The hsp60B gene of Drosophila melanogaster is essential for the spermatid individualization process.

The 60-kDa heat shock protein family (Hsp60) is found in prokaryotes, mitochondria, and chloroplasts. The Hsp60 proteins promote proper protein folding by preventing aggregation. In Drosophila melanogaster, the hsp60 gene is essential for a variety of developmental processes, beginning at early embryogenesis. In this study we show that an additional member of the Drosophila hsp60 gene family, hsp60B, is essential in male fertility. In males homozygous for a mutation of the hsp60B gene, developmental processes appeared normal throughout most of spermatogenesis, including spermatocyte growth, meiosis, and spermatid elongation. At these stages, mitochondria also displayed a differentiation process similar to wild-types. However, we found that the mutation disrupted a late stage of spermatogenesis, the spermatid individualization process. In this process, the individualization complex is assembled at spermatid nuclear heads, traverses along spermatid tails, and generates membranes for each of the spermatids in a cyst. Our analysis further shows that the individualization complex in sterile males displayed abnormal morphology as it was traveling along the spermatid tails. The Drosophila Hsp60 proteins are believed to be exclusively localized in the mitochondria. Our observation that the hsp60B mutation displayed no apparent defect in mitochondrial differentiation during spermatogenesis suggests that the Hsp60B protein may operate in a nonmitochondrial location.

Measurement of DNA damage in mammalian cells exposed in vitro to radiofrequency fields at SARs of 3-5 W/kg.

In the present study, we determined whether exposure of mammalian cells to 3.2-5.1 W/kg specific absorption rate (SAR) radiofrequency fields could induce DNA damage in murine C3H 10T(1/2) fibroblasts. Cell cultures were exposed to 847.74 MHz code-division multiple access (CDMA) and 835.62 frequency-division multiple access (FDMA) modulated radiations in radial transmission line (RTL) irradiators in which the temperature was regulated to 37.0 +/- 0.3 degrees C. Using the alkaline comet assay to measure DNA damage, we found no statistically significant differences in either comet moment or comet length between sham-exposed cells and those exposed for 2, 4 or 24 h to CDMA or FDMA radiations in either exponentially growing or plateau-phase cells. Further, a 4-h incubation after the 2-h exposure resulted in no significant changes in comet moment or comet length. Our results show that exposure of cultured C3H 10T(1/2) cells at 37 degrees C CDMA or FDMA at SAR values of up to 5.1 W/kg did not induce measurable DNA damage.

The role of Kupffer cells in non-alcoholic steatohepatitis of rats chronically fed with high-fat diet.

OBJECTIVE: To explore the role of Kupffer cells in non-alcoholic steatohepatitis (NASH) by means of rat model. METHODS: Nineteen male SD rats were randomized into model group (n=10) and normal group (n=9), with a high-fat diet and standard diet for 12 weeks, respectively. Routine histologic features of hepatic section were observed by HE staining. The number and shape of Kupffer cells in the liver were detected by immunohistochemistry and penetrated electron microscope, respectively. RESULTS: All rats of model group developed NASH, which was characterized by obesity and hyperlipidemia. Histopathological examination showed hepatocellular macrovesicular steatosis, lobular inflammatory cell infiltration and necrosis. Compared with normal group, the count of Kupffer cells in the liver was largely increased, and the Kupffer cells in the model group were activated to some extent. Furthermore, these changes of Kupffer cells were in accordance with the degree of steatosis, inflammation and necrosis in the liver of the model group. CONCLUSION: The number and activity of Kupffer cells are increased significantly in NASH induced by high-fat diet, and Kupffer cells might be involved in the pathogenesis of steatohepatitis.

Epidermal growth factor receptor-targeted therapy with C225 and cisplatin in patients with head and neck cancer.

C225, a human-mouse chimerized monoclonal antibody directed against the epidermal growth factor receptor (EGFr), has a synergistic effect with cisplatin in xenograft models. To determine the tumor EGFr saturation dose with C225 and the fate of infused C225, we conducted a Phase Ib study with C225 in combination with cisplatin in patients with recurrent squamous cell carcinoma of the head and neck. Using tumor samples, we assessed tumor EGFr saturation by antibody using immunohistochemistry studies, the EGFr tyrosine kinase assay, and detection of the EGFr/C225 complex formation by immunoblot. Potential candidates were screened for EGFr expression in their tumors, and 12 patients who had high levels of EGFr expression and tumors easily accessible for repeated biopsies (pretherapy, 24 h after first C225 infusion, 24 h before third C225 infusion) were entered at three different dose levels of C225 with a fixed dose of cisplatin. The median value of tumor EGFr saturation increased to 95% at the higher dose levels. EGFr tyrosine kinase activity was significantly reduced after C225 infusion, and EGFr/C225 complexes were also detected at higher doses of C225. The loading dose of C225 at 400 mg/m(2) with a maintenance dose at 250 mg/m(2) achieved a high percentage of saturation of EGFr in tumor tissue, and these doses were recommended for Phases II or III clinical trials. Six (67%) of nine evaluable patients achieved major responses, including two (22%) complete responses. Mild to moderate degrees of allergic reaction and folliculitis-like skin reactions were demonstrated. We conclude that infused C225 binds and significantly saturates tumor EGFr, which may render a high degree of antitumor activity, and provides a novel mechanism for targeting cancer therapy for patients who have EGFr expression in their tumors.

[The modified process for preparing natural organic polymer flocculant chitosan].

The modified process for preparing chitosan from crab or lobster shells was developed. In the decalcification stage, 10% HCl was used as soaking solution with addition of a small quantity of A as a promoter, and the mass ratio of reactants was 10% HCl:A:crab or lobster shells = 3.5:0.5:1, continuously stirring the crab or lobster shells at 30 degrees C for 3 h in place of simply soaking the crab or lobster shells at room temperature for 16-24 h in the previous process. In the deacetylation stage, 40% NaOH solution was used with addition of a small quantity of B as a promoter, and the mass ratio of reactants was 40% NaOH:B:chitin = 4:0.2:1, keeping reaction at 105 degrees C for 2 h in place of at 115 degrees C for 6 h in the previous process. By this new process, the cost of the raw materials used for preparing chitosan was cut down 49%, the preparation time was shortened by one half, and the main properties of this chitosan such as viscosity, deacetylation and molecular weight all approached or exceeded those of the Sigma' commercial chitosan (Chitosan C-3646).

[IFN-r/IL-4 expression in peripheral blood mononuclear cells of the patients with chronic severe hepatitis].

OBJECTIVE: To understand the expressions of interferon-r (INF-r) and interleukin-4 (IL-4) in peripheral blood mononuclear cells (PBMCs) of the patients with chronic severe hepatitis (CSH). METHODS: PBMCs from the patients with CSH were separated routinely and stimulated by PMA/Ionomycin/Monensin. The production of IFN-r/IL-4 by CD4+ T cells in PBMC was determined by fluorescence activated call sorting (FACS) analysis and fluorescence-quantitative PCR assay for quantification of HBV DNA. RESULTS: The percentage of IFN-gamma-producing T cells in CD4+ T cells was higher in patients with CSH (7.2%-26.3%) than in normal controls (2.2%-11.9%) (P<0.01). The percentage of Th1 cells increased significantly with the raise of hepatic inflammation activity. The percentage of IL-4-producing T cells in CD4+ T cells did not differ significantly between patients with CSH and normal controls. The quantification of HBV DNA decreased significantly with the increase of the percentage of IFN-gamma-producing T cells. CONCLUSIONS: Th1 cell is associated with hepatic inflammatory activity and IFN-gamma depresses HBV replication.

[The effect of composite Radix Salviae Militiorrhizae injection on myocyte apoptosis in acute myocardial infarction of rabbits].

OBJECTIVE: To study the effect of composite Radix Salviae Militiorrhizae injectiion on myocyte apoptosis in experimental acute myocardial infarction of rabbits. METHODS: The models of acute myocardial infarction were made with rabbits. The method of terminal deoxynucleotidyl transferase mediated dUTP nick end labeling was used to explore myocardial cell apoptosis in different injury area of acute myocardial infarction. The activities of plasma superoxide dismurase and the contents of serum malonic aldehyde were determined. RESULTS: When rabbits were in the condition of acute myocardial infarction, there were myocyte apoptosis in the area of ischemia reperfusion injury and the area of infarction. The number of myocyte apoptosis and the myocyte rate in control group and composite Radix Salviae Militiorrhizae injectiion treated group were higher than in sham-operated group (P < 0.001). In the area of ischemia reperfusion injury the number of myocyte apoptosis and the myocyte rate in composite salvia militiorrhiza-treated group were lower than in control group (P < 0.05). The activity of plasma superoxide dismurase in composite salvia militiorrhiza-treated group was higher than in control group (P < 0.05), but lower than in sham-orerated group (P < 0.001). The content of serum malonic aldehyde in composite Radix Salviae Militiorrhizae injectiion treated group were lower than in control group (P < 0.05), but higher than in sham-operated group (P < 0.01). CONCLUSION: Composite Radix Salviae Militiorrhizae injectiion could reduce the number of myocyte apoptosis in the area of ischemia reperfusion injury, which maybe explained by the increase of the activity of plasma superoxide dismurase and the decrease of the contents of serum malonic aldehyde.

Minimal prophylactic concentration of dietary zinc compounds in a mouse model of swine dysentery.

Dietary supplementation with 6000 mg of Zn2+/kg of feed has been shown to modify the clinicopathologic expression of Brachyspira hyodysenteriae infection in a laboratory mouse model of swine dysentery. However, this concentration impaired the body weight gain of the mice. The purpose of the present study was to determine a minimal prophylactic concentration of feed-grade zinc compounds that would not affect the growth of mice challenge-exposed with B. hyodysenteriae. A total of 440, 6- to 8-week-old, C3H/HeN mice were allocated randomly to groups and fed either a defined diet or a defined diet containing either 1000, 2000 or 4000 mg/kg ZnO, ZnSO4 or zinc-methionine for 7 days before intragastric inoculation with B. hyodysenteriae. From days 7 to 35 after inoculation, mice in each group were necropsied at weekly intervals for determination of body weight, presence of B. hyodysenteriae in the cecum, and histological assessment of cecal lesions. Only ZnO fed at 2000 mg/kg had a prophylactic effect against B. hyodysenteriae infection without affecting the body weight gain of the mice. The prophylactic effect of Zn2+ against infection with B. hyodysenteriae was also affected by the relative concentration of Fe2+ and Zn2+/Fe2+ ratio of the diet.

Histochemical study of transmyocardial laser channels in dogs.

OBJECTIVE: Laser-induced myocardial injury was investigated in a canine transmyocardial laser revascularization model. METHODS: A total of 23 channels were created in the left ventricles of 5 animals with a CO2 laser. Four animals were sacrificed immediately and one at week 2. Lactate dehydrogenase and succinate dehydrogenase in channel regions were examined with a Nitro-BT method staining. RESULTS: Immediately after the procedure, laser channels and surrounding injured myocardium could be stratified from the center to the border into 5 concentric zones, i.e., vaporized zone, carbonized zone, enzyme-disappeared zone, enzyme-decreased zone and enzyme-concentrated zone. Two weeks later, the phenomenon disappeared. CONCLUSIONS: Immediately after transmyocardial revascularization, there is a ring of enzyme-concentrated zone at the outermost region of injured myocardium. Two weeks later, enzyme activities of reversibly injured myocytes appear to have recovered.

Simian virus 40 large tumor antigen forms specific complexes with p53 and pRb in human brain tumors.

OBJECTIVE: To study the role of simian virus 40 (SV40) early region gene coding product large tumor antigen (Tag) expression and the interaction between Tag and tumor suppressors p53 and pRb in human brain tumorigenesis. METHODS: Tag was investigated by immunoprecipitation followed by silver staining and Western blot in 65 cases of human brain tumors and 8 cases of normal brain tissues. Tag-p53 and Tag-pRb complexes were screened in 18 and 15 Tag positive tumor tissues, respectively. RESULTS: Tag was found in all 8 ependymomas and 2 choroid plexus papillomas, 90% of pituitary adenomas (9/10), 73% of astrocytomas (11/15), 70% of meningiomas (7/10), 50% of glioblastomas multiforme (4/8), 33% of medulloblastomas (2/6). 5 oligodendrogliomas, 1 pineocytoma, and 8 normal brain tissues were negative for Tag. Tag-p53 complex was detected in all 18 Tag positive tumors. Tag-pRb complex was found in all 15 Tag positive tumors. CONCLUSION: SV40 Tag is expressed in human brain tumors and can form specific complexes with tumor suppressors p53 and pRb. The inactivation of p53 and pRb due to the formation of Tag-p53 and Tag-pRb complexes may be an important mechanism in the etiopathogenesis of human brain tumors.

[A clinical analysis of 798 cases of M protein positive patients].

OBJECTIVE: To improve the understanding of the diseases with positive M protein by analyzing 798 cases, so as to reduce the failure and error of diagnosis. METHODS: The relations of diseases with the types of the disease and age and sex of the patients were analyzed. The presenting symptoms of multiple myeloma(MM) and non-MM were compared. The difference of Ig content among each type of MM and the characteristics of M protein in non-MM were also studied. RESULTS: In 798 cases with positive M protein, there were 648 cases of MM (81.2%) and 34 cases of Waldenstrom's macroglobulinemia (WM) (4.3%). The remaining 116 cases (14.5%) were benign or secondary monoclonal gammopathies. The average age of the patients was 58.3 years, and there was no difference of disease incidence between men and women. There was a marked difference between MM and non-MM in the presenting symptoms which were mainly lumbago and bone pain in MM but fever, weight loss and fatigue in non-MM. 648 cases of MM included 290 cases of IgG type (44.8%), 143 of IgA type (22.1%), 138 of light chain type (21.3%), 45 of IgD type (6.9%), 30 of non-secretion type (4.6%) and 2 of double-clonal type (0.3%). There was no marked distinction between kappa and lambda chain except in IgD type. The average M protein content of 116 cases of benign or secondary monoclonal gammopathies was (11.3 +/- 6.5) g/L. 55 of the 116 cases were monoclonal gammopathy of undetermined significance (MGUS), which accounted for 6.9% of all the cases. 35 cases were non-Hodgkin's lymphoma (NHL) (4.4%). CONCLUSION: M protein was mainly discovered in MM; it may also be discovered in MGUS, NHL, WM and other diseases. It is essential to have a better understanding of these diseases.

[Pathomorphologic features of clinical stage A prostate carcinoma and causes for misdiagnosis and missed diagnosis].

OBJECTIVE: To study the pathomorphologic features and causes for misdiagnosis and missed diagnosis of clinical stage A prostate carcinoma. METHODS: Prostate samples from a series of 1 020 prostate resections performed in five Shanghai hospitals were obtained, from which 50 clinical stage A carcinoma were studied by immunohistochemistry, according to tumor differentiation and their volume, they were divided into stage A1 and A2 disease. The pathomorphologic features of stage A1 and A2 disease were compared, and the causes for misdiagnosis were analysed. RESULTS: Stage A1 cancer were well- or moderately well-differentiated, of low volume and tended to be multifocal; Stage A2 cancers were poorly differentiated, of high volume, diffuse infiltration and with high grade prostatic intraepithelial neoplasia. 8 cases were misdiagnosed of which 7 cases were stage A1 and misdiagnosed as benign proliferous small gland lesions and 1 case of A2 disease was misdiagnosed as epithelioid histiocytic reactive proliferation. CONCLUSIONS: Stage A1 cancer tended to be initiated in the transition zone and the central zone of the hyperplastic prostate. Stage A2 cancers were mostly middle grade to high grade cancers initiated in the peripheral region and then invaded the central area. The lower incidence of stage A carcinoma in China is related to the small amount of biopsy tissue, careless microscopic observation, or lack of the ability to diagnose stage A1 cancer.

[Isolation, purification and renaturation of recombinant-DNA-derived porcine somatotropin].

Large scale abstraction and isolation of bacterially synthesized, recombinant-DNA-derived, porcine growth hormone (r-pST) is described. The r-pGH is found in genetic engineering E. coli as the form of inclusion bodies. Pellet fraction which were mainly inclusion bodies, after cell breakage and centrifugation, were collected. Cell envelope components, such as protein, lipid, endotoxin and nucleic acids are selectively removed from the pellet fraction by an EDTA/lysozyme/deoxycholate extraction. Inclusion bodies were dissolved using 6 mol/L guanidine/HCl and air oxidation is then carried out in the presence of the guanidine/HCl. The Guanidine/HCl protein mixture were diluted by renaturation solution. Guanidine/HCl were removed by dialysis and then correctly refolded, oxidized r-pGH were obtained. Injection experiment of hypophysectomized rats proved r-pST with high native bioactivity was obtained.
