Effects of ground, steam-flaked, and steam-rolled corn grains on performance of lactating cows.

Five types of processed corn were compared to determine the effects of processing on lactational performance and nutrient digestibilities in dairy cows. Forty lactating Holstein cows averaging 160 +/- 60 d in milk were randomly divided into five groups on the basis of pretreatment milk yields and were fed diets containing 40% corn grain for 56 d. Treatments were 1) finely ground corn, 2) coarsely ground corn, 3) steam-flaked corn at a low density, 4) steam-flaked corn at a medium density, and 5) steam-rolled corn. Cows fed the diet containing steam-flaked corn at a medium density had a higher milk yield (37.1 kg/d) than did cows fed the diets containing coarsely ground corn, steam-flaked corn at a low density, or steam-rolled corn; cows fed the diet containing finely ground corn had an intermediate milk yield (35.5 kg/d). Efficiency of feed utilization was greater, and dry matter intake was lower, for cows fed the diet containing finely ground corn than for cows fed the other diets. The fat content of milk was higher for cows fed the diets containing coarsely ground corn and steam-rolled corn than for cows fed the diet containing steam-flaked corn at a medium density. Milk protein and SNF contents and yields of protein, lactose, and SNF did not differ among diets. Apparent starch digestibilities in the total digestive tract were lower for diets containing coarsely ground corn (87.4%) and steam-rolled corn (91.3%) than for the other diets (X = 96.3%). Milk yield was highest for cows fed the diet containing steam-flaked corn with a medium density. Fine grinding resulted in the greatest efficiency of feed utilization.

[An epidemiology study on common diseases among the elderly in Beijing].

In order to study the status and characteristics of some common diseases of elderly which seriously influence the Quality of Life of the elderly, 1434 elderly over 60 years old in the urban and rural areas of Beijing were investigated with a random sampling method. The results showed that prostate hyperplasia, deafness, cataract, osteoarthropathy, bone fracture and constipation among the elderly were 61.4%, 53.9%, 46.4%, 24.4%, 14.2% and 18.2% in the urban areas, and 65.7%, 64.7%, 44.4%, 14.9%, 9.1% and 23.0% in the rural areas respectively. The prevalence rates of osteoarthropathy (P < 0.01) and bone fracture (P < 0.01) were higher in the urban areas than in the rural areas (P < 0.01). In contrast, the prevalence rates of deafness (P < 0.01) and constipation (P < 0.05) of the elderly were higher in the rural than those in the urban. The prevalence rates of above diseases were lower when selfreporting than that by the medical examination, and also lower in the rural than in the urban (P < 0.01). Hence, the prevention and treatment of the common diseases of elderly should be strengthened, especially in the rural areas.

Trinucleotide repeat expansion of spinocerebellar ataxia (SCA1) found in a Chinese family.

OBJECTIVE: To investigate the gene mutation and the ratio of the spinocerebellar ataxia type 1 (SCA1) in Chinese patients with autosomal dominant spinocerebellar ataxia (ADSCA). METHOD: The family material and DNA samples were collected from thirteen families with ADSCA. To determine the characteristics of the CAG trinucleotide repeats in SCA1 gene, the PCR products of the Rep1 and Rep2 primers were analyzed, and the bands with CAG repeat expansion were cloned by PCR2. 1 vector and sequenced. RESULTS: One family was found to have an expanded CAG repeat in the 13 families with ADSCA. The clinically affected individual was heterozygous with one disease allele being 55 CAG repeats, whereas the mean size of the CAG repeats on 104 chromosomes generated from unrelated control Chinese individuals is 29.3 (ranging from 18 to 34). CONCLUSIONS: The frequency of the SCA1 mutation is about 7% in the 13 Chinese families with ADSCA, suggesting that this type of genetic defect is not the main cause involved in the pathogenesis of ADSCA in China. Since the mutation has also been found in Caucasian, Japanese, Malaysian, and Bangladeshi kindreds, it is suggested that this genetic defect may well have multiple origins in different ethnic groups.

[Research in the relationship between Chlamydia and ureaplasma and infertility].

Samples taken from cervix and male urethra of 768 infertilitas patients were studied, and samples taken from 226 normal fertile males and females served as control. The fluorescent monoclonal antibody technique and cell culture method were used for Chlamydia trachomatis (Ct) and microbiological culture method was used for Ureaplasma urealyticum (Uu) detection. The positive rates of Ct and Uu infections were 28.64% (220/768) and 36.59% (281/768) in infertilitas group, 5.2% (14/266) and 14.28% (38/266) in control group respectively. There was significant difference between the 2 groups (P < 0.01). The results suggest that infection with Ct or Uu is one of the factors causing infertility.

[Drug resistance study and detection of methicillin-resistant staphylococci].

MRSA and MRSE were identified by detecting oxacillin resistance of 107 strains of staphylococci from clinical sources. The positive rates of MRSA were 45.2% and 16.9%, respectively among the whole number of staphylococci examined. Antibiotic-resistant strains were detected with disks by Kirby-Bauer method. All MRSA and MRSE strains were multi-resistant. Resistant rates of MRSA and MRSE were higher than MSSA and MSSE. The positive rates of beta lactamase producing in MRSA and MRSE were significantly higher than MSSA and MSSE (P< 0.05 and P< 0.01) respectively. These results indicated that MRSA and MRSE were the main pathogens in the clinical infection. MRSA and MRSE detection are important for the early diagnosis and treatment of patients with infective diseases.

[Expression and significance of p53, c-erbB2, CEA proteins in colorectal adenoma and carcinoma].

Expression of p53, c-erB2, CEA proteins in 15 cases of normal colorectal mucosa, 40 adenoma and 40 carcinoma of colon were studied with immunohistochemistry. The positive rate of p53, c-erbB2, CEA in colorectal carcinomas were significantly higher than those in adenoma. The positive rate of p53 in adenoma with severe atypical hyperplasia was higher than that in the mild (P< 0.01), and was higher in colorectal carcinomas of poor differentiation that that in the well-differentiated ones (P< 0.05). The novel type monoclonal antibody of CEA Col-1 had a high sensitivity and specificity for the tissue of colorectal carcinoma in which the positive rate was 85%. The results suggested the alteration p53, c-erbB2 are involve in the tumorgenesis and development of colorectal carcinoma; and detection of p53, c-erbB2 and CEA proteins are helpful in distinguishing benign and malignant polyps of colon.

[Alterations of amino acids in rat cerebral cortex of infectious brain injuries].

Concentrations of amino acids such as glutamate(Glu), glutamine(Gln), gamma-aminobutyric (GABA), glycerin(Gly) in rat cerebral cortex were measured by o-phthaldialdehyde method to clarify alterations of these amino acids in infectious brain injuries(IBI). The results were that Gln and GABA in groups treated by bordetella pertussis suspension(BP) 4h were increased compared to those in the group treated by the normal saline(NS) or the operative control(OC) and a positive correlation with water content or Evans blue content, respectively. In the 24h BP group, Gln was still increased; GABA and Gly were decreased compared to those in the NS or OC group. The findings suggest that GLu, Gln, and Gly play an important role and GABA may be a marker of injury in pathogenetic mechanism of IBI induced by BP.

[Laboratory diagnosis of Chlamydia trachomatis and Ureaplasma urealyticum of chronic prostatitis].

Prostatic fluid taken from 1038 cases of chronic prostatitis were detected by fluorescent monoclonal antibody technique and isolating cultural method for Chlamydia Trachomatis (CT) and Ureaplasma Urealyticum (UU). The control group consisted of 80 cases. CT and UU were not found in control group. The positive rates of CT and UU infection were 28.4% and 33.0% respectively in the tested group whose cases were abacterial prostatitis. The results suggested that CT and UU are pathogens causing chronic abacterial prostatitis.

[Detection of interleukin-8 and nitric oxide in chronic prostatitis].

We detected levels of interleukin-8(IL-8) and nitric oxide(NO) in 62 prostatic fluids. The results were that microbiologic exam, IL-8 content, and NO level in control group were all negative; there was not significant difference on the microbiologic exam and NO level between the chronic prostatitis group and non-prostatitis group; there was significant difference between two groups of IL-8 positive ratio. These results suggest that the IL-8 detected in the prostatic fluid of patients with prostatitis may be a diagnostic indicator of prostatic infection.

[Alterations of intracellular free calcium concentration of cerebral neuronal cells in infectious brain injury in rats].

Intracellular free calcium concentration, [Ca2+]i, of cerebral synaptosomes loaded by Fura-2/AM was measured by fluorescent method to clarify alterations of [Ca2+]i in infectious brain injury (IBI) in rats. The results showed that [Ca2+]i, water content (WC), and Evans blue content (EB) in groups injected by Bordetella pertussis suspension (BP) were increased compared to those in groups injected by normal saline (NS) at 0.5 h, 4 h, and 24 h, respectively. Increased [Ca2+]i in BP groups showed time-dependent relationship and the highest level in the 24 h group. There was positive correlation (0.5 h, 4 h) and no correlation (24 h) between [Ca2+]i and WC or EB. It is suggested that [Ca2+]i accumulation of cerebral neuronal cells plays an important role in the pathogenetic mechanism of the acute and delayed IBI induced by BP.

[Effects of stimulation at different areas of nucleus raphe dorsalis on genioglossus and diaphragm activities].

Effects of electrical and chemical stimulation of the dorsal and ventral areas of the nucleus raphe dorsalis (dNRD and vNRD) on genioglossus and diaphragm activities were observed in 48 urethane-anaesthetized and vagotomized rabbits. (1) Long train electrical stimulation at the dNRD facilitated genioglossus and diaphragm activities. (2) Long train electrical stimulation delivered to the vNRD excited genioglossus activity but inhibited diaphragm activity. (3) Effects of microinjection of glutamate at the dNRD and vNRD were similar to the effects of electrical stimulation. The above results suggest that excitation of nucleus raphe dorsalis (NRD) increases genioglossus activity and reduces upper airway resistance. The dNRD and vNRD play different roles in modulating diaphragm activity.

An efficient and economic site-specific deuteration strategy for NMR studies of homologous oligonucleotide repeat sequences.

We describe herein the use of a 2H-labeling strategy to achieve specific assignments of considerably overlapped cross peaks in the 1H-NMR spectrum of a DNA trinucleotide repeat sequence. Our strategy focuses on site-specific 2H-labeling of base moieties to simplify the NMR spectral regions which contain the major portion of the structural information. To achieve efficient preparation of 2H8- or 2H6-labeled DNA and RNA nucleosides and nucleotides, the existing synthetic and purification procedures were significantly improved. Our experiments demonstrate that pyrimidine H6 deuteration reactions may be carried out using non-deuterated base reagents with DMSO-d6 as a 2H donor. These reactions are simple and economic to perform and produce base deuterated nucleosides and nucleotides in high yield. The 2H-labeled residues have been incorporated into oligonucleotides with minor modifications of the existing reaction conditions. Using the homologous CGG repeat sequence, d(CGG)5, as an example, the effectiveness of the site-specific base deuteration strategy is demonstrated. In the otherwise extensively overlapped spectra of d(CGG)5, 2H-labeling has permitted unambiguous identification of a sequential connectivity at a central CG step and confirmation of several other NOE assignments. This information is critical for elucidation of the structure and the folding of the CGG repeat sequences and will contribute to the intensive effort to understand the mechanisms of triplet expansion, which has been implicated in the development of a number of hereditary neurodegenerative diseases. In addition to the two dimensional spectral simplification in a key spectral region using site-specific 2H8/2H6-labeling, the potential applications of the prescribed strategy in homonuclear three dimensional experiments are also discussed.

Praja1, a novel gene encoding a RING-H2 motif in mouse development.

As part of a cloning strategy to identify genes involved in early mouse liver development we have isolated Praja1, a gene with similar sequences to the Drosophila melanogaster gene goliath (gl) which is involved in the fate of mesodermal cells ultimately forming gut musculatures, fat body, and the heart. Praja1 is a 2.1 kb gene encoding a putative 396 amino acid ORF and includes a COOH-terminal RING-H2 domain. Using the Jackson Laboratory BSS panel, we have localized Praja1 on chromosome X at 36 cM, which may be a candidate gene for mouse sla (sex linked sideroblastic anemia), near the X inactivation center gene, Xist. Northern blot analysis demonstrated three transcripts (3.1, 2.6 and 2.1 kb) in mRNA from adult mouse tissues brain, liver, and kidney as well as in mRNA from developing mouse embryos (days 7, 11, 15 and 17 post coitus, p.c.). In vitro transcription/translation yielded a product with an Mr of 59 kD. Immunohistochemical staining of in vitro liver explant cultures using a heterologous antibody against praja1 demonstrated cytoplasmic staining of cuboidal cells that have hepatocyte morphology and organization. The presence of the RING-H2 domain, a proline-rich region at the COOH-end, and regions rich in acidic amino acids, leads to the hypothesis that the Praja1 product is possibly involved in mediating protein-protein interactions, possibly as part of a protein sorting or transport pathway. This is strengthened by the similarity of Praja1 to rat Neurodap1, whose product has been shown to localize to the endoplasmic reticulum and golgi in brain.

A risk-assessment model for foot and mouth disease (FMD) virus introduction through deboned beef importation.

We present a risk-assessment model to assess the risk of introduction of foot and mouth disease (FMD) virus associated with deboned beef importation. The model was developed in accordance with the risk-reduction procedures proposed by the European Community for meat importation. The risk reduction procedures include farm-level inspection, ante-mortem inspection, post-mortem inspection, chilling and deboning. The risk assessment was based on the prevalence of FMD-infected cattle in herds as well as the prevalence of infected herds in the exporting country. Computer simulations were carried out to evaluate the probability of FMD virus introduction by importing 100 tons of deboned beef in relation to FMD prevalence, number of cattle selected from each herd, and sample sizes in ante-mortem and post-mortem inspections. The effects of the risk-reduction procedures on the probability of FMD virus introduction were examined.

Haplotype analysis of hemochromatosis: evaluation of different linkage-disequilibrium approaches and evolution of disease chromosomes.

We applied several types of linkage-disequilibrium calculations to analyze the hereditary hemochromatosis (hh) locus. Twenty-four polymorphic markers in the major histocompatibility complex (MHC) class I region were used to haplotype hh and normal chromosomes. A total of 169 hh and 161 normal chromosomes were analyzed. Disequilibrium values were found to be high over an unusually large region beginning 150 kb centromeric of HLA-A and extending nearly 5 Mb telomeric of it. Recombination in this region was approximately 28% of the expected value. This low level of recombination contributes to the unusually broad region of linkage disequilibrium found with hh. The strongest disequilibrium was found at locus HLA-H (delta = .84) and at locus D6S2239 (delta = .85), a marker approximately 10 kb telomeric to HLA-H. All disequilibrium methods employed in this study found peak disequilibrium at HLA-H or D6S2239. The cys282tyr mutation in HLA-H, a candidate gene for hh, was found in 85% of disease chromosomes. A haplotype phylogeny for hh chromosomes was constructed and suggests that the mutation associated with the most common haplotype occurred relatively recently. The age of the hh mutation was estimated to be approximately 60-70 generations. Disequilibrium was maintained over a greater distance for hh-carrying chromosomes, consistent with a recent mutation for hh. Our data provide a reasonable explanation for previous difficulties in localizing the hh locus and provide an evolutionary history for disease chromosomes.

Myocardial collagen changes and edema in rats with hyperdynamic sepsis.

OBJECTIVE: To determine if sepsis, which is accompanied by both systolic and diastolic myocardial dysfunction, involves changes in myocardial collagen, as myocardial collagen changes can affect both myocardial compliance and contractility. DESIGN: Prospective, randomized, controlled study. SETTING: Animal laboratory at a university-affiliated hospital. SUBJECTS: Male Sprague-Dawley rats, weighing 310 to 396 g. INTERVENTIONS: Cecal ligation and perforation (to induce sepsis) for 24 (n = 9) or 48 hrs (n = 9); sham laparotomy for 24 (n = 10) or 48 hrs (n = 9) with saline fluid resuscitation or normal control (n = 5) groups. MEASUREMENTS AND MAIN RESULTS: Collagen content and interstitial space were determined, using polarized light microscopy and a computer video densitometry system. At 24 and 48 hrs post surgery, heart rate and cardiac index were increased, and systemic vascular resistance index was decreased significantly in the sepsis vs. the sham rats. Collagen content was decreased significantly in the sepsis vs. the sham groups both at 24 and 48 hrs following surgery (1.83 +/- 0.79 [SD] % [24 hrs], 1.76 +/- 0.31% [48 hrs] vs. 2.83 +/- 0.73% [24 hrs], 2.25 +/- 0.72% [48 hrs]; p < .01). Interstitial space was increased significantly in the sepsis vs. the sham groups (13.9 +/- 3.5% [24 hrs], 15.6 +/- 5.2% [48 hrs] vs. 8.6 +/- 4.2% [24 hrs], 9.9 +/- 4.8% [48 hrs]; p < .01). CONCLUSIONS: Sepsis is accompanied by changes in myocardial collagen content and myocardial edema. These changes may contribute to the systolic and diastolic myocardial dysfunction, and particularly to the ventricular dilation, observed in sepsis.
