[The difference of thrombin-induced TF/uPAR mRNAs expressions in cultured endothelial cells and U937 cell line].

OBJECTIVE: To compare the effects of thrombin on the expression of the tissue factor (TF) and urokinase-type plasminogen activator receptor (uPAR) mRNA in the cultured vascular endothelial cells (VEC) and U937 cell line. METHODS: Reverse transcriptase-polymerase chain reaction (RT-PCR) was used to detect the mRNA levels of TF and uPAR genes in the cultured cells under stimulation of thrombin, inactivated thrombin (DFP-T), or at the presence of the inducers and anti-thrombin receptor antibody (ATR-Ab). RESULTS: In U937 cells, the TF and uPAR mRNA expression induced by thrombin could be fully quenched with the ATR-Ab, but in VEC, even at the presence of ATR-Ab, there still was a little stimulative effect of thrombin revealed. DFP-T had no effect on the expression of TF and uPAR mRNA in U937 cells, but could elicit a faint reaction in VEC. CONCLUSION: The induction effects on TF and uPAR mRNA expression in both of the cultured cells by thrombin were mainly dependent on the TR activation, which may not be the unique pathway for thrombin to induce TF and uPAR mRNA expression in VEC.

[Exploration of gene expression profiles of CD(34)(+) hematopoietic stem/progenitor cells based on large scale sequencing].

OBJECTIVE: To set up a large scale sequencing system and explore the gene expression profiles of CD(34)(+) hematopoietic stem/progenitor cells (HSPCs). METHODS: CD(34)(+) cells were isolated from umbilical cord blood and subjected to cDNA library construction. A primary profile of gene expression in HSPCs was emerged by EST sequencing and bioinformatics analyzing. RESULTS: Among 9,866 ESTs thus obtained, 7 476 meaningful ESTs were clustered into 2,060 unique sequence species (USSs), representing 1,054 known gene species and 1,006 unknown gene fragments. The 1054 known genes were divided into 8 categories: (1) hematopoiesis associated: 73, (2) chromatin structure and cell division/apoptosis: 91, (3) signal transduction and receptors: 111, (4) cell structure/mobility: 48, (5) cell/organism defense/homeostasis: 41; (6) Gene expression (transcription, translation and modification): 265, (7) metabolism: 192; and (8) unclassified: 233. CONCLUSION: A gene expression profile including 1,054 known genes and 1006 new gene fragments of HSPCs was primarily obtained, which may lay a basis for the further study on the molecular mechanism of hematopoiesis regulation and provide candidates for new gene cloning.

[Study of spontaneous echo contrast in patients with rheumatic mitral stenosis by transesophageal echocardiography].

Spontaneous echo contrast (SEC) is common in the areas of slow blood flow. This study was conducted to examine the correlation of SEC and left atrial appendage function, atrial fibrillation (Af) and thrombus in patients with rheumatic mitral stenosis. The transthoracic and multiplane transesophageal echocardiograms were performed on 117 patients(mean age 42.22 +/- 10.79 years). Left atrial size, left atrial appendage area, mitral valve area, left atrial appendage flow peak emphatic velocity and full velocity were measured. The presence of SEC and thrombus in the left atrium and appendage was noted. The results showed that among the patients, 57 had normal sinus rhythm, 60 had Af; 19 had thrombus, 98 had no thrombus; 71 had SEC and 46 had no SEC. Compared with the patients without SEC, the patients with SEC showed greater incidence of thrombus (28.76% vs 0, P < 0.05), higher rate of Af (69.01% vs 23.91%, P < 0.05), larger LAD (46.93 +/- 0.85 mm vs. 41.76 +/- 1.05 mm, P < 0.05), larger LAA area (8.06 +/- 0.32 cm2 vs 6.92 +/- 0.31 cm2, P < 0.05), smaller MVA area (1.07 +/- 0.23 cm2 vs 1.32 +/- 0.35 cm2, P < 0.05), lower LAA flow peak emphatic velocity (0.17 +/- 0.01 m/s vs 0.32 +/- 0.02 m/s, P < 0.05), and lower LAA full velocity (0.18 +/- 0.01 m/s vs 0.31 +/- 0.03 m/s, P < 0.05). These findings indicate that the presence of SEC is associated with significantly greater incidence of thrombus, higher rate of Af, larger LAD and LAA area, smaller MVA area, lower LAA flow peak emphatic velocity and full velocity. Atrial fibrillation, the diameter of left atrium and the area of mitral valve are independent predictors of SEC. SEC is significantly associated with LAA function and thrombus formation.

[The experimental study of intracellular actin in the scar fibroblasts].

OBJECTIVE: To study the relationship between intracellular actin and scar contracture. METHODS: Fibroblasts from 10 cases of hypertrophic scar and 5 cases of keloid were cultured in vitro. Total actin, filamentous actin(F actin), globular actin (G actin) and the ratio of F to G actin(F/G) were measured by densitometry after differential extraction and separation by polyacrylamide gel electrophoresis in the presence of sodium sulfate. RESULTS: Total actin, F actin, G actin and F/G in hypertrophic scar fibroblasts were 2.38 ng/10(4) cells, 0.98 ng/10(4) cells, 1.42 ng/10(4) cells and 0.68 respectively, while in keloid fibroblasts were 1.68 ng/10(4) cells. 0.46 ng/10(4) cells, 1.26 ng/10(4) cells, and 0.36 respectively. There was significant differences between two tissues fibroblasts in the items of total actin, F actin, G actin, and F/G (P < 0.01), while no significant difference in G actin (P > 0.05). CONCLUSION: Total intracellular actin, F actin, and F/G may play an important role in the scar contracture. The hypertrophic scar and keloid can be distinguished by the contents of total intracellular actin, F actin and F/G.

[Effects of human chorionic gonadotropin on cellular cAMP levels in human gastro-adenocarcinoma SGC-7901 cells].

The purpose of this study was to determine the effects of hCG on cellular cAMP levels in gastric adenocarcinoma cells(SGC-7901). Radioimmunoassay was used. The results showed that hCG obviously increased cAMP levels in SGC-7901 cells in a dose-dependent and time-dependent manner. When the concentration of hCG increased from 10(-11) mol/L to 10(-6) mol/L, the cellular cAMP levels increased from about 2.1 pmol/mg protein to about 10 pmol/mg protein and were significantly different from the control(P < 0.01). These results suggest that hCG possibly exerts its effects on gastric cancer cells by activating adenylyl cyclase/cAMP signal transduction pathway.

[Transthoracic three-dimensional echocardiography for determination of left ventricular mass in patients with dilated cardiomyopathy].

This study was performed to compare left ventricular mass (LVM) measurement by one- (1D), two- (2D) and 3D echocardiography with magnetic resonance imaging (MRI) in patients with dilated cardiomyopathy (DCM). 36 patients with DCM underwent imaging by conventional 1D and 2D echocardiography as well as transthoracic 3D echocardiographic data acquisition. Also patients were imaged with cardiac MRI. LVM was determined. There was no significant correlation (r = 0.42, P > 0.05) for measuring LVM between 1D echocardiography and MRI in patients with DCM. A significant correlation was obtained between 2D echocardiography and MRI (r = 0.64, P < 0.01) and between 3D and MRI (r = 0.78, P < 0.01) in determination of LVM. Compared with 1D and 2D echocardiography, the 3D analysis achieved a significantly higher agreement with the MRI results (1D; 399.2 g, 2D: 285.9 g, 3D: 172.6 g versus MRI: 199.1 g). Interobserver variability was 5.1% for measuring LVM by 3D echocardiography. These results suggest that LVM of patients with DCM can be accurately determined by transthoracic 3D echocardiography and this method could be used in clinical practice.

[Immune responses of a recombined live Salmonella typhimurium SL3261 expressing a multi-epitope antigen of HCV].

A multi-epitope antigen gene of hepatitis C virus(HCV) was fused to beta-galactosidase gene and introduced into attenuated Salmonella typhimurium SL3261 to construct HCV recombined live vaccine candidate SL3261 (pWR/PCX). when the oral live bacteria were used to immunize mice or rabbits, specific anti-GZ-PCX IgG was detected at week 6 and the strongest antibody responses happened at week 12 at a titer of 1:800 and 1:25,600 in mice and rabbits, respectively, which showed significant difference compared with those of SL3261 and blank controls. Anti-GZ-PCX sIgA in mice's intestine and anti-LPS antibody in sera were also detected. The oral live bacteria elicited obvious DTH reaction and proliferation response of peripheral lymphocytes by GZ-PCX antigen. The body weight of immunized mice slightly decreased but no other toxic effects was observed, which showed the safety of oral immunization. The study of oral live HCV multi-epitope vaccine might be able to provide a new route for the researches of HCV vaccines.

[Synthesis of droloxifene citrate and its new bioactivity].

AIM: To investigate a feasible synthetic procedure of droloxifene and study on its new bioactivities. METHODS AND RESULTS: Droloxifene was synthesized using methoxybenzene and phenylacetic acid as starting materials, via Friedel-Crafts acylation, alkylation, demethylation, etherification, Grignard addition, elimination-dehydration, conversing configuration and forming citrate, totally 8 steps, overall yield 14.7% (9.2%). By pharmacological test, droloxifene citrate shows two new bioactivities: (1) obvious effect on reversing the MDR of K562/A02 cells and modulating mdrl, GST pi and TopoII alpha expression. (2) inducing apoptosis in cultured rat luteal cell. CONCLUSION: The improved synthetic procedure is shorter than the reported method by two steps and has advantages of simple separation, purification and conversion of configuration; easily available starting materials and reagents; concise operation. Two new bioactivities on reversing MDR and inducing apoptosis of luteal cell offered a clue in new drugs research and widened clinic application of droloxifene.

[Interactions of insulin with dipalmitoylphosphatidylcholine liposomes].

AIM: To study the interactions of insulin with dipalmitoylphosphatidylcholine liposomes. METHODS: The liposomes were prepared by reverse-phase evaporation vesicle method. The entrapped efficiency, size and distribution of the liposomes were determined, and the influences of insulin on entrapped efficiency, size and distribution of the liposomes were investigated. The influences of liposomes on the fluorescence emission spectra of insulin and the calcein leakage from the liposomes entrapped calcein induced by insulin were measured. RESULTS: Insulin has little influence on the size and distribution of the liposomes while the sizes of the liposomes were about 170-190 nm. The insertion of tyrosine of insulin into dipalmitoylphosphatidylcholine liposomes membrane was not deep. The insulin disturbed the liposomes membrane, induced the calcein leakage from the calcein-loaded liposomes. CONCLUSION: Amphiphilic, example insulin, may disturb the intact membrane of liposome through the interaction either hydrophobic or hydrophilic. The attention should be paid to the entrapment process of peptides into liposomes.

[Effect of qiongyugao on the action of DDP in inhibiting the division of GLC-82 cell strain in vitro].

OBJECTIVE: To observe the effect of Qiongyugao (QYG) on the action of DDP in inhibiting the division of GLC-82 cell strain in vitro. METHODS: The method of serum pharmacology was used in observing the morphological change of the cancer cells. By accounting the number of the live cells of each day during 6 days respectively, the growth curve was drawn up. The effects of the three groups on the cancer cells apoptosis of GLC-82 cell strain were analysed by FACS. RESULLTS: The growth of the cancer cells in control group (normal saline) was much better than the other two groups; in chemical therapy group (DDP), the number of the live cells almost kept steady; the number of the live cells in combined group(DDP plus QYG) became even lower. QYG could enhance the action of DDP in inducing the apoptosis of GLC-82 cells strain remarkably. CONCLUSION: QYG can enhance the effects of chemotherapy in inhibiting cancer cells division and inducing their apoptosis.

Spatio-temporate expression of P53, Bcl-2 and Fas in the retina of RD, Rds and C3H mice.

PURPOSE: To investigate the expression status of p53, Fas and bcl-2 in the development of retinal degeneration in C3H, rd and rds mice. METHODS: Expression of p53, Fas and bcl-2 in the retina of rd, rds and C3H mice as well as in normal C3B mice in different periods was examined by immunohistochemical technique. RESULTS: The expression of Fas and p53 was not detected in normal C3B mice. Fas and p53 were expressed in the ganglionic layer in the early stage and then in the inner nuclear layer, while the scale and intense increased in the ganglionic layer. There is no difference of bcl-2 expression between the normal mice and mice with retinal degeneration. CONCLUSION: Fas and p53 may be involved in the retinal cell death in rd, rds, and C3B, but bcl-2 may not. It is unknown why p53 and Fas appeared first in ganglion layer but not in the outer nuclear layer where retinal cells death was noticed in early stage.

[Laryngostroboscopy in the monitoring of precancerous lesion of vocal cord and in canceration tracing].

OBJECTIVE: To evaluate the value of laryngostroboscopy in the monitoring of precancerous lesion of vocal cord and in the canceration tracing. METHODS: Fifty-two cases of precancerous lesion of vocal cord were examined under Laryngostroboscope. The modifications of mucosal wave and amplitude of vocal cord vibration were observed. All cases were monitored for 1-6 years. RESULTS: The mucosal wave was weak in 45 of 52 cases (86.3%) and the amplitude of vocal cord vibration was weak in 4 of 52 cases (9.6%) with precancerous lesions. However, the mucosal wave disappeared and the amplitude of vocal cord vibration weakened in all 14 cases with early glottic cancer. CONCLUSION: Laryngostroboscopy can be used in diagnosis of glottic precancerous lesion and distinguishing it from the early glottic cancer. It is also an effective means of canceration tracing.

[Partial laryngectomy plus radiotherapy versus partial laryngectomy alone for laryngeal carcinoma].

OBJECTIVE: To investigate the effect of radiotherapy in the combined treatment of laryngeal cancer with partial laryngectomy. METHODS: One hundred and seventy-one cases of laryngeal cancer were randomly separated into three groups of partial laryngectomy, partial laryngectomy with preoperative radiotherapy (dose 40-50Gy) and partial laryngectomy with postoperative radiotherapy(dose 51-70 Gy). The surgical procedures included cordectomy, vertical partial laryngectomy, horizontal partial laryngectomy and near total laryngectomy. RESULTS: The overall 5-year survival rate was 82% (123/150). The 5-year survival rates for surgery alone group (56 cases), surgery with preoperative radiotherapy group(65 cases) and surgery with postoperative radiotherapy group(50 cases) were 85.7%, 80.7% and 79.5% respectively. There was no significant difference among the groups(chi 2 = 0.703, P = 0.704). Analysis of survival rates revealed a significant difference among the different stages(chi 2 = 12.248, P = 0.007). All 171 cases except 3 that had near total laryngectomy achieved satisfactory phonation. CONCLUSION: Neither preoperative radiotherapy nor postoperative radiotherapy can improve the 5-year survival rate of laryngeal cancer with partial laryngectomy. All patients with partial laryngectomy got satisfactory phonation.

[The comparison of luminescence properties for Ce3+, Tb3+ ions in the fluorides].

The matrix K3AlF6 was prepared with a reaction of mixture of solution K2CO3 upon certain solution of Al(OH)3 plus HF (40%), dehydrating at 100 degrees C and annealing for 48 h at 600 degrees C. A series of phosphors for Ce3+, Tb3+ ions in complex fluorides KBF4 and K3AlF6 were synthesized by a solid reaction method. Their excitation and emission spectra were determined, compared with phosphors KAlF4: Ce, Tb, CaF2: Ce, Tb and AlF3: Ce, Tb. The results indicated that Ce3+ could only effectively sensitize Tb3+ in KAlF4 and enhance the emission of Tb3+ significantly. Ce3+ can not effectvely transfer energy to Tb3+ in matrixes CaF2, AlF3, KBF4 and K3AlF6.
