Generation of spatially periodic patterns by a mechanical instability: a mechanical alternative to the Turing model.

We have studied the generation of spatial patterns created by mechanical (rather than chemical) instabilities. When dissociated fibroblasts are suspended in a gel of reprecipitated collagen, and the contraction of the gel as a whole is physically restrained by attachment of its margin to a glass fibre meshwork, then the effect of the fibroblasts' traction is to break up the cell-matrix mixture into a series of clumps or aggregations of cells and compressed matrix. These aggregations are interconnected by linear tracts of collagen fibres aligned under the tensile stress exerted by fibroblast traction. The patterns generated by this mechanical instability vary depending upon cell population density and other factors. Over a certain range of cell concentrations, this mechanical instability yields geometric patterns which resemble but are usually much less regular than the patterns which develop normally in the dermis of developing bird skin. We propose that an equivalent mechanical instability, occurring during the embryonic development of this skin, could be the cause not only of the clumping of dermal fibroblasts to form the feather papillae, but also of the alignment of collagen fibres into the characteristic polygonal network of fibre bundles - which interconnect these papillae and which presage the subsequent pattern of the dermal muscles serving to control feather movements. More generally, we suggest that this type of mechanical instability can serve the morphogenetic functions for which Turing's chemical instability and other reaction-diffusion systems have been proposed. Mechanical instabilities can create physical structures directly, in one step, in contrast to the two or more steps which would be required if positional information first had to be specified by chemical gradients and then only secondarily implemented in physical form. In addition, physical forces can act more quickly and at much longer range than can diffusing chemicals and can generate a greater range of possible geometries than is possible using gradients of scalar properties. In cases (such as chondrogenesis) where cell differentiation is influenced by the local population density of cells and extracellular matrix, the physical patterns of force and distortion within this extracellular matrix should even be able to accomplish the spatial control of differentiation, usually attributed to diffusible 'morphogens'.

Sexual responsiveness in diabetic women.

Sexual responsiveness in 82 Type 1 (insulin-dependent) diabetic women was compared with that of 47 normal control subjects, using an interview method for rating various aspects of sexual response, and attitude questionnaires. The diabetic women were questioned about symptoms of autonomic neuropathy, and cardiovascular autonomic nerve function tests were performed. There were differences between the two groups in the reports of vaginal lubrication, but in most other respects the groups were similar. The diabetic women were not less orgasmic, and reported significantly fewer unpleasant feelings during sexual intercourse. They saw themselves and their husbands as less 'potent' than did the normal subjects. The possible enhancing effect of diabetes on the marriage of some diabetic women possibly counteracted the relatively mild physiological deficits involved. There was very little difference in sexual response between women with and without symptomatic autonomic neuropathy.

Mood, sexuality, hormones and the menstrual cycle. I. Changes in mood and physical state: description of subjects and method.

Mood and physical symptoms through the menstrual cycle were investigated in 55 women with normal ovulatory cycles. One-third had attended a clinic with severe premenstrual syndrome (clinic PMS group); the remainder were volunteers either with a history of PMS (non-clinic PMS group) or without (no PMS group). Each cycle was divided into six hormonally distinct phases on the basis of repeated hormone measurement. Self-ratings of "well-being" reached their maximum in the late follicular phase, declining throughout the luteal half of the cycle. This pattern was pronounced and statistically significant in the "clinic" and "nonclinic PMS" groups. In all three groups, "physical distress" increased during the second half of the cycle to reach a maximum in the late luteal phase. A clear temporal relationship was therefore demonstrated between mood, physical state, and hormonal phases of the cycle. It remains uncertain whether changes in the "clinic" group were extreme forms of a normal pattern or were qualitatively different.

Mood, sexuality, hormones, and the menstrual cycle. II. Hormone levels and their relationship to the premenstrual syndrome.

In women with premenstrual syndrome, negative changes start soon after ovulation gradually increasing as the corpus luteum develops, and reach a maximum during the last 5 days of the luteal phase. They decline rapidly once menstruation starts, disappearing within one or two days of ovarian steroids reaching baseline levels. Positive moods are at maximum when preovulatory estradiol reaches its peak. A comparison of hormone levels in women with high and low degrees of cyclical mood change showed no difference in progesterone, estradiol, testosterone, or androstenedione.

Mood, sexuality, hormones, and the menstrual cycle. III. Sexuality and the role of androgens.

Sexual interest and activity at different stages of the menstrual cycle was recorded by 55 women with normal ovulatory cycles. In women with marked cyclical mood change, there was an associated cyclical pattern of sexual feelings. Subjective sexuality independent of mood change, was maximal in the mid-follicular (i.e., postmenstrual) and late luteal (i.e., premenstrual) phases. Sexual activity was maximal in the mid-follicular phase. There was no evidence of a periovulatory increase in sexual interest or activity. Mean testosterone levels were correlated with masturbation frequency but not with sexuality involving the partner. A weak association between testosterone and life style (i.e., in full-time work or a housewife) was also evident.

The predictability of subnormal penetrating capacity of sperm in cases of unexplained infertility.

Multivariate discriminant analysis has been used to determine the predictability of sub-normal penetrating capacity in cases of unexplained infertility. The product of this analysis was the identification of 7 discriminating variables, all of which described various aspects of the post-capacitation movement characteristics exhibited by the spermatozoa, omitting all reference to the conventional parameters of semen quality. On the basis of these discriminating variables, 90.9% of samples exhibiting penetration rates within the normal range were correctly predicted to be functionally competent. The same variables correctly identified only 60% of samples with impaired penetrating capacity, indicating that in the remaining 40%, defects are present in the spermatozoa which are not reflected in their motility patterns.

The effects of sterilisation: a comparison of sterilised women with the wives of vasectomised men.

In a follow-up study, women sterilised by tubal diathermy were compared with a matched group of wives of vasectomised men. Semi-structured interviews were given to a random sample drawn from a representative population. The couples were young with small families and did not have a high proportion of unplanned pregnancies or terminations. They had previously used contraception, mainly the pill or sheath. Most couples were entirely satisfied with the operation. Both groups showed an increase in pre-menstrual symptoms but there was only slight evidence that menstrual loss was affected by female sterilisation. The vasectomy couples had a higher frequency of sexual intercourse, few sexual problems and tended to have more satisfactory marriages. They had had more discussion of their decision to have the operation and the implications of counselling are considered.

Androgen replacement with oral testosterone undecanoate in hypogonadal men: a double blind controlled study.

The effects of androgen withdrawal and replacement were investigated in six hypergonadotrophic and six hypogonadotrophic men with hypogonadism. A double blind cross-over design was used comparing testosterone undecanoate (T.U., Organon International), 160 mg daily by mouth, with placebo. There was a significant effect of T.U. on all measures of sexual interest and behaviour. Sexual interest increased within the first week of T.U. administration, ejaculation usually returning within the second week. Behavioural responses were similar in the hyper- and hypogonadotrophic groups. The rise in plasma testosterone during T.U. administration was modest, not reaching the normal range in several cases. 5 alpha dihydrotestosterone rose more substantially. There was a significant fall in sex hormone binding globulin, a rise in oestradiol and no significant change in gonadotrophin concentrations during T.U. administration.

Androgens and sexual behaviour in women using oral contraceptives.

Twenty women using oral contraceptives and complaining of impaired sexual function were compared with twenty women without sexual problems, matched for age and oral contraceptive. Whilst the sexual behaviour differed in the two groups, the plasma testosterone, androstenedione, oestradiol and SHBG concentrations were very similar. The total androgen levels were low in both groups. Plasma testosterone and oestradiol concentrations were correlated with measures of sexual interest in the no-problem group, but not in the problem group. Administration of exogenous androstenedione to women in the problem group, using a double blind cross-over comparison with a placebo, failed to improve their sexual function except in one case. The majority of women showed a rise in androgen and oestradiol between day 24 of one pill cycle and day 4 of the next. The possible behavioural indications of this pattern are discussed.

A method for the storage and retrieval of laboratory data on human lymphoblastoid cell lines.

A computer-based system has been developed for the handling and retrieval of data on long-term human lymphoblastoid cell lines. It permits accurate recording of a wide range of genetic markers and other defined characteristics for the donor of each culture and for individual aliquots of any cell line. The data is recorded in relation to the in vitro age of each aliquot studied and the programme is designed to permit both the sequential examination of a single cell line and the comparison of lines of different origins. It is hoped that, by the application of this type of system, the confusion which has arisen in relation to other long-term cell lines (and which threatens to develop in relation to human lymphoblastoid cell lines) may be avoided and that the exchange of information between laboratories may be facilitated.

Genetic analysis of drug resistance in Neisseria gonorrhoeae: production of increased resistance by the combination of two antibiotic resistance loci.

The studies reported here demonstrate that increased resistance of Neisseria gonorrhoeae to penicillin, tetracycline, and chloramphenicol results from the combined effect of two resistance loci. As shown by experiments with deoxyribonucleic acid from transformants carrying only a single resistance locus, transformants with an incresed level of resistance to penicillin result from the combination of a penicillin-specific locus, pen, and a multiple resistance locus, mtr. Similarly, transformants with an increased level of resistance to tetracycline result from the combination of mtr and a tetracycline-specific locus, tet. Transformants with an increased level of resistance to chloramphenicol result from the combination of mtr and a chloramphenicol-specific locus, cml. Deoxyribonucleic acid dilution experiments established that only a single dose of each of the two required resistance loci is necessary to give higher-level resistance. Higher-level-resistant transformants were not obtained when a double dose of one resistance locus or a combination of loci pairs other than mtr and pen, mtr and tet, or mtr and cml was introduced into a recipient. Combinations of the mtr and tet genes resulted in increased resistance to semisynthetic tetracyclines. The presence of the mtr and pen genes resulted in increased resistance to penicillinase-stable penicillins.

Human vaginal secretions: volatile fatty acid content.

Vaginal samples (682) were collected by a tampon method from 50 healthy young women. Samples were analyzed by gas chromatography. The volatile aliphatic acids increased during the late follicular phase of the menstrual cycle and declined progressively during the luteal phase. Women on oral contraceptives had lower amounts of volatile acids and did not show any rhythmic changes in acid content during the menstrual cycle. These same substances possess sex-attractant properties in other primate species.

Photostatic reporting in a hospital bacteriology laboratory.

A method of reporting results of tests performed in a hospital bacteriological laboratory is described in which a modified commercially available electrostatic copier is used. This device was designed for a hispital system in which more than one report per specimen is issued and in which rubber stamps are widely used and frequently changed. It has been working satisfactorily for more than six months. It increases accuracy and saves time and money. Attention to the clarity of reproduction is necessary. It is suggested that this system could be modified for other laboratory reporting systems.
