Arsenite activation of P13K/AKT cell survival pathway is mediated by p38 in cultured human keratinocytes.

BACKGROUND: Arsenic has been considered as a carcinogen. Recently the issue of arsenic in drinking water raised an unprecedented social concern on human health, and yet the molecular mechanisms through which arsenic induces cancer remain unknown. Activation of cell survival pathway leading to the activation of eNOS has been associated with various types of cancer. The objective of this study was to investigate the pathway leading to the activation of eNOS in response to arsenite using human keratinocytes. MATERIALS AND METHODS: Cultured keratinocytes (HaCat cells) were exposed to arsenite with or without pretreatment of various inhibitors. Western blot analysis was performed to determine the activation of p38, AKT, eNOS. EGFR tyrosine phosphorylation was detected by immunoprecipitation and Western blot analysis. pNPP assay was used to measure phosphatase activity in cell lysate. FACS analysis was performed for the determination of generation of reactive oxygen species. RESULTS: Arsenite induced the activation of AKT at both Ser473 and Thr308, and its downstream effector eNOS in cultured human keratinocytes. Arsenite also induced phosphorylation of p38. PI-3-kinase inhibitors, Wortmannin and LY294002 inhibited arsenite-induced phosphorylation of AKT and eNOS but had no effect on phosphorylation of p38. Interestingly, however, SB203580, a known p38 inhibitor, completely inhibited arsenite-induced phosphorylation of AKT and eNOS. Arsenite induced generation of reactive oxygen species and inactivated phosphatase activity, but did not activate EGF receptor tyrosine phosphorylation. CONCLUSIONS: Collectively, our data indicate that arsenite induces activation of AKT and eNOS, via PI-3-kinase and p38 pathway, likely bypassing the activation of EGF receptor in cultured human keratinocytes.

Effect of fetal spinal cord graft with different methods on axonal pathology after spinal cord contusion.

OBJECTIVE: To investigate the effect of fetal spinal cord (FSC) graft with different methods on axonal pathology and neurological function recovery after spinal cord injury (SCI). METHODS: Forty Wistar rats were divided into 4 groups. In Group A, the spinal cord was injured and hemisected. In Group B, fetal spinal cord (FSC) was transferred into the injured site. In Group C, after having done as Group B, the upper and lower spinal nerve roots were anastomosed. And in Group D, after having done as Group B, the pedicled omentum was transferred into the hemisection cavity. At 6 weeks after operation, light and electronic microscopes were used to examine the axonal pathology. The neurological function was assessed with inclined plane tests in the open field. The number of axons was quantitated by a computer image analysis system. RESULTS: A greater loss of axons was observed in Group A than that of other groups at 6 weeks. The sequence of the reduced rate of the axons was as following, Group A>Group B>Group C>Group D (P<0.05). The remaining axons were paralleled with the significant improvement in neurological function recovery of the rats. CONCLUSIONS: It indicates that FSC and pedicled omentum grafts after SCI can protect the axons and promote the neurological function recovery of the rats.

[Experimental study and application of extracellular matrix of conjunctiva].

OBJECTIVE: To investigate the characteristics and application of extra-cellular matrix (ECM) of conjunctiva. METHODS: Pieces of conjunctival ECM were made, and then the conjunctival defect was repaired with that in an experimental group. Control group 1 received the heterogeneous conjunctiva, and in control group 2, the refrigerated ECM was used for transplantation. All of them were photographed and examined by light microscope, electron microscope (EM), immunohistochemical examination and lymphocyte toxicity test. RESULTS: There were blood vessels growing into the grafts on the 3rd day in the experimental group. The grafts became mildly congestive and similar to normal conjunctival ones in the 4(th) week. But in control group 1, the grafts became white and necrotic in the second week; the conjunctiva was congestive, cicatricial and hyperplastic in the 4(th) week. Under the light microscope, the epithelium covered the most part of the region with the graft in the 2nd week, covered completely in the 4(th) week, and its appearance was as an approximately normal conjunctival epithelium in the experimental group. But the epithelium became ulcerative and a lot of lymphocytes infiltrated in the 2nd week in control group 1. Under the EM, the line of demarcation between the covering region of the regenerative conjunctival epithelium and the naked ECM was quite clear in the 1st week in the experimental group. And the ultrastructure of regenerative conjunctival epithelium was basically the same as an normal one in the 8(th) week. Both before and after transplantation, the immunohistochemical examinations verified that ECM was type I and IV collagen. The lymphocyte toxicity test showed that no obvious humoral immunoreaction existed. CONCLUSION: Conjunctival ECM has the characteristics of tissue activity and low antigenicity. It is an ideal conjunctival succedaneum in the plastic surgery of conjunctiva.

[The dynamic changes of serum TNFalpha level in scalded rats after the administration of rhGH during the early postburn stage].

OBJECTIVE: To examine the dynamic change of serum TNFalpha level in scalded rats after the administration of rhGH during the early postburn stage. METHODS: Forty-two male sanitary Sprague-Dawley rats were randomly divided into three groups, i.e. normal control (N), burn with early administration of rhGH (BA) and burn without rhGH (BN) groups. The rats were inflicted with 25% similar 30% TBSA deep partial thickness scalding on the back. rhGH in dose of 6 U/kg was administered subcutaneously via abdominal skin starting from the 1st postburn day (PBD). Rat serum TNFalpha levels were determined by ELISA in BA and BN groups at 3, 6 and 10 PBDs. RESULTS: The rat serum TNFalpha level in BA was lower than that in BN groups on 3 PBD. Whilst the TNFalpha levels in BA were much higher than those in BN groups on 6 and 10 PBDs. CONCLUSION: Administration of rhGH at early postburn stage might abate the early systemic postburn reaction.

[Cloning of HAL1 gene and characterization for salt tolerance tomato].

The HAL1 gene was cloned by PCR strategy and confirmed by sequencing. Its open read frame is 879 bp, encoding a peptide of 294 amino acids (32 kD Protein). A chimaeric construct of HAL1 and Npt II (neomycin phosphotransferase II) was constructed and introduced into commercial cultivars of tomato (Zhong SU No. 5: Lycopersicon escullentum) by Agrobacterium tumefacien-mediated gene transformation. Transformants were selected for their ability to grow and root on media containing kanamycin. Transformation was confirmed by analysis of PCR, Southern blot and RT-PCR. The salt tolerance of transgenic tomato is evaluated by comparing the fresh weight, dry weight, Na+, K+ content of transgenic tomato and control tomato. It is concluded that the over-expressing of HAL1 in tomato could enhance the salt tolerance of the transgenic tomato.

[Evaluation clinical long-term curative effect and analysis of its influence factors after restoration of teeth with wedge-shaped defects].

OBJECTIVE: To evaluate the long-term curative effect clinically and analysis of its influence factors after restoration of teeth with wedge-shaped defects. METHODS: Seventy five cases of 509 teeth with various degrees of dental wedge-shaped defects were randomly divided into two groups--the group restored by Dyract Cement and the group restored by 3M-Vitremer. Evaluation was made with the modified USPHS evaluation criterion after restoration for 1,2,4 years respectively. RESULTS: The successful rate dropped down from 2 year successful rate 85.5% to 4-year 27.7%. The new wedge-shaped defects were presented at the cervical margin of the original restoration in 1.8% (2 years later) and 6.6% (4 years later). There were a lot of influence factors on long-term curative effect. The failure rate in the group with occlusal interference was significantly higher than that without occlusal interference. CONCLUSIONS: The clinical successful rates are affected by the value of occlusal force on the restored teeth and the physical properties of the restorative materials. It is suggested that the process of restoration of dental wedge-shaped defect should include obviating occlusal factors as well as to improve the properties of restorative materials.

[Study on gas chromatographic method for the evaluation of residual solvents by using wide bore open tubular columns].

A GC method using wide bore open tubular columns was developed to evaluate residual organic solvents. Sixteen organic solvents commonly used for pharmaceutical synthesis were analysed. The effects of different polarity columns and extraction solvents on the assay of residual solvents were studied in this work. It showed that most solvents can be well separated from each other on non-polar or less polar wide-bore columns. Extraction solvents differed in their extraction recovery and peak shape. Satisfactory results can be obtained when the proposed method was employed and a suitable extraction solvent was selected. The procedures summarized in this paper can be used as a guidance for the evaluation of residual organic solvents in bulk pharmaceuticals.

[Recognition and quantitative contrast characteristic components for root of Chinese angelica].

A method of multi-component quantitative analysis in root of Chinese Angelica has been developed. The analytical mode was reversed-phase HPLC with photodiode array detector. Based on comparison of UV spectra, a software of UV spectra database to assist recognition has been established. The solvent system, a gradient program of mobile phase with 2-propanol/acetonitrile/water can separate a lot of liposoluble components in root of Chinese Angelica. UV detection wavelength was set at 202 nm and the flow rate was 1 mL/min on a Hypersil ODS column. Twenty-three components in two roots of Chinese Angelica were recognized using characteristic parameters of chromatography and UV spectroscopy. It is the base of quantitative contrast to recognize many characteristic components without standard components in root of Chinese Angelica. For two samples of root of Chinese Angelica both have 23 characteristic components and the concentrations of some components were similar and those of others were different. The method is convenient and reproducible. It can be used for the quality control of the root of Chinese Angelica without the use of standards.

[Study of unified analytical method of compatibleness for traditional Chinese medicines. I. Separation, recognition of characteristic components and fingerprint analysis for single and complex medicines].

It is difficult to separate, identify and quantify multi-component in complex traditional Chinese medicines (TCM). Many analytical methods for TCM have been established, but only a few components can be separated and quantified. For analyzing the multi-component and finding new method of compatibility analysis, a strategy--the multi-mode and multi-column chromatographic system and unified analytical method has been proposed by us. A part of the results from the study of astragalus root(A), Chinese angelica root(Ca.) and their complex prescription (Danggui Buxue Tang, DBT) with the analytical mode of reversed-phase HPLC/photodiode array detector as an example of the method for unified separation and recognition of characteristic components and fingerprint analysis was introduced. This is the first part of the results of compatibleness analysis for TCM.

[The effect of temperature on the enantiomeric resolutions on albumin and beta-cyclodextrin chiral stationary phases].

The effect of column temperature on enantiomeric resolutions of tryptophan, warfarin and ketoprofen was investigated on bovine and human serum albumin (BSA and HSA) stationary phases, which were synthesized with s-triazine as the activator. It was observed that the entropy change made a great contribution to the separation of those enantiomers on albumin chiral stationary phases. The column temperature for the maximal resolution of tryptophan on the BSA stationary phase prepared by this method was about 35 degrees C, which was not 24 degrees C as reported for the BSA stationary phase synthesized with glutaric dialdehyde as the activator. This results may come from the different conformation of the immobilized BSA and HSA due to the different coupling methods used. On the other hand, it was indicated that the resolution of enantiomers on beta-CD chiral stationary phase was mainly contributed from the change of enthalpy, which means the resolution of chiral solutes on albumin and beta-CD stationary phases has different thermodynamic behaviors.
