Development and characterization of wheat- Leymus racemosus translocation lines with resistance to Fusarium Head Blight.

Wheat scab (Fusarium Head Blight, FHB) is a destructive disease in the warm and humid wheat-growing areas of the world. Finding diverse sources of FHB resistance is critical for genetic diversity of resistance for wheat breeding programs. Leymus racemosus is a wild perennial relative of wheat and is highly resistant to FHB. Three wheat- L. racemosus disomic addition (DA) lines DA 5 Lr#1, DA 7 Lr#1 and DALr.7 resistant to FHB were used to develop wheat- L.racemosus translocation lines through irradiation and gametocidal gene-induced chromosome breakage. A total of nine wheat-alien translocation lines with wheat scab resistance were identified by chromosome C-banding, GISH, telosomic pairing and RFLP analyses. In line NAU 614, the long arm of 5 Lr#1 was translocated to wheat chromosome 6B. Four lines, NAU 601, NAU 615, NAU 617, and NAU 635, had a part of the short arm of 7 Lr#1 transferred to different wheat chromosomes. Four other lines, NAU 611, NAU 634, NAU 633, and NAU 618, contained translocations involving Leymus chromosome Lr.7 and different wheat chromosomes. The resistance level of the translocation lines with a single alien chromosome segment was higher than the susceptible wheat parent Chinese Spring but lower than the alien resistant parent L. racemosus. At least three resistance genes in L. racemosus were identified. One was located on chromosome Lr.7, and two could be assigned to the long arm of 5 Lr#1 and the short arm of 7 Lr#1.

A study of mechanisms involved in vasodilatation induced by resveratrol in isolated porcine coronary artery.

The present study was designed to investigate the acute relaxing effect of phytoestrogen resveratrol on isolated porcine coronary arteries and to determine the mechanisms underlying its vasodilatation. Rings of porcine coronary arteries were suspended in organ baths containing Krebs-Henseleit solution, and then isometric tension was measured. Resveratrol concentration-dependently relaxed arterial rings precontracted with 30 mM KCl. The IC(50) value of resveratrol was 38.67+/-3.21 microM. Incubation with N(omega)-L-nitro-arginine (L-NNA), endothelium removal or the presence of a potent inhibitor of protein tyrosine phosphatase sodium orthovanadate partly decreased the relaxation induced by resveratrol. However, the relaxation induced by resveratrol was unaffected by the estrogen receptor antagonist tamoxifen, the inhibitor of prostanoid synthesis indomethacin, the antagonist of beta-adrenoceptors propranolol or the protein synthesis inhibitor, cycloheximide. In addition, resveratrol significantly decreased the contractile responses of 5-HT, KCl and CaCl(2), and shifted their cumulative concentration-response curves to the right. These results suggest that the mechanisms of vasorelaxation induced by resveratrol are heterogeneous, two mechanisms participating partially in the relaxation of porcine coronary artery were detected in the study, one being the nitric oxide released from the endothelium, the other causing inhibition of Ca(2+) influx, but estrogen receptors were not involved in resveratrol-induced relaxation.

Superior neovascularization and muscle regeneration in ischemic skeletal muscles following VEGF gene transfer by rAAV1 pseudotyped vectors.

Recombinant adeno-associated virus serotype 2 (rAAV2) vector has been widely employed for gene therapy. Recent progress suggests that the new serotypes of AAV showed a better performance than did AAV2 in normal tissues. Here, we evaluate the potential role of human vascular endothelial growth factor (VEGF) gene transfer using rAAV vector pseudotyped with serotype 1 capsid proteins (rAAV1) in the treatment of muscle ischemia. In ischemic skeletal muscles, the rAAV1-LacZ vector allowed higher level, broader distribution, and long-lasting gene expression compared with the rAAV2-LacZ vector. Muscle VEGF165 production following the rAAV1-VEGF165 vector injection was 5-10 times higher than that following the rAAV2-VEGF165 vector injection. VEGF165 production mediated by the rAAV1-VEGF165 vector stimulated a large set of neovascularization with relatively mature vascular structures and enhanced muscle regeneration in the ischemic skeletal muscles. Thus, the rAAV1-VEGF165 vector mediated gene transfer may be a therapeutic approach to peripheral vascular diseases.

Synthesis and in vitro anti-hepatitis B and C virus activities of ring-expanded ('fat') nucleobase analogues containing the imidazo[4,5-e][1,3]diazepine-4,8-dione ring system.

As part of our structure-activity relationship studies, we report here the synthesis and in vitro anti-HBV and anti-HCV activities of a number of ring-expanded ('fat') nucleobases containing the imidazo[4,5-e][1,3]diazepine-4,8-dione ring system. One of the compounds, ZP-88, exhibited a good activity/toxicity profile against HBV by inhibition of the synthesis of extracellular virion release (EC(50)=1.7microM, CC(50)=286microM, SI=168) and intracellular HBV replication intermediates (EC(50)=8.4microM, CC(50)=286microM, SI=34) in cultured human hepatoblastoma 2.2.15 cells. By contrast, most of the compounds tested against HCV had only marginal activity/toxicity profile, although that was still better than that of the reference compound ribavirin.

The array and interfacial activity of sodium dodecyl benzene sulfonate and sodium oleate at the oil/water interface.

There is a close correlation between the interfacial activity and the adsorption of the surfactant at the interface, but the detailed molecular standard information was scarce. The interfacial activity of two traditional anionic surfactants sodium dodecyl benzene sulfonate (SDBS) and sodium oleate (OAS) were studied by experimental and computer simulation methods. With the spinning drop method and the suspension drop method, the interfacial tension of oil/aqueous surfactant systems was measured, and the influence of surfactant concentration and salinity on the interfacial tension was investigated. The dissipative particle dynamics (DPD) method was used to simulate the adsorption of SDBS and OAS at the oil/water interface. It was shown that it is beneficial to decrease interfacial tension if the hydrophobic chains of the surfactant and the oil have similar structure. The accession of inorganic salts causes surfactant molecules to form more compact and ordered arrangements and helps to decrease the interfacial tension. There is an osculation relation between interfacial density and interfacial activity. The interfacial density calculated by molecular simulation is an effective parameter to exhibit the interfacial activity.

Inelastic neutron scattering studies of the interaction between water and some amino acids.

The interaction between water and some of amino acids (glycine, L-glutamine, L-threonine, L-cysteine and L-serine) was studied by inelastic incoherent neutron scattering (IINS). The vibrational spectra of dry amino acids and amino acids with a water content (e.g., 1 mol water/1 mol amino acid) were recorded. Comparing the difference spectra obtained by subtracting the spectrum of dry sample from those of wet sample with the spectra of ice Ih, we obtained that the difference spectrum for serine changed greatly from normal ice spectrum; but on the other hand, the difference spectra for the other amino acids such as glycine, glutamine, threonine, and cysteine changed slightly. The results demonstrate that serine has stronger hydrophilic character than glycine, glutamine, threonine, and cysteine. This is the first time the hydrophilic or hydrophobic character of amino acids was studied by using inelastic neutron scattering techniques, which provides important information for theoretical modeling and force field refinement for the interaction between water and the amino acids studied here.

Biophysical studies of a ruthenium(II) polypyridyl complex binding to DNA and RNA prove that nucleic acid structure has significant effects on binding behaviors.

The interactions of a metal complex [Ru(phen)(2)PMIP](2+) {Ru=ruthenium, phen=1,10-phenanthroline, PMIP=2-(4-methylphenyl)imidazo[4,5-f]1,10-phenanthroline} with yeast tRNA and calf thymus DNA (CT DNA) have been investigated comparatively by UV-vis spectroscopy, fluorescence spectroscopy, viscosity measurements, isothermal titration calorimetry (ITC), as well as equilibrium dialysis and circular dichroism (CD). Spectroscopic studies together with ITC and viscosity measurements indicate that both binding modes of the Ru(II) polypyridyl complex to yeast tRNA and CT DNA are intercalation and yeast tRNA binding of the complex is stronger than CT DNA binding. ITC experiments show that the interaction of the complex with yeast tRNA is driven by a moderately favorable enthalpy decrease in combination with a moderately favorable entropy increase, while the binding of the complex to CT DNA is driven by a large favorable enthalpy decrease with a less favorable entropy increase. The results from equilibrium dialysis and CD suggest that both interactions are enantioselective and the Delta enantiomer of the complex may bind more favorably to both yeast tRNA and CT DNA than the Lambda enantiomer does, and that the complex is a better candidate for an enantioselective binder to yeast tRNA than to CT DNA. Taken together, these results indicate that the structures of nucleic acids have significant effects on the binding behaviors of metal complexes.

Spindlin1, a novel nuclear protein with a role in the transformation of NIH3T3 cells.

spindlin1, a novel human gene recently isolated by our laboratory, is highly homologous to mouse spindlin gene. In this study, we cloned cDNA full-length of this novel gene and send it to GenBank database as spindlin1 (Homo sapiens spindlin1) with Accession No. AF317228. In order to investigate the function of spindlin1, we studied further the subcellular localization of Spindlin1 protein and the effects of spindlin1 overexpression in NIH3T3 cells. The results showed that the fusion protein pEGFP-N1-spindlin1 was located in the nucleus and the C-terminal is correlated with nuclear localization of Spindlin1 protein. NIH3T3 cells which could stably express spindlin1 as a result of RT-PCR analysis compared with the control cells displayed a complete morphological change; made cell growth faster; and increased the percentage of cells in G2/M and S phase. Furthermore, overexpressed spindlin1 cells formed colonies in soft agar in vitro and formed tumors in nude mice. Our findings provide direct evidence that spindlin1 gene may contribute to tumorigenesis.

Organosulfur-functionalized Au, Pd, and Au-Pd nanoparticles on 1D silicon nanowire substrates: preparation and XAFS studies.

A hybrid preparative method was developed to prepare organosulfur-functionalized Au nanoparticles (NPs) on silicon nanowires (SiNWs) by reacting HAuCl(4) with SiNW in the presence of thiol. A number of organosulfur molecules-dodecanethiol, hexanethiol, 1,6-hexanedithiol, and tiopronin-were used to functionalize the Au surface. Size-selected NPs ranging from 1.6 to 7.5 nm were obtained by varying the S/Au ratio and the concentration of HAuCl(4). This method was further extended to the preparation Pd and Pd-Au bimetallic NPs on SiNWs. The morphology of the metal nanostructures was examined by transmission electron microscopy (TEM) and high-resolution transmission electron microscopy (HRTEM). The local structure and bonding of the SiNW-supported metal nanostructures were studied using X-ray absorption fine structures (XAFS) [including both X-ray near-edge structures (XANES) and extended X-ray absorption fine structures (EXAFS)] at the Au L(3)-, Pd K-, S K-, and Si K-edges. It was also found that the annealing of the thiol-capped Au NPs up to 500 degrees C transforms the surface of the thiol-capped NPs to gold sulfide, as identified using Au L(3)- and S K-edge XANES. We also illustrate that this preparative approach can be used to form size-controllable Au NPs on carbon nanotubes.

Three-dimensional vision with dual acousto-optic deflection encoding.

We report a new method of sensing a three-dimensional (3D) object surface with an arbitrary geometric shape. In this approach, the first-order beams diffracted from two acousto-optic deflectors (AODs) interfere with each other to form a spatial carrier that is used to encode the depth information from the 3D object surface. A direct digital synthesizer is utilized to control two AODs to generate sequentially spatial carriers with different spatial frequencies so that a modified temporal phase-unwrapping technique can be applied for decoding the shape information of the test surface. Preliminary experimental results are presented to demonstrate the effectiveness of this method.

Interactions among measles virus hemagglutinin, fusion protein and cell receptor signaling lymphocyte activation molecule (SLAM) indicating a new fusion-trimer model.

For measles viruses, fusion on the cell membrane is an important initial step in the entry into the infected cells. The recent research indicated that hemagglutinin firstly leads the conformational changes in the fusion protein then co-mediates the membrane fusion. In the work, we use the co-immunoprecipitation and pull-down techniques to identify the interactions among fusion protein, hemagglutinin and signaling lymphocyte activation molecule (SLAM), which reveal that the three proteins can form a functional complex to mediate the SLAM-dependent fusion. Moreover, under the confocal microscope, fusion protein and hemagglutinin protein can show the cocapping mediated by the SLAM. So fusion protein not only is involved in the fusion but also might directly interact with the SLAM to be a new fusion-trimer model, which might account for the infection mechanism of measles virus.

Expression of SLAM (CDw150) on Sf9 cell surface using recombinant baculovirus mediates measles virus infection in the nonpermissive cells.

Signaling lymphocytic activation molecule (SLAM; also known as CDw150) has been reported as the receptor of measles virus (MV) interacting with MV hemagglutinin (MVH). In this study, we developed a baculovirus-derived vector, the Bacmid-egfp, containing a reporter gene encoding the enhanced green fluorescent protein (EGFP) under the control of the promoter of very late polyhedrin gene from Autographa californica multiple nucleopolyhedrovirus (AcMNPV), and employed the recombinant baculovirus to express SLAM in Sf9 (Spodoptera frugiperda) cells and investigate SLAM function. The result showed that the integration of the EGFP expression cassette in the Bac-to-Bac system facilitated research with the system without introducing compromises due to its use. SLAM protein fused to His-tag was expressed in Sf9 cells through the modified Bac-to-Bac system. The expressed SLAM was identified as approximately 46 kDa, and it presented on the cell surface, as revealed by fluorescent immunochemical staining and confocal microscopic analysis. The pull-down assay proved that SLAM protein expressed in this system could interact with MVH protein. After incubating with MV vaccine strain S191, cell fusion was only observed in the Sf9 cells expressing both EGFP and SLAM from recombinant baculovirus rather than those expressing EGFP only from the modified viral vector. Furthermore, MV replicated and induced apoptosis in the Sf9 cells with SLAM expression.

Pathway analysis of coronary atherosclerosis.

Large-scale gene expression studies provide significant insight into genes differentially regulated in disease processes such as cancer. However, these investigations offer limited understanding of multisystem, multicellular diseases such as atherosclerosis. A systems biology approach that accounts for gene interactions, incorporates nontranscriptionally regulated genes, and integrates prior knowledge offers many advantages. We performed a comprehensive gene level assessment of coronary atherosclerosis using 51 coronary artery segments isolated from the explanted hearts of 22 cardiac transplant patients. After histological grading of vascular segments according to American Heart Association guidelines, isolated RNA was hybridized onto a customized 22-K oligonucleotide microarray, and significance analysis of microarrays and gene ontology analyses were performed to identify significant gene expression profiles. Our studies revealed that loss of differentiated smooth muscle cell gene expression is the primary expression signature of disease progression in atherosclerosis. Furthermore, we provide insight into the severe form of coronary artery disease associated with diabetes, reporting an overabundance of immune and inflammatory signals in diabetics. We present a novel approach to pathway development based on connectivity, determined by language parsing of the published literature, and ranking, determined by the significance of differentially regulated genes in the network. In doing this, we identify highly connected "nexus" genes that are attractive candidates for therapeutic targeting and followup studies. Our use of pathway techniques to study atherosclerosis as an integrated network of gene interactions expands on traditional microarray analysis methods and emphasizes the significant advantages of a systems-based approach to analyzing complex disease.

High-resolution numerical approximation of traffic flow problems with variable lanes and free-flow velocities.

This paper develops macroscopic traffic flow models for a highway section with variable lanes and free-flow velocities, that involve spatially varying flux functions. To address this complex physical property, we develop a Riemann solver that derives the exact flux values at the interface of the Riemann problem. Based on this solver, we formulate Godunov-type numerical schemes to solve the traffic flow models. Numerical examples that simulate the traffic flow around a bottleneck that arises from a drop in traffic capacity on the highway section are given to illustrate the efficiency of these schemes.

Age-dependent brain activation during forward and backward digit recall revealed by fMRI.

In this study, brain activation associated with forward and backward digit recall was examined in healthy old and young adults using functional MRI. A number of areas were activated during the recall. In young adults, greater activation was found in the left prefrontal cortex (BA9) and the left occipital visual cortex during backward digit recall than forward digit recall. In contrast, the activation in the right inferior frontal gyrus (BA 44/45) was more extensive in forward digit recall than in backward digit recall. In older adults, backward recall generated stronger activation than forward recall in most areas, including the frontal, the parietal, the occipital, and the temporal cortices. In the backward recall condition, the right inferior frontal gyrus (BA44/45) showed more activation in the old group than in the young group. These results suggest that different neural mechanisms may be involved in forward and backward digit recall and brain functions associated with these two types of recall are differentially affected by aging.

Preparation and evaluation of proline-based chiral columns.

Chiral stationary phases made of readily available proline peptides were prepared and evaluated for general chiral separation. With the proper structural elements, these columns demonstrated broad chiral selectivity. Among the 53 analytes tested, a tetraproline column resolved 31. The separations achieved for these analytes are comparable to those achieved on Whelk O2 column, while still inferior to those achieved on Daicel AD-H and OD-H columns. Number of proline units proves important for chiral recognition, because a control column made with a single proline unit is largely ineffective.
