[A study on the epidemiology and preventive measure on epidemic haemorrhage fever in army barrack area in Shandong].

OBJECTIVE: In order to guide the army and civilians to carry out the prevention on epidemic haemorrhage fever (EHF), it's necessary to elucidate the epidemic feature of natural epidemic foci and the risk factors of EHF in the army - stationed areas. METHODS: Samples were tested by pathogenic and serologic means before data was collected and analyzed by case - control or family - control studies. RESULTS: Cases belong to house - mouse type (91.48%), field - mouse type (7.21%) and unclassified (1.31%) coexist in the stationed area. In the two types of noticed cases, no significant difference in distributions of sex, profession and ages. Most house - mouse type cases occurred in the peak time from March to June and field - mouse type in November to next February. The type of EHF virus antigen of mice and the type of serum were both accordant with their host types. Dwelling in border area, living rooms with mice, dogs raising, injury of mouth tissue, eating food contaminated by mice, and household with mice living around were more frequently seen in cases themselves and in families of the cases than in controls of both groups (P < 0.05). The mice intensity could be under control to 1.0%, if comprehensive measure on mice control can be implemented. CONCLUSIONS: The stationed area is an epidemic area mixed with both house - mouse and field - mouse types, with majority of house - mouse type. It was house - mouse type virus infection that caused the epidemic area expanded and the number of cases increased radically these years. EHF was epidemic in spring and summer season in the stationed area mainly because the infections were among family members. Food contaminated by mice or close contact with mice played an important role. Mice eradication and prevention were the leading measures to prevent EHF, from our more than ten years of experiences.

[Investigation on natural foci of autumn-winter type tsutsugamushi disease in Shandong province].

OBJECTIVE: In order to provide basic data of tsutsugamushi disease in Shandong province, for the control and prevention of this disease in residents and army-men. METHODS: Aetiological, serological and epidemiological methods were used to investigate the natural foci of autumn-winter type tsutsugamushi disease. RESULTS: The peak of tsutsugamushi disease epidemic in Shandong province was in October with cases in this month accounted for 80% of all in a year, that showed autumn-winter type. The main reservoir hosts were Apodemus agrarius, Rattus norvegicus and Cricetulus triton, accounted for 82.87%, 85.47% and 13.04% respectively. It was confirmed that natural infection existed among Leptotrombidium (L.) scutellare, L. palpalis, L. linhuaikonense and Walchia pacifica. The main vector of transmission was Leptotrombidium scutellare. 41 strains of Rickettsia tsutsugamushi were isolated from the blood samples of patients, rodents and chigger mites but with weak toxicity. 90% of the serotypes of the strains belonged to Gilliam type. The distribution of the cases had a sporadic nature. More than 80% of the cases were young or middle aged peasants. Clinical symptoms were mild, with some differences in different areas. CONCLUSION: The natural foci type of tsutsugamushi disease in Shandong could be divided into plain and hilly types.

[Inhibition of monocytes adhesion to the intima of arterial wall by local expression of antisense monocyte chemotactic protein-1].

OBJECTIVE: To study the mechanism of monocyte recruitment in atherogenesis and to clarify the effect of monocyte chemotactic protein-1 (MCP-1) in this process. METHODS: Femoral arteries isolated from the rabbits which had been fed with a high cholesterol diet and locally perfused with MM-LDL within the artery beforehand, were used as the models. Antisense MCP-1cDNA was transferred into the arterial wall by injecting recombinant LNCX-anti-MCP-1/liposomal complex in the femoral sheath and the periarterial tissue. RESULTS: Expression of antisense MCP-1 mediated by recombinant LNCX plasmid/lipsomal complex gene transfer enabled to inhibit MCP-1 gene expression and adhesion of monocyte to the intima. CONCLUSION: MCP-1 plays an important role on the recruitment of monocytes in the arterial wall, which provides a potential clue in developing a gene therapy project for the prevention and treatment of atherogenesis.

[P15(INK4B) gene methylation in malignant hematopoietic diseases].

OBJECTIVE: To study the effect of operative region hypermethylation gene in human malignant hematopoietic tumors. METHODS: The abnormal methylation rate of P(15)(INK4B) gene 5'CpG island in 68 cases of malignant hematopoietic tumor samples were determined by methylation specific PCR using bisulfite modified DNA. RESULTS: The methylation rates of P(15)(INK4B) were 84%, 0, 50% and 75%, respectively, for 25 cases of acute myeloid leukemia (AML), 15 chronic myeloid leukemia (CML), 16 myelodysplastic syndrome (MDS) and 12 multiple myeloma (MM). P(15)(INK4B) gene was frequently methylated in patients with high risk MDS and early stage of MM. CONCLUSION: Hypermethylation of P(15)(INK4B) gene is one of the main causes of its inactivation. Hypermethylation of CpG island was closely related to the development of malignant hematopoietic diseases.

[The neutralizing effect of histidine-rich-polypeptides on LPS from suspected periodontal pathogens].

OBJECTIVE: Human parotid histidine-rich-polypeptides (HRPs) are a family of lowmolecular-weight, cationic polypeptides. HRP-1, HRP-3 and HRP-5 comprise 85%-90% of the total HRPs and are called major HRPs. There were many researches indicating the antimicrobial activities of HRPs. Recently, HRPs were reported to have an inhibitory action on the lipopolysaccharide (LPS) of E. coli, with HRP-5 being the most effective one among the three major HRPs. Since the LPS of oral gram-negative bacteria is thought to be one of the important etiological factors during the development of periodontal diseases, our experiment was aimed to investigate the neutralizing effect of human parotid HRP-5 on the LPS of anaerobic suspected periodontal pathogens, which have different chemical structures and biological activities compared with LPS of aerobic E. coli. METHODS: By using of preparative acid urea polyacrylamide gel electrophoresis (AU-PAGE), HRP-5 was purified from parotid saliva collected from healthy adults. Two stains of suspected periodontal pathogens, Porphyromonas gingivalis (P.g, 47-A) and Fusobacterium nuceatum (F. n, separated from subgingival plaque of a patient with adult periodontitis) were mass cultured. After harvested in the midlogarithmic phase, they were washed and lyophilized. The LPS of dried bacteria were extracted by the modified Westphal hot phenol-water procedures and purified by the enzyme digestion plus ultracentrifugation. Limulus test was applied to test the neutralizing effect of HRP-5 on the LPS-induced gelation of Limulus amoebocyte lysate. In brief, the standard LPS of E. coli, or extracted LPS of P. g or F. n, was preincubated with HRP-5 in a tube separately in room temperature for 10 minutes. Then the reagent of Limulus amoebocyte lysate was added in all the tubes, continued the incubation in 37 degrees C for one hour. After that, the gelation level of every tube was observed. RESULTS: LPS extracted from P. g and F. n both showed good purity and strong activities to induce gelation of Limulus amoebocyte lysate. The gelation induced by LPS (1 ng/ml) of these two anaerobic suspected periodontal pathogens were weakly inhibited by HRP-5 (10 micrograms/ml), similar to that observed with standard LPS of E. coli. To get a complete neutralizing effect on LPS, it may be important to increase the concentration of HRP-5. CONCLUSION: It appeared that HRPs could neutralize the endotoxic properties of LPS of suspected periodontal pathogens, therefore may contribute to periodontal health. The present investigation further confirmed that HRPs are important components of the host non-immune defense system.

[Enhancement of BCG-induced hBD-1 mRNA expression in human pulmonary gland epithelial cells].

Using reverse-transcription PCR (RT-PCR) and Northern blot analysis, the present authors found an enhanced expression of the human beta-defensin 1 (hBD-1) gene in the SPC-A-1 cells challenged with heat-killed M. Bovis Bacille Calmette-Guerin (BCG). The enhanced expression of hBD-1 mRNA was dose- and time-dependent. This result indicated a significant progress in the work of developing a new antimicrobial strategy that enhances mucosa antibiotic peptide expression for the prevention and treatment of mucosal infections.

[Studies on the correlation between production of L-malic acid and some cytosolic enzymes in the L-malic acid producing strain Aspergillus sp. N1-14].

The cytosol enzymatic study in the case of high L-malic acid(LMA) production of Aspergillus sp. N1-14' was reported. The activities of 4 kind enzymes that catalyse the CO2 fixation reactions have been detected, which are pyruvate carboxylase(PC), phosphoenolpyruvate carboxlase (PEPC), phosphoenolpyurvate carboxykinase(PCK) and malic enzyme(ME). With the exception of ME, the linear correlation was found between activities of three carboxlases and the production rate of LMA. The activity of malate dehydrogenase(MDH) was at the level of 2-3 exponential higher than that of the other analysed enzymes, while the activity of succinate dehydrogenase(SDH) was much lower, and as a discrepancy, SDH was in a positive correlation to the content of LMA in fermenting slurry(r = 0.9252). It is shown that the accumulated LMA acted as an activator of SDH. Through dynamic study, it is found that, in contrast with the slow and even increase of biomass, the content of cytosol protein(Cp) sharply fluctuated mainly due to the changes of aeration conditions. The data of the linear correlation coefficients(r) of activities of cytosol enzymes to Cp(PC r = 0.9563, PEPC r = 0.7688, PCK r = 0.7300, MDH r = 0.3920, SDH r = -0.2086) exhibited an inner law of protein synthesis. Experiment of increasing the amount of spore inoculum resulted in increase of LMA and decrease of SA. After fermenting 120 h in a 5 L stirred fermentor, with 3-fold of original spore inoculum 105.88 g/L of LMA was achieved, the overall productivity was 0.883 g/(L.h), the converting rate of glucose to LMA was 78.43%. This result supports the exist of a inner law of protein synthesis in the early period of LMA fermentation by Aspergillus sp. N1-14'.

[Study on erzhi pill effects on neuro-endocrinologic-immuno network of animal of deficiency of yin].

OBJECTIVE: To investigate Erzhi Pill effect on Neuro-Endocrinologic-Immuno network of animal of deficiency of Yin. METHOD: In the experiment through the determination of hemotic cortisol, insulin, testosterone, beta-endorphin, cholesterol, weight of adrenal and thymus, ConA-induced, spleelymphocyte proliferation and activity of natural killer cells (NKC). The changes of the animals of deficiency of Yin and the effects of the Erzhi Pill were investigated. RESULT: It showed that rats of deficiency of Yin, their hemotic, cortisol, insulin, testosterone, beta-endorphin, weight of adrenal and thymus decreased, cholesterol increased and mouse of deficiency of Yin, their lymphocyte proliferation and activity of NKC dcreased, which might be modulated with Erzhi Pill. Erzhi Pill could improve hemotic cortisol, insulin, testosterone, beta-endorphin, weight of adrenal and thymus, reduce hemotic cholesterol, improve lymphocyte proliferation and activity of NKC. CONCLUSION: This principle could improve animals of deficiency of Yin regulating Neuro-Endocrinologic-Immuno network and keeping homeostais.

[Study on inhibitory actions of san huang yi gan capsule (SHYGC) on HBeAg with seropharmacological method].

In this paper, the inhibitory actions of SHYGC on HBeAg in vitro were studied with the seropharmacological method. By Enzyme Linked Immuno Sorbent Assay (ELISA), it was found that the rabbit sera containing SHYGC have significant inhibitory effects on HBeAg, and they become stronger with the drug concentration in sera improved and the actions time prolonged, they decrease with the HBeAg concentration improved, and the effects of the sera containing hing-dose drugs equal those of Su Xiao Jing containing 250 micrograms/g effective chloric. The direct external inhibitory effects of SHYGC in original pharmaceutics are stronger than those of the sera containing drugs. It probably indicated that the active ingredients of SHYGC could not be digested and absorbed completely from gastrointestine, or were inactived by metabolism in vivo.

Experimental study of homium: YAG laser transmyocardial revascularization in acute ischemic settings with myocardial contrast echocardiography.

OBJECTIVE: To study the mechanism and effects of blood perfusion to the acute ischemic region of myocardium through Ho-YAG laser channels with myocardial contrast echocardiography. METHODS: To produce the model of acute myocardial ischemia, we partially ligated the left anterior decending (LAD) coronary artery of canine hearts between 1st. and 2nd. diagonal branches and then performed transmyocardial revascularization in this region with Ho - YAG laser. Myocardial contrast echocardiography was made with a new generation of ultrasound contrast agent and second harmonic imaging of this region before, after ischemia and after laser revascularization. Pictures were taken with "R" wave trigger skill. RESULTS: Acoustic density determing in the ischemia region (anterior wall) with MCE (myocardial contrast echocardiography) was obviously decreased (5.40 +/- 1.81) after the LAD was ligated,as compared with before (11.69 +/- 1.61, P < 0.01). It was increased remarkably after transmyocardial laser revascularization (TMLR) (11.2 +/- 2.01, P < 0.01) as compared with that when ischemia and approximated to that before ischemia (P > 0.05). There were no differences in acoustic density in the lateral wall (as control) among these comprehensive three periods (P > 0.05). Contrast in the laser region developed one cardiac cycle ahead of that in the non-ischemic normal region. CONCLUSION: Acute ischemic myocardium can be perfused by oxygenated blood from the left ventricle through Ho - YAG laser channels. Evidence of blood perfusion through laser channels during systolic phase was detected, and myocardial contrast ultrasonography using intravenous perfluorocarbon-exposed sonicated dextrose albumin may be regarded as a reliable method in the study of transmyocardial revascularization.

[Vibrational spectroscopic investigation of a series of heterometallic cubane-like M'-Mo (W) cluster compounds].

By combining the results of IR spectroscopic experiments with the molecular structures, the main stretching vibrations of a series of the heterometallic tetranuclear cluster compounds with [M3YS3M'] cluster core (M = Mo, W; Y = O, S; M' = Cu, Sn, Pb, Sb, Bi) have been assigned. The characteristic features and the bonding situations of the five configuration types are discussed. It is shown from the IR and Raman spectra that when M' is a transition metal such as Cu, the nu M's frequency of the [M3YS3M'] core has a value comparable to that of corresponding M'-S covalent bond thus indicating a close combination of M' with [M3YS3]. Whereas when M' is a main group metal, the nu M'-s vibration is always observed in a lower region than of the M'-S covalent bond because of a loose combination of M' with[M3YS3]. However, remarkable enhancement of M-M band strength due to the addition of M' is demonstrated by Raman spectra. Some comments for the M-M vibration of various structures of these compounds are also described.

[In situ time-resolved FTIR studies of HCOOH oxidation on Pt(100)/Sb electrodes].

The electrocatalytic oxidation of formic acid on Pt(100)/Sb electrodes of different coverage of Sb was investigated by using electrochemical techniques and in situ time-resolve FTIRS. The results demonstrated that the dissociative adsorption of formic acid was inhibited by the presence of Sbad on Pt(100) surface, as a consequence HCOOH can be oxidized directly into CO2 via reactive intermediates. The rate of oxidation of formic acid varied with the coverage of Sbad. It was revealed that at a Sbad coverage of 0.24 the Pt(100)/Sb electrode exhibited the highest electrocatalytic activity for HCOOH oxidation.

[In-situ FTIR spectroscopic studies of electro oxidation of ethanol in alkaline media at a nm-Pt/GC electrode].

The electro-oxidation of ethanol on a nm-Pt/GC electrode in alkaline solutions was investigated by using cyclic Voltammetry and in-situ FTIR spectroscopy. The results demonstrated that the main product of ethanol oxidation was CH3COO-, only small quantity of CH3CHO was determined simultaneously. In contrast with the dual path reaction mechanism for the oxidation of ethanol in acid media, the oxidation of ethanol in alkaline media was revealed via the intermediate process of dissociative adsorption.

[Comparison of FTIR microspectroscopy and diffuse reflectance spectroscopy for monitor of solid phase reaction].

17 FTIR micro transmission spectra of single resin beads of each sample, from polystyrene sulfonyl chloride resin changing to polystyrene sulfonyl amide resin in solid-phase reaction in water at 0 degree C, 20 degrees C, 40 degrees C, 60 degrees C, were recorded. The reaction was monitored by the means of FTIR microspectroscopy and diffuse reflectance spectroscopy upon the products. It was showed that the pressed single bead as a piece of biscuit was able to give higher quality spectrum. In comparison, 8 diffuse reflectance spectra of each sample were also collected. It seems that the diffuse reflectance spectroscopy is a more easy dealing with and effective method to control the whole reaction. However, the microspectroscopy is the first choice to check the difference between resin beads.

[In situ FTIR spectrocopic investigations of 1,3-butanediol oxidation on Sb and S modified Pt electrodes].

In the present paper, we studied the surface structure effect of Pt electrode modified with antimony and sulfur adatoms towards 1,3-Butanediol oxidation. The results demonstrated that the modification of Sb and S both inhibited the dissociative adsorption of 1,3-Butanediol into CO, which is the main source of self-poisoning in electrocatalysis of small organic molecules. At lower potentials, the principal oxidation pathway of 1,3-Butanediol on Pt/Sbad electrode is towards the production of C=O. However, at higher potentials, the products of CO2 increased. The presence of S(ad) inhibited the oxidation of 1,3-BD at low potentials(< 0.8 V), and yield mainly acid species as products at higher potentials(> 0.8 V).

[Study on mechanism of different PHAs during heating by FTIR].

In this paper, PHB, P(HO-co-HD), a various composition of P(HB-co-HV) copolymer and P(HB-co-HH) copolymer were studied both at normal temperature and in the process of increasing and decreasing temperature using FTIR spectroscopy technology. Results showed that FTIR spectra of PHAs with various compositions have distinguishing characteristics. Physical transformation occurred when PHAs were treated with heat denaturalization, and this physical process was reversible.

Efficient discovery of inhibitory ligands for diverse targets from a small combinatorial chemical library of chimeric molecules.

Living systems are mainly composed and regulated by compounds in four biochemical classes and their polymers-nucleotides, carbohydrates, lipids, and amino acids. Early combinatorial chemistry libraries consisted of peptides. The present report describes the general bioactivity and biophysical properties of a combinatorial chemical library that used glyco, nucleotidyl, and lipid building blocks. The resulting chimeric combinatorial library of 361 compounds had a confirmed cumulative hit rate of 0.16%, which is 8-fold higher than a commonly claimed industrial benchmark of 0. 02%. It produced 7 structurally confirmed hits for a third of 12 proprietary drug discovery projects, and these comprised a variety of molecular targets. Diversity analyses demonstrated that despite the small number of compounds, a wider range of diversity space was covered by this library of biochemical chimeras than by a branched tripeptide library of the same size and similar generic formula.
