouse PAI-1 promoter (-88/-59). Sequence analysis indicates that the P113 is highly homologous to HIP116/HLTF (human) and RUSH-1alpha (rabbit). The sequence homology and the fact that P113 contains seven motifs conserved in many DNA-dependent helicases/ATPases indicate that it is a new member of the SNF2/SWI2 protein family. A cysteine-rich motif, called RING finger, was found close to the C-terminus of P113. The expression pattern of P113 mRNA in rat tissues is significantly different from that of HLTF in human tissues. Affinity-purified P113 has an ATPase activity that is activated by DNA in a sequence-specific manner. Using Northern blot analysis and the PAI-1 promoter/luciferase system, we demonstrated that P113 is a transcription factor that activates the transcription of the PAI-1 gene in 30A5 cells.

Signaling through the ARK tyrosine kinase receptor protects from apoptosis in the absence of growth stimulation.

ARK (AXL) is the prototype of a distinctive family of receptor tyrosine kinases which contain in their extracellular domains features reminiscent of cell adhesion molecules. ARK is capable of homophilic binding, which results in a degree of receptor activation, but can also be activated by a heterophilic ligand, Gas6, a member of the family of vitamin K dependent proteins that is preferentially expressed in quiescent cells. Since a number of tissues and cell lines express both ARK and Gas6, we studied the effect of endogenous and exogenous Gas6 on the phenotype of ARK expressing cells. Here we show that constitutive expression of Gas6 in an NIH3T3 cell line that does not spontaneously express this protein does not result in cell transformation or uncontrolled growth, but protects from apoptosis induced by serum deprivation. Recombinant exogenous Gas6 was also capable of protecting cells from apoptosis at concentrations that did not result in significant induction of DNA synthesis. Activation of ARK phosphorylation and a weak but significant induction of MAP kinase activity accompanied the increased survival of cells treated with Gas6. The antiapoptotic effect of ARK signaling was confirmed by studies using fibroblasts from ARK knock-out mice, that showed that the absence of ARK resulted in higher levels of serum deprivation-induced apoptosis, that could not be rescued by the addition of Gas6. Interestingly ARK signaling protects from apoptosis induced by serum deprivation, myc overexpression, or by TNF alpha but not from u.v. irradiation or Staurosporine. These results suggest that a major function of Gas6-ARK signaling is that of increasing cell survival under conditions which do not allow cell proliferation.
