[Analysis of fusion points in hybrid genes and gene deletion for congenital red-green color vision defects].

OBJECTIVE: To investigate gene deletion and the fusion points of hybrid genes in congenital red-green color vision defects. METHODS: Genomic DNA was collected from 11 protans, 19 deutans and 5 normal controls. Promoter and exons 2-5 of the red and green pigment genes in these subjects were analyzed by using PCR-Heteroduplex-SSCP analysis. The origin and component of each individual gene were determined by comparison with the patterns of known sequence of the red and green visual pigment genes. RESULTS: Fourteen out of the 30 patients with red-green color vision defects were found to have hybrid gene. The fusion points of the hybrid gene were located in exon 1-intron 1(4 cases), introns 2-3(5 cases) and intron 4 (5 cases). CONCLUSION: The fusion point of a hybrid gene may occur in exon 1-intron 1 and intron 4 as well as in introns 2-3(including exon 3).

Twenty-three cases of atrophic rhinitis treated by deep puncture at three points in the nasal region.

Atrophic rhinitis is a disease which manifests itself mainly by anosmia due to dryness and atrophy of the nasal mucosa. There is no specific therapy for the disease at present. In the past few years, 23 cases of atrophic rhinitis were treated mainly by deep puncture at three points in the nasal region with satisfactory results. In order to find out the functional changes of the nasal mucosa, the mucociliary transport rate (MTR), surface temperature of the conchal mucosa, acid-base scale of nasal secretion, and volume of nasal secretion were determined before and after the treatment.

[Study on the relationship between the history of abnormal pregnancy and TORCH infection in pregnant woman].

OBJECTIVE: To investigate the relationship between the history of abnormal pregnancy and the Toxoplasma(TOX), Other(OTH), Rubella virus(RUV), Cytomegalo virus(CMV), Herpes simplex virus-II (HSV-II) (TORCH) infections in pregnant woman. METHODS: Enzyme-Linked immunosorbent assay(ELISA) combined with polymerase chain reaction (PCR) technique were used to detect TORCH infection in pregnant women with histories of abnormal pregnancies(study group, n = 54) and normal pregnant women (control group, n = 54). The fetal cord blood samples were also detected. RESULTS: In study group, the rates of previous TORCH infection: CMV 46.29%, RUV 16.29%, TOX 16.67%, HSV-II 29.63%. The rates of active infection: CMV 57.40%, RUV 59.26%, TOX 38.89%, HSV-II 46.29%. The rates of recurrent infection: CMV 38.89%, RUV 38.89%, TOX 11.11%, HSV-II 22.22%. These three kinds of infection rates in study group were significantly higher than that of control group (P < 0.01). The abnormal pregnant cntcomes in study group were significantly higher than that of control group(P < 0.01). The incidence of maternal-fetal vertical transmission in study group was 73.08%. CONCLUSION: TORCH series infection is one of the important causes of abnormal pregnant cutcomes. It is absolutely necessary to screen TORCH infection for women who had the histories of abnormal pregnancies in order to prevent birth defects and perinatal complications. ELISA combined with PCR technique is a valuable method for the diagnosis of TORCH infection.

[Study of the role of cyclin-dependent kinases (CDKs) in retinoic acid (RA) inducing HL-60 cell differentiation].

OBJECTIVE: To investigate the role of CDKs in the RA inducing HL-60 cell differentiation. METHODS: The effects of all-trans retinoic acid (ATRA) and arotinoid ethylester(AE) at the dosage of 5 x 10(-6) mol/L on the proliferation and differentiation of HL-60 cells were examined by NBT reduction test and cytometry analysis. Meanwhile, histone H1 kinase assay was used to observe the changes of CDK2 activities in HL-60 cells treatment with ATRA and AE. The amounts of both cyclin E/CDK2 and cyclin D1/CDK4 complexes were observed with immunoprecipitates. RESULTS: Both ATRA and AE inhibited the growth of HL-60 cells, arrested cells in G1/G0 phase, and induced cells to differentiation. The activities of cyclin E/CDK2 and the amounts of cyclin D1/CDK4 complexes were decreased in ATRA- or AE-treated HL-60 cells. CONCLUSION: The effects of RA on the proliferation and differentiation of HL-60 cells may be associated with significant decreases in the activities of CDKs.

[Expressions of cellular M-CSF and M-CSFR in hematopoietic cells].

OBJECTIVE: To study the distribution of cellular M-CSF and its receptor in abnormal hematopoiesis. METHODS: Six hematopoietic cell lines, 144 bone marrow (BM) and 160 peripheral blood (PB) samples were investigated with ABC immunoperoxidase assay. RESULTS: Cellular M-CSF and/or M-CSFR were positive in all the 6 hematopoietic cell lines, a few normal BM(4 cases) and PB (32 cases) samples, hematological malignancies and a few benign hematological disorder samples. M-CSF and M-CSFR were found mainly in granulocytic lineage cells, seldom in erythroid and megakaryocytic lineage cells and never in lymphocytes. M-CSF and M-CSFR usually presented in cytoplasma and membrane and in some cases in nuclei. CONCLUSION: Cellular M-CSF and M-CSFR is widely distributed in abnormal hematopoiesis, and is especially high in myeloid leukemias.

[Effects of interferon-alpha on the mRNA expression of procollagen type I and III of hepatic stellate cells and on the deposition of collagen type I and III in fibrotic liver of rats].

OBJECTIVE: To observe the effects of interferon-alpha on the proliferation and mRNA expression of procollagen type I and III and on the deposition of collagen type I and III in fibrotic liver of rats. METHODS: CCl4 was used to induce the liver fibrosis rat model. The stellate cells were isolated from liver and cultured in DMEM medium. The effects of interferon-alpha on the proliferation and collagen synthesis of stellate cells were determined with 3H-TdR, 3H-proline incorporating test. Total RNA of stellate cells was extracted and the cDNA of procollagen type I and III and of interstitial collagenase was labeled using Dig High Primer technique. The level of procollagen type I and III and interstitial collagenase mRNA were measured by Northern blot analysis. Deposition of collagen type I and III in fibrotic liver was evaluated with Dot blot. RESULTS: At the 4th, 24th and 48th hour, interferon-alpha inhibited the intake of 3H-TdR and 3H-proline by stellate cells significantly. The inhibition rates were 16%, 19% and 27%, respectively. The intensity of inhibition was proportional to the concentration of interferon-alpha. The expression level of procollagen type I and III mRNA in the interferon-alpha treated group was significantly lower than that in control group in all stages of fibrosis. At the 2nd and 6th week, the deposition of collagen type I and III in interferon-alpha treated group decreased significantly compared with that in control group. However, no difference was found at the 9th week. The expression of collagenase mRNA between interferon-alpha treated group and control group showed no difference. CONCLUSION: Interferon-alpha can inhibid the proliferation and collagen synthesis of stellate cells, down-regulate the expressions of procollagen type I and III mRNA and reduce the deposition of collagen type I and III in fibrotic liver. It has no effect on collagenase.

[The inhibiting effect of antisense thrombin receptor gene on the proliferation of pig vascular smooth muscle cell].

OBJECTIVE: To search for an effective approach to prevent the formation of restenosis after angioplasty. METHODS: A recombinant eukaryotic expression plasmid vector containing partial antisense thrombin receptor (ATR) gene named pcDNA3/ATR was constructed using recombinant DNA technique. Mini-pig aorta injury model was established by over-sized balloon catheter combined with high cholesterol diet feeding and its aortic smooth muscle cells(ASMC) were cultured. The effect of ATR gene expression in mini pig ASMC proliferation and growth factor gene expression were studied by 3H-TdR incorporation and Northern blot. RESULTS: The DNA synthesis in pig ASMC could be inhibited by ATR gene expression (The DNA synthesis in normal ASMC was lowered by 41.8%, and that in ASMC from injured artery was lowered by (50.3%). The mRNA and protein synthesis of TR could be down regulated by ATR gene expression. The mRNA expression of PDGF-A chain and bFGF stimulated by fetal calf serum (FCS) with thrombin were both downregulated in pig ASMC with expressed ATR gene. CONCLUSION: ATR gene expression can inhibit the proliferation of pig ASMC, and this is induced by its inhibiting effect on TR and finaly the signal transduction in ASMC.

[Effect of expression of c-myc gene on hyperplasia of smooth muscle cells in vein graft in rat].

OBJECTIVES: To analyse effect of expression of c-myc gene on hyperplasia of smooth muscle cells in vein graft and evaluate the effect of c-myc gene on intimal hyperplasia of vein grafts. METHODS: We transplanted the autologous internal jugular vein on iliac artery by end to end anastomosis in rat. The vein grafts were harvested at 30 min, 2 h, 6 h, 2 days, 1 week, 2 weeks and 4 weeks after grafting and were rapidly frozen in liquid nitrogen. Proteins of PCNA and c-myc were detected in the different time with method of immunohistochemical staining (SP) and c-myc mRNA with in situ hybridization. Staining result was measured by microscope (400X). RESULTS: Freshly harvested veins did not demonstrate PCNA, c-myc protein production and c-myc mRNA. Staining for c-myc mRNA was significantly higher at 2 h, 6 h compared with that at 30 min (P < 0.01). Staining for PCNA, c-myc protein production peaked at 1 week (44.8 +/- 8.2)%, (16.2 +/- 3.2)%, respectively) and was significantly higher than that at 6 h (3.2 +/- 0.7)%, (4.1 +/- 0.9)%, respectively) (P < 0.01). CONCLUSION: c-myc is expressed early after experimental vein grafting, with peak at 1 week paralleled with PCNA. This occurs during a period of maximal SMC proliferation, and increased expression of PCNA. Expression of c-myc gene may contribute to the induction and regulation of SMC proliferation.

Effect of Astragalin on matrix secretion and beta 1 integrin mRNA expression in human mesangial cells.

OBJECTIVE: To investigate the effect of Astragalin on human renal mesangial cells. METHODS: Cultured human mesangial cells were treated with Astragalin and Astragalin serum in different concentrations in the presence or absence of PDGF-BB, the proliferation and type IV collagen secretion of mesangial cells were measured by MTT assay and ELISA, and expression of beta 1 integrin gene was estimated by reverse transcription-polymerase chain reaction (RT-PCR) method, suspectively. RESULTS: After 72 hours Astragalin or Astragalin serum treatment, the proliferation of mesangial cells induced by PDGF-BB was inhibited significantly in a dose-dependent manner compared with untreated controls (P < 0.05 and P < 0.01). After 24 hours of Astragalin or Astragalin serum treatment, the secretion of type IV collagen protein in presence of PDGF-BB was significantly decreased and beta 1 integrin mRNA level decreased significantly compared with untreated control (P < 0.05, P < 0.01). CONCLUSIONS: Astragalin inhibits cell proliferation and matrix over-synthesis which might be mediated, at least, partly by decrease of beta 1 integrin gene over-expression. The study suggested that Astragalin might play a role in preventing the progression of chronic renal diseases.

[Evaluation of clinical value of technetium-99m(V) dimercaptosuccinic acid SPECT in diagnosis of parotid lump].

OBJECTIVE: To evaluate the clinical value of single photon emission computed tomography (SPECT) on classifying parotid lump as benign or malignant before treatment. METHODS: SPECT images of the parotid region were obtained in forty-five patients with parotid lump before surgery. We used technetium-99m(V) dimercaptosuccinic acid as imaging agent. The results were compared with their pathological diagnoses, clinical examination, and ultrasonography. RESULTS: The sensitivity, specificity and accuracy of Tc-99m(V)-DMSA SPECT imaging in determining malignancy of parotid lump were 93.7%, 72.4%, 80.0%, respectively. The uptake ratios of parotid lesions were higher in malignant tumors than benign tumors, cysts, and inflammatory lumps (P < 0.05). CONCLUSION: It is suggested that Tc-99m (V)-DMSA SPECT can be a useful aided method of preoperative diagnosis of parotid lumps. The uptake ratios can help increase the accuracy of diagnoses. For the benign tumors and inflammatory masses of parotid gland, the diagnoses had some false positives which may be reduced by combined clinical examination and ultrasonography.

[Study on antitumor activity and mechanism of Ganoderma polysaccharides B].

OBJECTIVE: To study the antitumor activity and mechanism of Ganoderma polysaccharides (GL-B). METHODS: Both in vivo and in vitro experiments were conducted in the study. Proliferation of tumor cells was detected by MTT method, tumor necrosis factor alpha (TNF alpha) was detected by biological assay, and interferon gamma (IFN gamma) by enzyme-linked immunosorbent assay (ELISA). The level of mRNA was detected with the method of reverse transcriptase-polymerase chain reaction (RT-PCR). RESULTS: (1) GL-B 50, 100, 200 micrograms/ml inhibited the growth of implanted sarcoma 180 in vivo significantly and dose-dependently. (2) GL-B directly adding to the cultured medium neither induced HL-60 apoptosis nor restrained its proliferation in vitro. (3) The macrophages and T lymphocytes cultured medium treated with GL-B significantly induced HL-60 apoptosis and inhibited its proliferation. GL-B significantly increased TNF alpha and IFN gamma release and their mRNA expression in the cultured medium. CONCLUSION: The anti-tumor activity of GL-B was derived from promoting mRNA expression of TNF alpha and IFN gamma, resulting in TNF alpha and IFN gamma release.

[Relationship between PDGF-A, bFGF and proliferation of smooth muscle cells in vein grafts].

OBJECTIVE: To detect the relationship between platelet-derived growth factor-A (PDGF-A), basic fibroblast growth factor (bFGF), and proliferation of smooth muscle cells in vein grafts. METHODS: An animal model of autogenous vein graft was established by transplanting internal branch of the jugular vein to the common iliac artery by end-to-end anastomosis. 25 rats were used. The grafted veins were harvested at 6 h, 2 d, 1 w, 2 w and 4 w respectively after the operation. The expression of PDGF-A and bFGF in different stages after grafting procedure was observed immunohistochemically. RESULTS: The expression of PDGF-A peaked at 1 w, paralleled with proliferation of SMC, and was significantly higher than that at 6 h (P < 0.01). The expression of PDGF-A diminished gradually. At 4 w, it was significantly lower than that at 1 w (P < 0.01). bFGF demonstrated bimodal pattern of the expression: the first peak of expression occurred by 2 d and the second peak by 2 w. The difference was significantly compared with other stages. CONCLUSIONS: The expression of PDGF-A 1 contributed to migration and proliferation of SMC. Except for contributing to proliferation of SMC, bFGF perhaps took part in endothelization.

[Effects of inflammatory infiltration on the formation of abdominal aortic aneurysm].

OBJECTIVE: To explore inflammatory infiltration and its effects on the formation of abdominal aortic aneurysm (AAA). METHODS: Immunohistochemical technique was used to detect lymphocyte (CD45 positive) and macrophage (CD68 positive) infiltration while in situ hybridization was applied to investigate the mRNA expression of Matrix Metalloproteinase-9 (MMP-9) in 20 cases of human AAA and 4 cases of normal human abdominal aortas. RESULTS: Inflammatory infiltration occurred in every case of AAA with various degree, and there was a parallel tendency between the degree of injury of abdominal aortic elastin and that of inflammatory infiltration in AAA. Inflammatory infiltration was not found in normal aortas. MMP-9 mRNA was detected in macrophages and lymphocytes in all 20 cases, in smooth muscle cells in 13, and not in normal abdominal aortas. CONCLUSIONS: Inflammatory infiltration attended and enhanced the formation of AAA through a complex of sequence of biochemical, cellular and immune events.

[The action of proglumide blocking gastrin on gastric cancer cells].

OBJECTIVE: To investigate the practicability of proglumide to treat gastric cancer. METHODS: MKN45 gastric cancer cell line was cultured and the effects on gastrin and gastrin receptor antagonist proglumide proliferative rate, cell dynamic cycle distribution and the concentration of cAMP in the cells were observed in vitro. RESULTS: Gastrin promoted the proliferation of MKN45 cells and shifted cells from phase G(0)/G(1) to phase S, G(2)/M as well as increased intracellular cAMP, while proglumide blocked these effects. CONCLUSIONS: Gastrin induces the proliferation and synthesis of DNA by its receptor. Proglumide may provide a new approach of non-cytotoxic treatment of gastric cancer.

[Effects of EB virus LMP on the differentiation and growth of human nasopharyngeal carcinoma cell line CNE1].

OBJECTIVE: To study the effects of EBV-LMP gene on differentiation and growth of human nasopharyngeal carcinoma (NPC) cell. METHODS: With NPC cell line (CNE1) as target, electroporation was used to transfect EBV-LMP gene and the vector into CNE1 cells. The proliferation changes of transfectants were measured in vitro by proliferation experiment. FCM method and transplantation in nude mice were used. RESULTS: LMP promoted the growth ability of NPC CNE1 cells in vitro. The average A ratio (3.98 +/- 0.11) in the treatment group was higher than in the control group (2.36 +/- 0.05) and also in the negative control group (2.75 +/- 0.07, P < 0.01). Clone forming test (25.2%, 378/1 500) in the treatment group was higher than in the control groups (11.2%, 168/1 500) and negative control groups (13.4%, 201/1 500). When FCM method was used, the keratin positive rate (82.7%) in the treatment group was markedly decreased as compared with the control (92.5%) and negative control groups (95.7%, P < 0. 05). Latent period (26.6 +/- 7.7) days and internal double time (2.51 +/- 0.18) days of transplant tumor of the treatment group were markedly shortened, tumor take rate (5/6) was increased, cells of the transplant tumor in the treatment group showed an image of small size, great allotype, and a trend of low differentiation. CONCLUSION: The EBV-LMP can promote the growth and inhibit the differentiation of NPC CNE1 cells. EBV-LMP may play an important role in the development of NPC and in studying its mechanism.
