Release and activity of allelochemicals from allelopathic rice seedlings.

3-Isopropyl-5-acetoxycyclohexene-2-one-1 (1), momilactone B (2), and 5,7,4'-trihydroxy-3',5'-dimethoxyflavone (3) were isolated and identified from an allelopathic rice accession PI312777. These three compounds at low concentrations could inhibit the growth of weeds Echinochloa crusgalli and Cyperus difformis associated with rice, especially mixtures of the compounds had stronger inhibitory activity than did individual compounds. Studies with hydroponic culture, continuous root exudates trapping system (CRETS), and direct resin adsorption methods showed that a total of 7.6 n moles 1, 2, and 3 were exuded from living roots of each seedling into the environment at 10 days after seedlings were transplanted. Furthermore, 1, 2, and 3 were found in the soil growing PI312777 seedlings at day 15 after seedlings emergence and reached a total of 39.5 microg/g soil at day 30. The results indicated that PI 312777 seedlings could release sufficient quantities of 1, 2, and 3 into the environment to act as allelochemicals inhibiting the growth of associated weeds. Investigations on the distribution of 1, 2, and 3 in PI 312777 plant, and its root exudates showed that the levels of 1, 2, and 3 were significantly higher in the shoots and root exudates than in the roots, and only trace 1 was observed in the roots. The results suggest that the roots of rice seedlings are not major site of synthesis or accumulation 1, 2, and 3, but a pathway for their release into the environment. The levels of 1, 2, and 3 in the root exudates were over 2-folds higher under direct resin adsorption than under hydroponic culture and CRETS, and hence, it is the preferred method to collect and identify active allelochemicals in rice exudates in future studies on rice allelopathy.

High-performance liquid chromatographic method for the bioequivalence evaluation of desloratadine fumarate tablets in dogs.

A simple HPLC method was developed for the determination of desloratadine in dog plasma and was used for evaluating the bioequivalence of desloratadine fumarate tablets and desloratadine tablets in dogs. Chromatographic separation was performed on a Hypersil CN column (150 mm x 5.0 mm, 5 microm) using a mixture of methanol, acetonitrile and phosphate buffer (pH 5.5; 0.01 mol/l) (35:35:30, v/v/v) as mobile phase delivered at a flow rate of 0.8 ml/min. The detection was set at 241 nm. The limit of quantitation was 5.0 ng/ml. The calibration range was from 5.0 to 800.0 ng/ml. Inter- and intra-day precision ranged from 1.8 to 3.8% and from 2.2 to 9.0%, respectively. The recovery of desloratadine from dog plasma ranged from 78.8 to 82.0%. The developed method was applied to the bioequivalence studies of desloratadine fumarate tablets (test preparation) and desloratadine tablets (reference preparation) in five dogs. Pharmacokinetic parameters t(max), C(max), AUC(0-t), AUC(0- infinity ), t(1/2) were determined from plasma concentration-time profiles of both preparations. The analysis of variance (ANOVA) did not show any significant difference between the two preparations and 90% confidence intervals fell within the acceptable range for bioequivalence. Based on these statistical inferences it was concluded that the two preparations exhibited comparable pharmacokinetic profiles and that desloratadine fumarate tablets was bioequivalent to desloratadine tablets.

Cytotoxicity of cord blood derived Her2/neu-specific cytotoxic T lymphocytes against human breast cancer in vitro and in vivo.

The Her2/neu oncogene encodes a transmembrane protein with homology to the epidermal growth factor receptor. Overexpression of this gene contributes to the aggressiveness of breast cancer and poor prognosis. Therefore, Her2/neu is an ideal target molecule for generating effective cytotoxic T lymphocytes (CTLs) against breast cancers. This study reports on the generation of Her2/neu-specific CTL from umbilical cord blood mononuclear cells (UCBC) using dendritic cells primed with Her2/neu-derived peptide (KIFGSLAFL, E75) for immunostimulation. The CTLs showed specific cytotoxicity to Her2/neu high expressing MDA-453 but not toward Her2/neu low expressing MDA-231 human breast cancer cells. Similarly generated CTLs stimulated with irrelevant peptide pulsed dendritic cells did not show significant cytotoxicity towards breast cancer targets. The phenotypes of cells in culture showed high percentage of CD3+, CD4+ and CD8+T cells as determined by flow cytometry. However, the antibody mediated blocking assay demonstrated that only HLA-Class I restricted CD8+ cells are involved in the cytotoxicity. Furthermore, in vivo studies showed that treatment of SCID mice bearing MDA-453 tumor with Her2/neu-specific CTLs resulted in significant inhibition of tumor growth compared to untreated tumor bearing control mice. These results demonstrate that human umbilical cord blood mononuclear cells are a good source for generating Her2/neu-specific CTLs against human breast cancer both in vitro and in vivo.

A novel protein-DNA interaction involved with the CpG dinucleotide at -30 upstream is linked to the DNA methylation mediated transcription silencing of the MAGE-A1 gene.

To understand the DNA-methylation mediated gene silencing mechanisms, we analyzed in cell culture of the promoter function of the MAGE-A1 gene, which is frequently demethylated and over-expressed in human hepatocellular carcinoma. We have established the correlation of the DNA methylation of the promoter CpG island with expression status of this gene in a panel of the established liver cancer cell lines. The crucial CpG dinucleotide(s) within the minimal promoter subjected to the control mediated by DNA methylation with profound biological functions was also delineated. Furthermore, a novel sequence-specific DNA-protein interaction at the -30 CpG dinucleotide upstream of the gene was found having a vital part to play in the DNA methylation mediated transcription silencing of the MAGE-A1 gene. Our results would not only provide new insights into the DNA methylation mediated mechanisms over transcription of the MAGE-A1 gene, but also pave the way for further defining the cross-talk among DNA methylation, histone modification and chromatin remodeling in detail.

[Effects of a novel recombinant polypeptide from the sea anemone Anthopleura sp. on left ventricular function in New Zealand rabbits with chronic congestive heart failure].

OBJECTIVE: To observe the effects of recombinant hk2a, a novel neurotoxin from the sea anemone Anthopleura sp. on the left ventricular function in New Zealand rabbits with chronic congestive heart failure(CCHF). METHODS: CHF model was established using New Zealand rabbits by abdominal aortic coarctation. The left ventricular ejection fraction (LVEF) was measured by Acuson ultrasound systems (Sequoia 512) at 0, 5, 15, 30, 45 and 60 min respectively after intravenous injection with a single dose of recombinant hk2a (20 microg/kg x b x w.). The response of the rabbits to hk2a treatment was observed in comparison with that of the control group. RESULTS: Recombinant hk2a immediately and significantly increased the left ventricular ejection fraction of the rabbit models of CCHF after intravenous injection (P<0.05), and its effects were maintained for more than 1 h without affecting the heart rate. CONCLUSION: recombinant hk2a significantly increases the left ventricular ejection fraction in New Zealand rabbits with CCHF.

Kinetics of aniline oxidation with chlorine dioxide.

For the first time, kinetics of aniline oxidation with chlorine dioxide (ClO2) were investigated systematically by detecting concentration of aniline with HPLC at regular intervals. Results showed that the reaction was first-order both in ClO2 and in aniline, and the oxidation reaction could be described as second-order reaction. Stoichiometric factor eta was experimentally determined to be 2.44. The second-order-reaction rate constant k was 0.11 L/(mol x s) under condition of pH 6.86 and water temperature (Tw) 287K. Reaction activation energy was 72.31 kJ/mol, indicating that the reaction could take place under usual water treatment conditions. The reaction rate constants in acidic and alkali media were greater than that in neutral medium. Chlorite ion could slightly increase reaction rate in acidic medium. p-aminophenol and azobenzene were detected by GC-MS as intermediates.

[Application of echocardiographic monitoring in percutaneous cardiopulmonary support for the treatment of acute fulminant myocarditis: report of one case].

OBJECTIVE: To observe dynamic echocardiographic changes in patients with acute fulminant myocarditis and assess the clinical value of echocardiography in treatment with percutaneous cardiopulmonary support (PCPS). METHOD PCPS through the femoral artery/vein was used in the treatment of a patient with acute fulminant myocarditis, with dynamic echocardiographic observation of the atrial and ventricular volumes, left ventricular wall thickness, pulsatile amplitude and cardiac functions before and after PCPS. The position of the catheter of PCPS was also under dynamic bedside monitoring. RESULTS AND CONCLUSION The patient showed significant echocardiographic changes, such as weakening of the wall movement, lowered ejection fraction, widening of the left ventricular internal diameter with mild pericardial exudation. After PCPS, the cardiac functions such as the heart movement, ejection fraction and the shortening rate of the short axis were greatly improved as observed with ambulatory echocardiography. This case suggests that ambulatory echocardiography can be instrumental in the management of cardiac emergencies.

[Effect of acute phosgene inhalation on antioxidant enzymes, nitric oxide and nitric-oxide synthase in rats].

OBJECTIVE: To study the effect of acute phosgene inhalation on the antioxidant enzymes, nitric oxide (NO) and nitric oxide synthase (NOS) in rats. METHODS: Phosgene was produced by decomposing bis (trichdomethyl) carbonate in the presence of N,N-dimethyl formamide. SD rats were randomly divided into two groups: control and phosgene exposure groups. In a special experimental device with equipment modulating the gas flow, phosgene exposed rats inhaled phosgene quantitatively for five minutes. Two hours later, all the rats were sacrificed and the ratio of wet weight to dried weight of lung (WW/DW) was calculated. Peripheral blood, serum and liver were collected to examine the activities of antioxidant enzymes including glutathione S-transferase (GST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px), NOS, and NO level. The total content of proteins were also determined. RESULTS: The WW/DW ratio of lung in phosgene exposure group was much higher than that in control group (P < 0.01). The activities of GST in serum and liver of phosgene exposure group increased significantly (P < 0.05). The activities of SOD, CAT, GSHPx and NOS in serum or blood and liver of phosgene exposure group were also increased significantly (P < 0.05). But the content of NO was significantly decreased (P < 0.01). CONCLUSION: Acute phosgene inhalation may cause a dramatically changes of several antioxidant enzyme activities, and acute injury of liver to some extent in rats. The latter is related to reactive oxygen species. But the elevation of antioxidant enzyme activities suggests that antioxidative treatment for acute phosgene poisoning should not be considered first.

Alternating shortcut nitrification-denitrification for nitrogen removal from soybean wastewater by SBR with real-time control.

A novel treating technology for nitrogen removal from soybean wastewater was studied. The process for nitrogen removal was achieved by alternating aeration and mixing, combined with real-time control strategies. Results showed that the COD and total nitrogen removal rates are more than 90% and 92% at COD and total nitrogen loads of 1.0 - 1.2 kg COD/(kgMLSS x d) and 0.20 - 0.27 kg TN/(kgMLSS x d), respectively. In addition, it could improve sludge settling property. SVI value is less than 70 g/ml during the whole cycles. The method not only may be adapted to treat soybean wastewater with high nitrogen, but also may be applied to treat other high nitrogen wastewater.

[Kinetics and mechanism of aniline oxidation with chlorine dioxide in water].

Reaction kinetics of aniline oxidation with chlorine dioxide in water were investigated systematically by detecting concentrations of aniline with HPLC at regular intervals. Results showed that the reaction was first-order with respect to both chlorine dioxide and aniline, and the entire reaction was of second-order. Under condition of pH 6.86 and water temperature (Tw) 287 K, the second-order reaction rate constant k was 0.11 L x (mol x s)(-1). Reaction activation energy was 73.11 kJ x mol(-1), revealing that the reaction could take place under usual water treatment conditions. The reaction rate constants in acidic and alkali media were greater than that in neutral medium. Effect of chlorite ion on the reaction rate was insignificant. P-aminophenol and azobenzene were detected by GC MS as intermediates. Oxidation pathways were proposed and discussed elementarily.

[Chemical mechanism of exotic weed invasion].

The ecological and economic impacts of exotic invasive weeds have been followed with interest. A successful invasion of exotic weeds depends not only on their bio-ecological traits, but also on some specific mechanisms of invasion. The clarification of the invasive mechanism of exotic weeds will be beneficial to predict and manage on them. In the process of establishment, reproduction and spreading, an exotic species has to become dominant in the interactions with native ones at new habitats, and then invades successfully. Among invasive mechanisms, the chemical interaction between exotic and native weeds should not be neglected. Previous studies showed that allelopathy plays an important role in exotic weed invasion. In fact, plants are able to defend or inhibit animals, plants and microbes from new habitats by using some specific chemicals. The chemical mechanism of exotic weed invasion might involve each aspect of plant chemical ecology. Therefore, chemical ecological characteristics of exotic weeds should be regarded as one of the most significant factors on predicting invasive species. The studies on chemical mechanism of exotic weed invasion will be one of the important aspects in future.

Correlation between methylation profile of promoter cpg islands of seven metastasis-associated genes and their expression states in six cell lines of liver origin.

BACKGROUND & OBJECTIVE: DNA methylation has been regarded as an important epigenetic signature reflecting the transcription state of DNA in cells. This study was to assess the correlation between methylation state of promoter CpG islands of metastasis-associated genes and their expression in 6 liver cell lines, including 5 cancerous. METHODS: Methylation specific polymerase chain reaction method (MSP) and DNA sequencing verification were used to analyze the methylation state of promoter CpG islands of 7 genes (ASPH, ENO3, ITGA9, LRP6, MTHFD2, OXCT, and SRP72) in 5 liver cancer cell lines (BEL-7402, SMMC-7721, Hep3B, HepG2, and HCCLM3), and 1 immortalized liver cell line (L-02). Expression of 6 genes in this list was assessed by the semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) method. RESULTS: The methylation state of genes was either unmethylated or heterozygously methylated in these 7 liver cell lines. Except for no expression of OXCT gene was detected by RT-PCR in both HepG2 and HCCLM3 cells where it was heterozygously methylated, there was expression of genes in all the remaining cases. CONCLUSION: Although expression state of genes in this study supported the general notion that hypermethylation state of promoter CpG islands of genes represents the silenced state of gene transcription, there were exceptions. Therefore, other mechanisms are likely to contribute to the observed expression state of these 7 genes in this study.

[Differences in plant hormone level of cucumber seedlings grown under sulfur or xenon lamp].

The primary mechanism of growth difference of cucumber (Cucumis sativus L.) seedlings cultured under sulfur lamp and xenon lamp in a phytotron was investigated. Compared with cucumber seedlings grown under xenon lamp, those under sulfur lamp were shorter, and the cell number in the middle hypocotyls epidermis and cortex of them were more (Fig. 1, Table 1). Endogenous hormone analysis indicates that the content of IAA and GA(3) of seedlings under sulfur lamp were 17% and 24% lower, while ABA content was 31% higher than that under xenon lamp (Fig. 2). Based on these results, it is suggested that the growth difference between cucumber seedlings grown under sulfur lamp and under xenon lamp might be related to the control of endogenous hormones.
