[Three dimensional observation on revascularization and study on bone metabolism of allogenic transplanted mandible].

In order to observe stereological correlation between new vessels and surrounding tissue, 6 rabbits were used for the following experiments. The mandible defect model was made by cutting 1 cm x 0.5 cm bone from both right and left side of mandible. Then, the left defect was repaired by the bone segment from the right side, and the right defect was repaired by frozen allogenic bone segment. One month later, the metabolism of the bone segments was observed by nuclein scintiphotography. The revascularization of the bone segments was observed by vascular corrosion cast method. It was shown that new vessels from host soft tissue could penetrate the periosteum of allogenic transplanted bone, along the absorbing path, ingrowth into the bone. The metabolism of the bone was active. It was suggested that the vessel growth from host to the graft is one of the main patterns of revascularization.

Clinical observation on treatment of 83 cases of posthemiplegic omalgia.

An analysis on 83 cases of posthemiplegic omalgia (shoulder pain) shows that the pathogenesis of the pain is closely related to the improper passive movement at the early stage of hemiplegia (62.7%). The large range of passive movement is a dangerous factor leading to omalgia. In the study of upper extremity complications, the incidence of shoulder-hand syndrome is relatively high (42.2%), and it is often accompanied by hand swelling (83.1%). The authors suggest that painless movement of the shoulder joint should be limited in a range of 90-120 degrees, massage be carried out immediately after acupuncture, and the affected upper extremity be moved passively during the needle retention. This therapeutic method is definitely effective for pasthemiplegic omalgia.

[The dose-response of cell proliferation and secretion of CA125 II antigen to follicle stimulating hormone in ovarian cancer cell lines].

OBJECTIVE: To study the dose-response of the proliferation and secretion of CA125 II antigen with follicle stimulating hormone (FSH) in epithelial ovarian cancer cell lines. METHODS: Two different kinds of ovarian cancer cell lines originated from different pathologic types-AO and 3AO were studied. Cancer cells were cultured in medium containing FSH in different concentrations. The proliferation rate of cells was detected by using the 3H-TdR intrusion technique. The accordant titer of CA125 II antigen in the culture medium was measured by enzyme linked immunoadsorbent assay technique. RESULTS: Proliferation rate of cancer cells (AO and 3AO) was increased apparently as increasing in the concentration of FSH in the culture medium. A positive correlation was found between them (P < 0.05). Proliferation rate of AO cells positively correlated with secreting quantity of CA125 II antigen (P < 0.05), however, no such correlation was found in 3AO cells. CONCLUSION: It suggested that FSH can stimulate the proliferation of epithelial ovarian cancer cell at high concentration and there is a accordant dose-response relationship between them. The secreting pattern of the CA125 II antigen varied by the pathological classification of the ovarian cancer.

[Isolation of antibacterial polypeptides of human cervical mucus].

The acid-soluble extract of human cervical mucus was obtained by solving mucus with 5% acetic acid in the presence of protease inhibitor. The antibacterial activity of the acid-soluble extract was analyzed by gel overlay technique. The result showed that two protein bands which were designated human cervical protein-1 (Hcp-1), human cervical protein-2(Hcp-2) were potently antibacterial against E. coli 25922 and S. aureus 25923. Tricine-SDS-PAGE analysis indicated that Hcp-1, Hcp-2 actually contained three and four protein bands respectively. The molecular weights of Hcp-1 were 6.7 kd, 10 kd, 15.4 kd; these of Hcp-2 were 4.4 kd, 6.7 kd, 9 kd, 15.4 kd. Our studies suggested that human cervical mucosa might secrete some currently-unknown antibacterial polypeptides which play an important role in the cervical defense against infection.

[Partially purified antibacterial polypeptides from granules of human large granular lymphocytes].

This was a study on the antibacterial activity of human large granular lymphocytes (LGL). In our previous work, three antibacterial fractions named HLP-1, HLP-2 and HLP-3 had been identified from the acid-soluble extracts of the granules of LGL by using the gel overlay technique. The present study demonstrated that HLP-1, HLP-2 and HLP-3 separated by preparative acid-urea polyacrylamide gel electrophoresis were potently bactericidal against E. coli MI-35P, S. aureus ATCC 25923, P. aeruginosa ATCC 27853 when tested for their antibacteiral activities by using both agarose radial diffusion assay and gel overlay technique. SDS-PAGE analysis showed that the HLP-3 was almost purified with the molecular weight of 7 kd. The HLP-1 and HLP-2 were more complex, and the ranges of molecular weight were 5.6-13 kd and 5.6-8.8 kd respectively. Our study suggests that the granules of human LGL might contain some low molecular weight antibiotic peptides which play an important role in human innate immunity.

[Changes of the gene expression of vascular endothelial growth factor in the lung of rats with chronic hypoxic pulmonary hypertension].

To observe the effect of chronic hypoxia on the gene expression of vascular endothelial growth factor (VEGF) in rat's lung, and the role of VEGF in the pathogenesis of hypoxic pulmonary hypertension, thirteen male Wistar rats were exposed to isobaric hypoxia for 3 weeks. The pulmonary artery pressure was measured by right cardiac catheterization. The serum level of VEGF was measured by Elisa. The VEGF cRNA was labeled with digoxigenin-UTP by in vitro transcription. The expression of VEGF mRNA in the lung was examined by hybridization in situ. The pulmonary artery pressure was significantly increased after hypoxic exposure. The serum level of VEGF in rats treated with hypoxia (420.3 +/- 73.1 pg/ml) was significantly increased in comparison with that of normal rats (322.2 +/- 58.1 pg/ml). The VEGF hybridization signals on the wall of pulmonary arteriole were significantly increased in rats with pulmonary hypertension. Chronic hypoxia can markedly increase expression of VEGF mRNA in the pulmonary arteriole and hence stimulate VEGF synthesis and secretion. The increase of VEGF may play a role in the developing process of hypoxic pulmonary hypertension.

[The examination of EBV-DNase gene fragment in the paraffin embedded NPC, pre-cancerous and high risk population nasopharyngeal tissues].

OBJECTIVE: To examine if EB virus enters nasopharyngeal epithelial cells in the course of the tumorigenesis in nasopharyngeal carcinoma (NPC). METHODS: Based on the annual investigation of Si-Hui county, a cancer-prevention base of the Sun Yet-sen University of Medical Sciences and Tumour Hospital outpatient clinic, more than 450 paraffin-embedded tissues with NPC, and with pre-cancerous lesions in high risk populations were collected for the detection of EBV-DNase gene fragment by an optimized PCR method. RESULTS: In 145 of the 149(97.3%) cases with invasive NPC, and 2 of 4 cases with in situ NPC, EBV-DNase gene was detected. However, in Sihui, Canton, a high-risk county for NPC, only 2 of the 155 subjects with nasopharyngeal precancerous state was the gene positive, and none of the 47 cases examined in the out-patient department of the Tumor Hospital with precancerous lesions in the nasopharynx was positive. CONCLUSION: EBV is seldomly detected in the non-malignant and precancerous lesions in the nasopharynx in high-risk populations. It implies that EBV is probably not etiologically involved in the genesis of NPC.

[Application of comparative genomic hybridization to hyperdiploid acute lymphoblastic leukemia].

OBJECTIVE: To evaluate the implication of comparative genomic hybridization (CGH) in leukemia study. METHODS: Genomic abnormalities in 14 ALL patients were assayed by CGH, and the results were compared with those of conventional karyotype analysis. RESULTS AND CONCLUSION: Regional and/or whole chromosome over-representation was found to be more frequent than under-representation (43 gains versus 6 losses), the most common gains involved being chromosomes 21 and X. Comparison between the results of CGH and conventional R-banding analysis showed that: 1. In 2 cases with trisomy, both the methods gave identical results. 2. In 8 cases, both the results were consistent excepting for minor discrepancies. 3. In 3 cases, including 2 each with triploidy and tetraploidy respectively, and one with chimeric karyotype of normal/+22, the results from the two methods were discrepant.

Visco-supplementation therapy in internal derangement of temporomandibular joint.

OBJECTIVE: To study if visco-supplementation therapy is useful to the internal derangement (ID) of temporomandibular joint (TMJ). METHODS: Sixty-three ID cases (69 TMJs) were studied by visco-supplementation therapy. The upper and/or lower articular cavities were irrigated with 5 ml normal saline and injected 0.3-1.0 ml 1% hyaluronate (HA) into articular cavity. If the symptoms of the disease still existed one week later, the therapy should be repeated for 1-2 times, once a week. The control group cases were injected 1 ml 2% lidocaine instead of HA. 8 other TMJs of 6 ID cases and 2 normal cadavers were studied with scanning electron microscopy (SEM) and light microscopy (LM). RESULTS: The visco-supplement therapy was useful to ID patients. The difference between the test group and control group had statistical significance (chi 2 = 6.6535, P < 0.01). SEM and LM showed that the condyle, disc and bilaminar region in ID were degenerated or destroyed. CONCLUSIONS: The friction between the articular surfaces in ID was increased and the bilaminar region could not retract the disc as in healthy TMJ. The visco-supplementation therapy can decrease the friction and resume the normal rheology of the diseased TMJs.

[Analysis of reasons failed focal debridement in 109 patients with spinal tuberculosis].

OBJECTIVE: To explore reasons failed focal debridement in treating spinal tuberculosis patients and methods to cope with them. METHOD: One hundred nine patients with spinal tuberculosis who failed initial focal debridement from 1987 to 1996 were reviewed. RESULT: The reasons of operational failure were found to be the use of unreasonable regimens and existence of drug resistant strains; inappropriate operational indication, approach and treatment during operation; lesions involving several vertebrae, skipping destruction and complicating with other kinds of extrapulmonary tuberculosis. CONCLUSION: The key to successful treatment of the disease is to choose appropriate operative indication and approach, discard spinal cord pressure and rebuild spinal stability on the basis of reasonable chemotherapy regimen.

[Treatment of thoracolumbar tuberculosis complicated with paraplegia].

OBJECTIVE: To explore characteristics of thoracolumbar tuberculosis complicated with paraplegia, relationship between efficacy and approach of operation, and to discuss indication and applicability of conservative treatment. METHOD: Forty-eight cases with thoracolumbar tuberculosis complicated with paraplegia were classified into two groups. Thirty-two cases in the operational group were treated with extrapleural, extraperitoneal approach and transpleural, extraperitoneal approach. Sixteen cases in the nonoperational group were treated conservatively. RESULT: The recovered cases in the operational and the nonoperational groups respectively accounted for 75% and 88%. CONCLUSION: Thoracolumbar tuberculosis complicated with paraplegia can be cured by either operative or conservative treatment, and the efficacy of operation directly related with operational approach.

[Clinical observation on nephrotic syndrome of lupus nephritis treated with integrated traditional Chinese and Western Medicine].

OBJECTIVE: To observe the clinical effect of integrated traditional Chinese and western medicine (TCM-WM) in treating nephrotic syndrome of lupus nephritis (NSLN). METHODS: One hundred and fifty-five patients of NSLN were randomly divided into the TCM-WM group and the control group (treated with western medicine alone). Changes of activity index (AI) of lupus nephritis, urinary protein (UP), plasma albumin (PA), blood lipid (BL) of the patients before and after treatment were observed and compared. And the difference on recurrence rate and adverse reaction rate between the two groups were assessed. RESULTS: The total effective rate of the TCM-WM group was 95%, significantly higher than that of the control group (79%, P < 0.05). The effect in improving AI, UP, PA and BL of the TCM-WM group was superior to that of the control group. The recurrence rate and adverse reaction rate of the former group were 8% and 34% respectively, which was much lower than those of the latter (38% and 68%, P < 0.05). CONCLUSION: TCM-WM treatment is obviously superior to treatment with western medicine alone.

[Clinical observation of forty cases in adolescent dysmenorrhea treated by traditional Chinese medicine].

OBJECTIVE: To observe the therapeutic effect of Tongjing powder (TP) and understand the change in pathophysiology of dysmenorrhea, the blood flow parameters of uterine arteries and hemorheological data before and after the TP administration. METHODS: The blood of both uterine arteries of patients was examined by color doppler flow imaging. The hemorheological data was examined by conical viscosimeter. The two groups were compared with each other. RESULTS: Pulsation index (PI), resistance index (RI), systolic peak value/diastolic peak value (A/B value) of dysmenorrhea patients and hemorheological data except erythrocyte sedimentation rate of dysmenorrhea patients were significantly higher than the normal control group. After treating with TP, no difference existed between two groups. CONCLUSIONS: The blood of dysmenorrhea patients appear to be viscous and adhesive, TP can improve the state of blood circulation and treat the dysmenorrhea effectively.

[Comparison of three antituberculous drugs in serum and cold abscesses of patients with spinal tuberculosis].

OBJECTIVE: Pharmacokinetics of INH, RFP and OFLX in serum and cold abscesses of patients with spinal tuberculosis was analyzed to provide reference to choosing clinical therapeutic regimen. METHOD: The aspiration specimens of abscesses and venous blood were collected from 8 patients with spinal tuberculosis at 0.5, 0.75, 1.0, 1.5, 2.0, 4.0, 6.0, 9.0, 12.0, 16.0 and 24.0 h after administration of antituberculous drugs. The specimens were assessed by high performance liquid chromatography (HPLC). The data were processed with software 3P87. RESULT: The Cmax of INH, RFP and OFLX in serum were 10.87 +/- 7.09 micrograms/ml, 9.98 +/- 3.53 micrograms/ml, and 5.29 +/- 0.72 micrograms/ml, while the Cmax of INH, RFP and OFLX in cold abscesses were 2.84 +/- 1.63 micrograms/ml, 0.57 +/- 0.26 microgram/ml and 3.19 +/- 1.29 micrograms/ml respectively. CONCLUSION: After administration, the Cmax of INH and OFLX in the cold abscesses of patients with spinal tuberculosis, reached the level beyond their MIC and appeared and disappeared more slowly than that in the serum; RFP was not easy to permeate into the cold abscesses, the Cmax of RFP in the cold abscesses just reached its MIC.

[Clinical features, diagnosis and treatment of solitary vertebral arch tuberculosis].

OBJECTIVE: To explore clinical features of vertebral arch tuberculosis, and to discuss the diagnosis and treatment of this illness. METHOD: Seventeen of 3,825 cases of spinal tuberculosis who were hospitalized and treated from 1956 to 1996 were reviewed, and their clinical features, diagnosis and treatment were analyzed and reported. RESULT: Sixteen cases of vertebral arch tuberculosis were in adolescence and prime of life, and another one was a 2-year-old child. Their symptoms in early stages were untypical, including local fixed pain and tenderness, and later, abscess, fistula and symptoms of nerve system appeared. All the cases were diagnosed through summarizing of their history, signs, imaging examination and bacteriology tests, and only one case was misdiagnosed. In one of 17 cases, antituberculosis chemotherapy was conducted alone and non-operation was performed. In the other 16 cases, focal debridement of vertebral arch was performed on the basis of antituberculous chemotherapy, and in 4 cases of them with symptoms of nerve system, laminectomy and spinal decompression was performed homochronously. All the cases were healed and no relapse occurred during follow-up. CONCLUSION: It is difficult to diagnose vertebral arch tuberculosis in early stages due to its occult symptoms, but the symptom of confined pain and manifestation in X-ray, CT and MRI are much useful for early diagnosis. Abscess puncture and acid-fast staining smear microscopy can usually be used to confirm diagnosis. And surgical intervention is an important measure for its treatment.

[Theoretical analysis of factors affecting heat exchange stability of human body with environment].

Life could not be normal without the heat produced by metabolism of human body being transmitted into environment. This paper discussed the ways of heat exchange of human body with the environment, and analyzed their effects on the stability of heat exchange theoretically. In addition, factors that affects the stability of heat exchange were studied. The results indicate that the environmental temperature is the most important factor.

[On the best time for harvesting Dendrobium candidum Wall. ex Lindl].

OBJECTIVE: To explore the theory on the best time for harvesting Dendrobium candidum so that the production of the herb can be directed more effectively. METHOD: Based on a component analysis in combination with mathematical calculation, a synthetical appraisal on the yield, contents of effective components and efficacy of D. candidum in different growing periods has been made, and different appraisal indexes and synthetical harvesting indexes have been worked out. RESULT: The harvesting indexes from the first year to the fourth year are as follows: 5.69, 9.54, 17.9, 22.2 (curing throat and eye diseases); 11.2, 14.6, 21.6, 17.6(strengthening immunity); 8.44, 12.1, 19.7, 19.9(the above-mentioned indexes and yield factor considered together). CONCLUSION: The best time for harvesting D. candidum is in the fourth year for curing throat and eye diseases; in the third or first year for strengthening immunity; and in autumn of the third year if the above-mentioned indexes and yield factor are to be considered together.

[Biological activity detection of mouse type I soluble interleukin-1 receptor].

OBJECTIVE: To comfirm that the recombinant sIL-1R had biological activity. METHODS: With biotin labed human IL-1 beta as ligand, protein blotting receptor-ligand binding assay and enzyme-linked immunological receptor-ligand binding assay were developed to detect the expression of sIL-1R I gene in insect cells (Sf9). Meanwhile, the blockage assay of IL-1 beta biological activity was used to detect the function of recombinant sIL-1R I. RESULTS: It showed that the recombinant sIL-1R I possessed immunological activity and ligand binding activity and could be secreted into the medium. CONCLUSIONS: The three assays could be used for detecting biological activity of recombinant sIL-1R I.
